Apolipoprotein C-III displacement of apolipoprotein E from VLDL: effect of particle size.

ApoC-III and apoE are important determinants of intravascular lipolysis and clearance of triglyceride-rich chylomicrons and VLDL from the blood plasma. Interactions of these two apolipoproteins were studied by adding purified human apoC-III to human plasma at levels observed in hypertriglyceridemic subjects and incubating under specific conditions (2 h, 37 degrees C). As plasma concentrations of apoC-III protein were increased, the contents in both VLDL and HDL were also increased. Addition of apoC-III at concentrations up to four times the intrinsic concentration resulted in the decreasing incremental binding of apoC-III to VLDL while HDL bound increasing amounts without evidence of saturation. No changes were found in lipid content or in particle size of any lipoprotein in these experiments. However, distribution of the intrinsic apoE in different lipoprotein particles changed markedly with displacement of apoE from VLDL to HDL. The fraction of VLDL apoE that was displaced from VLDL to HDL at these high apoC-III concentrations varied among individuals from 20% to 100% its intrinsic level. The proportion of VLDL apoE that was tightly bound (0% to 80%) was found to be reproducible and to correlate with several indices of VLDL particle size. In the group of subjects studied, strongly adherent apoE was essentially absent from VLDL particles having an average content of less than 50,000 molecules of triglyceride. Addition of apoC-III to plasma almost completely displaces apoE from small VLDL particles. Larger VLDL contain tightly bound apoE which are not displaced by increasing concentration of apoC-III.

Natural 13C abundance: a tool to trace the incorporation of dung-derived carbon into soil particle-size fractions.

During the decay of 13C enriched dung patches, the; delta 13C signal of surface soil (1-5 cm) increased with a temporary maximum after 42 d. To understand the underlying processes, we investigated the incorporation of dung-derived C into soil particle-size fractions. Dung, collected from beef steers fed on maize (delta 13C = -15.36/1000) or ryegrass (delta 13C = -25.67/1000), was applied in circular patches to a C3 pasture at North Wyke, UK. Triplicates were sampled from surface soil (1-5 cm) at 14, 28, 42, and 70 d after application, pooled, separated into fine (< 0.2 micron) and coarse clay (0.2-2 microns), silt plus fine sand (2-250 microns), and coarse sand (250-2000 microns), and analyzed for total C, N, and delta 13C. As particle-size diameter decreased, the C/N ratios decreased and delta 13C values increased at all plots due to increasing microbial alteration of soil organic matter. After dung application, ca. 60% of dung-derived C in soil was recovered in the 0.2-250 microns fractions during the whole experiment. The proportion of dung-derived C in the fine clay peaked 42 d after dung application, coinciding with the delta 13C maximum in the bulk soil and the maximum leaching rate measured in lysimeters at this time in another study at the same sites. The percentage of dung-derived C as particulate C in the coarse sand fraction increased until the end of the experiment. We conclude that incorporation of C into soil from decomposing dung patches involved both temporary sorption of leached dung C to < 0.2 micron fractions and continuous accumulation of particulate C (> 250 microns).

Particle size, satiety and the glycaemic response.

OBJECTIVE: This study investigated the hypothesis that the smaller the particle size of the food, the higher the glycaemic-insulin response and the lower the satiety rating. DESIGN AND SUBJECTS: Ten healthy subjects consumed equal carbohydrate portions of four test meals of equivalent nutritional composition based on four different grades of wheat: whole grains, cracked grains, coarse and fine wholemeal flour. RESULTS: The mean overall plasma glucose response (area under the curve: AUC) was highest for the fine flour meal (4205 +/- 451 mmol/l.120 min), followed by the coarse flour and cracked grain meals and lowest for the whole grain meal (1524 +/- 190 mmol/l.120 min). Insulin responses showed the same ranking but satiety responses were the reverse, the fine flour meal giving the lowest satiety response [AUC: 231.4 +/- 31.6 rating scale (RS) units.120 min] and the whole grain meal the highest response (AUC: 318.4 +/- 29.3 RS units.120 min). Total insulin responses (AUC) were significantly associated with subjective satiety when analyses on an individual basis (analysis of covariance coefficient = -0.024, P < 0.04) such that for every 1000 unit increase in AUC insulin, satiety decreased by 24 units (13% on average). CONCLUSION: The results suggest that the processing of cereals is not only a major determinant of the glycaemic and insulin response but also affects the levels of postprandial satiety.

