Synthesis and characterization of Sn(IV) complexes of lower rim 1,3-diacid derivative of calix[4]arene and their protective effects on tissue oxidative stress and essential metal concentration in lead exposed male Wistar rats.

The two Sn(IV) complexes synthesized using calix[4]arene-1,3-di-acid derivative were characterized by analytical, (1)H, (13)C and (119)Sn NMR, matrix assisted laser desorption ionization mass, and (119)Sn Mossbauer techniques and found that the complexes are tetranuclear possessing structurally two different types of tin centers. These complexes were evaluated for their protective value against blood and tissue oxidative stress in lead exposed male albino rats of Wistar strain. The results suggest that the two tin complexes significantly protect changes in lead induced biochemical variables indicative of heme synthesis pathway and exhibit only moderate effect on tissue oxidative stress. The beneficial effects could be attributed mainly to the ability of Sn(IV) complexes in preventing absorption of lead to the target sites/tissues.

Butyltin concentrations along the Japanese coast from 1997 to 1999 monitored by Caprella spp. (Crustacea: Amphipoda).

The concentrations of butyltins along the Japanese coastline were investigated from 1997 to 1999, 7 to 9 years after implementation of legislation limiting the use of tributyltin (TBT) in Japan. Seawater was sampled at 0.5 m depth, and Caprella spp. were collected from Sargassum spp. and aquaculture facilities from 18 areas within four broad areas along the coastline of Japan, i.e., the Pacific coast of northern Japan, the coast along the Sea of Japan, Tokyo Bay and adjacent areas, and western Japan. Butyltins (MBT, DBT and TBT) were detected in 32 of the 63 seawater samples with average concentrations of 4.6 ng MBT/l, 4.5 ng DBT/l and 6.8 ng TBT/l, respectively. Butyltin concentrations in seawater from western Japan indicate "hot spots" even in unpopulated areas. Butyltins (MBT, DBT and TBT) were detected in all samples of Caprella spp., varying from 2.3 ng BTs /g wet wt in C. penantis R-type from Tobishima Island in the Sea of Japan to 464 ng BTs /g wet wt in C. decipiens Mayer from Amakusa, western Kyushu. The BT concentrations in Caprella spp. form western Japan were significantly higher than those from other areas, including Tokyo Bay and adjacent areas, where large scale industry and international ports are located. These results indicate that butyltin contamination still remains even in unpopulated areas after the regulation on TBT usage, and that the regulation governing TBT usage since 1990 has not been effective enough to concede recovery of shallow water ecosystems around Japan.

An easy, non-destructive, probabilistic method to evaluate the imposex response of gastropod populations.

Endocrine disruption in marine populations is a growing concern. One of the best documented examples is imposex, the development of male characteristics in female gastropods, including the development of a penis and/or vas deferens, by exposure to organotins, mainly tributyltin. This syndrome has been an effective and widely used biomarker of organic tin contamination for almost 20 years. However, established imposex evaluation protocols sacrifice all the animals and repeated assessments can result in depletion of populations. To preserve animal lives as much as possible and to have a fast screening of areas subjected to organotin impacts, a non-destructive approach was tested. This approach uses reversible narcotization and a statistical criterion to evaluate the extent of imposex in the populations, instead of imposex indexes, which are based on evaluation of sacrificed, fixed animals. The data consist of two categories: males plus masculinized females in one category and normal females in another. The probability is then calculated that this sample could have been drawn from a normal population. This new method was tested in two contrasting areas of Brazil and with two different species, and the results compared to the conventional evaluation. Narcotization was completely reversed with a survival rate of 98%. Impacted sites could be identified with this technique in the same way as with the conventional one. The only limitation was that female sterility could not be verified. This disadvantage could well be compensated by the minimal mortality in the studied populations.

Attenuation of nociception in a model of acute pancreatitis by an NK-1 antagonist.

