When melatonin gets on your nerves: its beneficial actions in experimental models of stroke.

This article summarizes the evidence that endogenously produced and exogenously administered melatonin reduces the degree of tissue damage and limits the biobehavioral deficits associated with experimental models of ischemia/reperfusion injury in the brain (i.e., stroke). Melatonin's efficacy in curtailing neural damage under conditions of transitory interruption of the blood supply to the brain has been documented in models of both focal and global ischemia. In these studies many indices have been shown to be improved as a consequence of melatonin treatment. For example, when given at the time of ischemia or reperfusion onset, melatonin reduces neurophysiological deficits, infarct volume, the degree of neural edema, lipid peroxidation, protein carbonyls, DNA damage, neuron and glial loss, and death of the animals. Melatonin's protective actions against these adverse changes are believed to stem from its direct free radical scavenging and indirect antioxidant activities, possibly from its ability to limit free radical generation at the mitochondrial level and because of yet-undefined functions. Considering its high efficacy in overcoming much of the damage associated with ischemia/reperfusion injury, not only in the brain but in other organs as well, its use in clinical trials for the purpose of improving stroke outcome should be seriously considered.

[Evaluation of the OrthoPAT autologous transfusion system by experimental models simulating intra- and postoperative blood salvage].

OBJECTIVE: To evaluate the efficacy of the OrthoPAT (Haemonetics) system for blood salvage and for removing chemical or cellular debris, by experimental models simulating intra- and postoperative conditions. MATERIAL AND METHODS: Blood samples (20%-25% packed red cells) were prepared for the intraoperative model (n=8) and the postoperative model (n=22). Surgical compresses were soaked in some samples (n=5). Other samples were supplemented with hemolyzed blood (n=7). From others cytokines were removed and blood activated with bacterial liposaccharides (n=10) was added. The samples were analyzed before and after processing; tests included detection of free plasma hemoglobin (FPH), potassium ions (K+), glutamic oxalic transaminase (GOT), lactate dehydrogenase (LDH), proteins, and cytokines. RESULTS: In the intraoperative model 2935 (SD 260) mL of blood was processed. The concentration of packed red cells was 63% and 80% of the red cells were recovered. In the postoperative model 652 (35) mL was processed, the packed red cell concentration was 67% and 81% of the red cells were recovered. Reductions were observed in the concentrations of white blood cells (72%), platelets (88%), GOT and LDH (75%), and proteins and K+ (>95%). Fifty percent of the red cells were recovered in the surgical compresses model. In the hemolysis model, the K+ and FPH concentrations were reduced more than 95%. In the cytokine model, up to 90% of the interleukin 1beta, interleukin 6, and tumor necrosis factor content was removed from the activated blood samples. CONCLUSIONS: These findings suggest that the OrthoPAT system washes blood and salvages content effectively, recovering 80% of red cells. Moreover, its processing capacity (800-1000 mL x h(-1)) seems adequate for blood replacement in orthopedic surgery.

Prediction of velopharyngeal orifice area: a re-examination of model experimentation.

Warren has advanced a modification of the hydrokinetic equation for predicting velopharyngeal orifice area (Warren and DuBois, 1964). In the present work, this equation was subjected to extensive controlled model experimentation. The results of this experimentation show that, given the pressure differential across the orifice and the rate of airflow through the orifice, accurate predictions can be made of the area of the velopharyngeal port. These results were interpreted to provide strong support for both clinical and research use of the hydrokinetic equation for predicting velopharyngeal orifice area.

Disturbances in epithelial ionic secretion in different experimental models of colitis.

This paper studies the disturbances in ionic secretion in the colon of rats with different models of acute and chronic colitis measured as changes in short-circuit current. The aim was to verify whether the reported inhibition of basal and stimulated secretion in the trinitrobenzene sulfonic acid and mytomicin C models are applicable to experimental colitis as such. All models showed remarkable similarity in ion transport as determined in Ussing chambers, with downregulated basal as well as carbachol evoked secretion. The EC(50) of carbachol was unchanged in all cases. Iodoacetamide and oxazolone colitis models were notable exceptions in that the dose response curves for carbachol were unaltered compared to controls. The reason is unclear but seems to be unrelated to either interferon gamma or interleukin 4 levels or to the severity of the inflammatory response.

Relationship between contrast enhancement and diagnostic accuracy with the liver-specific CT contrast medium FP 736-04 in an experimental model of liver metastases.

