Evaluation of mating systems involving five breeds for integrated beef production systems: III. Integrated system.

Angus (A), Charolais (C), Hereford (H), Limousin (L), and Simmental (S) breeds were included in deterministic computer models simulating integrated cow-calf-feedlot production systems. Three mating systems were used: pure-breeding and two-and three-breed rotational crossbreeding. Breed information was taken from the literature. Herd sizes were unrestricted; however, 100 heifers were saved as replacements. Cows were removed for reproductive failure, age (greater than 10.5 yr), or death. Calves produced in the cow-calf segment were fed in a custom feedlot to four slaughter end points: 440 d, 457 d, 288-kg carcass weight, and low Choice. All animals were fed to requirements. Cull cows were slaughtered after weaning. Biological and economic efficiencies improved with crossbreeding; however, rankings of breed combinations depended on how efficiencies were measured (weight, lean, or value basis). Among purebreds, reproductive performance had a large influence on breed rankings at age and weight end points, whereas feedlot performance was important at the low Choice end point. Crossbred combinations involving British (A or H) and Continental (C or S) breeds were more efficient than other crossbred combinations at all end points. However, choosing specific breed combinations for integrated systems depends on slaughter end points, market end points (weight vs lean), and measures of efficiency (weight, lean, or value basis).

Evaluation of results for Schirmer tear tests conducted with and without application of a topical anesthetic in clinically normal dogs of 5 breeds.

OBJECTIVE: To evaluate, for clinically normal dogs, results of Schirmer tear tests in eyes without topical anesthetic (STT) and to detect differences associated with breed, sex, age, day, and time of day in eyes in which STT was performed after use of topical anesthetic (STTa). ANIMALS: 41 Beagles, 43 Labrador Retrievers, 25 Golden Retrievers, 26 English Springer Spaniels, and 22 Shetland Sheepdogs. PROCEDURE: Beagles had STT and STTa values measured twice daily for 5 days. Client-owned dogs of 4 other breeds had STT and STTa values measured once. RESULTS: Mean +/- SD values of Beagles for STT and STTa were 20.2 +/- 2.5 and 3.8 +/- 2.7 mm/min. Mean values for STT and STTa were as follows: Labrador Retriever, 22.9 +/- 4.1 and 9.6 +/- 3.8 mm/min; English Springer Spaniel; 20.7 +/- 3.2 and 5.4 +/- 3.4 mm/min; Golden Retriever, 21.8 + 3.7 and 8.8 +/- 3.1 mm/min; and Shetland Sheepdog, 15.8 +/- 1.8 and 3.6 +/- 2.8 mm/min. Overall mean values for STT and STTa were 20.2 +/- 3.0 and 6.2 +/- 3.1 mm/min. Differences for STT and STTa were detected among breeds, but significant differences were not associated with sex or age within each breed or in overall values for all dogs. CONCLUSIONS AND CLINICAL RELEVANCE: Results for the STT reported here compare favorably with reported values, except for results of Shetland Sheepdogs; however, results for the STTa differ dramatically from reported values. Clinicians should consider effects attributable to breed when evaluating results of STT and STTa in dogs.

Consequences of social conflict on plasma testosterone levels in rhesus monkeys.

Four adult male rhesus monkeys formed a new social group with 13 adult females. The male who became dominant [alpha] showed a progressive increase in plasma testosterone. The male who became subordinate to the other three males showed an 80% fall in testosterone from baseline levels. After 7 weeks, this group was introduced to a well-established breeding group, and all four males became subordinate to all members of the breeding group. All four males evidenced a fall in testosterone during the first week after introduction, and within 6 weeks their levels were approximately--80% of pre-introduction values. The alpha male of the breeding group showed a large increase in testosterone [238%] 24 hr after he successfully defended his group and became the dominant animal of the larger, newly-formed group. Thus, plasma testosterone levels appear to be significantly influenced by the outcome of conflict attendant to alterations in status of rhesus monkeys living in social groups.

Nutritive value of dried citrus pulp and its effect on milk yield and milk composition of lactating ewes.