Substance P (SP) acting at the NK-1 neurokinin receptor has a well-documented role in the transmission and maintenance of nociceptive information. SP is found in the majority of fibers innervating the pancreas, and it is up-regulated after pancreatic inflammation. The aim of this study was to investigate the role of the NK-1 receptors in the maintenance of pancreatic nociception. Using a newly developed rat model of acute pancreatic nociception that persists for 1 week, the NK-1 receptor expression in the spinal cord and pancreas was examined using immunohistochemistry and Western blotting procedures. The effects of a specific NK-1 antagonist, CP99,994, on the behavioral manifestations of pancreatic nociception were determined. The antagonist was administered intraperitoneally and intrathecally to differentiate peripheral and central effects. Injection of CP-100,263, the inactive enantiomer of CP-99,994 was used as a control for nonspecific effects of the antagonist. Immunohistochemistry and Western blotting analysis revealed an up-regulation of the NK-1 receptor occurs in the pancreas but not at the spinal cord level. The NK-1 antagonist was able to attenuate the nociceptive behaviors in rats with pancreatitis when applied intraperitoneally with a short duration of effectiveness. Intrathecal application of the antagonist was ineffective. These results suggest the involvement of pancreatic NK-1 receptors in the maintenance of nociception during pancreatic inflammation.

Developmental toxicity of dibutyltin dichloride in cynomolgus monkeys.

Dibutyltin dichloride (DBTCl) has been shown to be teratogenic in rats. The present study was conducted to determine the teratogenic potential of DBTCl given to pregnant monkeys during the entire period of organogenesis. Cynomolgus monkeys were dosed once daily by nasogastric intubation with DBTCl at 0, 2.5 or 3.8 mg/kg on days 20-50 of pregnancy, the whole period of organogenesis. The pregnancy outcome was determined on day 100 of pregnancy. In both DBTCl-treated groups, a significant increase in the incidence of pregnant females with soft stool and/or diarrhea, and with yellowish stool was observed. Maternal body weight gain at 3.8 mg/kg and food consumption at 2.5 and 3.8 mg/kg were decreased during the administration period. The survival rate of fetuses at terminal cesarean sectioning was decreased in the DBTCl-treated groups and significantly decreased at 2.5 mg/kg. There were no changes in the developmental parameters of surviving fetuses, including fetal body weight, crown-rump length, tail length, sex ratio, anogenital distance and placental weight, in the DBTCl-treated groups. No external, internal or skeletal malformations were found in the fetuses in any group. Although internal and skeletal variations were found, no difference in the incidence of fetal variation was noted between the control and DBTCl-treated groups. No effect on skeletal ossification was observed in fetuses in the DBTCl-treated groups. The data demonstrate that DBTCl is embryolethal but not teratogenic in cynomolgus monkeys.

Preparations and spectroscopic studies of organotin complexes of diclofenac.

The reactions of the potent and widely used anti-inflammatory drug diclofenac, HL, with diorganotin(IV) oxides were studied. The dimeric tetraorganodistannoxane complexes [Me(2)LSnOSnLMe(2)](2), [Bu(2)LSnOSnLBu(2)](2), [Ph(2)LSnOSnLPh(2)](2) and the dibutyltin complex [Bu(2)SnL(2)], have been prepared and structurally characterized in the solid state by means of vibrational and 119Sn Mössbauer spectroscopy. Determination of lattice dynamics by temperature-dependent 119Sn Mössbauer spectroscopy. From the variable-temperature Mössbauer effect, the Debye temperature was determined. The complexes have been characterized in solution by NMR (1H and 13C) spectroscopy. Vibrational, Mössbauer, and NMR data are discussed in terms of the proposed structures.

New organotin(IV) ascorbates: synthesis, spectral characterization, biological and potentiometric studies.

