Response mechanisms of thermionic detectors with enhanced nitrogen selectivity.

The response mechanisms of a thermionic detector with enhanced nitrogen selectivity operating in an inert gas environment were investigated. According to accepted theory, the analyte has to contain electronegative functional groups in order for negative ions to be formed by the extraction of electrons from the thermionic source. This leads to a selective detector response for compounds containing nitro groups or multiple halogens. However, in the tests described here, polycyclic aromatic nitrogen hydrocarbons (PANHs), acridines, and carbazoles were used as reference substances. These compounds contain no electronegative functional groups. None of the investigated acridines exhibited any response from the detector, but carbazoles generated a strong structure-related detector response. By examining partial charges for all hydrogens of all individual carbazoles and acridine, it was demonstrated that the acidic hydrogen atom attached to the nitrogen heteroatom of the carbazoles has a strong influence on the detector response. Ionization of carbazoles may occur by dissociation of the nitrogen-hydrogen bond during contact with the thermionic surface. Support for this theory was provided by the linear relationship between the relative detector response and the deprotonization energy of the carbazoles (coefficients of determination of 0.90 and 0.98 for linear and quadratic models, respectively, were obtained). Further, there appeared to be no linear relationship between the detector response and electron affinity of the carbazoles, (R2 value, 0.32). Thus, the mechanism involved in ionization of the carbazoles is probably not direct electron transfer from the thermionic surface to the carbazoles. Principal component analysis (PCA) showed that the thermal conductivity of chemically inert detector gases also has an influence on the detector response. The investigated gases were helium, neon, nitrogen, carbon dioxide, and argon. It was found that thermal conductivity can be used to rank the detector response for the carbazoles, and there was no discernible response when helium, which has the highest thermal conductivity, was used as the detector gas.

Mutagenicity of nitracrine analogues in Salmonella typhimurium: mutational specificity and activation by bacterial enzymes and rat-liver S9.

The mutagenic potential of 9-[(3-dimethylaminopropyl)amino]-acridine and its 1-(nitracrine), 2-, 3- and 4-nitro derivatives was studied in several strains of Salmonella typhimurium, using a plate-incorporation assay. In view of the potential importance of DNA binding and nitro group reduction, binding constants and redox potentials were determined. The parent compound was mutagenic only in the frameshift tester strain TA1537, and this effect was not enhanced by the plasmid pKM101. Each of the nitroarenes showed significant activity in S. typhimurium strains TA1537 and TA1538, and this mutagenicity was enhanced by the presence of plasmid pKM101. All caused reversion of the base-pair substitution allele in hisG46 in strain TA100 and this effect was either largely or totally dependent upon the plasmid. The frameshift mutagenic effects of the 3-nitro and 4-nitro compounds appeared to be little dependent upon the classical nitroreductase which is deficient in strain TA98NR, or transacetylase enzyme lacking in strain TA98/1,8-DNP6, whereas the activity of the 1-nitro compound depended partly on that nitroreductase enzyme, and that of the 2-nitro compound on having both activities present. In the latter two cases, the mutagenic effects of the compounds could not be restored by the addition of an S9 mix of mammalian enzymes. Mutagenicity data were compared with physicochemical parameters. The results do not distinguish between the view that the orientation of the nitro group with respect to the aromatic plane dictates mutagenic potential, and the earlier view that the nitro group redox potential is important. However, the extraordinary mutagenic potency of the 1-NO2 derivative is not explainable by its physicochemical properties alone.

Synthesis and antitrypanosomal evaluation of E-isomers of 5-nitro-2-furaldehyde and 5-nitrothiophene-2-carboxaldehyde semicarbazone derivatives. structure-activity relationships.

Several novel semicarbazone derivatives were prepared from 5-nitro-2-furaldehyde or 5-nitrothiophene-2-carboxaldehyde and semicarbazides bearing a spermidine-mimetic moiety. All derivatives presented the E-configuration, as determined by NMR-NOE experiments. These compounds were tested in vitro as potential antitrypanosomal agents, and some of them, together with the parent compounds, 5-nitro-2-furaldehyde and 5-nitrothiophene-2-carboxaldehyde semicarbazone derivatives, were also evaluated in vivo using infected mice. Structure-activity relationship studies were carried out using voltammetric response and lipophilic-hydrophilic balance as parameters. Two of the compounds (1 and 3) displayed the highest in vivo activity. A correlation was found between lipophilic-hydrophilic properties and trypanocidal activity, high R(M) values being associated with low in vivo effects.