RATIONALE AND OBJECTIVES: The authors sought to establish the optimal level of liver enhancement at computed tomography (CT) with a hepatocyte-specific contrast medium in an experimental model of liver metastases. MATERIALS AND METHODS: Anesthetized nude rats (n = 18) injected intraportally with cells from human colonic adenocarcinoma and Sprague-Dawley rats (n = 11) without metastases were used. A fractional intravenous infusion of contrast material was given in escalating doses up to a total of 4.0 mL per kilogram of body weight. Mean attenuation of the liver was calculated for each accumulated dose, and the overall detection rate was determined on a lesion-by-lesion basis. Sensitivity, specificity, and positive and negative predictive values were also established for each dose. RESULTS: Liver metastases were diagnosed with increasing reliability for enhancement values of up to 30.0 HU +/- 5.2 (mean +/- standard deviation), which corresponded to a dose of 0.5 mL/kg. CONCLUSION: In this model, optimal reliability in the diagnosis of liver metastases with CT was achieved at an enhancement value of approximately 30 HU.

The rabbit as an experimental model for regional chemotherapy. 1. Intra-arterial hindlimb infusion.

Regional delivery of chemotherapy to a tumour or tumour-bearing region has pharmacokinetic advantages over the systemic route. The applications of an animal model for regional drug delivery are outlined. The technique for intra-arterial infusion in the rabbit hindlimb is described. The use of the implantable VX2 rabbit carcinoma as a model for solid human tumours may be studied by this method. Pharmacokinetic data obtained with the model allow comparison between systemic and regional routes of delivery. The distribution of the cancerostatic plant toxin ricin following regional delivery has been investigated using this experimental model.

Rabbit cranial sutures in vitro: a new experimental model for studying the response of fibrous joints to mechanical stress.

An organ culture system has been developed whereby mechanical stress can be applied to cranial sutures under controlled experimental conditions. The application of a continuous tensile mechanical stress (30 g) to cranial sutures from newborn rabbits (1--2 days) was accompanied by a significant increase in the incorporation of 3H-leucine and 3H-proline into suture protein. The specific activities of 3H-hydroxyproline indicated that mechanical stress produced a two-fold increase in the incorporation of 3H-proline into collagen. However, the proportion of the total radioactivity recoverable in collagen (45.63 +/- 2.33% for nonstressed; 40.58 +/- 2.17% for stressed sutures) was not significantly different. These data suggest that the increase in collagen synthesis occurs as part of a general stimulation of protein synthesis, and do not support the view that mechanical stress is the principal mechanism regulating the turnover of collagen in fibrous joints. These initial studies demonstrate that an in vitro experimental model has considerable potential for investigating the morphological and metabolic response of fibrous joints to mechanical deformation.

[Asporogenic anaerobic thoracic empyema--an experimental model for anaerobic thoracic empyema in rabbits].

Sterile pleural effusion was induced by intrapleural administration of turpentine, and empyema was induced by direct intrapleural inoculation of bacteria in rabbits. Experimental models of anaerobic thoracic empyema were successfully made in this study, using Escherichia coli, Peptostreptococcus asaccharolyticus, Bacteroides fragilis, using the following methods; (1) a single bacterium inoculation of, a) E. coli, b) P. asaccharolyticus, c) B. fragilis, (2) simultaneous inoculations with an aerobe and an anaerobe, a) E. coli + P. asaccharolyticus, b) E. coli + B. fragilis. (3) an anaerobic inoculation 5 days after an aerobic inoculation, a) P. asaccharolyticus inoculation 5 days after E. coli inoculation, b) B. fragilis inoculation 5 days after E. coli inoculation. The developments of several parameters (sialic acid levels, glucose levels, oxidation-reduction potential levels, bacterial counts and WBC counts in the pleural fluid and so on) were measured in each group. There was little difference of the rate of incidence of empyema between each group. Empyema was highly induced even by a single anaerobe inoculation. The following tendencies were observed in development of parameters: (1) sialic acid levels and oxidation-reduction potential levels were decreased only in the conditions being associated by B. fragilis infections. (2) in mixed infections of E. coli and anaerobes, bacterial counts of E. coli increased in number in the late phases of the infections probably by the influences of anaerobes. The experiments of groups (2) and (3) were designed for demonstrations of biphasic infections between aerobes and anaerobes, but it turned out that quite unexpected results were seen in these models.

A comparison of the effects of troglitazone and vitamin E on the fatty acid composition of serum phospholipids in an experimental model of insulin resistance.

