Histological, histochemical and biochemical effects of cadmium chloride in female koel (Eudynamys scolopacea).

Effects of single subcutaneous injection (1.5 mg/100 g body weight) of cadmium chloride were studied in the ovary, oviduct, thyroid and adrenal gland of Indian koel (Eudynamys scolopacea). Treatment resulted the significant decrease (p less than 0.001) in the weight of ovary and oviduct. The stromal tissue showed hyperaemia and profused haemorrhage leading to some cellular destruction. A marked degeneration was noticed in the lamina propria of magnum. No remarkable change occured in thyroid histology. An increment of alkaline phosphatase activity was pronounced in ovarian tissue as well as in magnum after the injection. An overall reduction of periodic acid Schiff reaction was noticed in both ovary and oviduct after the salt treatment. The experiment caused a significant reduction of eponephrine content (p less than 0.001) in hypertrophied adrenal medulla. The cortical tissues, however, unaltered in their histomorphology. The loss of total amount of cholesterol (p less than 0.05) from the adrenal gland after the experimentation was recorded. It has been suggested that the augmentation of epinephrine secretion suppressed the gonadal acitivty.

[Histochemical study on effect of radix Salviae miltiorrhizae on growth of isolated cells from embryonic chicken frontal bone cultured in vitro].

12-day embryonic chicken frontal bone digested with trypsin to prepare the suspension of isolated bone cells. 3 x 10(6) cells were harvested altogether. The cells were divided equally into five parts. Then the Eagle medium and 0.1%, 0.2%, 0.4% and 0.8% Radix Salviae Miltiorrhizae in Eagle medium were added respectively and cultured in 5% CO2 incubator. It was observed under the inverted microscope every day. At the 26th day of culture, the cells were studied. The specimens were stained with H. E., Alcian Blue-Sirius Red, Alizarin Red S staining and alkaline phosphatase-acid phosphatase reaction for comparison. It was found that the maturation of the osteoblast-like cells could be accelerated by Radix Salviae Miltiorrhizae. Secretion of the collagenous substance, positive alkaline phosphatase reaction and deposition of mineral on the collagenous substance, forming bone nodules were found to be enhanced. But unduly high concentration of Radix Salviae Miltiorrhizae could lead to inhibition of osteoblast-like cell growth. The optimal concentration of Radix Salviae Miltiorrhizae was 0.2% in culture medium.

[Changes in succinate dehydrogenase activity in the cerebral cortex as an effect of aminazin (histochemical and electron-cytochemical study)].

The activity and localization of succinate dehydrogenase (SDH) in the cortex of rats was studied before and after aminazin administration (15 mg/kg). Changes in the enzyme activity according to photometry occurred in 2 directions: a general drop in the activity of neurons and neuropile, a decrease in the amount of neurons reacting to SDH. Ultrastructurally the enzyme was detectable in the mitochondria of the cell body, adjacent processes and in the processes of cells circumscribing the vessels. In most of the neuropile mitochondria the product of the reaction was not found. Under the influence of aminazin there appeared dark neurons not containing organellas with products of reaction. There were also light neurons in the mitochondria without products of reaction and lipofucsin granules containing products of reaction. The above-mentioned changes permit to explain the drop in the SDH activity, found on photometry of histochemical changes.

[Neurohumoral, autacoid and transductional mechanisms in the cardiovascular effects of vanadate: histochemical correlations].

Rabbits given 1 ppm of vanadate in drinking water for twelve months showed (a) increased plasma levels of catecholamines (b) reduction of the arterial concentration of nitric oxide (c) lower activity of urine kallikrein and higher activities of urine kininases I and II and enkephalinase (d) reduced cardiac inotropism and augmented total peripheral resistance, with unchanged blood pressure levels (e) accumulation of the metal in the aorta and cardiac ventricles. Monoaminooxidase and glucose-6-phosphate dehydrogenase activities were increased by vanadate in both kidney and liver and that of NADH-diaphorase in the kidney, in which NADPH-diaphorase activity was reduced. Some of the above results were also obtained in rats given 10 and 40 ppm of vanadate in drinking water for six-seven months; these animals showed arterial hypertension and reduced activity of Na, K-ATPase in the kidney. Vanadium appears to act on the cardiovascular function through selective neurohumoral, autacoidal and transductional mechanisms only in part depending on the species.