Ab initio and DFT study on 1,4-dinitroglycoluril configurational isomers: cis-DINGU and trans-DINGU.

cis-1,4-Dinitroglycoluril (1,4-dinitrotetrahydroimidazo[4,5-d]imidazole-2,5-(1H,3H)-dione, C(4)H(4)N(6)O(6), known as DINGU in the literature) is an important explosive regarded as one of the potential ingredients for LOVA (low vulnerability ammunition) applications. However, there is no study concerning trans-DINGU, to the best of our knowledge. Thus, in the present study the structural and electronic properties of the configurational isomers of DINGU (cis- and trans-isomers) have been investigated by performing density functional theory (DFT) calculations at B3LYP/6-31G(d,p), UB3LYP/6-31G(d,p) levels and also ab initio calculations at RHF/6-31G(d,p) and UHF/6-31G(d,p) levels. The optimized geometries, vibrational frequencies, electronic structures, and some thermodynamical values for the presently considered DINGU isomers have been obtained in their ground states. Comparing the calculated energy data (total electronic energy, heat of formation values and homolytic bond dissociation values of N-NO(2)) and the calculated bond lengths, cis-DINGU was found to be a more stable molecule than the trans-isomer.

Characterization of anti-apoptotic action of TCDD as a defensive cellular stress response reaction against the cell damaging action of ultra-violet irradiation in an immortalized normal human mammary epithelial cell line, MCF10A.

It was originally shown by Woerner and Schrenk [Woerner, W., Schrenk, D., 1998. 2,3,7,8-Tetrachlorodibenzo-p-dioxin suppresses apoptosis and leads to hyperphosphorylation of p53 in rat hepatocytes. Environ. Toxicol. Pharmacol. 6, 239-247] that TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin) acts as an antagonist against the action of UV-irradiation to induce apoptosis in rat primary hepatocytes. Since prevention of apoptosis has been shown to promote carcinogenesis, we have decided to investigate this phenomenon in a human mammary gland epithelial cell line, MCF10A. We found that, in this cell line, TCDD can antagonize apoptosis that was induced by a variety of treatments, such as UV- and gamma-irradiation, growth factor starvation and trypsinization, or by the addition of H(2)O(2), TGFbeta, and staurosporine. Furthermore, other agents that are known to elicit defensive cellular responses, such as LPS, Fe(3+), nitric oxide and hypoxia could also antagonize UV induced apoptosis just as in the case of TCDD. In addition, we found that, in this cell line, such anti-apoptotic action of TCDD resembles that of exogenously added EGF or TGF alpha. To study the basic mechanism of such an action of TCDD, we tested a variety of diagnostic agents to reverse the effect of TCDD. Antagonists of TCDD which were found to be effective in this way were (a) inhibitors of c-Src kinase, such as PP-2 and CGP77675, (b) those known to block the action of TGF alpha, such as anti-TGF alpha antibody, and alpha(1)-antitrypsin, (c) PD98059, a specific inhibitor of ERK activation, but not SB202190 (an inhibitor of p38 MAPK activation) or SP600125 (a JNK inhibitor) and (d) Ah receptor antagonists, alpha-naphthoflavone and 1, 10-phenanthroline. These results support the notion that TCDD acts as an anti-apoptotic agent by mimicking the action of EGF through activation of the c-Src/ERK signaling pathway.

A rapid and specific method for the determination of oxytocinase (cystine aminopeptidase) activity using continuous flow analysis and S-benzyl-L-cysteine-p-nitroanilide as substrate.

1. An automated method is described for the assay of pregnancy serum cystine aminopeptidase (Oxytocinase, E.C. 3.4.1.10) using continuous flow analysis. S-Genzyl-L-cysteine-p-nitroanilide, the most highly specific substrate so far available for oxytocinase is employed in this assay. Comparison with the manual method from which it has been developed, gave a correlation coefficient of 0.98. Reaction of oxytocinase on the substrate liberated p-nitroaniline, the absorption of which is measured at 410nm. 2. Blank values are eliminated by the incorporation of a dialyzer in the manifold design and thereby effectively increasing the rate of assay sample to 40 per hour. The lag phase (a) of the continuous flow analysis is 0.17 min while the exponential phase (b) is 0.31 min. Using a 2:1 sample to wash ration the carry-over is 3% at a 95% of steady states.
