Effect of kanglemycin C on lymphocyte proliferation induced by tetradecanoylphorbol acetate and ionomycin.

AIM: To compare the suppressions of kanglemycin C (Kan) with that of cyclosporine (Cyc) on lymphocyte proliferations induced by tetradecanoylphorbol acetate (TPA) with ionomycin (IM), and concanavalin A (Con A). METHODS: Cell proliferation was quantified with 3-(4, 5-dimetyl-thiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) or [3H]thymidine ([3H]TdR) incorporating method. Calcineurin (CN) activity was measured with molybdate dye colorimetry. RESULTS: Kan (8, 40, 80, and 400 nmol.L-1) strongly suppressed splenocyte proliferation induced by TPA and IM, and the suppressive effect of Kan gradually decreased along with the increasing concentrations of TPA (1-100 micrograms.L-1) and IM (125-500 micrograms.L-1). But, the suppression of Cyc on the splenocyte proliferation induced by TPA was mild. Cyc suppressed CN activity of mouse splenocytes stimulated by Con A stronger than Kan. Moreover, Kan and Cyc strongly suppressed spleen enriched T-cell proliferation induced by Con A and TPA + IM, and the suppression of Kan on proliferation was partly attenuated by exogenous IL-2. CONCLUSION: Kan competitively suppressed the proliferation induced by TPA and IM, and Cyc mainly suppressed IM part of the proliferation induced by TPA and IM.

[Effect of WT1 gene expression on cell growth and proliferation in myeloid leukemia cell lines].

OBJECTIVE: To explore the effect of WT1 antisense oligonucleotide(AS-oligo) on cell proliferation and apoptosis in myeloid leukemia cell lines. METHODS: K562 and HL-60 cells were cultivated with WT1 AS-oligo. The cell proliferation, apoptosis, cell cycle and gene expression were examined by MTT colorimetry, FACS and RT-PCR. RESULTS: WT1 AS-oligo could inhibit the proliferation of K562 cell and induce apoptosis of K562 and HL-60 cells. On the contrary, the growth of HL-60 cells and the expression of WT1, mdm2 and bcl-2 genes were unaffected. CONCLUSION: WT1 gene is related to the proliferation and apoptosis of leukemic cells. WT1 gene could suppress cell apoptosis independent of status of p53 and bcl-2 genes. It might play an role in leukemogenesis.

Phenolic compounds in two Solidago L. species from in vitro culture.

The regenerated S. virgaurea and S. graminifolia, obtained previously by micropropagation from shoot tips and the callus tissue of S. virgaurea, were analysed for phenolic compounds. Preliminary used 2D-TLC methods (fingerprinting) showed the presence of phenolic glucoside--leiocarposide (S. virgaurea only), C-glycosyflavones--schaftoside and isoschaftoside (S. graminifolia), flavonoids and caffeoylquinic acids, similar to the ones reported earlier for naturally growing plants with the exception of the callus tissue. The latter contained predominantly caffeoylquinic acids, which were isolated and identified by NMR as 5-O-caffeoylquinic and 3,5-di-O-caffeoylquinic acids. The content of leiocarposide (HPLC) and flavonoids (colorimetry) in S. virgaurea was determined.

OBJECTIVE: To study the color distribution space of natural anterior tooth crowns of people in southwest China and effects of sex and age on tooth color. METHODS: The subjects studied consisted of 162 persons aged from 20 to 73 years and born in southwest China. A total of 1,944 healthy anterior teeth were measured in-vivo with a fiber-optical spectrophotometry system. RESULTS: Statistical analysis showed that the color distribution space of Chinese tooth crowns was wide. The value of L* varied from 42.383 to 67.428, the value of a* from -4.750 to 1.371, the value of b* from 1.88 to 20.268, the value of x from 0.298 to 0.393, the value of y from 0.332 to 0.392, and the value of Y from 19.383 to 34.025. The color difference between the upper and lower limits of 95% confidence was 31.667 CIE delta E units. The difference in colorimetry between male and female was not significant. The results showed that in three sections of teeth (incisal, middle and cervical) the values of L* and a* were negatively related to age (r < 0, P < 0.01); the value of b* was positively related to age just at the cervical and middle sites of teeth (r > 0, P < 0.01) and was not related to age at the incisal site of teeth (P > 0.05). CONCLUSION: The color distribution space of Chinese anterior tooth crowns is wide. There is no difference in the anterior tooth color between male and female subjects. The values of L* and a* and negatively related to age except the value of b*.

Effect of recombinant human basic fibroblast growth factor on acute inflammation in mice and rats.