The aim of this study was to investigate the effects of troglitazone (TRO)--a new insulin-sensitizing agent--on some metabolic parameters in an experimental model of hypertriglyceridemia and insulin resistance, hereditary hypertriglyceridemic rats, and to compare its effects with those of vitamin E, an antioxidant agent. Three groups of the above rats were fed diets with a high content of sucrose (70% of energy as sucrose) for four weeks. The first group was supplemented with TRO (120 mg/kg diet), the second one with vitamin E (500 mg/kg diet), and the third group served as the control. Vitamin E supplementation did not lower serum triglycerides (2.42 +/- 0.41 vs. 3.39 +/- 0.37 mmol/l, N.S.) while TRO did (1.87 +/- 0.24 vs. 3.39 +/- 0.37 mmol/l, p < 0.01). Neither TRO nor vitamin E influenced the serum levels of free fatty acids (FFA). Both drugs influenced the spectrum of fatty acids in serum phospholipids--TRO increased the levels of polyunsaturated fatty acids (PUFA) n-6 (36.04 +/- 1.61 vs. 19.65 +/- 1.56 mol %, p < 0.001), vitamin E increased the levels of PUFA n-3 (13.3 +/- 0.87 vs. 6.79 +/- 0.87 mol %, p < 0.001) and decreased the levels of saturated fatty acids (32.97 +/- 0.58 vs. 51.45 +/- 4.01 mol %, p < 0.01). In conclusion, TRO lowered the level of serum triglycerides but vitamin E did not have this effect in hypertriglyceridemic rats. Compared with TRO, vitamin E had a different effect on the spectrum of fatty acids in serum phospholipids.

[A new experimental model of pulmonary hypertension in dog using an adjustable pulmonary artery band].

In order to study pathogenesis, reversibility, and treatment of pulmonary hypertension in congenital heart disease, a new experimental model to induce pulmonary vascular diseases has been designed. Previous models by other investigators were anastomosis between a systemic artery and pulmonary artery of one lung lobe, and these could not provide constant induction of pulmonary vascular diseases. Furthermore the postoperative mortalities in their experiences were quite high and acute iatrogenic pathological changes such as bleeding, edema and emphysema developed in the pulmonary vasculature and parenchyma of the anastomosed lobe. Use of our adjustable pulmonary artery band has eliminated such unwanted complications. The surgical procedure includes an end-to-side anastomosis of the left main pulmonary artery to the descending thoracic aorta, and banding of the anastomosed pulmonary artery using the adjustable band. The band was gradually loosened by extrathoracic manipulation during three to four weeks observation after operation. This technique provided dogs to survive sufficiently long enough and with a high success rate, and to produce extremely advanced pulmonary vascular diseases. Of the twenty dogs operated such, eighteen survived the first postsurgical week and thirteen dogs developed hypertensive pulmonary vascular diseases. Development of the medial hypertrophy was evident at one week and gradually advanced. In eight weeks, intimal cellular proliferation (Grade 2 of the Heath & Edwards' classification) became apparent. In twelve weeks, the intimal proliferation became fibrous and acellular (Grade 3) and caused complete obstruction of the lumen of some muscular arteries. In twenty weeks specimen, typical plexiform lesions (Grade 4) and dilatation lesions (Grade 5) involved the small muscular arteries in the whole left lung.(ABSTRACT TRUNCATED AT 250 WORDS)

Intraarticular sodium hyaluronate injections in the Pond-Nuki experimental model of osteoarthritis in dogs. II. Morphological findings.

The effect of the intraarticular sodium hyaluronate (HA) injection on the osteoarthritic knee joint has been evaluated in dogs using an experimental model of osteoarthritis induced by sectioning the anterior cruciate ligament. Seven weeks after surgery, the damage, graded according to Mankin's scale, was significantly reduced in knee joints treated with HA from the second week postsurgery compared to untreated joints. When intraarticular HA therapy was initiated after the seventh week, osteoarthritis progression was still reduced compared to controls. Both morphology and morphometry showed a beneficial effect of HA on the cartilage response to the damage, as well as a clearcut inhibitory effect on the development of the fibroblastlike cell layer on the articular cartilage in untreated joints. The beneficial effects on the cartilage integrity and response to osteoarthritic damage might be related to a primary effect of HA on the cartilage surface. However, these effects do not exclude the possibility that, in addition, HA might act on the synovial membrane by limiting the synovial reaction.

