Thermal behavior and elastic properties of dimyristoyl phosphatidylcholine bilayers under the effect of pentoxifylline.

We have investigated the effect on dimyristoyl phosphatidylcholine bilayers of pentoxifylline, a derivative of xanthine by using two optical techniques, quasi-elastic light scattering (QLS) and Fourier transform infrared spectroscopy (FT-IR). The results show that in the presence of pentoxifylline, the bilayer phase transition point is lowered and that the elastic modulus is decreased. The FT-IR results indicate strong interactions in the aqueous interface regions of the bilayers. We discussed these results comparatively with those obtained from flavonoid derivatives whose effect was analogous and previously studied.

Coimmobilization of urease and glutamate dehydrogenase in electrochemically prepared polypyrrole-polyvinyl sulfonate films.

Immobilization of urease and glutamate dehydrogenase enzymes in electrochemically prepared polypyrrole-polyvinyl sulfonate films (PPY-PVS) was carried out using physical adsorption and electrochemical entrapment techniques. Detailed studies on optimum pH, Fourier transform infrared spectroscopy, cyclic voltammetry, and scanning electron microscopy of the enzymes in the immobilized state were conducted. The value of the apparent Michaelis-Menten constant was experimentally determined to be 2.5 and 2.7 for physically adsorbed and electrochemically entrapped urease in PPY-PVS films, respectively.

Detection of specific sugars in dairy process samples using multivariate curve resolution.

Dairy process monitoring by application of multivariate curve resolution using alternating least squares is presented. Alternating least squares was used for resolving Fourier transform infrared spectral data from a dairy batch process in which lactose is enzymatically hydrolyzed to glucose and galactose. It was possible to extract four compounds (fat, lactose, and two other sugar components) from the spectral data obtained from nine process runs. Subsequently, the pure spectra obtained in this way were used to monitor the content of these compounds in two new process runs. In this way, alternating least squares made it possible to follow the hydrolysis process by Fourier transform infrared spectroscopy without the need for reference analyses. When the results were correlated to reference results for lactose, the accuracy was similar to that obtained when a partial least squares regression was performed on the same data; lactose correlation was 0.980 when alternating least squares was used and was 0.987 when partial least squares was used.

Bioactivity of tape cast and sintered bioactive glass-ceramic in simulated body fluid.

A common ceramic processing technique, tape casting, was used to produce thin, flexible sheets of bioactive glass (Bioglass 45S5) particulate in an organic matrix. Tape casting offers the possibility of producing three-dimensional shapes, as the final material is built up layer by layer. Bioactive glass tapes were sintered together to form small discs for in vitro bioactivity testing in simulated body fluid (SBF). Four different sintering schedules were investigated: 800, 900, and 1000 degrees C for 3 h; and 1000 degrees C for 6 h. Each schedule produced a crystalline material of major phase Na2Ca2Si3O9. Tape cast and sintered bioactive glass-ceramic processed at 1000 degrees C formed crystalline hydroxyapatite layers after 20-24 h in SBF as indicated by Fourier transform infrared spectroscopy, Scanning electron microscopy, and EDS data. FTIR revealed that the greatest amount of hydroxyapatite formation after 2 h was observed for samples sintered at 900 degrees C. The differences in bioactive response were likely caused by the variation in the extent of sintering and, consequently, the amount of surface area available for reaction with SBF.

Effect of process parameters on the characteristics of porous calcium phosphate ceramics for bone tissue scaffolds.

Porous hydroxyapatite (HA) has already been widely used as a bone substitute due to its similarity with the mineral part of the bone. In this work, cylindrical tablets with micro and macro porosity were produced from stoichiometric and deficient hydroxyapatites by using naphthalene as porosifier agent. The influence of the processing parameters such as Ca/P ratio of start material, calcination temperature, and naphthalene content on the characteristics of porous calcium phosphate tablets was evaluated. Three mineral phases-HA, alpha-TCP (alpha tri-calcium phosphate), and beta-TCP (beta tricalcium phos-phate)-with variable contents were identified by x-ray diffraction (XRD) and Fourier-transformed infrared spectroscopy (FT-IR). Image analysis and density measurements were used to characterize sample porosity. As expected, the total porosity of the calcinated material is not dependent on the stoichiometry of the precursor hydroxyapatite. For calcium-deficient hydroxyapatite, the increase in naphthalene content contributes to stabilize alpha-TCP phase, altering the relative phases content.