AIM: To investigate the anti-inflammatory effects of recombinant human basic fibroblast growth factor (rh-bFGF). METHODS: Several inflammation models such as croton oil-induced ear swelling, carrageenan-induced hind paw edema, and acute peritonitis in rats or mice were prepared. Superoxide dismutase (SOD) activity was measured by hydroxyamine method, nitric oxide (NO) concentration by Griess reaction assay, nitric oxide synthase (NOS) activity by NADPH-diaphoras stain assay, N-acetyl-beta- D-glucosaminidase (NAG) activity by colorimetry, prostaglandin E2 (PGE2) production by radioimmunoassay (RIA), malondialdehyde (MDA) content by thiobarbituric acid (TBA) fluorescence technique, and protein content by Coomassie brilliant blue method in peritoneal exudate in rats. RESULTS: Recombinant human bFGF 2, 4 kU/kg im inhibited croton oil-induced ear swelling and carrageenan-induced paw edema in mice. In addition, rh-bFGF 2, 4 kU/kg im reduced neutrophil counts in the rat peritoneal exudate, and lessened protein content in peritoneal exudate in rats and mice. In the rat peritonitis induced by carrageenan, rh-bFGF 4 kU/kg decreased the MDA and NO levels, inhibited the NOS activity, augmented the SOD activity, and lowered the production of PGE(2) in exudate. However, rh-bFGF had no effect on NAG content. CONCLUSION: Recombinant human bFGF has an anti-inflammatory effect and its mechanisms are related to the inhibition of NOS activity, reduction of NO, MDA, and PGE(2) content, and increase of SOD activity.

Inhibitory effect of aminoguanidine on bleomycin-induced pulmonary toxicity in rat.

AIM: To observe the inhibitory effect of aminoguanidine (AG) on the pulmonary toxicity induced by bleomycin A5 (BLM-A5) in rat. METHODS: The contents of hydroxyproline, nitrite/nitrate (NO2-/NO3-), and malondialdehyde (MDA), which reflect fibrosis, nitric oxide (NO) production, and hyperoxidative injury of lung, were investigated by colorimetry. Histologic and morphometric examination of lung was also carried out on histological sections stained with hematoxylin and eosin (HE). RESULTS: (1) The content of hydroxyproline in lung and MDA in out-going pulmonary blood (OPB) increased from 14 d to 30 d after intratracheal administration of BLM-A5. Collapsed alveoli and fibrotic areas were seen and a lot of fibroblasts appeared in the lung interstitum of the rats 30 d after BLM-A5. (2) The NO2-/NO3- content increased in supernatant of alveolar macrophage culture and in OPB. (3) The increment of hydroxyproline in lung and MDA in OPB induced by BLM-A5 was alleviated by aminoguanidine (AG, 20 mg.kg-1 . d-1, ip). AG also reduced the histologic and morphometric changes in lung interstitum. (4) The augment of NO2-/NO3- in OPB was blocked by AG. CONCLUSION: AG had inhibitory effect on pulmonary injury and fibrosis induced by BLM-A5, and the inhibitory effect was related to the decrease of NO production in lung.

[The effects of blocking intrarenal renin-angiotensin system on the expression of transforming growth factor-beta1 mRNA and extracellular matrix components].

OBJECTIVE: To explore the possible mechanism of benazepril in diminishing the progression of the renal diseases. METHODS: Experimental glomerulosclerosis in rats was induced by adriamycin. The treated group was given benazepril (4 mg x kg(-1) x d(-1)). Intrarenal angiotensin converting enzyme (ACE) activity and angiotensin II (AngII) concentration were measured with colorimetry and radioimmunoassay respectively. Immunohistochemistry staining was employed for transforming growth factor beta1 (TGF beta1) and extracellular matrix (ECM) examination. To observe the expression of TGFbeta1 mRNA in the kidney, in situ hybridization was performed. RESULTS: In the rats treated with benazeapril, ACE activity and Ang II concentration in renal tissue were significantly inhibited. In the meantime, expression of TGFbeta1 mRNA and TGFbeta1 protein were down-regulated, accompanied by decreasing accumulation of ECM (compared with the untreated rats, P < 0.01). CONCLUSION: Benazepril plays an important role in down-regulating the expression of TGFbeta1 and decreasing the accumulation of ECM by blocking intrarenal renin-angiotensin system.

[Determination of blood iron and the total fixation capacity of transferrin by a simple automatic technic without dialysis].

An automated method without dialysis for determining serum iron is described. It is an adaptation of the Lauber procedure to the Technicon Auto Analyser I: serum iron is split from its protein combination by exposure to an acetate pH 5.8 buffer, in the presence of a detergent (Teepol 710) which prevents any precipitation of proteins. Iron is maintained in ferrous state by addition of sodium dithionite and is quantitated with bathophenanthrolin colorimetry. Serum samples are analysed at a rate of 60/H. It is necessary to run a blank for each serum sample. Between 20 and 500 mug iron/100 ml the color intensity agrees with Beer's law. The reproducibility is good, and the variation coefficient less than 1.2 per cent. Our results are not significatively different of those obtained with the ICSH standard method. Bilirubin does not interfere. Iron added is satisfactorily recovered. Our method can also be used for determining the transferrin iron binding capacity.

[Evaluation of colorimetric method for analysis of glycated hemoglobin].

The level of glycated hemoglobin (GH) measured by the Diabetic test colorimetric method did not depend on the compensation. On the other hand, its level was increased in diabetes mellitus decompensation in comparison with subcompensation, if isoelectrofocussing of the studies hemolysates was carried out prior to colorimetry. Study of colorimetric migration of hemolysates showed that they were represented by a background which could distort the results.

