It has been valued a new quick latex method (Staphaurex Wellcome) for the identification of the aureus Staphylococcus. This shown simultaneously the Clumping Factor (CF) and the A protein; the production of the free Coagulase (CL) and the Thermonuclease (TNase), has been utilized as methods of remarking. On 120 bacterial stocks isolated by many biological materials, belonging to the family of the Micrococcaceae, 56 stocks was identified as aureus. St. and 64 as not aureus St. For what concerns the quick Staphaurex method, it has shown a sensitivity of 83.4, with 8 false negatives on 120 cases, while it proved very satisfactory as specification with one false positive.

The nucleotide sequences of the 3' leader and 5' trailer regions were determined for genomic RNA of bovine respiratory syncytial virus (BRSV) strain A-51908. The leader and trailer sequences are '45' and '161' nucleotides in length, respectively. The functionality of BRSV leader and trailer sequences and their recognition by HRSV and ovine respiratory syncytial virus (ORSV) proteins were examined with a in vitro transcribed BRSV genomic RNA analog carrying the bacterial chloramphenicol acetyl transferase (CAT) gene under the control of BRSV transcription signals. Upon transfection into BRSV, HRSV or ORSV infected cells, the BRSV minireplicons were 'rescued' such that the reporter gene was expressed, the minigenome was replicated and packaged into micrococcal nuclease resistant-infectious minireplicons. The passage of infectious minireplicons could be blocked by a polyclonal BRSV neutralizing antiserum. Bovine parainfluenza virus-3, a heterologous paramyxovirus was inactive in rescuing BRSV genomic RNA analog. Mutational substitution of the G residue at position 4 of leader sequence in the BRSV genomic RNA analog, with an A or U residue inhibited its transcription and replication, while replacement with a C residue had no significant effect on rescue. These results show that the cis-acting elements of BRSV are functional and are also recognized by the proteins of HRSV and ORSV. The helper virus complemented rescue system developed here will be useful for characterizing the cis-acting elements of BRSV.
Prions were purified from scrapie-infected hamster brains and incubated for 24 hr at 65 degrees with 2 mM Zn2+ or 5 mM Mg2+; no loss of infectivity was observed. Bacteriophage M13, tobacco mosaic virus (TMV), potato virus X, and potato spindle tuber viroid were all inactivated by divalent metal ions under these conditions. Prions also resisted inactivation by prolonged digestions with DNase I, RNases A and T1, and micrococcal nuclease. Prions were resistant to psoralen photoadduct formation using high concentrations of psoralens; in contrast, M13 bacteriophage was inactivated by low concentrations of all these psoralens. Hydroxylamine failed to inactivate prions even after lengthy exposures to concentrations as high as 1 M, while TMV and M13 were both inactivated. Sonication of prions failed to decrease infectivity even though rod-shaped aggregates were disrupted while both M13 and TMV lost infectivity.
