Improved separation of motile sperm in asthenospermia and its application to artificial insemination homologous (AIH).

A method is described for the separation of motile sperm from semen by "migration' into a diluent, providing a large area of interface between the semen and the diluent and resulting in a good yield of motile sperm (58%), with a mean percentage of motility of 95%. The method provides a specimen free of seminal plasma and debris and suitable for intrauterine insemination in less than two hours. The motile fraction was used for AIH treatment of 20 couples with a mean of 4 years' infertility, where the female factor had been excluded and the male was asthenospermic. After a mean of six cycles, three pregnancies were achieved. Improvement of the percentage of motility alone did not, in this study, give a significant improvement in conception rates when compared with accepted cycles with the placebo.

Synchronization of lactating dairy cows with prostaglandin F2 alpha: insemination on observed oestrus versus timed artificial insemination.

The effect of two insemination policies after synchronization of oestrus on reproductive performance in two groups of cows was investigated. Oestrus was synchronized by two treatments with prostaglandin F2 alpha (PGF2 alpha) at a 14-day interval. Cows in Group 1 (n = 234) were inseminated twice on appointment 66 and 90 h after the second treatment. Cows in Group 2 (n = 222) were intensively watched for signs of oestrus after the synchronization protocol and inseminated on observed oestrus. Cows with abnormal discharge after synchronization were excluded from breeding and inseminated later during the study period. Service rate within 1 week after synchronization was higher in Group 1 than in Group 2 (89.3 versus 67.1%). Conception rates on first service did not differ between groups (33.2 versus 30.0%). Days to first service and days open were shorter in Group 1 (P < 0.05). The number of cows culled for infertility did not differ between the groups. Endometritis 14-20 days post-partum decreased the percentage of cows pregnant at the end of the study period in both groups but did not have a significant effect on conception rates and days open. It is concluded that additional inseminations required in the timed artificial insemination protocol were economically justified by the reduction in days open in comparison with insemination on observed oestrus after synchronization.

Artificial insemination: an overview.

This article presents an overview of the indications for and techniques of artificial insemination, including artificial insemination of husband's/partner's sperm and artificial insemination of donor sperm. Important issues related to psychologic, legal, and ethical concerns are addressed, as well as long-term ramifications and implications for nursing.

Serum PP12, PP14, SP1 and hCG values in the 28 days after the LH-surge in patients who do and do not conceive after artificial insemination or in vitro fertilization.

Serum levels of PP12 (somatomedin binding protein), PP14 (beta lactoglobulin homologue), Schwangerschaftsprotein 1 (SP1) and of human chorionic gonadotropin (hCG) were simultaneously measured in patients being treated for infertility in the 28 days after the LH-surge. PP14 levels were similar in the 14 days after the LH-surge in the patients who conceived when compared with those who did not and a high PP14 level was only indicative of pregnancy at 21 days after the LH-surge. hCG and SP1 levels behaved similarly in pregnant subjects. PP12 levels did not change significantly in the 28-days after the LH-surge.

A survey of patient attitudes towards artificial insemination by donor.

The psychological characteristics of a consecutive series (n = 58) of women attending an AID clinic have been examined. Women were assessed prior to treatment on a number of attitudinal and acceptability measures, as well as required to complete personality and martial adjustment inventories. At the time of each insemination, over 6 successive cycles, state anxiety was assessed. Following an interval of 6 months women were reassessed on an acceptability measure. The results showed generally positive attitudes at the outset. No systematic trends in anxiety were identified, with no differences between women becoming pregnant and those continuing in treatment. At follow-up women acknowledged the strain involved in timing the insemination to coincide with ovulation, with both pregnant and nonpregnant women attributing outcome to their psychological and emotional state at the time. The implications of these results are considered in terms of the characteristics of women requesting AID and the ways in which clinics can attend to the psychological states of the patients.
