Proportions of mitotic and apoptotic cells in a range of untreated experimental tumours.

The quantitative aspect of apoptosis in experimental tumours is often neglected. In this study, the apoptotic and mitotic indices for a range of tumours have been determined at light microscope level. It has been found that the apoptotic levels fall into a consistent range for all tumour types and agree well with those described by previous workers. It is suggested that these might be basic parameters of tumour expansion, as relevant to growth kinetics as mitotic levels.

Microwave absorption by normal and tumor cells.

Energy levels exist in mammalian cells which result in the absorption of microwaves between 66 and 76 gigahertz. Many of these energy levels occur when water molecules associate with the various chemical groups of macromolecules. The absorption spectra of cells between 66 and 76 gigahertz, therefore, is determined by the structure of in vivo water lattices, and these seem to reflect indirectly the structural makeup of macromolecules or macromolecular complexes. Tumor cells absorb 66-, 68-, and 70-gigahertz microwaves less strongly and 69-, 72-, and 75-gigahertz microwaves more strongly than normal cells. These differences in the strength of attenuation at each frequency suggest that either the ratio of RNA to DNA or the relative number of certain types of chemical groups in tumor cells is different from that in normal cells.

Purification and properties of L-asparaginase from Serratia marcescens.

The purification and properties of a tumor inhibitory l-asparaginase from Serratia marcescens are described. The following properties of the enzyme were examined: kinetics of the enzyme reaction, catalytic activity as a function of pH, boundary sedimentation velocity, electrophoresis on polyacrylamide gel, immuno-electrophoresis against homologous and heterologous antisera, immunodiffusion, blood clearance rate in mice, and inhibition of the 6C3HED lymphoma in C3H mice. Complete regression of this tumor was obtained with a smaller dose of the enzyme from S. marcescens than with enzyme from Escherichia coli. The reason for this difference was not evident from a comparison of several properties of the two enzymes.

In vitro and in vivo observations on a murine C-type virus.

From 40 discrete mouse tissue culture cell lines examined by electron microscopy or complement fixation, or both, for the presence of detectable virus, one (NCTC 4705), initiated and maintained on chemically defined medium, was chosen for a more extensive study. Virus-like particles (100 to 110 mmu), morphologically similar to previously reported immature and mature C-type leukemia virus particles, were found budding from the plasma membrane and free in the intracellular spaces of cells in tissue culture and in fibrosarcomas resulting from intramuscular implants of these tissue cultures. Complement-fixation tests for group reactive murine leukemia antigens were positive, with titers consistently higher to a broadly reactive anti-serum than to anti-Friend, anti-Moloney, or anti-Rauscher sera. The 4705 virus was neutralized by Gross antiserum, but not by the F-M-R antisera. When injected into DD, BALB/c, or C3H/He newborn mice, the virus thus far has manifested no leukemogenicity, though virus from tumor extracts and tissue culture medium has been shown to be capable of infecting C3H and Swiss mouse embryo tissue cultures and successfully replicating in them. The role of the virus in accelerating or inducing neoplastic transformation in NCTC 4705 is still not known. When it was introduced into NCTC [ill], a non-neoplastic cell line in other respects similar to NCTC 4705, 4823 manifested no signs of neoplastic transformation after harboring the virus more than 300 days in vitro.

Effects of some low molecular weight sheep pineal fractions and melatonin on different tumors in rats and mice.

With these preliminary experiments attention is drawn to a possible relation between tumor development and the pineal. The influence of melatonin and some low molecular weight pineal fractions on the growth and incidence of different experimental tumors was compared. Some pineal fractions which have an effect on the activity of hypothalamus on anterior hypophyses of male rats and mice in vitro may be active in some of these tumor models.

In vitro cultivation of human tumor tissues. II. Morphological and virological characterization of three cell lines.

Nineteen human tumors, mostly of sarcomatous nature, were cultured in vitro. Three cell lines were isolated and further characterized: MG-57 derived from a giant cell tumor, MG-63 derived from an osteosarcoma and MG-72 derived from a xanthohistiocytoma. The cell lines varied in morphology and growth pattern. An abnormal karyotype with marker chromosomes was present in Mg-63 and MG-72. None of the cell lines spontaneously produced detectable C-type virus particles. Stimulation with IUDR and dexamethasone also failed to induce detectable particle release.

Chemotherapy-induced malignancies in rats after treatment with cisplatin as single agent and in combination: preliminary results.

6 groups of 24-50 BD IX rats, each, were treated with cis-diamminedichloroplatinum (CDDP)-(II) alone or in combination chemotherapy. The animals were either bearing transplanted adenocarcinomas of the stomach or large bowel, or were treated without having any tumors, to study the long-term consequences of the chemotherapy. CDDP was given within 3 weeks either as 9 X 1 mg/kg body weight, or 9 X 2 mg/kg. Autopsy findings revealed fibrosarcomas of the kidneys and/or leukemias in each of the six groups, while the 77 animals of the control group, which did not receive chemotherapeutic agents, did not show any malignancies in postmortem examinations. In one group of rats, which received CDDP in combination with 1-methyl-1-nitroso-urea (MNU), 50% of the animals that survived chemotherapy for more than 100 days died of malignancies, mainly of fibrosarcomas of the kidneys and leukemias. In another group, 24% of the animals, which survived more than 100 days after chemotherapy with CDDP alone, had developed myeloic leukemias. We therefore may presume that a carcinogenic risk in cancer chemotherapy with CDDP is also present in humans.

Studies on the antitumour effects of N-137.

The antitumour properties of the drug N-137 were assessed in vivo in two murine T lymphoma models and two naturally metastatic hamster fibrosarcomas of Herpesvirus hominis aetiology. N-137 therapy caused a significant delay in the subcutaneous growth rate of both lymphomas (EL4 and TLX9) and in many cases completely prevented tumour appearance when administered at high doses. The antitumour effect observed in both systems was shown to be dose dependent. In contrast, N-137 therapy failed to influence the growth of two hamster fibrosarcomas (HSV-333-2-26 Met A and Met B lines), and drug administration prior to or following resection of Met B tumours failed to influence the development of natural metastases as measured by monitoring animal survival.

Tumors and dental and ocular abnormalities after treatment of infant rats with adriamycin.

In rats repeatedly treated subcutaneously as infants with adriamycin in 2 cycles of 4 treatments each, the induction of ocular and dental abnormalities and tumors was studied. Cataracts appeared from 18 to 26 days in 80% of CD rats treated with 1.15 mg/kg/day of adriamycin and from 28 to 104 days in 55% of Wistar-Lewis rats given 0.75 mg/kg/day adriamycin. Abnormal growth of incisors was observed in 30% of the CD rats and in 44% of the Wistar-Lewis rats. At lower doses, no such abnormalities were found. At about 1 year after treatment, 100% of the CD rats treated with 0.75 mg/kg/day adriamycin and about 60% of the Wistar-Lewis rats treated with 0.75 and 0.5 mg/kg/day adriamycin developed tumors, which were histologically classified.
