[The vanishing gender?].

The literature data indicating a significant impairment of the basic parameters of male reproductive function in the last decades are analyzed and summarized. This impairment is expressed in the reduction of spermatogenic activity, increased incidence of genital malformations and testicular tumors. The listed changes are reviewed in comparative aspect and include examples of some species of wild animals. The data on the factors responsible for occurrence of pathological changes in male reproductive system are analyzed.

Hormonal contraception in Chinese men: variations in suppression of spermatogenesis with injectable testosterone undecanoate and levonorgestrel implants.

AIM: To explore the causes of the difference in spermatogenic suppression between responders and non-responders in Chinese men treated with levonorgestrel (LNG) implants plus testosterone undecanoate (TU) injectable. METHODS: The 16 Chinese volunteers treated were divided into two groups in regard to the sperm count during the treatment period, 7 men in the responder group (Group R), including 6 azoospermia and one severe oligozoospermia, and the remaining 9 in the non-responder group (Group N), including 4 oligozoospermia and 5 with sperm counts greater than 20 x 10(6)/mL. The differences in serum profiles of FSH, LH, T, LNG and T/LH ratio were compared between the two groups and the correlation between the seminal fluid parameters and serum reproductive hormones was analyzed. RESULTS: The serum FSH level was lower in Group R than that in Group N (P<0.05), while the serum LH and LNG levels were higher in Group R than those in Group N (P<0.05). The sperm density (P<0.01, r=0.235), motility(P<0.01, r=0.326) and vitality (P<0.01, r=0.219) showed significantly positive correlation with the serum FSH level. CONCLUSION: The blood LNG and T levels, the degree of FSH inhibition and/or the sensitivity of the pituitary-testis axis to exogenous steroids, as well as the individual spermatogenetic potential and the functional status of the Leydig cells may be factors bringing about individual differences in spermatogenic suppression in Chinese men treated with LNG and TU.

[Dynamic morbidity of children as a evaluation criterion for renovation of aluminum manufacturing industries].

Negative pollution effects from atmospheric discharges by aluminium facilities exerted on population health can be traced by the parameters of the reproductive function in women, physical development of newborns, general and differential morbidity of children aged below one year as well as by anthropometric signs in birth, morbidity of children and adults, mortality, including due to oncology. The introduction of modern technologies including the preliminarily fire anode treatment and the use of highly effective methods of purification of industrial wastes cut the concentration (in atmospheric air) of anhydrous hydrogen fluoride and of solid fluorides as well as of aluminium to maximum permissible concentration; it also essentially reduced the content of benzapilene. A better atmospheric air observed yet in the course of renovation contributed to a lower morbidity of children, aged below one year, as well as to the prevalence of diseases affecting the eyes, respiratory and digestive organs, skin and subcutaneous cellular tissue; it also cuts the number of congenital anomalies versus the data obtained in a neighboring district.

Seasonal variation in fertility, fecundity and litter sex ratio in laboratory and wild stocks of house mice (Mus domesticus)

Data compilations were made for three parameters pertaining to reproduction in a domestic strain (14 years) and a wild stock (4 years) of commensal house mice: (a) the percentage of females mated that produced litters; (b) the average number of pups per litter; and (c) the sex ratio of the pups in the litters. Fecundity and fertility varied seasonally in both domestic and wild mice. More females become pregnant and litter sizes were larger in the spring, summer, and fall months than during the winter season. Sex ratios also varied seasonally with more male biased litters produced during the 4 winter months. There appear to be seasonal shifts in productivity for both stocks of mice and these seasonal trends have not been altered by domestication under laboratory conditions. In spite of the fact that house mice are generally opportunistic, it is possible that there has been selection in the mice for shifting rates of production in relation to the best seasons of the year in terms of climate and resource availability. The higher production of males during summer months may be geared toward greater success in dispersal at that season and a higher probability that the males can find a territory and mate successfully in summer rather than winter. These results have potential implications for animal breeders and for those who maintain mouse colonies to produce animals for scientific investigations.

[Nature of the estrous cycle, incidence of anovulation, chromosome gametopathies and fetal death in female rats with 2,4-dichlorophenoxyacetic acid (2,4-D) before pregnancy].

