[The importance of the substrate for the induction of tyrosine-alpha-ketoglutarate-transaminase in RLC-cells (author's transl)].

1. Dexamethasone phosphate causes approximately a threefold increase of the tyrosine-alpha-ketoglutarate transaminase in the culture of RLC cells. 2. The induction of the enzyme depends on the presence of L-tyrosine. Omission of L-leucine or L-tryptophan, respectively, has no effect. 3. Omission of L-tyrosine influences the activity of the lactate dehydrogenase and malate dehydrogenase not at all and that of the glucose-6-phosphate-dehydrogenase only to a small extent. 4. In the absense of L-tyrosine an superinduction takes also place by actinomycin.

[Induction of hepatic tyrosine transaminase in rats by phenothiazine derivatives and analogs].

We have showed induction of tyrosine-alpha-ketoglutarate transaminase in hepatic cytosol of Rats (Wistar strain) five hours after intraperitoneal administration of tricyclic compounds (phenothiazine, iminodibenzyl, thioxanthene, thiophenylpyridylamin, dibenzocycloheptadiene, dibenzoxepin derivatives). Chemical structure of these molecules is very important: sulfur atom (phenothiazine, thioxanthene), some substituants like chlorine (chlorpromazine, chlorprothixene) and 2'-dimethylaminopropyl chain (promethazine) increase this inductive effect.

Immunohistochemical localization of glutamine transaminase K, a rat kidney cysteine conjugate beta-lyase, and the relationship to the segment specificity of cysteine conjugate nephrotoxicity.

Rat kidney glutamine transaminase K is a major rat kidney cysteine conjugate beta-lyase and is a key enzyme in the nephrotoxicity of some cysteine conjugates. However, it has not been demonstrated that the beta-lyase is present in the target cells. Furthermore, although all segments of the proximal tubule are affected by high doses of nephrotoxic cysteine conjugates, the S3 segment is the most sensitive. Because heterogeneous distribution of the beta-lyase could account for the enhanced sensitivity, antibody raised against rat kidney cysteine conjugate beta-lyase has been prepared and used to investigate the distribution of the enzyme in kidney and other tissues. The data show that the enzyme is highest in rat kidney, consistent with enzyme activity data. By immunohistochemical staining, no enzyme is present in the glomeruli or distal tubular elements of the kidney. The enzyme is present only in the target cells, the renal proximal tubular epithelium. However, the distribution of the beta-lyase within the proximal tubule is not consistent with the hypothesis that a higher concentration of the enzyme in the S3 segment accounts for the greater sensitivity of S3 to nephrotoxic cysteine conjugates compared to S1 and S2. Several alternative hypotheses are discussed.

[The effect of starvation on GABA-transaminase and glutamate decarboxylase activity on mitochondria of dog limbic structures during postnatal ontogenesis].

Activities of GABA-transaminase (GABA-TA) and glutamate decarboxylase (GDC) were estimated in mitochondria of limbic cortex (L1 and L2 areas), hypothalamus, hippocampus, amygdala and midbrain reticular formation (RF) of 3 months and 1 year old dogs during 5, 12 and 20 days of starvation. Total activity of GABA-TA and GDC was distinctly decreased in mitochondria of limbic cortex, hippocampus, hypothalamus and amygdala. At the same time, after 5 days starvation activity of GDC, in presence of 0.1 mM PALP or without it was increased in RF mitochondria of 3 months old dogs, while GABA-TA was activated in the mitochondria of 1 year old dogs. The rate of GDC activation by PALP was dissimilar in certain structures of brain limbic areas during various periods of starvation in 3 months old dogs, which occurred due to difference of the GDC mitochondrial forms in synaptic structures of the dog brain limbic system. Activity of GABA-TA in 3 months and 1 year old dogs as well as activity of GDC, in presence of PALP or without it, in 3 months old dogs were distinctly decreased in mitochondria of all the dog brain limbic structures studied after long-term deprivation within 12 and 20 days. The decrease in activity of GABA-TA and GDC was related to duration of starvation as well as to morpho-functional alterations and adaptation of the brain structures studied during postnatal ontogenesis.

Changes in the electroencephalographic and gamma-aminobutyric acid transaminase and succinic semialdehyde dehydrogenase in the allergen induced rat brain.

