[Effects of manganese, zircon and lithium alone on rat liver lipid peroxidation].

Lipid peroxide (LPO) in rat liver was detected by malondiadehyde (MDA) colorimetry. The effect of manganese, zircon and lithium alone on lipid peroxidation in rat liver was also studied. The results showed that manganese and zircon at the doses of (9.862-1.972) x 10(-4) and (0.1972-9.862) x 10(-5) nmol/L respectively decreased LPO in rat liver(P < 0.01). Lithium inhibited lipid peroxidation at the dose of (19.72-1.972) x 10(-4) nmol/L, and induced lipid peroxidation at higher concentration.

[Detection of serum tumor supplied group of factors and its clinical significance].

BACKGROUND & OBJECTIVE: Tumor supplied group of factors (TSGF) is the first kind of tumor marker in vitro which was primarily obtained the approval by Chinese government to land the market, there was no any reports about it in abroad before. In order to know its specificity and sensitivity, we designed this study to detect the TSGF in serum of the patients with malignancies and evaluate its clinical significance for diagnosis. METHODS: Serum TSGF in 523 cases with malignancies and 400 cases of normal control were detected with chemical colorimetry. The diagnostic accuracy was evaluated. RESULTS: Mean levels of serum TSGF in the patients without malignancies (group A), the patients with malignancies (group B), the patients after curative treatment for malignancies (group C) and the normal control (group D) were 66.57, 71.24, 60.78, and 53.56 u/ml, respectively. After the statistical analysis, differences between the group A and group B, group A and group D, group B and group C, group B and group D, group C and group D were found (P < 0.05). But there was no obvious difference between the group A and group C (P > 0.05). The sensitivity was 63.86%; the specificity was 90.89%; the truly negative rate was 87.01%; and the falsely negative rate was 13.00%. CONCLUSIONS: Serum TSGF increased in malignancies and may be helpful for screening and early detection of cancer.

Serum iron determination in patients receiving therapy with iron dextran ("Imferon").

Imferon (Lakeside Labs., Inc.), an iron dextran complex, is widely used for parenteral therapy with iron. Colorimetry and atomic absorption spectroscopy give different values for serum iron concentration in patients receiving such therapy. Values determined by atomic absorption spectroscopy were usually much higher than those obtained by either a manual or an automated colorimetric procedure, and also were higher than the total serum iron-binding capacity of the same sera. This difference is attributed to circulating iron being present both as iron dextran as as transferrin-bound iron. We conclude that atomic absorption spectroscopy should be used to determine serum iron in patients receiving Imferon.

[Protective effect of ginsenosides on human peritoneal mesothelial cell against lactate-based peritoneal dialysis solutions--induced injury in vitro].

OBJECTIVE: The aim of this research was to observe the influence of ginsenosides (GS) on viability and proliferation of human peritoneal mesothelial cell (HPMC) which were inhibited by L-PDS in vitro. METHOD: Mesothelial cells were isolated from human omental specimens by trypsin disaggregation and established a stable HPMC culture model. The viability and capacity of proliferation of HPMC were assessed by lactate dehydrognase (LDH) release and tetrazolium salt colorimetry assay (MTT assay). RESULTS: Compared with the control group, the release of LDH in L-PDS groups (2.5%, 4.25% glucose) significantly increased. But there was no difference between the two glucose concentrations. As HPMC pre-exposed to L-PDS prolonged, the viability of cells decreased gradually. GS decreased the release of LDH and increased the cells viability (P < 0.01), as well as enhanced the proliferation of HPMC. CONCLUSION: The results suggest that GS has some protective effects on cell viability and proliferation of HPMC inhibited by L-PDS in vitro.

Inhibitory effect of meloxicam on human polymorphonuclear leukocyte adhesion to human synovial cell.