[Combined effect of human recombinant interferon-beta and interferon-gamma against an experimental model of peritonitis carcinomatosa in nude mice].

The development of useful therapy for the peritonitis carcinomatosa of gastrointestinal cancer is an important theme in cancer therapy. We developed an experimental model of peritonitis carcinomatosa in nude mice transplanted intraperitoneally human colon cancer cells. In this study, we investigated combined effect of human recombinant interferon-beta (rIFN-beta) and recombinant interferon-gamma (rIFN-gamma) in vivo using this model. The result indicate that each rIFN-beta and rIFN-gamma showed a significant prolonged survival compared with the control group (mean survival days: 41.8 +/- 5.0 days). Furthermore, combined administration of rIFN-beta and rIFN-gamma showed a marked prolonged survival (mean survival days: 114.0 +/- 7.4 days) compared with rIFN-beta or rIFN-gamma alone. These results suggest that a combined treatment of rIFN-beta and rIFN-gamma may be useful against peritonitis carcinomatosa of gastrointestinal cancer.

The hamster cheek pouch: an experimental model of corneal vascularization.

To gain insight into factors that might be responsible for the normal avascularity of the cornea and for its vascularization in certain pathologic states, an experimental model was designed in which corneal vascularization could be studied under controlled conditions in hamster cheek pouch chambers. Normal corneal tissue, as well as corneas that had been altered in a variety of ways (eg, boiled, autoclaved, freeze-thawed) were implanted into hamster cheek pouch chambers. The fate of the transplanted tissue was observed at regular intervals by direct visualization within the hamster cheek pouch at various magnifications and by light and electron microscopy. This report reviews observations on more than 300 such experiments. Normal and injured corneal autografts, allografts and xenografts and nonviable (autoclaved, boiled or freeze-thawed) corneas commonly became vascularized in the cheek pouch. When this occurred, a similar morphologic sequence of events preceded and accompanied the growth of blood vessels into the cornea. Vascular invasion was generally preceded by the formation of granulation tissue around the cornea. This was followed by a leukocytic, and frequently a fibroblastic, infiltration of the cornea. When cells did not invade the transplanted cornea, the cornea invariably remained avascular. In the present model, a swollen cornea was not a sufficient stimulus for corneal vascularization. The data suggest that under certain circumstances leukocytes may produce one or more factors which stimulate directional vascular growth. The findings are viewed in terms of current concepts on corneal vascularization.

An experimental model of leptomeningeal metastases employing rat mammary carcinoma cells.

Five percent of patients dying with breast cancer have leptomeningeal metastases (LM) but current therapy is of only marginal benefit. Therefore, an experimental model of LM from breast cancer was developed to facilitate the development of novel therapies. Cell suspensions of 13762 MAT BIII rat mammary carcinoma cells are injected into the cisterna magna of adult, female Fischer 344 rats under general anesthesia. 10-12 days after the injection of 2 x 10(5) viable cells, animals develop neurologic signs, including ataxia, paralysis and spontaneous rotation. Histologically, tumor cells can be seen in the subarachnoid space over the surface of the brain and spinal cord and within the ventricles. Tumor cells do not invade the brain parenchyma. Collections of tumor cells are extensively infiltrated by macrophages and CD8-positive (suppressor/cytotoxic) T cells, but by few CD4-positive (helper) T cells. MAT BIII cells therefore provide a model of LM from breast cancer with a reproducible clinical course and histologic features. The tumor elicits a cellular immune response and can be useful in exploring biologic therapies for leptomeningeal metastases.

Apoptosis and nitric oxide in an experimental model of osteoarthritis in rabbit after hyaluronic acid treatment.

OBJECTIVE: this study determine the effect of hyaluronic acid on chondrocyte apoptosis, as well as variations in nitric oxide levels in an experimental model of osteoarthritis elicited anterior cruciate ligament section (ACL) in a rabbit model with two differentiated developmental periods. METHODS: apoptosis and the quantification of nitric oxide (NO) were studied in two groups of 16 animals each. All animals had both knees operated, but only the right knees were treated with hyaluronic acid (HA). In the first group hyaluronic treatment was performed five weeks after osteoarthritis induction (short term group, ST) and in the second group, 10 weeks after induction (long term group, LT). The animals in both series were sacrificed two weeks after the last dose of HA. Flow cytometry by means of Annexin labelling and the TUNEL method were used for the study of apoptosis, NO levels were measured in cultured cartilage and in the supernatant of the enzymatic digestion of the cartilage. RESULTS: regarding apoptosis measurement, a significant reduction in apoptosis levels was observed in both series as compared to untreated knees. NO production was lower in knees treated with HA, with significant differences after cartilage digestion. CONCLUSION: The administration of HA has been effective ameliorating the damage associated with the process of osteoarthritis induced by experimental surgery as evidenced by decreased apoptosis (TUNEL method), the results more promising in the earlier phases of the disease.