In a digestibility experiment, six adult wethers of the Karagouniko breed were used to determine the nutritive value of dried citrus pulp. The rations consisted of 800 g of hay and 75, 150, 225, 300, 375, and 450 g of citrus pulp. The apparent digestibilities of the DM, OM, CP, ether extract, crude fiber, and N-free extract for dried citrus pulp were 78.6, 87.2, 52.7, 82.0, 93.2, and 83.1%, respectively. Energy content was estimated to be 1.66 Mcal of NE(L)/kg of DM. In a second experiment, 26 lactating ewes of the Karagouniko breed were used to study the nutrient utilization of dried citrus pulp for milk yield when citrus pulp was used as a replacement for cereal grains. The ewes were divided into two groups immediately postweaning and fed daily 700 g of alfalfa hay, 300 g of wheat straw, and 580 or 550 g of concentrates with or without 30% citrus pulp, respectively. The inclusion of citrus pulp in rations for ewes had no significant effect on milk yield and composition but decreased the C4 to C10 fatty acids. Citrus pulp is a valuable, high energy by-product that can partly replace cereal grains in sheep rations without adverse effect on milk yield or composition.

Song as an indicator of parasitism in the sedge warbler.

We studied female choice and reproductive success in a marked population of sedge warblers Acrocephalus schoenobaenus, from 1995 to 1996. Three genera of parasitic blood protozoans, namely Haemoproteus sp. Trypanosoma sp. Plasmodium sp., were identified from blood samples taken from all breeding adults. Relatively high prevalence values of 19.5% in 1995 and 37.5% in 1996 were associated with increased levels of white blood cells relative to the number of red blood cells. Compared with nonparasitized males, parasitized males had significantly lower repertoire sizes in both years of the study; in one year, they also spent less time in song flights and weighed less. They also provisioned their broods at a lower rate. Parasitized females produced the same clutch size as nonparasitized females, although their broods were smaller at 7 days old. We suggest that haematozoan infections may reduce the expression of sexually selected song traits. Furthermore, such infections may influence the standard of parental care provided by males, although further research is needed to determine whether this is mediated through genetic resistance to parasitism or the effects of parasitism upon immediate body condition. Autosomal genetic control of the activity of a new variant ornithine transcarbamylase in chicken kidney.

The mode of inheritance of the gene for chick kidney ornithine transcarbamylase (OTC), found previously as a genetic variant, was investigated. White Leghorn B line males homozygous for the allele for the variant OTC gene were selected using the California Gray breed, having a near-absolute deficiency of the enzyme. Then further crosses of the two breeds were made. The mean value of the OTC level of F1 progeny was about 170 units. Chicks from the backcross generation were divided into two groups, of high activity and low activity, in a ratio of 1:1. F2 chicks were divided into three groups: one-fourth of the chicks were classified as a "super high" group, one-half were "high," and the remaining one-fourth were "low"; the mean values for OTC level were 356.7, 196.4, and 15.6 units, respectively. From these results, it was suggested that the variant OTC represents a simple autosomal incompletely dominant trait.

Brassinolide improves embryogenic tissue initiation in conifers and rice.

Somatic embryogenesis (SE), the most promising technology for the large-scale production of high-value coniferous trees from advanced breeding and genetic engineering programs, is expected to play an important role in increasing productivity, sustainability, and the uniformity of future U.S. forests. To be successful for commercial use, SE technology must work with a variety of genetically diverse trees. Initiation in loblolly pine ( Pinus taeda L.), our main focus species, is often recalcitrant for desirable genotypes. Initiation percentages of loblolly pine, Douglas-fir [ Pseudotsuga menziesii (Mirb.) Franco], and Norway spruce ( Picea abies L., Karst.) were improved through the use of brassinolide. Brassinosteroids, which include brassinolide, are a relatively new group of natural plant growth regulators that are found in many plant species. They have been shown to have diverse, tissue-specific, and species-specific effects, including the stimulation of cell elongation and ethylene production and increasing resistance to abiotic stress. In our media, brassinolide was effective at concentrations ranging from 0.005-0.25 micro M. Using control medium (no brassinolide) and brassinolide-supplemented (0.1 micro M) medium, we achieved improved initiation percentages in loblolly pine, Douglas-fir, Norway spruce, and rice-15.0% to 30.1%, 16.1% to 36.3%, 34.6% to 47.4%, and 10%, respectively. Brassinolide increased the weight of loblolly pine embryogenic tissue by 66% and stimulated initiation in the more recalcitrant families of loblolly pine and Douglas-fir, thus compensating somewhat for genotypic differences in initiation. Initiation percentages in loblolly pine were improved through the combination of modified 1/2-P6 salts, 50 mg/l activated carbon (AC), adjusted levels of Cu and Zn (to compensate for adsorption by AC), 1.5% maltose, 2% myo-inositol (to raise the osmotic level, partially simulating the megagametophyte environment), 500 mg/l casamino acids, 450 mg/l glutamine, 2 mg/l alpha-naphthaleneacetic acid, 0.63 mg/l 6-benzylaminopurine, 0.61 mg/l kinetin, 3.4 mg/l silver nitrate, 10 micro M cGMP, 0.1 micro M brassinolide, and 2 g/l Gelrite. Across 12 open-pollinated families of loblolly pine, initiation percentages ranged from 2.5% to 50.7%, averaging 22.5%.