[Stabilization of two patients with brittle asthma by inhaled beclomethasone dipropionate with small particle size].

UNLABELLED: Two patients with brittle asthma whose bronchial obstruction was less variable during treatment with HFA-beclomethasone (HFA-BDP) solution aerosol than with other previous treatments are presented here. In order to evaluate whether this improvement was related to the smaller particle size of the new formulation (MMAD 1.1 mu vs 4 mu with the CFC-formulation) both patients participated in a prospective case study sequence. METHOD: During a 4 week run-in both patients inhaled 200 micrograms of HFA-BDP (Ventolair) BID from the Autohaler followed by 4 weeks of treatment with 500 micrograms CFC-BDP (Aerobec) BID from the Autohaler in study phase 1 and 4 weeks of treatment with 200 mcg HFA-BDP (Ventolair) BID from the Autohaler in study phase 2. During the entire study period other concomitant medications remained unchanged. The dose of CFC-BDP was chosen to be 2.5 times higher than the HFA-BDP dose to get approximately comparable amounts of intrabronchial deposition. During the study Peak-Flow and concomitant medications were recorded daily. RESULTS: Both patients showed significantly higher Peak-flow values during treatment with Ventolair (HFA-BDP) than during treatment with AeroBec (CFC-BDP). P-values were p < 0.0001 and p < 0.005 for patient 1 and 2 respectively. CONCLUSION: At a comparable intrabronchial dose these two cases of brittle asthma showed significant improvements in control of bronchial obstruction with a BDP-formulation of smaller particle size. This is an indicator that smaller airways in the periphery of the lung participate in the inflammatory process leading to bronchial obstruction and that deposition of inhaled steroids in this region could have therapeutic advantages.

Particle size study of nine metered dose inhalers, and their deposition probabilities in the airways.

This study deals with the particle size measurement of nine aerosol metered dose inhalers. Calibration was made possible by the use of a laser particle velocimeter (aerodynamic Particle Sizer from TSI). The count median aerodynamic diameters (CMAD) show little variation, from 0.63 to 0.73 micron, with standard deviations (sigma g) between 1.2 and 1.8. Aerodynamic diameter aerosol diagram analysis showed multimodal mass distribution for all the tested dose inhalers. Calculations for the airway deposition probabilities (extrathoracic, tracheobronchial and alveolar) refer to the studies made by W. Stahlhofen and co-workers. As most aerosol metered dose inhalers have a predominantly bronchial therapeutic destination, the deposition at the bronchial level could be enhanced with the following parameters: inspired volume of 1500 ml, inspiratory time of 2 sec, aerosol mass median aerodynamic diameter (MMAD) of 7.5 microns, with a monodispersed distribution. The respective influences of the excipients and propellents used for the aerosolization of these dose metered inhalers are also discussed.

Leaching of metals from sewage sludge during one year and their relationship to particle size.

Leaching of metals from sewage sludge can lead to their accumulation in topsoil and can also contaminate groundwater. Our objectives were to document the metal leachates and the size distribution of leached particles from sewage sludge and to identify possible correlations with physical factors. Results from monthly lysimeter sampling showed an initial release followed by decline for most metals. Cadmium, Ca, Sr, Li, Mn, Ni and Zn showed a "cyclic" behaviour. Filtration revealed that this "cyclicity" had no correlation to the size of released particles, but Al, Cr, Fe, Cu, Ag and Pb were clearly related to release of coarser particles most of the year. Total metal amounts leached during one year, relative to original sludge content, had the order Na>Ca=Mg>Mn>Sr>Zn>K>Li=Ni>Cd>Co>Rb>Ag>Cr>Ba=Cu>Ga>Al=Pb=Fe. There were no simple correlations between monthly measured leachate concentrations and precipitation, temperature or pH of precipitation. Occasional leachate sampling might give misleading values for metals with "cyclic" behaviour.

