Total synthesis of (+)-acanthodoral by the use of a Pd-catalyzed metal-ene reaction and a nonreductive 5-exo-acyl radical cyclization.

[reaction: see text] The first total synthesis of the antibiotic acanthodoral (1) has been achieved from 3-methyl-2-cyclohexen-1-one in 19 steps in 2.1% overall yield. The synthesis features the use of a Pd-ene reaction in the presence of CO to form the endocyclic alkene 8, a nonreductive acyl radical cyclization reaction, and a ring contraction reaction by the Wolff rearrangement. (+)-Acanthodoral has also been synthesized starting from (+)-S-2,2-dimethyl-6-methylenecyclohexanecarboxylic acid.

Ru(III)-catalyzed cyclization of arene-alkene substrates via intramolecular electrophilic hydroarylation.

[reaction: see text] We herein report that RuCl(3)/AgOTf has proven to be a hydroarylation catalyst with an efficiency and scope superior to previously known methods. This catalyst demonstrated consistent performance with arene-ene substrates of diverse structural features, providing good to excellent yields of cyclization products (chromanes, tetralins, terpenoids, dihydrocoumarins).

An expeditious Nazarov cyclization strategy toward the hydroazulene core of guanacastepene A.

The hydroazulene core of guanacastepene A has been synthesized in five steps from commercially available starting materials using a classical Nazarov cyclization to install the stereochemistry in the cyclopentanone diastereoselectively. In the presence or absence of Lewis bases, a hydroazulenone or a spirocyclic ketone generated via a novel Wagner-Meerwein rearrangement is obtained with excellent selectivity and yield.

Practical asymmetric approach to medium-sized carbocycles based on the combination of two ru-catalyzed transformations and a lewis Acid-induced cyclization.

[Reaction: see text] Ruthenium-catalyzed coupling of allyl ethyl ether to optically active 1-trimethylsilyl-1-alkyn-3-ols, followed by in situ ketalization and Lewis-acid-induced cyclization, affords enantiomerically pure 1,5-oxygen-bridged eight- and nine-membered carbocycles. Opening of the oxygen bridge under basic or electron transfer conditions provides optically pure medium-sized carbocycles, products that are difficult to construct using other currently available methodologies.

Radical cyclization approach to spirocyclohexadienones.

Cyclization of an aryl radical at the ipso position of a p-O-aryl-substituted acetamide or benzamide generates oxindoles or quinolones bearing spirocyclohexadienone rings. This versatile reaction is applied to formal syntheses of the vasopressin inhibitor SR121463A and aza-galanthamine.

Improving catalyst scope: use of multiple aniline substrates to optimize a palladium-catalyzed bisdiene cyclization.

[reaction: see text] Combinatorial screening of five catalyst precursors and nine ligands with three substituted aniline trapping reagents uncovered a catalyst system that promotes efficient palladium-catalyzed cyclization-trapping with a series of substituted anilines of varying steric and electronic character. The results of the parallel optimization study illustrate the interdependency of the key reaction variables.

Tin-free radical cyclizations for the synthesis of 7-azaoxindoles, 7-azaindolines, tetrahydro[1,8]naphthyridines, and tetrahydro-5H-pyrido[2,3-b]azepin-8-ones.

[reaction: see text] Compounds containing a pyridine nucleus fused to a saturated nitrogen-containing ring, including 7-azaoxindoles, 7-azaindolines, tetrahydro[1,8]naphthyridines, and tetrahydro-5H-pyrido[2,3-b]azepin-8-ones, were prepared in good yield starting from various 2,6-dichloropyridines. The method hinges on a free-radical xanthate-mediated cyclization or intermolecular addition/cyclization sequence for the construction of the new fused rings.

alpha,5-didehydro-3-picoline diradicals from skipped azaenediynes: computational and trapping studies of an aza-Myers-Saito cyclization.

[reaction: see text] On the basis of density functional calculations, the isomerization of skipped azaenediynes (C-alkynyl-N-propargylimines) to azaenyne allenes and subsequent rapid aza-Myers-Saito cyclization to alpha,5-didehydro-3-picoline were predicted. We prepared the N-propargylimine of 1-phenyl-3-tri(isopropyl)silylprop-2-yn-1-one, which undergoes proto-desilylation and isomerization to an azaenyne allene when treated with tetrabutylammonium fluoride. In the presence of 1,4-cyclohexadiene, this azaenyne allene affords 6-phenyl-3-picoline and other products corresponding to the trapping of an alpha,5-didehydro-3-picoline diradical.
