Rat liver DNA damage by the non-hepatocarcinogen 3,2'-dimethyl-4-aminobiphenyl.

The intestinal carcinogen 3,2'-dimethyl-4-aminobiphenyl (DMAB) and the hepatocarcinogen N-2-acetylaminofluorene (AAF) were administered intraperitoneally to male F344 rats to determine if both would produce DNA damage in the liver. Hepatocyte primary cultures (HPC) were prepared and DNA-repair was assessed in vitro by autoradiography. Hepatocytes from both DMAB- and AAF-exposed rats displayed DNA repair synthesis. These results confirm other observations that non-hepatocarcinogens can produce DNA damage in the liver, and that they can be detected in both in vitro and in vivo HPC/DNA repair assays.

Mutagenicity of three aromatic amines in the presence of fractions from various tissues.

The mutagenic activity of 2-aminofluorene, 4-aminobiphenyl and 3,2'-dimethylaminobiphenyl towards Salmonella typhimurium strain TA1538 was tested in the presence of post-mitochondrial fractions of liver, kidney, intestine, colon and testicle from untreated and pretreated rats. The metabolic activation capacity of these various fractions was very different. In the experimental conditions, no relationship was found between the mutagenic activity and the carcinogenicity of the three aromatic amines.

Accumulation of phenols in isolated hepatocytes after pretreatment with methylglyoxal.

The effects of a single intraperitoneal injection of methylglyoxal (50-800 mg/kg body wt.) in mice were investigated in the liver after 24 h. The administration of methylglyoxal (400 mg/kg body wt.) resulted in an increase in aniline hydroxylase activity in liver microsomes. At the same time an accumulation of p-amino-phenol, the hydroxylated product of aniline, was observed in isolated hepatocytes upon addition of aniline similarly to conditions (starvation, diabetes mellitus, pyrazole pretreatment) when aniline hydroxylase was induced. Methylglyoxal also decreased the reduced glutathione content in the liver, while the activity of serum glutamate pyruvate transaminase was increased, suggesting the onset of liver injuries. It is assumed that the increased oxidation of aniline hydroxylase combined with decreased glutathione levels after methylglyoxal treatment favours the formation of potentially hazardous phenol derivatives in the liver.

Influence of long-term ethanol treatment on rat liver aniline and p-nitrophenol hydroxylation.

The present study investigates the influence of long-term ethanol (EtOH) treatment of rats [10% (v/v) for 1, 4, 12, and 36 weeks] on hepatic microsomal cytochrome P450 (P450) content and liver aniline and p-nitrophenol hydroxylation. Total P450 per liver was stimulated after EtOH treatment for 1, 4, and 12 weeks. In the case of longer EtOH treatment no additional stimulation in P450 content was observed. Aniline and p-nitrophenol hydroxylase activity increased in direct relation with the duration of EtOH consumption. The stimulation of both enzymatic activities was different. In comparison to controls, in rats treated with 10% (v/v) EtOH for 1, 4, 12, and 36 weeks, an increase in nitrocatechol formation (1.1-, 1.2-, 2.2-, and 2.8-fold, respectively) was found. In contrast, no effect was observed on the metabolism of aniline after 1 and 4 weeks of EtOH consumption. Aniline hydroxylation increased after 12 and 36 weeks of EtOH treatment only. Addition of EtOH in vitro had an inhibitory effect on both aniline and p-nitrophenol hydroxylation. With liver microsomes from controls as well as EtOH-treated rats the inhibition of p-nitrophenol hydroxylation was competitive in nature (Ki = 5.6 mM and Ki = 5.9 mM). In contrast, there was a competitive inhibition of aniline hydroxylation with liver microsomes from controls only. With microsomes from EtOH-treated rats a mixed inhibition was found.

Effects of Aroclor 1254 treatment on the in vitro hepatic metabolism of toluene, aniline and aminopyrine in hybrid sunfish.

The in vitro metabolism of the volatile aromatic hydrocarbon toluene by enzymes associated with the 12,000 g supernatant fraction of hybrid sunfish (Lepomis macrochirus X L. cyanellus X L. gibbosus) liver homogenates was studied. Aminopyrine demethylase (APDM) and aniline hydroxylase (AH) activities were measured. Intramuscular injections of Aroclor 1254 (a polychlorinated biphenyl) produced significant increases in APDM and AH activities (P less than or equal to 0.1, ANOVA). There were no significant changes in the metabolism of toluene, liver wet weights, or liver protein concentrations following treatment.

Arylamine-DNA adduct conformation in relation to mutagenesis.