[Particle size determination by radioisotope x-ray absorptiometry with sedimentation method].

The possibility of radioisotope X-ray absorptiometry to determine the particle size of powder in conjunction with sedimentation was investigated. The experimental accuracy was primarily determined by Cow and X-ray intensity. where Co'=weight concentration of the particle in the suspension w'=(micron/rho)l/(mu/rho)s-rhol/rhos rho; density micron/rho; mass absorption coefficient, suffix l and s indicate dispersion and particle, respectively. The radiosiotopes, Fe-55, Pu-238 and Cd-109 have high w-values over the wide range of the atomic number. However, a source of high micron value such as Fe-55 is not suitable because the optimal X-ray transmission length, Lopt is decided by the expression, micronlLopt approximately 2/(1+C'ow') by using Cd-109 AgKX-ray source, the weight size distribution of particles from the heavy elements such as PbO2 to light elements such as Al2O3 or flyash was determined.

An improved stereologic method for three-dimensional estimation of particle size distribution from observations in two dimensions and its application.

Single enzyme-altered hepatocytes; altered hepatic foci (AHF); and nodular lesions have been implicated, respectively in the processes of initiation, promotion, and progression in rodent hepatocarcinogenesis. Qualitative and quantitative analyses of such lesions have been utilized both to identify and to determine the potency of initiating, promoting, and progressor agents in rodent liver. Of a number of possible parameters determined in the study of such lesions, estimation of the number of foci or nodules in the liver is very important. The method of Saltykov has been used for estimating the number of AHF in rat liver. However, in practice, the Saltykov calculation has at least two weak points: (a) the size class range is limited to 12, which in many instances is too narrow to cover the range of AHF data obtained; and (b) under some conditions, the Saltykov equation generates negative values in several size classes, an obvious impossibility in the real world. In order to overcome these limitations in the Saltykov calculations, a study of the particle size distribution in a wide-range, polydispersed sphere system was performed. A stereologic method, termed the 25F Association method, was developed from this study. This method offers 25 association factors that are derived from the frequency of different-sized transections obtained from transecting a spherical particle, thus expanding the size class range to be analyzed up to 25, which is sufficiently wide to encompass all rat AHF found in most cases. This method exhibits greater flexibility, which allows adjustments to be made within the calculation process when NA((k,k)), the net number of transections from the same size spheres, was found to be a negative value, which is not possible in real situations. The reliability of the 25F Association method was tested thoroughly by computer simulation in both monodispersed and polydispersed sphere systems. The test results were compared with the original Saltykov method. We found that the 25F Association method yielded a better estimate of the total number of spheres in the three-dimensional tissue sample as well as the detailed size distribution information. Although the 25F Association method was derived from the study of a polydispersed sphere system, it can be used for continuous size distribution sphere systems. Application of this method to the estimation of parameters of preneoplastic foci in rodent liver is presented as an example of its utility. An application software program, 3D_estimation.exe, which uses the 25F Association method to estimate the number of AHF in rodent liver, has been developed and is now available at the website of this laboratory.

Gemfibrozil treatment increases low-density lipoprotein particle size in Type 2 diabetes mellitus but does not alter in vitro oxidizability.