Gibberellin inhibitors improve embryogenic tissue initiation in conifers.

Somatic embryogenesis (SE), the most promising technology to multiply high-value coniferous trees from advanced breeding and genetic engineering programs, is expected to play an important role in increasing productivity, sustainability, and uniformity of future forests in the United States. For commercial use, SE technology must work with a variety of genetically diverse trees. Initiation in loblolly pine (LP; Pinus taeda L.), our main focus species, is often recalcitrant for desirable genotypes. Initiation of LP, slash pine (SP; Pinus elliottii), Douglas-fir (DF; Pseudotsuga menziesii), and Norway spruce (NS; Picea abies) were improved through the use of paclobutrazol, a gibberellin synthesis inhibitor. Paclobutrazol was effective at concentrations ranging from 0.25 mg/l to 3.0 mg/l (0.85-10.2 microM) and optimal in LP at 1.0 mg/l. Using control media (no paclobutrazol) and 0.33-1.0 mg/l paclobutrazol, initiation percentages in LP, SP, DF, and NS were improved from 37.7% to 44.2% (across experiments), 19.3% to 28.5%, 16.9% to 23.7%, and 38.8% to 48.5%, respectively. Other gibberellin inhibitors such as flurprimidol, chlormequat-Cl, and daminozide also caused statistically significant increases in LP initiation when added to the medium at concentrations of 0.34, 10.0, and 1.0 microM, respectively. No detrimental effects on subsequent embryo development were observed when 29 new initiations from medium without GA inhibitor and 28 new initiations from medium containing paclobutrazol were tracked through culture capture, liquid culture establishment, cotyledonary embryo development, and germination.

Seasonal changes in body weight, fat depots, and plasma levels of thyroxine and growth hormone in free-living great tits (Parus major) and willow tits (P. montanus).

Annual changes in body weight, fat depots, and plasma levels of thyroxine (T4) and growth hormone (GH) were studied in free-living great tits and willow tits. Birds were collected during six ecologically well-defined periods of the year. Special attention was given to the nonreproductive part of the year. T4 showed simple unimodal cycles in both species and both sexes, with high levels during the warmer part of the year, and low levels during the winter and spring periods. Although increasing levels were temporarily separated between the two species, they were in both cases correlated with the onset of gonadal regression and moult. Plasma levels of GH fluctuated in a much more complex pattern, and no obvious and consistent correlation to any extrinsic or intrinsic factor was found. Body weights and fat depots both showed seasonal variations that varied slightly between the two species. Values, with the exception for breeding females, were generally the highest during the autumn, winter, and spring periods.

Morphometric study of the recurrent laryngeal nerve in young 'normal' horses.

Quantitative measurements were made on cross-sectional preparations of the distal part of the recurrent laryngeal nerve (RLN) from nine young mixed-breed horses to establish reference values for the total number of myelinated fibres, mean fibre diameter and percentage of thickest fibres (over 9.5 microns) and to delineate diameter distribution curves. The total number of myelinated fibres, mean fibre diameter and percentage of thickest fibres for the left RLN were significantly lower than those of the right RLN (P < 0.005). The distribution of fibres was unimodal. The fibre diameter ranged from 1 micron to 17 microns. Approximately, 95 per cent of fibres had a diameter larger than 5 microns.

Physical characteristics of the blood plasma of Labeo ruddi and Labeo rosae (Pisces: Cyprinidae).