The effect of herbicide--2,4-D, injected before pregnancy begins, on the main parameters of the reproductive function of the female rats has demonstrated that chronic administration in doses 1 mg/kg and 12 mg/kg per day for 2 months results in certain disturbances in the estrus cycle, manifesting as prolongation of the diestrus phase and in changes of estrus and meta-estrus++ duration, as well as in an essential increase in rates of anovulatory cycles. Single administration of the preparation during the preovulatory period (50 mg/kg) produces certain disturbances in the chromosomal complex, manifesting as appearance of numerical chromosomal aberrations. When the administration is acute (10 mg/kg and 50 mg/kg) an essential increase of the embryonal death takes place, in the first case--at the expense of ++pre-implantational, and in the second--both at the expense of pre- and ++post-implantational death. At chronic administration increasing rate of the intrauterine death takes place mainly at the expense of ++post-implantational death of embryos.

Loglinear models, sexual behavior and HIV: epidemiological implications of heterosexual transmission.

An analysis is presented of sexual behaviour data from a "random" sample (n = 780) of the adult population of England and Wales. Sex stratified frequency distributions of the reported number of sex partners/time were analysed for heterosexuals, demographic characteristics associated with the number of sex partners/time were identified, and epidemiological parameters (the basic reproductive rate of HIV and the doubling time of the epidemic) were calculated. These analyses suggest that the size of the group at risk for acquiring the virus by heterosexual transmission may be large and that the age of first sexual intercourse (for males and females) is decreasing in younger cohorts. Members of the potential heterosexual at-risk group may be identified by demographic variables such as marital status (males and females) and age (females only), but not by socioeconomic class. The epidemiological implications of our results for the heterosexual transmission of HIV are discussed.

[Hypogonadotropic form of amenorrhea. Principles of ovulation induction].

Analysis of the clinico-anamnestic and endocrine parameters of the reproductive system of 45 patients with hypogonadotropic amenorrhea helped single out three types of this condition. Drug doses and schemes of their administration to induce ovulation were selected individually with due consideration for the initial functional status of the reproductive system. The authors defined the basic principles of ovulation induction in patients with hypogonadotropic amenorrhea: the patients should be carefully selected according to WHO classification, with due regard for their clinico-anamnestic data and the function of the reproductive system (hormonal functional test); drug doses for substitution therapy and protocols of their administration should be selected individually, with consideration for the degree of hypophyseal-gonadal insufficiency; daily double (ultrasonic and hormonal) monitoring is needed for the correction of ovulation induction protocols; the choice of the optimal time of administration of the "ovulatory" dose should be based on the findings of double monitoring indicating follicle size 19-20 mm and the maximal activity of steroidogenesis (350 to 400 pmol/liter estradiol per follicle). The possibility of using lutrelef, an analog of gonadotropin releasing hormone, for ovulation induction in patients with the hypothalamic form of gonadotropic amenorrhea was studied. The drug was administered in a pulsed mode using Zykloma+ device (Ferring, Germany). The advantages of a physiological principle of substitution therapy were demonstrated, although the induction of cycles was not appreciably improved by this method as against substitution therapy with human menopausal gonadotropin.

Models relating the timing of intercourse to the probability of conception and the sex of the baby.

The probability that conception will result from intercourse on particular days of the menstrual cycle is of biological, demographic, and personal interest to many. We describe an existing model for conception as related to the timing of intercourse, and develop a method for fitting it by means of the expectation-maximization (EM) algorithm, using widely available software (GLIM). We then generalize the model to allow for effects on its parameters due to reproductively toxic exposures. A further extension allows one to address the question of whether the pattern of intercourse in a conception cycle has an influence on the sex of the resulting baby. We illustrate the methods by application to data from a prospective study of couples trying to begin a pregnancy.

Semen preparation with the Sperm Select System versus a washing technique.

OBJECTIVE: To compare sperm migration through sodium hyaluronate with simple washing as methods for preparing sperm for IUI. DESIGN: Ten normal semen specimens were prospectively collected and samples were prepared by simple washing and by migration into sodium hyaluronate using the Sperm Select System (Select Medical Systems, Williston, VT). The semen and each treatment group were evaluated for sperm concentration, percent motile, viability, acrosomal status, longevity, and computer-aided semen analysis (CASA) parameters. SETTING: University reproductive endocrinology facility. RESULTS: The recovery of motile sperm was significantly higher for the washing method (mean +/- SEM 75% +/- 7%) than for the hyaluronate method (10% +/- 1%). The number of motile sperm recovered by migration into hyaluronate was independent of the percentage of motile sperm in the semen specimen and positively correlated with sperm concentration. The hyaluronate method produced greater percentages of motile, viable, and morphologically normal sperm, with lower proportions of premature acrosome reactions, higher sperm velocity, and greater linearity. CONCLUSIONS: The Sperm Select System method of sperm separation provides a highly uniform specimen with improved sperm quality. However, the recovery of motile sperm is considerably lower than for simple washing methods.