The aim of this study was to determine the effect of the lipid modifying agent gemfibrozil on lipid and coagulation risk factors in patients with Type 2 diabetes mellitus (Type 2 DM). Twenty-six subjects with Type 2 DM and dyslipidaemia were treated for 24 weeks with either gemfibrozil 600 mg orally twice daily or placebo in a double-blind randomized trial. Lipid profiles, fibrinogen, Factor VII, and plasminogen activator inhibitor-1 (PAI-1) were measured by routine laboratory methods. Low density lipoprotein (LDL) size was determined by gradient gel electrophoresis and the resistance of LDL to copper-induced oxidation was assessed by measuring absorbance at 234 nm. Gemfibrozil significantly reduced total cholesterol (-0.9 (-0.48, -1.32) mmol l(-1); p < 0.05) and triglycerides (-2.7 (-1.55, -1.35) mmol l(-1); p < 0.001) vs placebo. The fall in triglyceride was reflected by a fall in VLDL cholesterol levels in the gemfibrozil treated group vs placebo (-1.31 mmol l(-1); p < 0.001). LDL-cholesterol level did not change but LDL particle size increased by 0.5 nm (0.01, 0.93); P < 0.02. The increase in particle size was inversely correlated with the change of triglyceride level (r = -0.79, p < 0.0001) but did not result in any reduction of susceptibility to copper-induced oxidation. There were no significant changes in the coagulation parameters studied. Because of its ability to correct the lipid abnormalities associated with Type 2 DM particularly hypertriglyceridaemia, gemfibrozil provides a useful therapeutic option in the management of diabetic dyslipidaemia but it does not alter in vitro oxidizability of LDL.

Low-density lipoprotein particle size, triglycerides, and high-density lipoprotein cholesterol as risk factors for coronary heart disease in older Japanese-American men.

Decreased low-density lipoprotein (LDL) particle size is associated with coronary heart disease (CHD) risk among middle-aged Caucasian populations, and has been consistently correlated with increased plasma levels of triglyceride and decreased levels of high-density lipoprotein (HDL) cholesterol. This study examines whether these risk factors predict CHD among older Japanese-American men. With use of the Honolulu Heart Program Lipoprotein Exam 3 (1980 to 1982) as baseline, and 12-year follow-up for CHD events, a nested, case-control study was designed. One hundred forty-five incident CHD cases were identified and matched to 2 controls each. LDL particle diameter (size) was determined by gradient gel electrophoresis. A 10-angstrom (A) decrease in LDL size at baseline was associated with increased risk of incident CHD (relative risk 1.28, 95% confidence interval 1.01 to 1.63). After adjustment for baseline risk factors, the LDL size association was no longer statistically significant (relative risk 1.13, 95% confidence interval 0.86 to 1.49). When principal components analysis was used to define a composite variable for LDL size, triglycerides, and HDL cholesterol, this component predicted CHD independent of smoking, alcohol consumption, physical activity, body mass index, hypertension, diabetes, and beta-blocker use (p <0.01). Therefore, this prospective analysis of data from older, Japanese-American men demonstrated that decreased LDL size is a univariate predictor of incident CHD, and that a composite risk factor of LDL size, triglyceride, and HDL cholesterol was a risk factor for CHD independent of other risk factors.

Particle size and magnetic field-induced optical properties of magnetic fluid nanoparticles.

Magnetite nanoparticles with diameters of 7, 9, and 12 nm have been prepared by a chemical coprecipitation method. The transmission of light through magnetic fluid containing these nanoparticles has been investigated as a function of film thickness with wavelength between 400 and 750 nm, and applied magnetic fields up to 275 Oe. The transmission threshold shifts to the lower wavelength side with decreasing magnetic fluid film thickness as well as the particle size. For a given film thickness, the transmittance increases with increasing magnetic field for films with a particle size of 7 and 9 nm, but decreases in the 12-nm film. This is attributed to the competition between the van der Waals and dipole-dipole interaction.

Broadband dielectric and resistivity spectroscopy of WO(3) x H(2)O in the range of 10(3)-10(10) Hz: particle size effect.