A considerable body of evidence has indicated that local conformational alterations induced by DNA adducts may provide the molecular basis for differences in mutational specificities exhibited by structurally similar adducts. To elucidate the relationships between adduct structure and mutation induction, the ability of several single-ring arylamines present in tobacco smoke (i.e., methylanilines, dimethylanilines, and ethylanilines) to form DNA adducts was investigated. In all cases, the major adducts were C8-substituted deoxyguanosine derivatives, which is consistent with what has been observed with more carcinogenic arylamines, such as 2-aminofluorene and 4-aminobiphenyl. Spectroscopic and theoretical data on the adducts indicated conformational differences depending upon the location of the alkyl substituents. Adducts containing alkyl groups ortho to the amino function (e.g., 2-methylaniline) had a greater percentage of syn conformers about the glycosyl bond than those not bearing such groups. Arylamines with ortho alkyl substituents tend to be more mutagenic and tumorigenic than analogues not containing an ortho alkyl substituent. This increase in biological activity may be due in part to the greater propensity of ortho alkylated adducts to adopt a syn conformation.

Oxyaniliniums as acetylcholinesterase inhibitors for the reversal of neuromuscular block.

A series of oxyanilinium-based AChE inhibitors have been synthesised and tested for the reversal of vecuronium-induced neuromuscular block. Several compounds, for example 2-hydroxy- and 2-methoxy-N,N-dimethyl-N-allylanilinium bromide (3 and 6) showed comparable reversal potencies to edrophonium and clean in vivo cardiovascular profiles.

Oxidative DNA damage by an N-hydroxy metabolite of the mutagenic compound formed from norharman and aniline.

Norharman (9H-pyrido[3,4-b]indole), which is a heterocyclic amine included in cigarette smoke or cooked foodstuffs, is not mutagenic itself. However, norharman reacts with non-mutagenic aniline to form mutagenic aminophenylnorharman (APNH), of which DNA adducts formation and hepatocarcinogenic potential are pointed out. We investigated whether N-OH-APNH, an N-hydroxy metabolite of APNH, can cause oxidative DNA damage or not, using 32P-labeled DNA fragments. N-OH-APNH caused Cu(II)-mediated DNA damage. When an endogenous reductant, beta-nicotinamide adenine dinucleotide (NADH) was added, the DNA damage was greatly enhanced. Catalase and a Cu(I)-specific chelator inhibited DNA damage, suggesting the involvement of H(2)O(2) and Cu(I). Typical -*OH scavenger did not inhibit DNA damage. These results suggest that the main reactive species are probably copper-hydroperoxo complexes with DNA. We also measured 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) formation by N-OH-APNH in the presence of Cu(II), using an electrochemical detector coupled to a high-pressure liquid chromatograph. Addition of NADH greatly enhanced 8-oxodG formation. UV-VIS spectra and mass spectra suggested that N-OH-APNH was autoxidized to nitrosophenylnorharman (NO-PNH). We speculated that NO-PNH was reduced by NADH. Cu(II) facilitated the redox cycle. In the presence of NADH and Cu(II), very low concentrations of N-OH-APNH could induce DNA damage via redox reactions. We conclude that oxidative DNA damage, in addition to DNA adduct formation, may play an important role in the expression of genotoxicity of APNH.

Monooxygenase activity of human hemoglobin: role of quaternary structure in the preponderant activity of the beta subunits within the tetramer.

Catalysis of para hydroxylation of aniline was measured for human ferrihemoglobin and various derivatives in a reconstituted system consisting of the appropriate hemoprotein (at 4 microM heme), reduced nicotinamide adenine dinucleotide phosphate (NADPH), cytochrome P-450 reductase, and aniline under atmospheric O2. The isolated subunits of hemoglobin (alpha 3+ and beta 3+4) were prepared by treatment with p-(hydroxymercuri)benzoate. Semihemoglobin (alpha heme2 beta 02) was prepared from ferrihemoglobin and apohemoglobin. Converse valency hybrids alpha 3+2(beta 2+-CO)2 and (alpha 2+-CO)2 beta 3+2 were prepared from appropriately ligated alpha and beta subunits. After chromatography, the hemoglobin derivatives were characterized by visible and 1H NMR spectroscopy and electrophoresis. At the same concentration of aniline, the alpha and beta subunits were much less active than the normal tetramer. alpha-Semihemoglobin and the alpha 3+2(beta 2+-CO)2 hybrid also displayed lower hydroxylase activity. The (alpha 2+-CO)2 beta 3+2 hybrid was about as active as normal alpha 3+2 beta 3+2. This result suggests that the activity of tetrameric hemoglobin primarily involves the beta subunits. Also transfer of the beta subunits from the beta 4 molecular environment to the alpha 2 beta 2 state enhances their monooxygenase activity approximately 15-fold. The hemoglobin derivatives were differently susceptible to substrate inhibition, the beta 4 species being most sensitive. Estimates of Vmax from the linear portions of the corresponding Lineweaver-Burk plots showed agreement within a factor of 2.5 for all of the hemoglobin derivatives, suggesting that the intrinsic O2-activating capacities of the derivatives are similar.(ABSTRACT TRUNCATED AT 250 WORDS)

