[Study of Chinese herbal medicine in treating ascites and their mechanism in regulating lymphatic stomata].

OBJECTIVE: To study the therapeutic effect of Chinese herbal medicines (CHM) in treating ascites to elucidate its mechanism in regulating the lymphatic stomata and promoting the absorption of ascites from the peritoneal cavity. METHODS: Using scanning electron microscope (SEM) and computerized image processing and quantitative analysis assays, the CHM extract consisting of Atractylodes macrocephala, Salvia miltiorrhiza, Codonopsis pilosula, Alismatis orientale and Leonurus heterophyllus were studied. RESULTS: Intraperitoneal injection of nitric oxide (NO) supplier or CHM administration could cause the average area of lymphatic stomata obviously enlarged (P < 0.05), and the open numbers significantly increased (P < 0.01) in normal healthy mice. When L-notroarginine, a NO synthetase suppressor, was injected after CHM administration, it was found that the regulating effect of CHM on lymphatic stomata was inverted obviously, i.e. the average area and the density of lymphatic stomata were markedly reduced (P < 0.01). CONCLUSION: CHM might treat ascites through increasing the endogenous NO concentration to open the lymphatic stomata and in turn to conduct the peritoneal water through lymphatic path.

A new form of polyagglutination related to Cad.

Four phenotypes of Cad (Cad 1-4) have been characterized by a continuum of polyagglutinability and reactivity with lectins, with the strongest Cad+ red blood cells (RBCs) being polyagglutinable because of the presence of anti-Cad (anti-Sda) in most normal sera. Over a period of 7 years, a French male blood donor's RBCs demonstrated polyagglutinability with 50 percent to 70 percent of normal adult sera. The reactivity was characteristic of anti-Sda (refractile agglutination at 4 degrees C, 20 degrees C, 37 degrees C, and anti-human globulin test), and was inhibitable by two examples of Sd(a+) urine, but not by Sd(a-) urine or dialysate from Sd(a+) urine. The donor's RBCs reacted 1+ with Glycine max, but did not react with Dolichos biflorus, Leonurus cardiaca, Salvia horminum, or Arachis hypogaea. The first four of these lectins were reactive with five of five Cad+ RBCs, including one example of Cad 4 RBCs. Polybrene(R) aggregated the donor RBCs. Dilutions of nine samples of anti-Sda reacted more strongly with the donor RBCs than with normal RBCs. Even though lectin studies failed to classify this donor's RBCs as Cad, the persistent polyagglutinability and serologic characteristics are consistent with Cad and demonstrate the heterogeneity of this antigen.

The effect of i-mu-ts'ao on a partially purified prostaglandin E 9-ketoreductase from swine kidney.

The enzyme system, prostaglandin E 9-ketoreductase, which catalyzes the reduction of prostaglandin E to form prostaglandin F has been partially purified from swine kidney. This NADPH-linked enzyme is studied spectrophotometrically. The KM of this enzyme for prostaglandin E2 was found to be 180 microM. Studies with the partially purified enzyme indicate that prostaglandin E 9-ketoreductase is affected by a Chinese herbal medicine i-mu-ts'ao (leonurus heterophyllus sweet). An increase in the concentration of i-mu-ts'ao aqueous extract may influence the conversion of prostaglandin E2 into prostaglandin F2 alpha. This finding offers a possible explanation for the physiological role of i-mu-ts'ao when it is treated as a medicine.

[Antioxidant properties of a series of extracts from medicinal plants].

Investigation of antioxidant properties of some plants was carried out. A group of plants affected human central nervous system was studied in detail. Efficiency of plants as antioxidants was tested by the influence of their extracts on the yield of photochemiluminescence of Gly-Trp solutions. Antioxidant properties were examined under conditions when their own absorption was minimized. Riboflavin as additional sensitizer was used in this experiment for superoxide generation. The antioxidant effect was evaluated with regard to single dose of plant extracts and their concentration in human organism. The effect decreases in the following consequence: Hypericum > Eleutherococcus > Rhodiola > Leonurus > Aralia > Valeriana > Echinopanax > Schizandra > Panax gin-seng.

[Photosensitizing and photoprotective properties of extracts from groups of medicinal plants].

Investigation of photosensitization and photoprotection induced by plant extracts was carried out. A group of plants affected human central nervous system was studied in detail. Efficiency of plants as photoprotectors and photosensitizers was tested in the frame of the influence of their extracts on the yield of photochemiluminescence of Gly-Trp solutions. Photosensitization was studied under irradiation with light lambda > 280 nm and lambda > 320 nm, as well as with monochromatic light lambda=313, 365, 405 and 436 nm. All of the plants studied acted as photoprotectors in low concentration and as photosensitizers in high concentration. The efficiency of photoprotection and photosensitization was evaluated with regard to single dose of plant extracts and their concentration in human organism. The effect decreases in the following consequence of plants: Leonurus > Hypericum > Aralia > Schizandra > Echinopanax > Eleutherococcus > Valeriana > Panax ginseng. Photosensitization is due to the components of plant extracts which have strong absorbtion at the high wavelength range. The mechanism of photosensitization was suggested. Singlet oxygen generated by photoexcited compounds is the main species resulted in chemiluminescence. Superoxide radicals does not contribute significantly to the chemiluminescence formation.

Anti-oxidative stress effects of Herba leonuri on ischemic rat hearts.

Our current study was to test the hypothesis that the extract of Herba leonuri (HL) would have antioxidant and cardioprotective effects on ischemic myocardium. The extract of HL (400 mg/kg/day) was administered orally (daily) starting from 1 week before and continuing until 3 weeks after myocardial infarction (MI). Surviving rats were sacrificed at different time points to obtain left ventricles for biochemical assays. Our study demonstrates for the first time that HL does have antioxidant effects both in vitro and in vivo. The antioxidant effects of HL are exerted only under the condition of oxidative stress, by selectively preserving the activities of superoxide dismutase and glutathione peroxidase, as well as depressing the formation of malondialdehyde, especially in the acute phase of acute MI. Its effects of scavenging free radicals and inhibiting the formation of reactive oxygen species may play a key role in protecting the endogenous antioxidant system from oxidative stress in vivo.

[Effect of motherwort herb on the myoelectric activity of uterus in rats].

OBJECTIVE: To investigate the effect of motherwort herb on the myoelectric activity of uterus in rats. METHOD: A pair of bipolar Ag-AgCl electrode was implanted on the serosal surface of the uterus in rats, motherwort herb was injected intra-abdominal cavity in rats in dose of 0.1 ml, 0.2 ml and 0.4 ml respectively, and the changes of uterine myoelectric activity were observed. RESULT: Motherwort herb significantly increased the frequency and average amplitude of uterus slow waves of in rats. The frequency and maximum amplitude of single waves also obviously increased. The time course of outbreak waves was prolonged, the series intervals were reduced and the maximum amplitude in each outbreak was increased. CONCLUSION: The exciting effect of motherwort herb on the uterine smooth muscle may have something to do with the alteration of the ion concentration in relation to myoelectric activity, resulting in the increase of myoelectric activity of pacesetter cells as well as in the acceleration of depolarization of spike activity.

[The effect of tofisopam and tinctura leonuri on the color-discrimination function in young humans].

Enhanced anxiety state is accompanied by limitation of the color-discrimination function of retina in young humans. Chronic administration of tofisopam (grandaxin) or tinctura leonuri decreased anxiety and significantly improved the color discrimination function of retina with respect to all four colors studied. It is suggested that this improvement is related to the drug action upon the GABAergic processes both in the retina and in the related cerebral structures.