The interest in studying the electrical properties of WO(3) x H(2)O powders is made absolutely necessary because their infrared modulation properties depend on their morphologies and electronic populations. Broadband dielectric and resistivity spectra of WO(3) x H(2)O powders were recorded in a frequency range of 10(3)-10(10) Hz at temperatures varying between 200 and 300 K. Complex resistivity and permittivity diagrams have permitted thermal behavior of both dc-conductivity and permittivity to be obtained. A dielectric relaxation is found, attributed to water molecules motions. The role of the powder morphology has been investigated on two types of compounds: the first one being constituted by nanometric particles and the second by micrometric particles. Strong differences are observed in the thermal behaviors of the dc-conductivities (activation energies). Particle size effect is evidenced, giving rise to stronger electron localization on the nanometric particles. The permittivity values and the dynamical behavior of the structural water are also influenced by the particle size effect. A strong interaction between moving polarons and water molecules has been determined.

[Fractal characteristics of particle size distributions of mangroves soils in Yingluo Bay].

Based on fractal theory, the fractal characteristics of particle-size distributions of mangrove soils in Yingluo Bay (21 degrees 28'N, 109 degrees 43'E) were studied. The results show that the fractal dimensions of the soils ranged from 2.6837 to 2.8834, and decreased in the order of sand loam < light loam < medium loam < heavy loam < light clay. The fractal dimensions of the soils on exterior beach were lower than those on middle and inner beaches. There was a significant positive linear relationship between fractal dimension and soil salinity and organic matter content. The major factors that influenced the fractal dimensions of the soils were community type, soil texture, beach position, soil salinity, and soil organic matter content.

Relation of anthropometric parameters to the functional assessment of HDL particle size in three ethnic cohorts.

The purpose of this study was to characterize the association between HDL particle size (assessed by fractional esterification rate in apo B depleted plasma (FERHDL)), and anthropometric measures in men and women of Chinese, European and South Asian origin and to determine if ethnic background is a modifier of this relationship. A convenience sample of apparently healthy men and women of Chinese (n=56), European (n=66) or South Asian (n=76) descent were recruited and assessed for body mass index, waist circumference, blood pressure, lipids, insulin, glucose and FERHDL. Univariate correlations with FERHDL were determined within each ethnic-gender group. Regression analysis was used to determine if ethnicity was a significant modifier of the relationship between FERHDL and waist circumference. FERHDL was significantly correlated with various anthropometric measures within the ethnic and gender groups. The relationship between waist circumference and FERHDL was significantly different between the European, and Chinese and South Asian groups combined in women, but not in men. After adjustment for age, body mass index and insulin, ethnicity was no longer a significant predictor. However, ethnic background (European vs. Chinese/South Asian) was a significant predictor (P=0.034) of the relationship between FERHDL and waist circumference adjusted for age, gender, body mass index and insulin in the entire cohort. Increasing adiposity in Chinese and South Asian men and women is associated with a less protective HDL particle profile. Ethnic background (Asian compared to European) is a significant modifier of the relationship between FERHDL and waist circumference.

Measurement of ultrafine particle size distributions from coal-, oil-, and gas-fired stationary combustion sources.

Currently, we have limited knowledge of the physical and chemical properties of emitted primary combustion aerosols and the changes in those properties caused by nucleation, condensation growth of volatile species, and particle coagulations under dilution and cooling in the ambient air. A dilution chamber was deployed to sample exhaust from a pilot-scale furnace burning various fuels at a nominal heat input rate of 160 kW/h(-1) and 3% excess oxygen. The formation mechanisms of particles smaller than 420 nm in electrical mobility diameter were experimentally investigated by measurement with a Scanning Mobility Particle Sizer (SMPS) as a function of aging times, dilution air ratios, combustion exhaust temperatures, and fuel types. Particle formation in the dilution process is a complex mixture of nucleation, coagulation, and condensational growth, depending on the concentrations of available condensable species and solid or liquid particles (such as soot, ash) in combustion exhausts. The measured particle size distributions in number concentrations measured show peaks of particle number concentrations for medium sulfur bituminous coal, No. 6 fuel oil, and natural gas at 40-50 nm, 70-100 nm, and 15-25 nm, respectively. For No. 6 fuel oil and coal, the particle number concentration is constant in the range of a dilution air ratio of 50, but the number decreases as the dilution air ratio decreases to 10. However, for natural gas, the particle number concentration is higher at a dilution air ratio of 10 and decreases at dilution air ratios of 20-50. At a dilution air ratio of 10, severe particle coagulation occurs in a relatively short time. Samples taken at different combustion exhaust temperatures for these fuel types show higher particle number concentrations at 645 K than at 450 K. As the aging time of particles increases, the particles increase in size and the number concentrations decrease. The largest gradient of particle number distribution occurs within the first 10 sec after dilution but shows only minor differences between 10 and 80 sec. The lifetimes of the ultrafine particles are relatively short, with a scale on the order of a few seconds. Results from this study suggest that an aging time of 10 sec and a dilution air ratio of 20 are sufficient to obtain representative primary particle emission samples from stationary combustion sources.