AIM: To investigate the effect of meloxicam on human polymorphonuclear leukocyte (PMN) adhesion to human synovial cell (HSC), and to explore its mechanism. METHODS: MTT colorimetry was used to determine the adhesion effect of PMN to HSC. Cell-ELISA and RT-PCR methods were used to determine the expression of ICAM-1 and VCAM-1. Nuclear transcription factor-kappa B (NF-kappa B) was measured by electrophoretic mobility shift assay (EMSA) method. RESULTS: Meloxicam was found to effectively inhibit TNF-alpha (50 u.mL-1 for 12 h) and IL-1 beta (50 u.mL-1 for 12 h)-induced adhesion of PMN to HSC (IC50 3.38 x 10(-7) mol.L-1 and 3.56 x 10(-6) mol.L-1, respectively) in a concentration-dependent manner. ICAM-1 protein and mRNA expression induced by TNF-alpha (50 u.mL-1) were inhibited by meloxicam at 1 x 10(-6)-1 x 10(-5) mol.L-1. The activation of NF-kappa B was also inhibited by meloxicam at 1 x 10(-6)-1 x 10(-5) mol.L-1. CONCLUSION: These results suggest that meloxicam inhibit TNF-alpha stimulated PMN-HSC adhesion and expression of ICAM-1 by suppressing the activity of NF-kappa B.

[Effect of herbal compound 861 on expression and activity of nitric oxide synthase in hepatic stellate cells].

OBJECTIVE: To study the effect of herbal compound 861 (HB861) on expression and activity of nitric oxide synthase (NOS) in hepatic stellate cells (HSC), and to explore the feasibility of its application in preventing and treating the early portal hypertension. METHODS: HSC of HSC-T6 cell line (1 x 10(5)/ml) were cultured in dish with 95% O2 plus 5% CO2 under 37 degrees C for 24 hrs, then divided into 5 groups, 6 dishes in each group. Group A was the blank control group. To Group B-E, HB861 2 mg/ml, HB861 4 mg/ml, HB861 8 mg/ml, HB861 4 mg/ml + NW-Nitro-L-Arginine Methyl Ester (L-NAME)4 mg/ml were added separately, and continuously cultured for 24 hrs. NOS activity was measured using colorimetry, NO level was determined by nitrate reductase technique. The cells were fixed by 4% paraformaldehyde for 2 hrs for test HSC-T6 iNOS expression by immunocyto-chemical method. RESULTS: HB861 in 2 mg/ml, 4 mg/ml and 8 mg/ml could increase HSC-T6 NOS activity from 1.7 +/- 0.1 to 2.5 +/- 0.3, 3.5 +/- 0.4 and 3.7 +/- 0.9 respectively (P < 0.01), the NO levels in supernatant were increased in parallel from 56.1 +/- 4.8 to 90.7 +/- 4.6, 99.7 +/- 4.1 and 109.0 +/- 2.7 respectively (P < 0.01). L-NAME could not inhibit the effect of HB861 in increasing the synthesis and secretion of NO by activated HSC-T6. Immuno-cyto-chemical study showed that there was iNOS expression in cytoplasm, and which could be increased by HB861. CONCLUSION: The activated HSC-T6 showed positive iNOS expression, suggesting it could produce NO. HB861 could markedly increase HSC-T6 iNOS expression and NOS activity, enhance the NO synthesis and secretion, it also could inhibit the contractility of activated HSC by way of increase HSC to secrete NO, so as to lower the resistance in hepatic sinusoid, therefore would play important role in preventing and treating of early portal hypertension.

[Inhibitory effects of hydrocortisone on human polymorphonuclear leukocyte adhesion to human synovial cell].