Preparation and processing of vitreoretinal instrumentation and equipment as a risk factor for silicone oil emulsification.

PURPOSE: To investigate possible sources for the induction of silicone oil emulsification in patients' eyes. METHODS: The contaminants on a ready-to-use standard set of vitreoretinal instruments cleaned and sterilized in an eye clinic were determined. The determination of detergents was carried out according to a standardized procedure, which uses ultrapurified water to rinse the equipment in question, followed by a measurement of the conductivity. Silicone oil remnants were determined using Fourier transformed infrared spectroscopy. RESULTS: Ionic components of detergents and remnants of silicone oil could be detected on instrumentation deemed sterile, clean, and ready-to-use. CONCLUSION: During routine cleaning and sterilization of vitreoretinal instruments and accessories, remnants of silicone oil and detergents can persist and trigger emulsification of silicone oil, which came into contact with these contaminated devices during instillation of the endotamponade.

Radiation effects on polypropylene for sterilization.

In order to improve the resistance of gamma-radiation sterilization of polypropylene (PP), the formulations of the additives such as antioxidants, crosslinking agent, and trans-stilbene oxide (StO) have been carried out. The irradiated PP and ethylene-propylene copolymer samples with control and additives were characterized by mechanical tester, colorimetry, and Fourier transform infrared spectroscopy with total attenuated reflectance mode (FTIR-ATR). Crosslinking agent and StO formulated PP showed remarkable radiation resistance and minimum discoloration. Also, radiation resistance of ethylene-propylene copolymers with 3% of ethylene contents was better than that of PP homopolymers in the case of no additives. The proposed mechanisms of radiation stabilization with additives are also discussed.

Influence of P2O5 on crystallinity of apatite formed in vitro on surface of bioactive glasses.

Two sol-gel glasses with 80 mol % SiO2 were prepared in the system SiO2-CaO-P2O5; the first one had 3 mol% P2O5 in its composition, and the second one was P2O5 free. The in vitro behavior of glasses was studied by soaking them in simulated body fluid for 7 days at 37 degrees C. After the in vitro test, the study by Fourier transform infrared spectroscopy, scanning electron microscopy, energy dispersive spectroscopy, electron diffraction, and transmission electron microscopy showed an apatite-like layer had formed on the surface of both glasses. However, for identical soaking time, the apatite crystals formed on the surface of the glass containing P2O5 in the composition were larger. Therefore, the presence of P2O5 in the sol-gel glass composition promotes the crystal growth of the apatite.

Physicochemical and structural properties of oat globulin polymers formed by a microbial transglutaminase.

Oat globulin was polymerized by a microbial transglutaminase (TG), and some physicochemical and functional properties of polymers were studied. Reversed-phase HPLC revealed that the number of epsilon-(gamma-glutamyl) lysine isopeptide bonds formed after 4 h of enzyme incubation was 2.21 micromol/g of protein. SDS-PAGE showed that the oat globulin acidic polypeptides (AP) were more susceptible to polymerization than the basic polypeptides (BP), and the reactivities of both AP and BP were enhanced by the addition of other substrate proteins. Differential scanning calorimetry showed that both the denaturation temperature and denaturation enthalpy were decreased after TG treatment. Fourier transform infrared spectroscopy revealed marked increases in the intensity of two intermolecular beta-sheet bands associated with aggregate formation but little conformational changes in the polymerized protein. TG incubation led to progressive changes in flow properties of oat globulin dispersions, indicating enhanced pseudoplasticity and increased viscosity and yield stress.

Photoreaction of aromatic compounds at alpha-FeOOH/H2O interface in the presence of H2O2: evidence for organic-goethite surface complex formation.

The photoreactions of aromatic compounds were investigated in aqueous dispersions of alpha-FeOOH(goethite)/H(2)O(2) at neutral pHs. It was found that aromatic compounds could undergo rapid decomposition and mineralization (even to 100% yield) in the presence of both alpha-FeOOH and H(2)O(2) under UV irradiation, and the degradation rates of the organics were related to their sorption ability of the surface of alpha-FeOOH and were in the following order: salicylic acid approximately m-hydroxylbenzoic acid>p-hydroxylbenzoic acid approximately benzoic acid>p-biphthalic acid>phenol>benzenesulfonic acid. The interactions of salicylic acid and benzenesulfonic acid with goethite were examined by Fourier transform infrared spectroscopy (FTIR), and electron spin resonance (ESR) was applied to detect the intermediate radicals formed in the photoreaction. The effect of pH and foreign ligands on the degradation of salicylic acid, decomposition of H(2)O(2), and formation of dissolved iron was also examined.

