[Simultaneous measurement of 32P and a 33P double-labelled sample by liquid scintillation counter and its application to phosphorus compounds isolated by paper chromatography].

Simultaneous determination of 32P and 33P radioactivities was carried out with the liquid scintillation counter (Aloca LSC 670). For the quenching correction, the channel ratio method was used, and the proper gain and window level were set to get the maximum performance number or Klein-Eisler value. Using this method the radioactivities of the 32P and 33P double labelled compounds which were separated with paper chromatography were determined. New index, RSA (relative specific activity, i.e. 32P/33P) was proved to be effective as well as specific activity for the studies of the transport and metabolism of phosphorus compounds in the plant roots.

Effect of phosphorus deficiency on levels of phosphorus compounds in spirodela.

When Spirodela plants are transferred to a phosphate-deficient medium, growth slows down immediately, and ceases after 14 days. During this time, inorganic phosphate content falls from 30 to 0.7 mumoles/g fresh weight of tissue, phosphate ester content from 3.5 to 0.6 mumoles/g, phospholipid content from 3.5 to 1.2 mumoles/g, and residual phosphate (mainly RNA) content from 7.5 to 2.0 mumoles/g. Relative proportions of the various phosphate esters, and relative proportions of the various phospholipids, are not markedly affected by phosphate deficiency. Turnover rates of phosphate esters are somewhat higher in phosphate-deficient tissue. In control tissue, inorganic phosphate is present in 2 pools; a metabolic (12%) and a non-metabolic pool (88%). In phosphate-deficient tissues, most of the inorganic phosphate (>90%) is in the metabolic pool. Non-metabolic phosphate is presumably stored in the vacuole, and is not readily accessible to the tissue, so that growth normally occurs at the expense of external phosphate. During deficiency, growth is limited by the rate at which phosphate can be transported through the tonoplast and tissue to the growing point. Growth ceases when the supply of non-metabolic phosphate is exhausted. Metabolic phosphate is presumably located in the cytoplasm: it can not be used for growth. Nor can the plant respond to deficiency by making some phosphorus compounds at the expense of others. In this respect, phosphorus deficiency and nitrogen deficiency are dissimilar.

Bioavailability of dissolved organic phosphorus compounds to typical harmful dinoflagellate Prorocentrum donghaiense Lu.

The bioavailability of dissolved organic phosphorus (DOP) compounds to harmful alga Prorocentrum donghaiense Lu and its alkaline phosphatase activity (APA) were studied. Results showed that P. donghaiense could utilize the test DOP compounds such as glucose-6-phosphate (G-6-P), adenosine triphosphate (ATP) and ribonucleic acid (RNA) to sustain its growth. Nutrition between the test DOP compounds and orthophosphate was comparable. P. donghaiense could utilize intracellular phosphorus to sustain growth under depletion of dissolved phosphorus. Variation of APA in different test DOP compounds was addressed; the controlling mechanism of APA in different DOP media was discussed.

Molecular Structures of Three-, Four-, and Five-Coordinate Phosphorus Compounds Containing Salicylate Ligands(1).

The new cyclic compound 2,2'-sulfurylbis(4-methyl-6-tert-butylphenyl) methyl 2-benzoate phosphite, O(2)S[(t-Bu)MeC(6)H(4)O](2)(OC(6)H(4)CO(2)Me)P (3), containing a salicylate ligand was synthesized from 2,2'-sulfurylbis(4-methyl-6-tert-butylphenyl) chlorophosphite and methyl salicylate in the presence of triethylamine in ether solution. X-ray analyses of bis(methyl salicylate-O)phenylphosphine, (OC(6)H(4)CO(2)Me)(2)PPh (1), and bis(methylsalicylato-O)phenyl(tetrachlorophenylene-1,2-dioxy)phosphorane, (O(2)C(6)Cl(4))(OC(6)H(4)CO(2)Me)(2)PPh (2), as well as that for 3 were obtained. The phosphane 1 has a pseudo trigonal bipyramidal (TBP) structure due to coordination of a carbonyl oxygen atom at an axial site. The cyclic phosphorane 2 and the phosphite 3 lack any coordination from salicylate ligands. This results in a TBP geometry and a pyramidal geometry respectively for 2 and 3. Comparisons with X-ray structures for carboxylate-containing phosphorus compounds exhibiting oxygen coordination show the formation of four- and five-membered cyclic systems. Thus, 1 appears to be the first example of formation of a six-membered ring via carbonyl oxygen coordination. Reference is made to the tyrosyl-tRNA synthetase system where it is proposed that carbonyl oxygen atom coordination is a likely occurrence in the transition state on the basis of the analysis presented in this work.