AIM: To investigate the inhibitory effects of hydrocortisone on human polymorphonuclear leukocyte (PMN) adhesion to human synovial cell (HSC), and to explore its mechanism. METHODS: MTT colorimetry was used to determine the adhesion effect of PMN to HSC. Cell-ELISA and RT-PCR methods were used to determine the expression of adhesion molecular ICAM-1 and VCAM-1. EMSA method was also used to observe the activity of nucleus transcription factor-kappa B (NF-kappa B). RESULTS: Hydrocortisone inhibited TNF-alpha (50 U.mL-1 for 12 hours) and IL-1 beta (50 U.mL-1 for 12 hours)-induced adhesion of PMN to HSC (IC50 2.05 x 10(-6) mol.L-1 and 2.13 x 10(-7) mol.L-1, respectively) in a concentration-dependent manner. Adhesion molecular VCAM-1 and ICAM-1 protein and mRNA (rather than ICAM-1) expression in HSC induced by TNF-alpha (50 U.mL-1) were inhibited significantly by hydrocortisone at 1 x 10(-6)-10(-5) mol.L-1. The activity of NF-kappa B was also extensively inhibited by hydrocortisone at 1 x 10(-6)-10(-5) mol.L-1. CONCLUSION: Hydrocortisone inhibited TNF-alpha-stimulated PMN-HSC adhesion, and expression of VCAM-1 by suppressing the activity of NF-kappa B.

[The effects of leptin on proliferation and function of human osteoblast].

OBJECTIVE: To observe the effect of leptin on osteoblast. METHODS: Human osteoblast primary culture was carried out, and the morphology and function of osteoblast were observed. The effects of different levels of leptin on osteoblast in different days were assessed by MTT colorimetry. Osteocalcin production was measured also. RESULTS: Human osteoblasts were fusiform in shape and were positive for alkaline phosphatase by histochemical staining, positive for osteocalcin by immunofluorescence staining, and positive by Alizarin Reds staining after mineralized upon supplementation with ascorbate and beta-glycerophosphate. On the first, second and third days, the proliferation of osteoblast, cultured with different concentrations of leptin, had no changes. The leptin-stimulated synthesis of osteocalcin of cells was found to be dose-dependent (P < 0.05), but not time-dependent (P > 0.05). CONCLUSION: The above data indicated that there were no evidences for the effects of leptin on the proliferation of human osteoblast, but leptin could enhance the function of human osteoblast.

The epidemiology of hyperuricemia in children of Taiwan aborigines.

OBJECTIVE: To explore the factors influencing serum uric acid concentrations and the prevalence of hyperuricemia in aboriginal children from an area in Taiwan with a high prevalence of gouty arthritis. METHODS: A cross sectional study of the Bunun tribe was conducted in central Taiwan from March to May 2001. Children aged 4-13 were requested to fill out a structured questionnaire with the assistance of their parents. A total of 414 children (mean age, 8.9 +/- 2.1 yrs) were recruited. Uric acid was determined by colorimetry using the uricase method. Since no previous study has investigated serum uric acid concentrations in these aboriginal children, hyperuricemia was defined as uric acid > 416.5 micromol/l (7 mg/dl) in boys and > 357 micromol/l (6 mg/dl) in girls. RESULTS: The mean concentration of serum uric acid was 368.9 +/- 83.3 micro mol/l. Ninety of 224 girls (40.2%) and 56 of 190 boys (29.5%) were hyperuricemic. Children with hyperuricemia had significantly higher body mass index (BMI), blood pressure, and triglyceride and creatinine concentrations than those with normal uric acid concentrations. After adjustment for age, sex, lipid profile, and blood pressure, hyperuricemia was found to be significantly associated with serum creatinine (OR 2.40, 95% CI 1.91-3.04), BMI (OR 1.24, 95% CI 1.11-1.40), and a family history of gouty arthritis in parents (OR 2.01, 95% CI 1.02-3.96). CONCLUSION: BMI, a positive family history of gouty arthritis in parents, and creatinine level correlated with hyperuricemia in aboriginal children in Taiwan.

[Effects of calcium channel antagonist on gastrin-induced proliferation of HT29 colon carcinoma cells].