Monooxygenase activity of human hemoglobin: NMR demonstration of different modes of substrate binding corresponding to different activities of hemoglobin derivatives.

In the accompanying paper [Ferraiolo, B. L., Onady, G. M., & Mieyal, J. J. (1984) Biochemistry (preceding paper in this issue)] we reported different aniline hydroxylase activities for ferrihemoglobin, its isolated subunits, and the converse pair of valency hybrids alpha 3+2(beta 2+-CO)2 and (alpha 2+-CO)2 beta 3+2 in a reconstituted system containing reduced nicotinamide adenine dinucleotide phosphate (NADPH) and cytochrome P-450 reductase. To investigate the molecular basis for the different activities, 1H NMR T1 relaxation studies of aniline were performed in the absence and presence of each of the hemoglobin (Hb) species. The paramagnetic contribution of the ferric heme iron atoms of each Hb derivative to the enhanced relaxation of the proton nuclei of aniline was determined relative to control experiments in which the hemoproteins had been converted fully to the corresponding (carbonmonoxy)ferrous forms, which are diamagnetic. According to the known distance dependence of the paramagnetic effect and the relative changes in T1 for the upfield and downfield signals in the spectrum of aniline, it was ascertained that aniline binds in the same manner to the beta-ferric hybrid and to ferrihemoglobin. These two forms displayed equivalent hydroxylase activities that were the highest among the Hb derivatives for the same aniline concentration. The T1 changes observed with the alpha-ferric hybrid suggest a different orientation for aniline in that complex. The T1 data for the isolated subunits alpha 3+ and beta 3+4 would indicate that overall binding of aniline includes a component of direct aniline-heme ligation in each case.(ABSTRACT TRUNCATED AT 250 WORDS)

Catalysis of the photo-Fries reaction: antibody-mediated stabilization of high energy states.

A conformationally constrained hapten is presented that is capable of catalyzing the first antibody-mediated photo-Fries rearrangement. In this reaction, absorption of light energy by a diphenyl ether substrate results in homolytic C-O bond cleavage followed by recombination to yield biphenyl-derived products. The most proficient antibody studied converts 4-phenoxyaniline 15 into 2-hydroxy-5-aminobiphenyl 16 under high-intensity irradiation at a rate of 8.6 microM/min. These results support a recent hypothesis stating that immunization with conformationally constrained haptens provides higher titers for the acquisition of simple binding antibodies; however, in this case, conformational constraint does not ensure the development of more efficient catalysts. Using the obtained antibodies, the presence of products resulting from escape of free radicals from the solvent cage can be suppressed, altering the excited state energy surface such that free radicals are funneled into the formation of the desired biphenyl product. However, studies also show the inactivation of the antibodies as a result of photodecay of the biphenyl product. Using an isocyanate scavenging resin, the photodecay product could be removed and the inactivation of the antibody drastically reduced. Furthermore, despite the observed photodecay, turnover of the antibody was present; this represents the first case in which true turnover of a photochemical reaction using a catalytic antibody could be observed.

Synthesis of diindolocarbazoles by Cadogan reaction: route to ladder oligo(p-aniline)s.

Symmetric and nonsymmetric diindolocarbazoles were successfully synthesized for the first time by a Cadogan ring closure using N-alkyl-2,7-disubstituted carbazole precursors. Cyclization reaction on N-alkyl-2,7-di(2'-nitrophenyl) carbazole derivatives is not regioselective and produced a separable mixture of symmetric and nonsymmetric diindolocarbazoles. A carbazole derivative with methyl protective groups at the 1- and 8-positions was therefore used to obtain a symmetric ladder oligo(p-aniline) (compound 22). Optical and electrochemical properties of compound 22 indicate that its neutral semiconducting form is stable in air. This novel class of electroactive ladder oligomers should create new opportunities in micro- and nanoelectronics.