Electroencephalographic activity and gamma-Aminobutyric acid Transaminase together with Succinic semialdehyde dehydrogenase activity changes produced by sensitization with Prosopis juliflora pollen allergen were studied in the cerebral cortex and hypothalamus of the rat brain. Electrical activity of EEG recording begins to appear on 3rd day after sensitization with maximum increase in activity was found on day 9 and decreased after that. A sudden increase in electrical activity was produced in 9th day sensitized rat with 10 min after giving challenging dose intravenously. The measurement of enzymatic activity of GABA-T and SSA-DH showed decrease and increase in 3, 9, 15 and 30 days sensitized rat hypothalamus and cerebral cortex whole homogenate and mitochondrial fractions. A maximum changes in enzymatic activity was found in 9th day sensitized rat with significant alterations after giving sudden stress as challenging dose. These changes in EEG activity and GABA-ergic neurotransmitter in allergenic rats showed the immunoregulatory role of nervous system mediated via GABA shunt.

In vivo action of enzyme-activated irreversible inhibitors of glutamic acid decarboxylase and gamma-aminobutyric acid transaminase in retina vs. brain.

The effects of low s.c. doses of gamma-acetylenic gamma-aminobutyric acid (GAG) on glutamic acid decarboxylase (GAD) and gamma-aminobutyric acid transaminase (GABA-T) activities, as well as of gamma-vinyl GABA (GVG) and gabaculine on GABA-T activities, were examined using preparations from retina and several other regions of rat central nervous system (CNS). GAG, in doses of 5 to 50 mg/kg, inactivated retinal GAD to a significantly greater degree than GAD from any other CNS region studied. Retinal GABA-T activities were also differentially inactivated by 1 to 50 mg/kg of GAG, 50 mg/kg of GVG, or 1 and 5 mg/kg of gabaculine. GAG, in doses of 25 and 50 mg/kg, more completely inactivated GAD and GABA-T in frontal cortex than in other brain regions. Frontal cortical GABA-T was not differentially inactivated by 10 and 50 mg/kg of GVG or 1 and 5 mg/kg of gabaculine. The effects of GAG on retinal GABA enzymes were long-lasting and not reversed by dialysis. The GAD and GABA-T activities from 1:1 mixes of control and GAG-treated retinal preparations were comparable to the means of the GAG-treated and control activities. The effects documented in this study, therefore, probably reflect irreversible in vivo changes. After peripheral administration, GAG, GVG and gabaculine might reach higher levels in the retina than in the brain. Alternatively, the differential effects of these compounds might be due to the relative proportions of catalytically active GABA enzymes in different CNS regions. On the basis of the foregoing results, the retina might be a particularly suitable region of the CNS for enzyme-activated irreversible inhibitors to label catalytically active enzymes of GABA metabolism.

Effect of chronic exposure to static magnetic field upon the serum glutamic pyruvic transaminase activity GPT and morphology of the cardiac muscle, skeletal muscles, kidneys, cerebellum and lung tissue in guinea pigs.

In a cycle of investigations concerning the pathogenesis of functional changes caused by the influence of magnetic field of induction occurring in laboratory and industrial conditions glutamic pyruvic transaminase activity in external blood and morphological picture of cardiac muscle, skeletal muscles, kidneys, cerebellum and lung tissue in guinea pigs were examined. Static homogeneous magnetic field as low as 0.005 T produced a statistically significant decrease in GPT activity. The animals were exposed to the magnetic field action for seven weeks 1 hour a day, 7 days a week. The range of observed changes of enzyme activity were determined by the duration of magnetic field. No morphological changes were observed.

[Effect of thyrotropin releasing hormone (TRH) on GABA (gamma aminobutyric acid) metabolism in mouse and rat brains: as to the activities of GAD (glutamic acid decarboxylase), GABA-T (GABA-transaminase) and GABA re-uptake].

It has been reported that thyrotropin-releasing hormone (TRH) improves the ataxia of cerebellar type. The mechanism of action is unclear. As well recognized, GABA (gamma aminobutyric acid) is an important neurotransmitter in cerebellar system. So, if TRH acts on cerebellum, it is expected that the GABA metabolism will be modified by in vivo or in vitro TRH application. The purpose of this experiment is to clarify whether or not TRH affects on GABA system in cerebellar system. The first experiment was to determine the effect of TRH on the two GABA related enzyme activities, that is, GAD (glutamic acid decarboxylase) and GABA-T (GABA-transaminase). TRH was intraperitoneally injected at a dose of 5 mg/kg. In mouse brains, the two enzyme activities of hindbrains increased after 60 minutes. Next experiment assaying GAD activities at two parts of hindbrain revealed that the increase in hindbrain observed above was due to marked increase in brain-stem (p less than 0.001), but not in cerebellum itself in which the GAD activities decreased (p less than 0.05). On the other hand, in the forebrains, the same dose of TRH failed to change both GAD and GABA-T activities. In order to ascertain the effect more precisely, we assayed GAD activities at seven parts of the brain of Wistar male rats. By this experiment, it was found that GAD activities increase at two portions, namely, at thalamo-midbrain after 30 minutes and at pons-medulla after 180 minutes of TRH injection (p less than 0.05, in both). Other five portions, including cerebellum, showed no significant change of GAD activities.(ABSTRACT TRUNCATED AT 250 WORDS)

Identification of human red cell glutamate-pyruvate transaminase (GPT) phenotypes by isoelectric focusing.