OBJECTIVE: This study aimed to assess the effects of nifedipine on gastrin-induced proliferation of HT29 colon carcinoma cells and inquire into the possible mechanisms. METHODS: Flow cytometry was used to monitor the cytoplasmic free calcium; MTT colorimetry was used to determine the proliferation of HT29 cells. RESULTS: The results showed that 2.5 x 10(-6) mol/L pentagastrin (PG) induced a quick rise of intracellular free calcium ([Ca2+]i) (P < 0.01). 10(-5) mol/L nifedipine can significantly inhibited the rise of [Ca2+]i induced by pentagastrin (P < 0.01), in parallel, at growth assay we demonstrated that 10(-5)-10(-6) mol/L nifedipine could obviously block the increase in cell number elicited by 2.5 x 10(-6) mol/L PG. CONCLUSION: These data indicate that nifedipine can stop the influx of Ca2+ and hence inhibit the pentagastrin-induced proliferation.

[Effect of ginkgolide B on platelet-activating factor induced activation of rat polymorphonuclear leukocytes].

AIM: To investigate the effect of ginkgolide B on PAF-induced adhesion, chemotaxis and degranulation of rat polymorphonuclear leukocytes (PMNs). METHODS: The adhesion of rat PMNs to rat synovial cells (RSC) was measured with MTT colorimetry. The chemotaxis of PMNs was quantified with Boyden chamber method. The degranulation of rat PMNs was evaluated by determining the activity of released beta-glucuronidase. RESULTS: In comparison with control, ginkgolide B at the concentration of 10 mumol.L-1 significantly inhibited the adhesion of PMNs to RSC by 71.74%. At the final concentration of 1-1,000 nmol.L-1, ginkgolide B dose-dependently inhibited the chemotaxis of PMNs stimulated with 10 nmol.L-1 platelet-activating factor (PAF), the IC50 was 4.84 nmol.L-1. At the final concentration of 0.01-10 mumol.L-1, ginkgolide B decreased the release of beta-glucuronidase in PMNs induced by 1 mumol.L-1 PAF in dose-dependent manner. The IC50 was 3.56 mumol.L-1. CONCLUSION: Ginkgolide B was found to significantly inhibit PAF-induced adhesion, chemotaxis and degranulation in rat polymorphonuclear leukocytes. These effects might be considered a part of the mechanisms underlying the antiinflammatory action of ginkgolide B.

Dissolution, bioavailability and ulcerogenic studies on piroxicam-nicotinamide solid dispersion formulations.

Solid dispersions with different ratios of Piroxicam and Nicotinamide (as a carrier) were prepared by fusion method. Drug-carrier interactions in the solid state were ascertained using phase-diagram, X-ray diffraction (XRD) and Differential scanning colorimetry (DSC). Select ratios of the formed piroxicam-nicotinamide solid dispersions and their corresponding physical mixtures were formulated into tablets and capsules. The dissolution characteristics of the formed solid dispersions, physical mixtures and their formulations were compared with pure piroxicam in 900 ml of 0.1 N HCl (pH 1.2) at 37 +/- 0.5 degrees C, 50 rpm using USP XXI paddle apparatus. Solid dispersion capsule PNC1 (containing 1:5--drug-carrier ratio solid dispersion) and the corresponding physical mixture capsule PNC1A were used for bioavailability studies in healthy human volunteers. Further, ulcer indices of the prepared solid dispersion and their corresponding physical mixture were compared with pure piroxicam in rats. The DSC, XRD and phase diagram indicated the formation of solid dispersed system. The prepared dispersion showed better dissolution than the corresponding physical mixture and pure drug. The fabricated formulation complied with the pharmacopoeial limits for physico-chemical properties and the solid dispersion capsules PNC1 and PNC2 (containing 1:9 ratio of drug to carrier solid dispersion) showed 3 and 6.6 fold increase and 3.2 and 8 fold increases in dissolution rate compared to the corresponding physical mixture capsules (PNC1A and PNC2A), respectively. The solid dispersion capsule PNC1 showed significantly higher absorption and dissolution and consequently better bioavailability than the corresponding physical mixture capsule (PNC1A) in healthy human volunteers. The ulcer indices of the prepared dispersions were significantly lower than the corresponding physical mixture and pure drug.
