Energetics, mechanics and molecular engineering of calcium cycling in skeletal muscle.

During muscle contraction and relaxation, Ca2+ moves through a cycle. About 20 to 40% of the ATP utilized in a twitch or a tetanus is utilized by the SR Ca2+ pump to sequester Ca2+. Parvalbumin is a soluble Ca2+ binding protein that functions in parallel with the SR Ca2+ pump to promote relaxation in rapidly contracting and relaxing skeletal muscles, especially at low temperatures. The rate of Ca2+ dissociation from troponin C, once thought to be much more rapid than the rate of relaxation, is likely to be similar to the rate of cross-bridge detachment and to the rate of muscle relaxation under some conditions. During the past fifty years, great progress has been made in understanding the Ca2+ cycle during skeletal muscle contraction and relaxation. Nonetheless, there are still mysteries waiting to be unraveled.

Molecular engineering of biotin-glutamic acid decarboxylase 65 fusion protein (Biotin-GAD65) for non-radioactive GAD65 antibody assay.

We constructed biotinylated fusion proteins that linked to three detection tags to GAD65 at the N-terminus, and expressed them in an E. coli expression system. The Biotin14-GAD65 protein exhibited the strongest binding to both the GAD65 antibody and the streptavidin among the three constructs. We describe the optimal conditions using a Biotin14-GAD65-based immunoassay for the detection of GAD65 antibody.

Molecular engineering of proteins and polymers for targeting and intracellular delivery of therapeutics.

There are many protein and DNA based therapeutics under development in the biotechnology and pharmaceutical industries. Key delivery challenges remain before many of these biomolecular therapeutics reach the clinic. Two important barriers are the effective targeting of drugs to specific tissues and cells and the subsequent intracellular delivery to appropriate cellular compartments. In this review, we summarize protein engineering work aimed at improving the stability and refolding efficiency of antibody fragments used in targeting, and at constructing new streptavidin variants which may offer improved performance in pre-targeting delivery strategies. In addition, we review recent work with pH-responsive polymers that mimic the membrane disruptive properties of viruses and toxins. These polymers could serve as alternatives to fusogenic peptides in gene therapy formulations and to enhance the intracellular delivery of protein therapeutics that function in the cytoplasm.

[Molecular engineering of hemoglobin for transfusion].

The search for a safe alternative to conventional blood transfusion has been directed towards either the use of synthetic perfluorochemicals or the biochemical manipulation of highly purified stroma-free Hb solutions prepared from outdated bank blood. However when using human blood, one does not eliminate the risks of viral infections. A novel source of Hb appeared with recent biotechnology techniques enabling one to synthesize recombinant Hb from microorganisms (E coli or S. cerevisiae) whose genome has been modified to contain globin genes. Normal human Hb A in solution, i.e. outside the red cells, is not suited for direct usage as a blood substitute because i) its high oxygen affinity, due to the absence of 2,3 DPG in the plasma, precludes sufficient O2 unloading to the tissues; ii) at low concentration, relative to that in the red cells, tetrameric Hb dissociates into dimers which escape the circulatory system by renal filtration or rapidly oxidize to the non functional metHb form. Expression of alpha- and beta-globins in Escherichia coli and in Saccharomyces cerevisiae enables one to introduce appropriate mutation(s) in the globin genes resulting in the expression of a synthetic Hb with low oxygen affinity, resembling that of normal whole blood; functional Hb has also been produced in a soluble form either in E. coli or in yeast. The coexpression of beta globin chains and alpha globin subunits linked by a peptide bond results in the direct synthesis of stabilized and fully functional Hb tetramers. Lastly, dilute haemoglobin solutions are prone to autooxidize and the rate of oxidation appears to be inversely proportional with the oxygen affinity of the heme groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Consequences of molecular engineering enhanced DNA binding in a DNA repair enzyme.

Facilitated one-dimensional diffusion is a general mechanism utilized by several DNA-interactive proteins as they search for their target sites within large domains of nontarget DNA. T4 endonuclease V is a protein which scans DNA in a nonspecifically bound state and processively incises DNA at ultraviolet (UV)-induced pyrimidine dimer sites. An electrostatic contribution to this mechanism of target location has been established. Previous studies indicate that a decrease in the affinity of endonuclease V for nontarget DNA results in a decreased ability to scan DNA and a concomitant decrease in the ability to enhance UV survival in repair-deficient Escherichia coli. This study was designed to question the contrasting effect of an increase in the affinity of endonuclease V for nontarget DNA. With this as a goal, a gradient of increasingly basic amino acid content was created along a proposed endonuclease V-nontarget DNA interface. This incremental increase in positive charge correlated with the stepwise enhancement of nontarget DNA binding, yet inversely correlated with enhanced UV survival in repair-deficient E. coli. Further analysis suggests that the observed reduction in UV survival is consistent with the hypothesis that enhanced nontarget DNA affinity results in reduced pyrimidine dimer-specific recognition and/or binding. The net effect is a reduction in the efficiency of pyrimidine dimer incision.