Long GnRH-agonist protocol in an IVF program. Is it appropriate for women with normal FSH levels and high FSH/LH ratios?

OBJECTIVE: To determine whether subjects with an elevated ratio of follicle-stimulating hormone (FSH) to luteinizing hormone (LH) but normal basal FSH levels should be regarded as poor responders to controlled ovarian hyperstimulation. STUDY DESIGN: One hundred twenty-five women undergoing in vitro fertilization (IVF) for the first time were recruited in this retrospective cohort study. Women over 40 years old or having serum basal FSH > 10 mIU/mL were excluded. RESULTS: Various cutoff values for the FSH/LH ratio were chosen, and the ratio demonstrated that pregnancy rates were apparently higher in patients with the long protocol than with the short one if they had an FSH/LH ratio < 3.0 (48.5% vs. 25.8%, P = .034), < 2.5 (53.3% vs. 28.6%, P = .030) or < 2.0 (57.8% vs. 21.7%, P = .005). Pregnancy rates were similar with the long and short protocols in patients with FSH/LH > or = 3.0 (57.1% vs. 70%, P = .521), FSH/LH > or = 2.5 (40% vs. 53.8%, P = .435) or FSH/LH > or = 2.0 (40% vs. 55.6%, P = .281). CONCLUSION: This study failed to demonstrate that FSH/LH was a useful parameter for predicting reproductive outcome in IVF programs and for patient selection for the long or short gonadotropin-releasing hormone agonist protocol.

Reproductive and neurobehavioural effects in three-generation toxicity study of piperonyl butoxide administered to mice.

Piperonyl butoxide (PB) was administered continuously to mice from 5 weeks of age in the F0 generation to weaning of the F2 generation. PB was administered in the diet at levels of 0 (control), 0.1, 0.2, 0.4 and 0.8%. Selected reproductive, developmental and behavioural parameters were measured. Litter size and litter weight were reduced in higher-dosed groups, and the body weight of the pups in the lactation period was reduced in dosed pups in each generation. The survival index at postnatal day 21 of the group receiving 0.8% PB was reduced in each generation. The developmental and behavioural parameters in the lactation period were little different from those of the controls, apart from olfactory orientation in the F1 generation. However, in the F2 generation mice, surface righting, cliff avoidance and olfactory orientation were adversely affected in treatment groups. The results suggest that PB had adverse effects on reproductive, developmental and behavioural parameters of mice, with increasing effects in subsequent generations of offspring.

Reproductive soundness of captive southern and northern white rhinoceroses (Ceratotherium simum simum, C.s. cottoni): evaluation of male genital tract morphology and semen quality before and after cryopreservation.

White rhinoceroses suffer from a low rate of reproduction in captivity. This study examines the role of male reproductive function as a contributing factor. We used ultrasonography to image accessory sex glands, testis and epididymis. Electroejaculation provided 36 ejaculates from 21 rhinoceroses. Based on the percentage of progressively motile spermatozoa, semen was categorized in three groups, high (I: >75%), intermediate (II: 50-70%) or low (III: <50%) quality. Only 52% of the males showed high semen quality. Ejaculates in the high motility category also had the highest proportion of morphologically intact spermatozoa. Both semen parameters, sperm motility and morphology, were found to positively correlate with size of the accessory sex glands. The semen category was associated with group size suggesting that the social status influenced functional reproductive parameters. Change of territorial status (n = 1) improved semen quality. Testicular fibrosis was characterized as a sign of reproductive ageing in all males older than 15 years of age (n = 13); although, this ageing process did not notably affect semen parameters. Furthermore, for the benefit of assisted-reproduction and genetic banking programs protocols for the storage of cooled semen and the cryopreservation of spermatozoa were designed using different cryodiluents. This report provides basic data for the evaluation of reproductive components and of breeding management in male rhinoceros. Our results indicate that low rate of reproduction in captivity can be attributed to reduced male reproductive fitness. Changes in management of white rhinoceroses may positively affect male reproductive function.