New organotin(IV) ascorbates of the general formulae R(3)Sn(HAsc) (where R = Me , n-Pr, n-Bu and Ph) and R(2)Sn(Asc) (where R = n-Bu and Ph) have been synthesized by the reaction of R(n)SnCl(4-n) (where n = 2 or 3) with monosodium-l-ascorbate. The bonding and coordination behaviour in these complexes are discussed on the basis of UV-Vis, IR, Far-IR, (1)H and (13)C NMR, and (119)Sn Mossbauer spectroscopic studies. L-Ascorbic acid acts as a monoanionic bidentate ligand in R(3)Sn(HAsc) coordinating through O(1) and O(3). The Mossbauer studies together with IR and NMR studies suggest that for these polymeric derivatives, the polyhedron is trigonal bipyramidal around tin with three organic groups in the equatorial positions. In R(2)Sn(Asc), L-ascorbic acid acts as dianionic tetradentate ligand and a polymeric structure with octahedral geometry around tin with trans organic groups has been tentatively proposed. The complexes have been assayed for their anti-inflammatory and cardiovascular activity. Ph(2)Sn(Asc) has been found to show the highest activity among the studied complexes. It is suggested on the basis of potentiometric studies of Me(2)Sn(IV) and Me(3)Sn(IV) systems with L-ascorbic acid that under physiological conditions (pH = 7.0) Me(2)Sn(HAsc)(OH) (approximately 60%), Me(2)Sn(OH)(2) (approximately 40%) and Me(3)Sn(HAsc) (approximately 60%), Me(3)Sn(OH) (approximately 40%), respectively, are existing, which may be responsible for their biological activities.

Organotin(IV) complexes of thiohydrazones: synthesis, characterization and antifungal study.

Some new organotin(IV) complexes with salicylaldehyde aniline-N-thiohydrazone (L1) and cinamaldehyde aniline-N-thiohydrazone (L2) of the type (p-ClC6H4)3Sn[L] Cl and (p-ClC6H4)2Sn[L]Cl2 have been synthesized (where L = L1 and L2). The complexes and ligands were characterized by elemental analysis and spectral (UV-vis, IR and 1H NMR) studies. In all the complexes, ligands act as bidentate, coordination through sulphur and azomethane nitrogen. Complexes are 1:1 metal ligands complexes. Antifungal studies of some complexes against Rhizoctonia bataticola fungal strain have been carried out.

An experimental and theoretical vibrational spectroscopic study of [AsPh(4)](2)[Sn(dmit)(3)] x Me(2)CO.

As shown previously by X-ray structure determinations, [tris(1,3-dithiole-2-thione-4,5-dithiolato)stannate(IV)](2-) salts, [Q](2)[Sn(dmit)(3)], contain isolated cations and dianions. While the tin centres generally having octahedral geometries, the overall shapes of the dianions of these complexes in the solid state can differ with conformations varying from T, Y to asymmetrical arrangements. We now report, as a follow up to our earlier study on the Y-shaped complex, [NEt(4)](2)[Sn(dmit)(3)], an experimental and theoretical study of the vibrational spectra of solid solvated {[AsPh(4)](2)[Sn(dmit)(3)] x Me(2)CO}, in which the dianion has a T-shaped conformation. The infrared and Raman spectra, recorded from 4000 to 150 cm(-1), have been analysed by different ab initio calculations based on restricted Hartree-Fock (RHF) and density functional theory (DFT-Beck3LYP). The calculations were carried out on isolated dianions and cations with the 6-31G and 6-31G(d) basis sets and effective core potentials of Steven, Bash and Krauss (SBK). Fundamentals, overtones and combinations have been assigned. Generally, the Y- and T-shaped dianions exhibit similar infrared/Raman spectra, apart from differences in the C=C and the symmetrical M-S stretching frequencies: such differences can be used diagnostically to distinguish the overall shape of the tris(chelated)metallate dianion.

Vibrational spectra and ab initio analysis of tert-butyl, trimethylsilyl, trimethylgermyl, and trimethylstannyl derivatives of 3,3-dimethylcyclopropene VIII. 3,3-Dimethyl-1,2-bis-(trimethylstannyl)cyclopropene.