Dynamics of p-nitroaniline molecules in micoporous aluminophosphate AlPO4-5 studied by solid-state NMR.

Dynamics of deuterated p-nitroaniline (pNA-d) molecules in the micropores of AlPO4-5 has been investigated by means of solid-state NMR. The adsorbed amounts of pNA-d were 5.0 and 10.1 mass % of the total mass. We have measured 13C magic-angle-spinning (MAS) and 2H NMR spectra of the guest molecules and 31P and 27Al MAS NMR spectra of the host framework. The pNA-d molecules distribute rather inhomogeneously in the channel, and do not coordinate to Al strongly like H2O. The intermolecular hydrogen bonds are formed between a part of the guest molecules only when the loading level is high. The 2H NMR spectra are successfully analyzed, elucidating the orientation and the motion of the guest molecules. The molecular axis of pNA-d is inclined to the channel axis, and the molecular plane is perpendicular to the inner wall. The guest molecule jumps among 12 sites or 12 orientations. This motion is faster in the sample of 5.0 mass % than in the sample of 10.1 mass %, suggesting that the guest-guest interaction hinders the motion. The mean residence times of the molecules are estimated from the analysis of the 2H NMR spectra, which are affected by the size of the nanospace as well as the property of the adsorbed site.

32P-Postlabeling of N-(deoxyguanosin-8-yl)arylamine adducts: a comparative study of labeling efficiencies.

32P-Postlabeling is an extremely powerful technique for the detection of DNA adducts. Typically, the quantitation of DNA adducts by (32)P-postlabeling is achieved by relative adduct labeling, via comparison of the radioactivity incorporated into the adducts to that associated with the normal nucleotides. This approach is based on a number of assumptions, the foremost being that normal and adducted nucleotide 3'-phosphates are converted to 3', 5'-bisphosphates with similar efficiencies. To evaluate labeling efficiencies for specific DNA adducts, we conducted a comparative study of the kinetics of phosphorylation by T(4) polynucleotide kinase using 2'-deoxyguanosine 3'-phosphate (dG3'p) and a series of N-(deoxyguanosin-8-yl)arylamine 3'-phosphate adduct standards (dG3'p-C8-Ar, Ar being 4-aminobiphenyl, 3- and 4-methylaniline, and 2,4- and 3,4-dimethylaniline). Phosphorylation of dG3'p and the dG3'p-C8-Ar adducts followed Michaelis-Menten kinetics. The apparent turnover numbers were 40-240-fold lower when labeling dG3'p-C8-Ar adducts compared to that when labeling dG3'p. The apparent specificity constant calculated for dG3'p-C8-4-aminobiphenyl (1.4 microM(-)(1) min(-)(1)) was approximately 4-fold lower than that (5. 4 microM(-)(1) min(-)(1)) found for dG3'p. Apparent specificity constants for the monoarylamine adducts were even lower (0.043-0.23 microM(-)(1) min(-)(1)) and decreased in the following order: 4-methylaniline > 3,4-dimethylaniline > 3-methylaniline > 2, 4-dimethylaniline. Similar experiments conducted with dG3'p-C8-Ar standards for 2-methylaniline and 2,3-dimethylaniline produced very poor and irreproducible labeling. These results indicate that (32)P-postlabeling of dG3'p-C8-Ar adducts is less efficient than that of dG3'p and suggest that normal nucleotides will be labeled preferentially to the arylamine adducts under kinetically controlled conditions. The data also indicate a further decrease in labeling efficiency upon substitution ortho to the amino group (e.g., 2, 4-dimethylaniline). In addition, the ATP concentrations required for optimal labeling were found to be substantially higher than those used in typical (32)P-postlabeling assays. Since the high specific activity of carrier-free [gamma-(32)P]ATP precludes increasing the ATP concentration to a significant extent, these data emphasize the need for using highly efficient adduct enrichment procedures when conducting (32)P-postlabeling analyses of DNA adducts.

Pyridine nucleotide involvement in rat hepatic drug metabolism: pyridine nucleotide kinetics of aniline parahydroxylation.

Apparent kinetic constant (NADPH) have been calculated for NADPH-cytochrome P450 reductase and parahydroxylase in the presence of aniline with hepatic microsomes from both phenobarbital pre-treated and untreated rats. The addition of NADH gave similar stimulation of both aniline parahydroxylation and NADPH-cytochrome P450 reductase activity in the presence of aniline. It is proposed that the increase in aniline metabolism with NADH is due to an increase in the NADPH-cytochrome P450 reductase activity which is rate limiting in microsomes from both phenobarbital and untreated rats.