Active immunization against leptin fails to affect reproduction and exerts only marginal effects on glucose metabolism in young female goats.

Approximately 150 days before expected breeding time, 12 female goats (3 months of age) were actively immunized against ovine leptin. Booster injections were given throughout the following year. Control animals (n = 6) were sham-immunized. After the first observed oestrus, a buck was introduced and goats were mated. Blood samples were collected twice weekly and frequent blood sampling series were performed on days -15, 76, 153 and 286 relative to the first immunization. Nine of the immunized goats developed titres within 3 months and had elevated serum concentrations of leptin compared with controls (p < 0.0001). Hematological parameters and blood chemistry were not affected by the immunization. No differences were detectable in all reproductive parameters recorded. Serum insulin was higher in immunized goats during the frequent blood sampling series of day 287 after the first immunization. Glucose metabolism was investigated during pregnancy using hyperglycaemic and euglycaemic/hyperinsulinaemic clamps. None of the parameters derived from the clamp studies was different (p > 0.05) between the two groups. During the hyperglycaemic clamp there was a trend (p < 0.15) towards increased insulin concentrations in immunized animals whereas glucose infusion rates were not different between the groups. This indicates decreased insulin sensitivity in immunized goats. Our study describes the ontogenesis of serum concentrations of leptin during growth, puberty and first pregnancy and parturition for the caprine species. The effects of the immunization were not detectable or only marginal and the approach aimed at therefore not effective to investigate leptin action in detail.

Sperm chromatin structure assay parameters as predictors of failed pregnancy following assisted reproductive techniques.

The predictive value of sperm chromatin integrity for pregnancy outcome following in-vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) was studied in 24 men attending a university-based assisted reproductive techniques laboratory using the flow cytometric sperm chromatin structure assay (SCSA). The SCSA is a measure of the susceptibility of sperm DNA to low pH-induced denaturation in situ. The mean percentage of spermatozoa in the neat sample demonstrating DNA denaturation was significantly lower in the seven men that initiated a pregnancy (15.4 +/- 4.6, P = 0.01) than in the 14 men who did not initiate a pregnancy (31.1 +/- 3.2). No pregnancies resulted if > or =27% of the spermatozoa in the neat semen sample showed DNA denaturation. These data demonstrate that SCSA parameters are independent of conventional semen parameters. Furthermore, the SCSA may allow physicians to identify male patients for whom IVF and ICSI will be unlikely to result in pregnancy initiation.

Fitness parameters and DNA effects are sensitive indicators of copper-induced toxicity in Daphnia magna.

This study compared the effects occurring at molecular and population levels in Daphnia magna exposed to copper concentrations in the range of 15-120 microg/l. The qualitative and quantitative modifications arising in random amplified polymorphic DNA (RAPD) profiles as a measure of DNA effects were compared with a number of key ecological fitness parameters, namely, the age-specific survival, age-specific fecundity, net reproductive rate, and intrinsic rate of population increase. Results suggested that growth, reproduction, and most of the fitness parameters as well as genomic template stability (a qualitative measure reflecting changes in RAPD profiles) were significantly affected at copper concentrations of 90 and 120 microg/l. Among the fitness parameters, the age-specific fecundity and net reproductive rate were the most sensitive parameters of toxicity. Changes in RAPD patterns generally occurred at copper concentrations of 90 and 120 microg/l, but with one primer, changes significantly arose at all copper concentrations. Overall, molecular and population parameters compared well and represented a sensitive means to measure toxicity induced by copper in Daphnia magna. In conclusion, the measurement of parameters at both molecular and population levels is valuable for investigating the specific effects of agents interacting with DNA. Ultimately, this methodology may allow the ecotoxicological examination of the link between molecular alterations and measurable adverse effects at higher levels of biological organization.

Effect of seasonal host reproduction on host-macroparasite dynamics.

The impact of seasonal host reproduction on the population dynamics of host-macroparasite interactions is considered. We modify the classic Anderson and May model so that parameters associated with host reproduction are periodic functions of time with a period corresponding to a year. This allows us to compare our findings with those already well documented. If, in the absence of any seasonality, a stable steady-state solution exists annual reproduction gives rise to stable annual population cycles. Moreover, the parameter domain for which there is stability is increased by the seasonality. However, if the life span of the free-living stages is reasonably long, and the continuous model has limit cycle solutions, complex behavior can be observed in the seasonally forced case. Results also indicate that if seasonal effects are ignored, regulation of the hosts by the parasite population is overestimated.