The quantum mechanical force fields of 3,3-dimethyl-1,2-bis-(tert-butyl)cyclopropene (I), 3,3-dimethyl-1,2-bis-(trimethylsilyl)cyclopropene (II), 3,3-dimethyl-1,2-bis-(trimethylgermyl)cyclopropene (III), and 3,3-dimethyl-1,2-bis-(trimethylstannyl)cyclopropene (IV) were calculated at the HF/3-21G*//HF/3-21G* level. The scale factors which were optimized previously for the HF/3-21G*//HF/3-21G* quantum mechanical force field of 3,3-dimethyl-1-(trimethylsilyl)cyclopropene were used for correction of the force fields of these molecules. Good agreement between the frequencies calculated from these scaled force fields and the well-analyzed and assigned experimental frequencies of II and III suggests the transferability of these scale factors and the possibility of the spectroscopically accurate prediction of the vibrational spectrum of IV. Some regularities in the changes of the vibrational frequencies were found for this molecular series.

Organotin(IV) complexes of thiohydrazides and thiodiamines: synthesis, spectral and thermal studies.

Organotin(IV) complexes of tribenzyltin(IV) chloride and di(para-chlorobenzyl)tin(IV) dichloride with thiohydrazides have been reported. The ligands synthesized were bidentate coordinating through sulphur and terminal nitrogen atoms. These form 1:1 metal-ligand complexes. The following organotin(IV) complexes have been synthesized: (C(6)H(5)CH(2))(3)Sn(L(1))Cl, (p-ClC(6)H(4)CH(2))(2)Sn(L(1))Cl(2), (C(6)H(5)CH(2))(3)Sn(L(1))Cl, (p-ClC(6)H(4)CH(2))(2)Sn(L(2))Cl(2), (C(6)H(5)CH(2))(3)Sn(L(3))Cl, (p-ClC(6)H(4)CH(2))(2)Sn(L(3))Cl(2), where (L(1)): 2-phenylethyl N-thiohydrazide, (L(2)): N-(2-phenylethyl-N-thio)-1,3-propane diamine, (L(3)): N-(2-phenylethyl-N-thio)-1,2-ethane diamine. The complexes were synthesized by directly mixing, refluxing and stirring the ligands with organotin(IV) chlorides in a suitable solvent. The complexes were found to be pure and were characterized by elemental analysis, electronic, infrared, (1)H and (13)C NMR spectroscopy. These complexes were also studied for their thermal decomposition by thermogravimetry (TG) and differential thermal analysis (DTA). Various kinetic and thermodynamic parameters, viz. activation energy (E(a)), order of reaction (n), apparent activation entropy (S(#)) and heat of reaction (DeltaH) have been determined by using Horowitz-Metzger method. It was observed that these complexes are highly stable and the thermal degradation of these complexes is a spontaneous process. The ligands and their tin complexes have also been screened for their fungitoxicity activity and found to be quite active in this respect.

Differential sensitivity of three cyanobacterial and five green algal species to organotins and pyrethroids pesticides.

In this work, five organotins and pyrethroids pesticides were tested to examine their effects on the three cyanobacteria Anabaena flos-aquae, Microcystis flos-aquae, Mirocystis aeruginosa and on the five green algae Selenastrum capricornutun, Scenedesmus quadricauda, Scenedesmus obliqnus, Chlorella vulgaris, Chlorella pyrenoidosa through 96 h acute toxicity tests. The results indicated that: (1) the decreasing order of the average acute toxicity to cyanobacteria and green algae of five dissimilar organotins and pyrethroids pesticides was: fentin hydroxide > cyhexatin > azocyclotin > fenbutatin oxide > beta-cyfluthrin. (2) Wide variations occurred in response to the tested pesticides among the eight individual species of cyanobacteria and green algae. The sensitivity of various species of algae exposed to fenbutatin-oxide varied over one order of magnitude, exposed to cyhexatin/fentin-hydroxide/beta-cyfluthrin varied over two orders of magnitude and exposed to azocyclotin varied three orders of magnitude. (3) In contrast with the sensitivity of cyanobacteria and green algae, cyanobacteria were much less sensitive to beta-cyfluthrin than green algae. The pollutants may result in a shift of green algal and cyanobacterial group structure, especially in a shift from dominance by green algae to dominance by cyanobacteria, and may sustain cyanobcterial blooms during the special period. Thus, the decreasing order of the aquatic ecological risk was: beta-cyfluthrin > fentin hydroxide > cyhexatin > azocyclotin > fenbutatin oxide. There was a strong variance between toxicity and ecological risk, i.e. "low toxicity" does not automatically imply "low ecological risk". The toxicity of pyrethroids pesticides was lower than that of organotins pesticides, whereas the aquatic ecological risk of pyrethroids pesticides was higher than that of organotins pesticides.