Fasting insulin and apolipoprotein B levels and low-density lipoprotein particle size as risk factors for ischemic heart disease.

CONTEXT: Epidemiological studies have established a relationship between cholesterol and low-density lipoprotein cholesterol (LDL-C) concentrations and the risk of ischemic heart disease (IHD), but up to half of patients with IHD may have cholesterol levels in the normal range. OBJECTIVE: To assess the ability to predict the risk of IHD using a cluster of nontraditional metabolic risk factors that includes elevated fasting insulin and apolipoprotein B levels as well as small, dense LDL particles. DESIGN: Nested case-control study. SETTING: Cases and controls were identified from the population-based cohort of the Quebec Cardiovascular Study, a prospective study conducted in men free of IHD in 1985 and followed up for 5 years. PARTICIPANTS: Incident IHD cases were matched with controls selected from among the sample of men who remained IHD free during follow-up. Matching variables were age, smoking habits, body mass index, and alcohol consumption. The sample included 85 complete pairs of nondiabetic IHD cases and controls. MAIN OUTCOME MEASURES: Ability of fasting insulin level, apolipoprotein B level, and LDL particle diameter to predict IHD events, defined as angina, coronary insufficiency, nonfatal myocardial infarction, and coronary death. RESULTS: The risk of IHD was significantly increased in men who had elevated fasting plasma insulin and apolipoprotein B levels and small, dense LDL particles, compared with men who had normal levels for 2 of these 3 risk factors (odds ratio [OR], 5.9; 95% confidence interval [CI], 2.3-15.4). Multivariate adjustment for LDL-C, triglycerides, and high-density lipoprotein cholesterol (HDL-C) did not attenuate the relationship between the cluster of nontraditional risk factors and IHD (OR, 5.2; 95% CI, 1.7-15.7). On the other hand, the risk of IHD in men having a combination of elevated LDL-C and triglyceride levels and reduced HDL-C levels was no longer significant (OR, 1.4; 95% CI, 0.5-3.5) after multivariate adjustment for fasting plasma insulin level, apolipoprotein B level, and LDL particle size. CONCLUSION: Results from this prospective study suggest that the measurement of fasting plasma insulin level, apolipoprotein B level, and LDL particle size may provide further information on the risk of IHD compared with the information provided by conventional lipid variables.

Particle-size analysis of pharmaceutical powders.

An automated electrolytic sensing zone (electrozone) method was developed to determine the particle-size distribution of milled and micronized pharmaceutical powders. The powdered drugs obeyed log-normal statistics, and the distributions were well defined by thier geometric volume mean diameter and the geometric standard deviation. The results show that accurate data can be obtained between 2 and 80 micron with a precision of approximately 0.5 micron. Pulse-width analyses were performed to determine the feasibility of using a pulse-width discrimination program. However, in this case, the program discriminates against real particles and, therefore, its usefulness is limited. Milled and micronized materials are described adequately by a spherical diameter, and the automated electrozone system described is an excellent method for quality control purposes.