Isoenzymes of human red cell glutamate-pyruvate transaminase (GPT) were resolved by isoelectric focusing (IEF) of hemolysates in polyacrylamide gels at pH 5.0-7.0. The bands of enzyme activity required both alpha-ketoglutarate and L-alanine in the staining mixture for visualization, indicating that the bands were not lactate dehydrogenase or glutamate dehydrogenase. Phenotyping of 41 individuals by IEF, including types GPT 1, 2A, 1-2A, 1-2B, and 2A-2B, agreed with the typing results obtained by electrophoresis in starch gels and in polyacrylamide gels at acid and alkaline pH. Analysis of one kindred demonstrated autosomal codominant transmission of the rare GPT*2B gene through 3 generations. IEF facilitates phenotyping by permitting identification of the GPT types on a single gel with a considerable reduction in time and cost. Although no new variants were found in this investigation, IEF may be more powerful for the recognition of presently undetected variants of GPT.

gamma-Aminobutyric acid transaminase (GABAT) polymorphism among ethnic groups in Singapore--with report of a new allele.

gamma-Aminobutyric acid transaminase (GABAT, E.C.2.6.I.19) was phenotyped by starch-gel electrophoresis in post-mortem liver samples from 650 unrelated subjects of either sex, comprising 289 Chinese, 177 Indians, 140 Malays, and 44 from other racial groups from Southeast Asia. The estimated gene frequencies of GABAT1 and GABAT2 were found to be .5779 and .3806 in Chinese, .5678 and .3955 in Indians, and .6214 and .3250 in Malays. The frequency of GABAT1 was .5909 in the mixed group of other races. There was no significant difference in the phenotypic distribution between sexes. A new slow (less anodal) variant (GABAT3) has been observed in low frequency in all the groups (.0415, .0367, 0536, and 0536, and .0568 in Chinese, Indians, Malays, and the mixed-group, respectively). The distribution of GABAT phenotypes was at Hardy-Weinberg equilibrium in all the ethnic groups studied.

The conversion of L-cystathionine into the cyclic ketimine form by heated rat liver extracts containing cystathionase and transaminase activities.

Rat liver homogenates heated for 10 min at 60 degrees C incubated with L-cystathionine yield cystathionine ketimine which was identified by its typical UV spectrum and by cochromatography with authentic samples on the amino acid analyzer. Alanine and alpha-amino butyric acid have been also detected among the final products. The reaction is due to heat stable gamma-cystathionase and transaminases present in the extracts. Cystathionase produces alpha-keto butyric acid and pyruvic acid which are then used for the transamination of the remaining cystathionine to yield the ketimine. This is the first report indicating the occurrence in a mammalian tissue of an enzymatic system using cystathionine for reactions differing from the traditional transulfuration to cysteine.

[Serum alkaline phosphatase and transaminase activity in cows with clinical ketosis].

Studied was the activity of the alkaline phosphatase and the transaminases in the serum of clinically normal and ketosis-affected cows as well as cows with an affected liver. The changes were followed in the activity of SGOT and SGPT that had set in as a result of five-day starving and the injection of prednisolon at the rate of 0.5 mg/kg in the course of four days. The activity of serum alkaline phosphatase in ketosis-affected cows proved unchanged--1.99 +/1 0.35 VE. A moderate increase was established in cows with liver affections-- 3.73 +/- 0.35 VE. The hyperenzymemia in ketosis-affected animals, in animals with liver injuries as well as in cows subjected to starving or prednisolon treatment was substantiated by the higher activity of SGOT. Dependable higher values of SGOT were established in the serum of freshly calved cows as compared to cows of advanced lactation. The higher activity of SGOT in animals with ketosis is associated with the physiologic status, starving, enzyme induction, inadequacy of the cell energy balance--all limiting the possibility of it being ascribed with preference to the tissue proliferation or destruction, which is to be expected with cows having an affected liver.

A comparison between the morphologic changes in the livers of hamsters and rats after galactosamine treatment and their correlation with altered serum transaminase levels.