1. The physical composition of blood plasma of Labeo ruddi and Labeo rosae males were determined during the breeding and post breeding phases. 2. The mean values for most blood electrolytes, pH and osmolality of the two species showed the same variation as was found for other cyprinids. 3. The ability to handle stress may be reflected in the presence of certain electrolytes in the blood and efforts are made to relate differences in the concentration of these parameters to interspecies differences between L. ruddi and L. rosae.

Have we entered a 'post-model' era in plant biology?

Models such as arabidopsis (Arabidopsis thaliana) have underpinned genomic and physiological research in plant science. Advances in genome sequencing, pangenomics, and genome editing have prompted claims of a 'post-model' era, with model-crops and crops such as rice and bread wheat combining agricultural relevance with experimental tractability. We argue that the 'simplicity-to-complexity' approach remains valid, although model systems have evolved. Arabidopsis remains indispensable for interpreting multi-omics data, testing developmental hypotheses, and generating mechanistic insights difficult to obtain in crops. Linking these strengths to model-crops adds translational value by bridging discovery and breeding, while niche models such as Brachypodium distachyon and legumes address grass cell wall biology and nitrogen fixation. Future progress depends on diverse species with complementary strengths across fundamental and applied plant biology.

Pregnancy-associated glycoproteins (PAG) detection in milk samples for pregnancy diagnosis in dairy goats.

In ruminants, glycoproteins synthesized in the outer layers of the trophoblast enter the maternal bloodstream. The assay of their concentration in plasma or in serum can be used for pregnancy diagnosis and to indicate feto-placental health status. In this study, concentrations of the pregnancy-associated glycoprotein (PAG) were investigated in the milk of dairy goats. The mean concentrations of PAG in milk were significantly higher in pregnant than in non-pregnant goats throughout the sampling period. Positive milk PAG levels were found in all pregnant animals from Day 32 after breeding, but remained at negative values in nonpregnant ones. This study reports for the first time a pregnancy diagnosis based on milk concentrations of PAG, that could be of great use on the farm.

[Effect of increasing the omega-3 fatty acid in the diets of animals on the animal products consumed by humans].

As shown by huge amount of assays in human as well as in animal models, w-3 polyunsaturated fatty acids play important role in the development and maintenance of different organs, primarily the brain, and could be useful in the prevention of different pathologies, mainly the cardiovascular diseases, and, as proposed recently, some psychiatric, dermatological or rheumatological disorders. For ALA, the major and cheapest source for human is rapeseed oil (canola oil), and walnut "noix de Grenoble" oil). The actual goal is first to identify which foods are naturally rich in w-3 fatty acids, and, second, to determine the true impact of the formulations (enriched in w-3 fatty acids) in chows used on farms and breeding centres on the nutritional value of the products and thus their effect on the health of consumers, thanks to quantities of either ALA, or EPA or DHA or both. This concern fish (in proportion of their lipid content, mainly mackerel, salmon, sardine and herring), eggs (wildly naturally rich in w-3 fatty acids, both ALA and DHA, or from laying hen fed ALA from linseed or rapeseed), meat from birds, mammals (from the highest concentration : rabbit, then pig and monogastrics, then polygastrics such as beef, mutton and goat) \; in butter, milk, dairy products, cheese (all naturally poor in w-3 fatty acids)... Indeed, the nature of fatty acids of reserve triglycerides (found in more or less large amounts depending on the anatomical localisation, that is to say the butcher's cuts) can vary mainly as a function of the food received by the animal. EPA and DHA are mainly present in animal's products. The impact (qualitative and quantitative) of alterations in the lipid composition of animal foods on the nutritional value of derived products (in terms of EPA and DHA content) eaten by humans are more important in single-stomach animals than multi-stomach animals (due to their hydrogenating intestinal bacteria). The intestinal physiology of birds results in the relatively good preservation of their dietary w-3 fatty acids. The enrichment in eggs is proportional to the amount of w-3 fatty acids in the hen's diet and can be extremely important. Including ALA in fish feeds is effective only if they are, like carp, vegetarians, as they have the enzymes required to transform ALA into EPA and DHA \; in contrast, it is probably less effective for carnivorous fish (75 % of the fish used for human), which have little of these enzymes : their feed must contain marine animals, mainly fish or fish oil. Analysis of the published results shows that, under the best conditions, feeding animals with extracts of linseed and rapeseed grains, for example, increases the level of ALA acid by 20 to 40-fold in eggs (according to the low or high level of ALA in commercial eggs), 10-fold in chicken, 6-fold in pork and less than 2-fold in beef. By feeding animals with fish extracts or algae (oils), the level of DHA is increased by 20-fold in fish, 7-fold in chicken, 3 to 6-fold in eggs, less than 2-fold in beef. In practise, the effect is considerable for fish and egg, interesting for poultry and rabbit, extremely low for beef, mutton and sheep. The effect on the price paid by the consumer is very low compared to the considerable gain in nutritional value.