[A cross-sectional study on the effects of microcystin in drinking water to the health of human beings].

OBJECTIVE: To understand the effects of microcystin (MC) in drinking water on health of human beings. METHODS: In Taixing, a cross-sectional study was conducted in two middle schools with different types of drinking water, from which 111 and 92 students were randomly selected and investigated. Blood samples from all individuals were collected and tested for serum HBsAg, HBeAg, anti-HBe, anti-HBs, anti-HBc with enzyme linked immunoabsorbent assay and for serum alanine aminotransferase (ALT), gamma-glutamyltransferase (gamma-GT) and alkaline phosphatase (ALP) contents by colorimetry. These levels of enzymes in sera were compared after the students were stratified according to the status of HBV infection. RESULT: With the exclusion of the effect of HBV infection, three enzymes levels in sera between two groups of students showed significant differences (P < 0.01), much higher in exposure group than those in control group. CONCLUSION: MC was possibly harmful to human beings in drinking water and induced the contents of some enzymes from the liver of human beings to increase in sera.

[Effects of humic acid on lipid peroxidation in arsenosis prevalent areas].

OBJECTIVE: To explore the causes of blackfoot disease in Taiwan. METHODS: Experiments to induce lipid peroxidation by arsenic and humic acid were made in vitro. Arsenic was determined for the water sampled from the arsenosis prevalent areas in Inner Mongolia and blackfoot disease prevalent areas in Taiwan and humic acid determined for the extracts of the coal rich in arsenic sampled from Guizhou Province in China by MDA-TBA colorimetry. RESULTS: Experiments found that humic acid could induce lipid peroxidation caused by sodium salt of unsaturated fatty acids in vitro in a weak and unstable manner, which could be promoted by 0.05 mmol/L of Fe(+ +) and not by 1 mmol/L As(2)O(3). The extent of lipid peroxidation caused depended on the sources, constituents and structure of humic acid samples, those from Inner Mongolia ranked the highest, those from Guizhou and Taiwan the medium and the commercial humic acid (Aldrich Co. in the US) the lowest. The experiments also found that lipid peroxidation caused by commercial humic acid presented a dynamic process in the existence of trace amount of iron, and humic acid could decompose the products of lipid peroxidation. CONCLUSION: The experiments suggested the relations of humic acid and arsenic to iron and other transition elements in the blackfoot disease prevalent areas and areas with an environment rich in arsenic and humic acid.

Inhibitory effect and mechanism of action of sanggenon C on human polymorphonuclear leukocyte adhesion to human synovial cells.

AIM: To examine the effect of sanggenon C on human polymorphonuclear leukocyte (PMN) adhesion to human synovial cell(HSC), and explore its mechanism. METHODS: Adhesion of PMN to HSC was measured by MTT colorimetry. Cell-ELISA and RT-PCR methods were used to examine the expression of adhesion molecules ICAM-1 and VCAM-1. Activation of nuclear factor-kappa B(NF-kappaB) was measured by electrophoretic mobility shift assays(EMSA) method. RESULTS: Sanggenon C effectively inhibited TNF-alpha (50 kU/L for 12 h) and IL-1beta (50 kU/L for 12 h) induced adhesion of PMN to HSC (IC50 27.29 nmol/L and 54.45 nmol/L, respectively) in a concentration-dependent manner. Adhesion molecule VCAM-1 surface protein and mRNA expression induced by TNF-alpha 50 kU/L were significantly inhibited by sanggenon C, nevertheless, for ICAM-1 only surface protein expression being inhibited. The activation of NF-kappaB was also extensively inhibited by sanggenon C. CONCLUSION: Sanggenon C inhibited TNF-alpha -stimulated PMN-HSC adhesion and expression of VCAM-1 by suppressing the activation of NF-kappaB.

[The effects of DiBaiRen decoction on the proliferation of human keratinocytes and its in vitro anti--oxidation role].

OBJECTIVE: To investigate the effects of DiBaiRen decoction on the proliferation of human keratinocytes and its in vitro anti -- oxidation role. METHODS: Human keratinocytes were cultured in vitro in the media containing gradient diluted DiBaiRen decoction. The cell proliferation rates at different culture times were recorded. Cellular oxidation injury inflicted by superoxide anion and hydroxyl anion was produced by adding hypoxanthine -- xanthine oxidase and hydrogen peroxide to the culture media. MTT colorimetry was employed to determine cellular activity. The antagonistic effects of DiBaiRen decoction on cellular oxidation injury were analyzed. RESULTS: In the case that DiBaiRen decoction was added to the culture in concentrations of 30 ml/L to 100 ml/L, there exhibited obvious accelerated proliferation of the keratinocytes, with evident shortening of the cell fusion time. In addition, the oxidation injury of cultured keratinocytes caused by hypoxanthine -- xanthine oxidase and hydrogen peroxide was ameliorated or eliminated by the addition of DiBaiRen decoction, and the effects were dose -- dependent. CONCLUSION: DiBaiRen decoction might possess anti -- oxidation effects and could promote the in vitro proliferation of cultured human keratinocytes, and it might be beneficial in wound protection and epithelization.