Organotin speciation in Bizerte lagoon (Tunisia).

For the first time, organotins have been assessed in samples collected from Bizerte lagoon, in Tunisia, during two seasons (summer and winter). The organotin distribution was studied in marine sediments and mussels tissues of this lagoon. Butyl-, phenyl- and octyltins were determined using a rapid speciation analytical method based on one-step ethylation/extraction with sodium tetraethylborate in aqueous phase. Simultaneously to the ethylation, the extraction was performed by either liquid/liquid extraction (LLE) or head-space solid phase microextraction (HS-SPME). Gas chromatography with pulsed flame photometric detection (GC-PFPD) was used to perform quantitative determination. The technique has been validated using biological and sediment reference materials. The different samples from Bizerte lagoon were found to be moderately contaminated, especially by butyltins. This pollution was attributed to industrial activities, which are very important in this area. Organotins appeared accumulated in both sediments and mussels, while significant degradations of triorganotins to monosubstituted ones was observed in water.

Toxicity and bioaccumulation of tributyltin and triphenyltin on oysters and rock shells collected from Taiwan maricuture area.

The present study was undertaken to evaluate the toxicity of tributyltin (TBT) on oysters (Crassostrea gigas) and bioaccumulation of TBT and triphenyltin (TPhT) on oysters and rock shells (Thais clavigera) from mariculture areas of Taiwan. When treated with concentrations of 0.08, 0.40, 2.00, 10.00 and 50.00 microg TBT/L, the 48-, 72-, 96- and 120-h LC50s of oysters were 44.6, 18.4, 17.9 and 14.3 microg TBT/L, respectively. In the bioaccumulation experiments, oysters and rock shells were exposed to various concentrations of organotins, i.e. A: control, B: 0.40 microg TBT/L, C: 0.40 microg TPhT/L, and D: 0.20 microg TBT/L + 0.20 microg TPhT/L. In general, TPhT was faster accumulated than TBT in both oysters and rock shells and oysters had a higher elimination capability than rock shells. Additionally, greater bioaccumulation and elimination rates had been observed in female oysters than males. To rock shells, the bioaccumulation rate of organotins in imposex females was greater than males and females.

Fate of organotins in sewage sludge during anaerobic digestion.

Adsorption onto sewage sludge is an important process for the elimination of tributyltin (TBT) from wastewater. However as the disposal of sewage sludge to agricultural land is a significant route for recycling biosolids, there exists an issue as to whether the potential long-term build-up of organotins in agricultural soil is acceptable, from a human health and environmental point of view. For the sustainable use of biosolids in agriculture it is essential to control and reduce the quantities of persistent pollutants such as organotins in sewage sludge. In this study, a sampling program was designed to establish the levels of TBT (and other organotins) in sewage sludge and their reduction during anaerobic treatment and processing prior to disposal. Experiments were also undertaken to assess the fate of TBT in laboratory scale anaerobic digesters where the influence of digester operating parameters could be evaluated. Organotin concentrations were determined using capillary gas chromatography with flame photometric detection. The results demonstrated that the majority of TBT remained concentrated in the solid phase (sewage sludge). Concentrations of TBT in sewage sludge were approximately 18 mg kg(-1) (dry weight) and both laboratory experiments and fieldwork demonstrated that degradation of TBT during anaerobic digestion of sludge was minimal.