Galactosamine caused changes in the livers of rats that were similar to those seen in human viral hepatitis. In hamsters the changes were characteristic of a typical toxic hepatitis. The response by the liver to galactosamine was very variable in both the rat and the hamster and there was a degree of correlation between the raised transaminase levels and the liver cell damage.

Fasting-induced changes of tyrosine transaminase activity in rat tissues.

Tyrosine transaminase activity in liver, kidney, intestine, stomach, skin, adipose tissue, striated muscle and brain in fed and 24-hour fasted rats, has been studied. Maximal activity has been found in liver, with only fractional activity in the other tissues. 24 hour fasting induced significant decrease in liver and adipose tissue activity, while no changes have been detected in the other tissues. The possible implications of these facts are discussed.

[Therapeutic trial of gamma-vinyl GABA, an inhibitor of GABA transaminase, in tardive dyskinesias induced by neuroleptics].

Oral gamma-vinyl GABA, an irreversible inhibitor of GABA-transaminase, was given to 10 patients with neuroleptic-induced tardive dyskinesia (7 chronic simple schizophrenics and 3 chronic paranoid schizophrenics). Dyskinesia was reduced in 8 of 10 cases. Aggravation of dyskinesia and major psychomotor sedation was observed in two elderly patients with senile dementia. In addition, with regard to schizophrenic symptoms, gamma-vinyl GABA appeared to have beneficial effects on affective withdrawal and retardation, while hallucinations seemed to be aggravated.

Use of gamma-aminobutyric acid (GABA)-transaminase inhibitors and a GABA uptake inhibitor to investigate the influence of GABA neurons on dopamine-containing amacrine cells of the rat retina.

The effects of inhibiting gamma-aminobutyric acid-transaminase (GABA-T) with aminooxyacetic acid or with gabaculine and inhibiting GABA uptake with nipecotic acid on dopamine (DA) synthesis in the retina of light-exposed rats were studied. 3,4-Dihydroxyphenylalanine (Dopa) accumulation after the inhibition of L-aromatic amino acid decarboxylase with NSD-1015 was used as an index of DA synthesis. The GABA-T inhibitors significantly increased GABA levels in the retina but had no effect on retinal Dopa accumulation in the light. Dopa accumulation was significantly inhibited at a high dosage by nipecotic acid alone and by low dosages of nipecotic acid in rats pretreated with either aminooxyacetic acid or gabaculine. The dosages of nipecotic acid which inhibited Dopa accumulation in the light also inhibited [3H]GABA uptake in the retina. The inhibitory effects of gabaculine and nipecotic acid on Dopa accumulation appeared to occur at least partially via GABA receptors because their action was significantly reversed by the GABA antagonist bicuculline methiodide. These experiments thus suggest that endogenous GABA can suppress the light-evoked increase in DA synthesis in the rat retina and support previous studies which concluded that an inhibitory GABA input may at least partially regulate the activity of the DA neurons in the retina of rats.

[Urinary transaminase in the diagnosis of kidney disorders in liver cirrhosis].

In examinations of 72 patients with liver cirrhosis and chronic hepatitis the transaminase activity of the urine (T-uria) was found in 55.6% of the cases, above all in liver cirrhosis. The T-uria increases parallel to the progressing portal hypertension to the hepatocellular insufficiency and to the hepatorenal syndrome. It is reduced in positive dynamics of the hepatic process. The T-uria has an essential prognostic value. In the terminal phase of the liver cirrhosis the T-uria is perhaps connected with disturbances of the hepatic haemodynamics.

Changes in the activity of gamma-amino butyric acid transaminase and succinic semialdehyde dehydrogenase in the cobalt and iron experimental epileptogenic foci in the rat brain.

GABA-transaminase (GABA-T) and succinic semialdehyde dehydrogenase (SSA-DH) activities were measured in the mitochondrial fractions from the cobalt- and FeCl3-induced chronic epileptogenic foci in the rat brain. Electroencephalographically, the FeCl3 epileptogenic focus remained active for a duration longer than that of the cobalt focus. In both the foci SSA-DH activity showed significant increases which were concomitant with the EEG epileptiform activity. In cobalt focus, the GABA-T activity fell whereas, in the FeCl3 focus it was unchanged. In cobalt focus fall in GABA-T activity seemed to be concomitant with EEG epileptiform discharge. The measurements of the enzyme activities in the mirror (secondary) foci showed that, except for a brief stimulation of SSA-DH activity in the mirror focus in FeCl3 epileptic animals, the enzyme activities remained unchanged. Possible significance of the observed enzymatic changes in the physiology of epileptogenic focus is discussed.