Canine models of ocular disease: outcross breedings define a dominant disorder present in the English mastiff and bull mastiff dog breeds.

Progressive retinal atrophies (PRA) are a heterogeneous group of inherited eye diseases common to both dogs and man. Over 100 individual canine breeds display some sort of retinal degeneration, making the dog an extremely valuable resource both for finding the genetic determinants of inherited blindness and for developing naturally occurring animal models that mimic human disease. Progressive retinal atrophies within the English mastiff displayed an ambiguous mode of inheritance. By conducting outcross matings between affected English mastiffs and normal animals from other breeds, the mode of inheritance was confirmed as dominant. This directed candidate gene analysis and led to identification of two synonymous mutations and one nonsynonymous mutation within the canine rhodopsin gene. The nonsynonymous mutation (T4R) is the cause of PRA in the English mastiff, and a test was developed to investigate its presence in 17 additional breeds. Testing of PRA-affected animals from 16 breeds revealed that none carry the T4R mutation, indicating a different cause of PRA. Analysis of two affected bull mastiffs revealed one heterozygote (+/T4R) and one homozygous normal individual (+/+). These findings suggest that the genetic origin of PRA is often breed specific and underline the value of outcross mating to circumvent problems that act to mask the mode of inheritance.

Evaluation of genetic diversity in Japanese Brown cattle population by pedigree analysis.

The Japanese Brown is the second most common domestic beef breed in Japan. However, nowadays this breed is facing reduction in numbers because of pressure from a profitable domestic breed. This breed is uniformly characterized by its brown coat colour, but is comprised of two isolated sub-breeds, Kumamoto and Kouchi, each possessing a different gene pool. Pedigree analyses were carried out for the two sub-breeds using the pedigree records of animals born from 1970 to 2000. The effective population size has been found to be consistently reducing during the last three decades in both sub-breeds. The current effective sizes were estimated to be 25.5 and 6.0 for the Kumamoto and Kouchi sub-breeds, respectively. The estimate of the effective number of founders (N(ef)) in the Kumamoto sub-breed decreased from 152.1 to 74.4; that of non-founders (N(enf)), from 41.7 to 5.3; and that of founder genome equivalents (N(ge)), from 32.7 to 4.9. The corresponding changes in the Kouchi sub-breed were from 108.2 to 79.4, 16.2 to 4.1, and 14.1 to 3.9. Increasing differences between the two genetic diversity indices in the sub-breeds indicate that the greater part of the reduction of genetic diversity can be attributed to genetic drift that accumulated in the non-founder generations. A comparison with published estimates for several cattle breeds suggests the extremely limited genetic diversity of Japanese Brown. In addition to the avoidance of further reduction of genetic diversity, it will be important to counteract the process of breed decline by establishing a production system to efficiently utilize the unique characteristics of this breed and by developing links between the breed and products with market value.

Initial sensitivity of rat inbred strains to acute alcohol.

The ethanol sensitivity of eight inbred strains and one heterogeneous stock of rats has been determined by the assessment of "sleep time" and blood alcohol levels upon awakening. In addition, levels of liver alcohol and aldehyde dehydrogenases and phenobarbital-inducible aldehyde dehydrogenase were determined. These data are presented as baseline values for correlation and selective breeding studies using the heterogeneous rats.

Annual circulating testosterone levels in captive and free-ranging male armadillos (Dasypus novemcincuts).

Mean circulating concentrations and seasonal changes in plasma testosterone over 3 years were similar in free-ranging and captive male armadillos. The highest concentrations were seen in the summer months, i.e. at the time of maximum breeding, but the relatiely high values at other times and the inability of ACTH to restore dexamethasone-suppressed testosterone concentrations confirms that testicular function continues throughout the year. There was a gradual rise in plasma testosterone to adult levels from the 5th to the 9th month of life in captive-born males, indicating that sexual maturity is reached within the first year of life and that captivity is not deleterious to Leydig cell function.