The effect of EDTA on the histochemical myofibrillar ATPase reaction.

The effects of different salts and concentrations of ethylene diamine tetraacetic acid (EDTA) on the histochemically demonstrable myofibrillar ATPase reaction have been investigated. Serial frozen sections from the vastus lateralis muscle of rats, rabbits and guinea pigs were treated with cold (4 degrees) dipotassium EDTA solution and then incubated for myofibrillar ATPase activity. The results showed that all type I muscle fibres exhibited enhanced ATPase activity while type II fibres demonstrated either very low, moderate or intense reaction suggesting the existence of three different sub-populations. None of these effects was observed if the sections were immersed in magnesium chloride solution after preincubation in cold diK+ EDTA. Preincubation in cold non-ionic EDTA, disodium or tetrasodium EDTA did not affect the regular ATPase reaction pattern, while preincubation in warm (19 degrees--21 degrees C) solutions of the same salts resulted in reversal of the regular pattern of ATPase activity. The present histochemical data emphasise the importance of K+, Mg++ and temperature during preincubation of muscle fibres in EDTA before the demonstration of myofibrillar ATPase activity. Moreover, preincubation in a cold solution of diK+ EDTA may be valuable in the histochemical detection of distinct sub-populations of type II muscle fibres and certainly distinguished between type II white and type II red.

A histochemical study on the central effect of monoamine precursors.

A histochemical investigation has been made on the central monoamine neurons after the administration of monoamine precursors and peripheral decarboxylase inhibitor in experimental animals and the results are summarized as below: (1) Catecholamine fluorescence in the brain reduces and yellowish indoleamine fluorescence manifests after the treatment with 5HTP alone and Ro4-4602 plus 5HTP. These findings support the view that catecholamine is likely to be replaced by indoleamine. (2) Ro4-4602 does not cause directly any change in central catecholamine neurons except in median eminence where the inhibitor causes a decrease in dopamine fluorescence. The findings indicate that the inhibitor may act in the hypothalamus as same manner as in the peripheries, because the hypothalamus is presumably lacking of blood brain barrier. (3) L-Dopa alone and L-Dopa plus Ro4-4602 enhance dopamine fluorescence in the substantia nigra and the median eminence, especially an increase in the extra-neuronal fluorescence can be seen in the latter. This indicates the releasing of amines from cell body into extraneuronal spaces.

[Experimental study of the hepatotoxic effect of toluol. I. Histological and histochemical studies].

The hepatotoxic effect of the analytically pure toluol and a toluol being used in the Chemical Factory "Tiszai" was investigated. Single toxic- and various subtoxic doses of toluol were administered to male rats of CFY intraperitoneally and subcutaneously. The body weight and the weight of the liver were registered. Histological and histochemical study of the liver have also been carried out. After an acute and subacute toluol intoxication decrease of the PAS-positive and Bestcarmin positive material in the liver and increase of the activity of SDH were observed. The degree of the damage to the hepatic tissue depends on the doses of toluol and on the duration of the administration. It is stressed that a severe damage to the whole liver was not seen in any of the experimental animals. There was not any difference noticed between the lesions caused by the different types of toluol.

Correlation of HPV antigen and type of genital warts with atypia.

Five clinical types of genital warts were identified, studied histopathologically and stained for human papilloma virus (HPV) antigen. HPV antigen was found in all degrees of koilocytosis and it was present more often in the hyperplastic warts than in other types of genital warts. However, as reported earlier, the presence of HPV antigen did not decrease proportionately as the degree of atypia increased.

Effect of elastase on the histochemical demonstration of alkaline phosphatase activity in chick pecten.

The effect of elastase on alkaline phosphatase activity in the chick pecten capillaries was studied electron histochemically. The pecten was treated with elastase before incubation in the medium for alkaline phosphatase. Inactive alkaline phosphatase in the chick pecten capillary under dark adaptation could be demonstrated electron histochemically after treatment with elastase. Under light adaptation, the elastase-treated pecten demonstrated more intense alkaline phosphatase activity in the same localization as in the untreated pecten. Biochemical data also supported histochemically demonstrated enhancement effect of elastase on alkaline phosphatase activity in the chick pecten oculi. Elastase is expected to be useful for demonstrating enzymatic activity which is otherwise hard to detect.