Molecular engineering of myoglobin: the improvement of oxidation activity by replacing Phe-43 with tryptophan.

The F43W and F43W/H64L myoglobin (Mb) mutants have been constructed to investigate effects of an electron rich oxidizable amino acid residue in the heme vicinity on oxidation activities of Mb. The Phe-43 --> Trp mutation increases the rate of one-electron oxidation of guaiacol by 3-4-fold; however, the peroxidase activity for F43W/H64L Mb is less than that of the F43W single mutant because the absence of histidine, a general acid-base catalyst, in the distal heme pocket suppresses compound I formation. More than 15-fold improvement versus wild-type Mb in the two-electron oxidation of thioanisole and styrene is observed with the Phe-43 --> Trp mutation. Our results indicate that Trp-43 in the mutants enhances both one- and two-electron oxidation activities (i.e., F43W Mb > wild-type Mb and F43W/H64L Mb > H64L Mb). The level of (18)O incorporation from H2(18)O2 into the epoxide product for the wild type is 31%; however, the values for F43W and F43W/H64L Mb are 75 and 73%, respectively. Thus, Trp-43 in the mutants does not appear to be utilized as a major protein radical site to form a peroxy protein radical in the oxygenation. The enhanced peroxygenase activity might be explained by the increase in the reactivity of compound I. However, the oxidative modification of F43W/H64L Mb in compound I formation with mCPBA prevents us from determining the actual reactivity of the catalytic species for the intact protein. The Lys-C achromobacter digestion of the modified F43W/H64L mutant followed by FPLC and mass analysis shows that the Trp-43-Lys-47 fragment gains a mass by 30 Da, which could correspond two oxygen atoms and loss of two protons.

Molecularly engineered nanocomposites: layer-by-layer assembly of cellulose nanocrystals.

Cellulose nanocrystals are promising as a new class of reinforcing material for the preparation of nanostructured composites. We report here the preparation of cellulose nanocrystal multilayer composites with poly(diallyldimethylammonium chloride) using layer-by-layer assembly (LBL) technique. The LBL assembly was characterized with UV-Vis spectroscopy and ellipsometry. The average thickness of a single bilayer was found to be 11 nm. AFM and SEM characterization revealed uniform coverage and densely packed cellulose crystal surface.

Molecular engineering of C60-based conjugated oligomer ensembles: modulating the competition between photoinduced energy and electron transfer processes.

A series of novel and soluble C60-(pi-conjugated oligomer) dyads were synthesized, starting from suitably functionalized oligomer precursors (i.e., dihexyloxynaphthalene, dihexyloxynaphthalene-thiophene, and dihexyloxybenzene-thiophene). A systematic change in the nature of the oligomeric component allowed (i) tailoring the light absorption of the chromophore by shifting the ground-state absorption from the ultraviolet to the visible region and (ii) varying the oxidation potential of the donor. The resulting electro- and photoactive dyads were examined by electrochemical and photophysical means. In general, both singlet-singlet energy transfer and intramolecular electron transfer were found to take place and, most importantly, to compete with each other in the overall deactivation of the photoexcited oligomer. The selection of polar solvents in combination with the dihexyloxybenzene-thiophene donor shifted the reactivity from an all energy (1a; dihexyloxynaphthalene) to an all electron-transfer scenario (1d, dihexyloxybenzene-thiophene). Encouraged by the favorable electron-transfer properties of dyad 1d, we prepared photodiodes by embedding 1d between asymmetric metal contacts, which showed external monochromatic efficiencies (IPCE) close to 10% at the maximum absorption of the molecule.

Molecular engineering. 8. Kinetic and conformational studies of resorcin [4]arene-based C4 tetraoxatetrathiahemicarceplexes: carceroisomerism and twistomerism.

New C(4v) tetraoxatetrathiahemicarcerands and their six hemicarceplexes containing DMF, DMA, DMSO, or NMP were synthesized and characterized. Their conformations, kinetic properties, carceroisomerism, and twistomerism were studied by VT, 2D COSY, NOESY, and ROESY (1)H NMR experiments. The decomplexation rates of DMF or DMA were very slow with high activation energy barriers (73 and 104 kJ mol(-1), respectively) and the complexed guests feel more constriction than their free liquid state. The largest isomerization energy barrier of carceroisomers was 15.4 kcal mol(-1), and the isomerization energy barriers of twistomers are significantly larger than those of carceroisomers.

Advances in radioimmunotherapy in the age of molecular engineering and pretargeting.

Now that radioimmunotherapy is an approved method for the treatment of certain types of non-Hodgkin's lymphoma, investigators are turning to new approaches to further improve radionuclide targeting in hopes of expanding the use of this technology. A number of innovative recombinant proteins have been developed with more favorable pharmacokinetic and targeting properties than standard whole IgG, which conceivably could improve the therapeutic index for cancer treatment. Pretargeting methods also are coming of age, with preclinical and early clinical studies in a variety of cancers illustrating how this alternative approach can enhance the therapeutic window several-fold of what has been possible with directly radiolabeled IgG. This review will discuss some of these promising new developments.