Interfacial molecular recognition between polysaccharides and calcium carbonate during crystallization.

Based on the basic principles of biomineralization, using beta-cyclodextrin, hepatin and soluble starch as organic matrices, employing the biomimetic method, the crystallization of CaCO(3) was studied by Fourier transform infrared spectroscopy (FT-IR), X-ray powder diffraction analysis (XRD), scanning electronic microscopy (SEM), thermal gravity-differential thermal analysis and conductivity analysis. The results show that polysaccharides have an effect on the crystallization of CaCO(3). CaCO(3) crystals also have an effect on polysaccharides. The possible mechanism of this interaction is discussed.

Studies on steroids in fetuses and neonates: identification of 16-dehydropregnenolone in the circulation of pre-term neonates.

Unconjugated steroids were extracted with dichloromethane from the serum of pre-term neonates. Conjugated steroid fraction in the residue was separated and purified by high performance liquid chromatography. The purified steroid fraction was subjected to enzyme (arylsulfatase from Helix pomatia) or acid hydrolysis. Liberated steroid was analyzed by gas chromatography-mass spectrometry (GC/MS) and gas chromatography-Fourier transformation infrared spectroscopy (GC/FT-IR). All of the results obtained from GC/MS and GC/FT-IR analysis were identical with those of authentic 16-dehydropregnenolone (3 beta-hydroxy-5,16-pregnadien-20-one, 16-DHP). This is the first report to demonstrate the presence of 16-DHP in human blood.

Structure and organization of the human antimicrobial peptide LL-37 in phospholipid membranes: relevance to the molecular basis for its non-cell-selective activity.

The antimicrobial peptide LL-37 belongs to the cathelicidin family and is the first amphipathic alpha-helical peptide isolated from human. LL-37 is considered to play an important role in the first line of defence against local infection and systemic invasion of pathogens at sites of inflammation and wounds. Understanding its mode of action may assist in the development of antimicrobial agents mimicking those of the human immune system. In vitro studies revealed that LL-37 is cytotoxic to both bacterial and normal eukaryotic cells. To gain insight into the mechanism of its non-cell-selective cytotoxicity, we synthesized and structurally and functionally characterized LL-37, its N-terminal truncated form FF-33, and their fluorescent derivatives (which retained structure and activity). The results showed several differences, between LL-37 and other native antimicrobial peptides, that may shed light on its in vivo activities. Most interestingly, LL-37 exists in equilibrium between monomers and oligomers in solution at very low concentrations. Also, it is significantly resistant to proteolytic degradation in solution, and when bound to both zwitterionic (mimicking mammalian membranes) and negatively charged membranes (mimicking bacterial membranes). The results also showed a role for the N-terminus in proteolytic resistance and haemolytic activity, but not in antimicrobial activity. The LL-37 mode of action with negatively charged membranes suggests a detergent-like effect via a 'carpet-like' mechanism. However, the ability of LL-37 to oligomerize in zwitterionic membranes might suggest the formation of a transmembrane pore in normal eukaryotic cells. To examine this possibility we used polarized attenuated total reflectance Fourier-transform infrared spectroscopy and found that the peptide is predominantly alpha-helical and oriented nearly parallel with the surface of zwitterionic-lipid membranes. This result does not support the channel-forming hypothesis, but rather it supports the detergent-like effect.

Physicochemical characterization of solid dispersions of indomethacin with PEG 6000, Myrj 52, lactose, sorbitol, dextrin, and Eudragit E100.

The purpose of this study was to prepare and characterize solid dispersions of indomethacin with polyethylene glycol (PEG) 6000, Myrj 52, Eudragit E100, and different carbohydrates such as lactose, mannitol, sorbitol, and dextrin. Indomethacin is a class II substance according to the Biopharmaceutics Classification System. It is a poorly water soluble antirheumatic agent. The goal was to investigate whether the solid dispersion can improve the dissolution properties of indomethacin. The solid dispersions were prepared by three different methods depending on the type of carrier. The evaluation of the properties of the dispersions was performed using solubility measurements, dissolution studies, Fourier-transform infrared spectroscopy, and x-ray powder diffractometery. The results indicate that lactose, mannitol, sorbitol, and especially Myrj 52 are suitable carriers to enhance the in vitro dissolution rate of indomethacin at pH 7.2. Eudragit E100, Myrj 52, and mannitol increase the dissolution properties at pH 1.2. The data from the x-ray diffraction showed that the drug was still detectable in its solid state in all solid dispersions except solid dispersions with dextrin and high amounts of mannitol. However, the results from infrared spectroscopy together with those from x-ray diffraction showed well-defined drug-carrier interactions for dextrin coevaporates.