Epidemiology of male reproductive function: a field searching for tools.

BACKGROUND: An increase in the frequency of disorders of the male human reproductive organs has been described over recent decades. Neither its causes nor its consequences on fecundity, the ability of the couples to produce a live child, are clearly known. This lack of knowledge may partly be due to methodological difficulties specific to reproductive epidemiology. METHODS: We discuss the relevance and limits of some markers of the male aspects of human reproduction, focusing on semen parameters and fecundability, a measure of the probability of pregnancy. RESULTS: Semen parameters are associated with fecundability, although they have a relatively low sensitivity to detect couples with low fecundability. The study of semen parameters proved central to describe the influence of environmental factors on the male side of reproductive function. The main limitation of semen studies is low participation rates and the possible selection biases ensuing. Fecundability can be estimated by collecting waiting time to pregnancy. Its assessment in retrospective studies often excludes the least fecund, those couples remaining childless, which entails a bias and a decrease in statistical power. The prospective approach and an approach relying on the enrollment of a cross-sectional sample of the couples currently trying to obtain a pregnancy (current duration approach) do not have these limitations. Although it has never been used, the assessment of fecundability using the current duration approach is promising both for aetiologic research and monitoring. CONCLUSION: Most of the potential markers of male reproductive function are not assessed in the general population of France, with the exception of the incidence rate of testis cancer, which is currently increasing. We present some alternatives for a monitoring system of reproductive function.

Reproductive characteristics of Ethiopian highland sheep. I. Genetic parameters of testicular measurements in ram lambs and relationship with age at puberty in ewe lambs.

A study was conducted to estimate heritabilities of, and genetic correlations among, body weight (WT) and testicular measurements - scrotal circumference (SC), testicular diameter (TD), testicular length (TL) and epididymal diameter (ED) - in ram lambs between 6, 9 and 12 months old, and relationship of the testicular traits with age at puberty in ewe lambs (AP). Two fat-tailed sheep breeds, the Horro and Menz, indigenous to the Ethiopian highlands were studied. Experimental lambs were produced by mating 250 oestrus-synchronized ewes of each breed to 10 sires in a single-sire mating system over three mating periods which produced, for this study, a total of 361 ram lambs and 148 ewe lambs, with substantial pedigree information, in two dry seasons (October/November 1992 and 1993) and one wet season (June/July 1993). Mean AP was 322.7 days. Breed was not significant (P0.05) for all testicular measurements, except ED at 9 months for which the Horro had a higher (P<0.01) value (20.6, s.e. 0.38 versus 19.3 s.e. 0.30). Horro ewe lambs had their first oestrus 28 days earlier (P<0.05) than the Menz. Heritability estimates were obtained from a sire model (M1) and from an individual animal model with additive direct and maternal effects (M2). The maternal effect was not fitted in M2 for AP. Repeatability and heritability estimates were also obtained by analysing records at the three ages in a repeated measures model (M3). M1 and M2 heritability estimates were generally low to moderate but were associated with large standard errors. Heritability estimates for testicular measurements were generally similar to those for WT. Among the testicular measurements, SC had the highest heritability estimates at 6 (0.45, s.e. 0.24 for M1) and at 12 months (0.41, s.e. 0.21 and 0.41, s.e. 0.20 for M1 and M2, respectively). Model 3 heritability estimates were 0.29 (s.e. 0.14), 0.24 (s.e. 0.12), 0.09 (s.e. 0.07), 0.11 (s.e. 0.08) and 0.12 (s.e. 0.08) for WT, SC, TD, TL and ED, respectively. Only repeatability estimates for WT (0.34, s.e. 0.02) and SC (0.25, s.e. 0.03) were significant. Heritability of AP was estimated at 0.16 (s.e. 0.21) by M1 and 0.14 (s.e. 0.19) by M2. Genetic correlations of AP with testicular measurements, especially with SC (-0.57, s.e. 0.38 and -0.78, s.e. 0.33, at 9 and 12 months, respectively) and TD (-0.72, s.e. 0.33 and -0.83, s.e. 0.48), were desirable and generally high. Adjustment of testicular measurements for liveweight consistently resulted in a reduction in estimates of heritability and genetic correlations. SC was suggested as the most appropriate selection candidate for the genetic improvement of both male and female reproductive performance in these breeds.
