Flow cytometric method to measure urea-resistant alkaline phosphatase from blood neutrophils for use in the prenatal screening of Down's syndrome.

The histochemical measurement of urea-resistant alkaline phosphatase from maternal blood neutrophils is known to have a high detection rate for the prenatal detection of Down's syndrome pregnancies. However, because the histochemical method is laborious and subjective to use, it has not gained widespread acceptance in prenatal screening programmes. We present a simple and objective method for the measurement of urea-resistant alkaline phosphatase by flow cytometry. The method should allow the design of larger studies aimed at evaluating the role of neutrophil urea-resistant alkaline phosphatase in the prenatal screening for Down's syndrome.

[Two forms of alkaline phosphatase in blood serum in liver diseases].

A fraction whose electrophoretic mobility in polyacrylamide gel corresponded to the liver isoenzyme was produced due to trypsin hydrolysis of the high-molecular alkaline phosphatase of blood serum of patients suffering from liver diseases and of liver homogenates. In the presence of positively and negatively charged micelles of detergents the electrophoretical mobility of high-molecular alkaline phosphatase in agarose changed essentially as compared to that in the presence of the noncharged detergent micelles. Similar changes in mobility were not observed for the liver isoenzyme. Due to the autolysis of liver homogenates the activity of the zone corresponding to the liver isoenzyme in the electrophoretic mobility increases and that of high-molecular alkaline phosphatase decreases. Similar changes were observed in sera as well, however, their rate is lower.

[Diagnostic significance of isoenzymes and isoforms of alkaline phosphatase in blood serum of patients with multiple sclerosis on the clinical form of the illness].

In multiple sclerosis there takes place a gradual destruction of most organs and tissues of the sick organism. With progression of the illness changes are noticeable in their functional development. A biochemical mode of assessment of functioning of organs and tissues is analysis of alkaline phosphatase, which is performed with high dilution. Activity of alkaline phosphatase was measured together with its isoenzymes and isoforms in blood serum of 101 patient with disseminated sclerosis depending on the clinical form of the illness. Measurement of blood serum alkaline phosphatase isoenzymes and isoforms can be a marker of the demyelinating process in multiple sclerosis.

The source and clinical significance of serum alkaline phosphatases in neoplastic blood diseases.

Total serum alkaline phosphatase activity and its isoenzymes were studied in 126 patients sent to hospital with a neoplastic blood disease. They comprised 63 patients with a lymphoreticular neoplastic disease, 17 with immature stem-cell leukaemia, 11 with a plasmocytoma, 16 with lymphocytic leukaemia, 4 with polycythaemia and 9 with the myelofibrosis syndrome. Evaluation of the serum alkaline phosphatase isoenzymes significantly enhances the efficiency of the diagnosis and the organ specificity of the enzymes in these conditions. It is particularly important in the above states for studying liver and bone involvement, both from the aspect of the organic localization of the disease and from the aspect of evaluation of the success of therapy.

[Isoenzymes of esterase and alkaline phosphatase in blood serum and adipose tissue of rats after partial pancreatectomy].

A decrease in activities of isoenzymes of aryl-, carboxyl esterases and alkaline phosphatase was shown, by means of agar gel electrophoresis, in blood serum and in most the isoforms of perinephric adipose tissue of rats within 3 days after partial pancreatectomy. In subsequent periods (6, 11 and 14 days) a normalization in the enzymatic activity and an increase in content of blood serum cholinesterase activity occurred.