Preparation of PVA/PEI ultra-fine fibers and their composite membrane with PLA by electrospinning.

Ultra-fine fibers of poly(vinyl alcohol)/polyethylenimine (PVA/PEI) were prepared by electrospinning of their blend solutions in water. Effects of PVA/PEI mass ratio and the polymer concentration on the fiber morphology were discussed by analysis of scanning electron micrographs. Results showed that uniform ultra-fine fibers could be obtained from an 8% PVA/PEI solution with 75:25 mass ratio. It was supposed that the introduction of PVA could promote electrospinning of PEI by weakening the intermolecular interaction and increasing solution viscosity. A composite membrane of PVA/PEI with poly(D,L-lactide) (PLA) was produced by co-electrospinning simultaneously from the aqueous 8% PVA/PEI (75:25) solution and a 20% PLA solution in N,N-dimethylformamide in two separated syringes. Fourier transform infrared spectroscopy, X-ray diffraction and X-ray photoelectron spectroscopy verified the existence of PVA/PEI and PLA in the fibrous membrane. We attempted to incorporate PEI with PLA as ultra-fine fibers to diminish the acidic inflammation caused by biodegradation of PLA. The fibrous composite membrane of PVA/PEI-PLA could provide better biocompatibility and would be used as drug-delivery carriers or tissue-engineering scaffolds.

Effect of lead on the sorption of 2,4,6-trichlorophenol on soil and peat.

The effect of lead on the sorption of 2,4,6-trichlorophenol (2,4,6-TCP) on soil and peat was investigated using a batch equilibration method. Lead markedly diminished the sorption of 2,4,6-TCP, and 2,4,6-TCP had little effect on lead sorption. Peat was a more effective adsorbent for 2,4,6-TCP than soil. The desorption hysteresis of 2,4,6-TCP verified the presence of high-energy sorption sites. Mechanisms of lead suppression effect on the 2,4,6-TCP sorption included the following: Firstly, lead accelerated the aggregation of colloids, the aggregates covered the surface in part and shrunk the pore sizes of the adsorbents, hence decreased the sorption of 2,4,6-TCP. Secondly, X-ray absorption and Fourier transform infrared spectroscopy study suggested that lead competed with 2,4,6-TCP for carboxylic, phenolic and Si-OH groups of organic matter and clay minerals. Such competition was partly responsible for the overall suppression effect of lead on the sorption of 2,4,6-TCP.

Biological evaluation of aerosol-gel-derived hydroxyapatite coatings with human mesenchymal stem cells.

The bioactive properties of hydroxyapatite (HAP) are evaluated for applications involving the enhancement of biocompatible prostheses by seeding human pluripotent mesenchymal stem cells (MSCs). The in vitro response of human MSCs seeded on aerosol-gel HAP coatings is addressed in this work. The processing of the HAP coatings has been carried out by the aerosol-gel technique using calcium nitrate and triethylphosphate as starting precursors. The characterization of the coatings was carried out by using transmission electron microscopy, Fourier transform infrared spectroscopy, scanning electron microscopy, energy disperse X-ray microanalysis, and surface force microscopy, which confirmed the high performance of the HAP coatings. In vitro tests show that human MSCs adhere to aerosol-gel-derived HAP coatings and show proliferation signals on these surfaces.

Structure and conformational stability of the enzyme I of Streptomyces coelicolor explored by FTIR and circular dichroism.

The bacterial phosphoenolpyruvate (PEP): sugar phosphotransferase system (PTS), formed by a cascade of several proteins, couples the translocation and phosphorylation of specific sugars across cell membranes. The structure and thermal stability of the first protein (enzyme I, EI) of the PTS in Streptomyces coelicolor is studied by using far-UV circular dichroism (CD) and Fourier transform infrared spectroscopy (FTIR) at pH 7.0. The deconvolution of FTIR spectra indicates that the protein is mainly composed by a 35% of alpha-helical structure and 30% of beta-sheet. The thermal denaturation curves, as followed by both techniques, show only a midpoint at 330 K. This thermal denaturation behaviour is different to that observed in other members of the EI family.