An experimental model of human leukemic meningitis in the nude rat.

An experimental animal model of meningeal leukemia was developed in the nude rat, rnu/rnu, using the human-derived acute lymphoblastic leukemia cell line HPB-ALL. Anesthetized rats were placed in a modified stereotaxic frame and then injected intrathecally, at the level of the cisterna magna, with human leukemic cells. Cerebrospinal fluid and tissue samples from brain, spinal cord, heart, liver, kidney, spleen, bone marrow, and cervical lymph nodes were subjected to histopathologic examination and molecular genetic screening by clonotype primer-directed polymerase chain reaction (CPD-PCR). Ninety-three percent of animals (n = 14) developed signs of meningeal irritation leading to death 30 to 63 days postinjection (median, 36.0 days, mean, 38.7); death occurred between 30 and 39 days in 77% of all animals. Leukemic cells progressively infiltrated the pericerebellar and pericerebral subarachnoid space and infiltrated the Virchow-Robin (perivascular) space. The infiltrating meningeal leukemia closely resembled the pathologic presentation in the human condition. By CPD-PCR, leukemic cells were first detected in cerebrospinal fluid (CSF) on day 4 postinjection, were variably present over the ensuing 17 days, and were consistently detected after day 21. At terminal stages, CPD-PCR tissue surveys showed leukemic DNA in all brains and spinal cords and rarely in cervical lymph nodes, but leukemic DNA was not detected in any other tissue screened. Leukemic meningitis was reliably produced with a predictable survival time. Intrathecal administration of leukemic cells was an efficient means of transmitting leukemic meningitis and it compartmentalized the disease to the central nervous system (CNS), eliminating potential complications of systemic illness. The use of human-derived cell lines may render this model more relevant to the development of future therapeutic strategies to treat leukemia and lymphoma that invade the CNS.

Novel experimental models of human cancer metastasis in nude mice: lung metastasis, intraabdominal carcinomatosis with ascites, and liver metastasis.

The ability of RPMI 4788 cells, a human colon cancer cell line, to produce experimental metastases in the lung, intraperitoneal cavity, and liver was studied in nude mice. Injection of 2 X 10(6) tumor cells into the tail vein of nude mice produced metastatic lung tumors, and an intraportal injection of 5 X 10(6) cells produced metastatic liver tumors. An intraabdominal carcinomatosis with ascites was formed after an i.p. injection of 5 X 10(6) tumor cells. The nude mice with lung metastasis or intraabdominal carcinomatosis always died within a few weeks. Macroscopic observation showed that the number of lung metastatic nodules on day 21 after tumor inoculation was 311.3 +/- 78.2 (mean +/- SD) in BALB/C nude mice, and 187.5 +/- 26.7 in ICR nude mice. In survival experiments, the mice with intraabdominal carcinomatosis showed a mean survival of 29.0 +/- 1.7 (mean +/- SD) days in BALB/C nude mice and 43.6 +/- 6.1 days in ICR nude mice. These novel experimental models of metastases in nude mice produced by injection of RPMI4788 cells had high reproducibility and may be useful not only for the study of the metastatic process but also for testing anticancer drugs.

Correlation between ossification and inflammation using a rat experimental model.

In seronegative spondylarthropathies both inflammation and ossification can be demonstrated. Inflammation is a hallmark of diseases associated with antigen HLA-B27 in ankylosing spondylitis, Reiter's syndrome, and acute uveitis. Ossification is traditionally considered the end product of inflammation, but clinical examination does not show that this is always the case in man. The relationship between inflammation and ossification is not demonstrated in experiments on spinal involvement in adjuvant arthritis in the rat. Using that experimental model, we tested the efficacy of 3 nonsteroidal antiinflammatory drugs (indomethacin, naproxen, and phenylbutazone) given at dosages comparable to those employed in clinical practice, but at a lower level than those used by drug companies in animals. Results show that the drug exhibiting almost no antiinflammatory activity in the rat at the dosage used, phenylbutazone, was the most powerful inhibitor of ossification. Another mechanism of local osteogenesis must be sought to explain such a phenomenon.
