An IgM antibody is a potent immunosuppressive agent that inhibits B cell proliferation.

The results of this study demonstrate that the culture supernatant of lipopolysaccharide (LPS)-stimulated murine spleen cells is able to inhibit the growth of freshly isolated B lymphocytes. The inhibition is specific for B cells because the suppression of the LPS, Fc-fragment of human IgG, dextran sulfate, and anti-mu induced proliferation of B, but not the Concanavalin A response of T lymphocytes could be shown. The cells producing the inhibitor do not adhere to plastic, and are Thy-1 negative but surface Ig positive, i.e. they are B lymphocytes. The regulatory substance is heat resistant, sensitive to trypsin treatment, and has a high molecular weight of approximately 1000 kDa. Moreover, it is specifically adsorbed on, and can be eluted from, anti-mu and anti-Ig immunoaffinity columns. Thus, it seems to be an IgM antibody. Non-specific effects were excluded by the ineffectiveness of poly- and monoclonal IgM proteins. IgM-IgG complexes were also excluded. Thus, these results suggest the existence of a novel IgM antibody mediated control mechanism regulating B cell growth during polyclonal activation.

Effect of the new immunosuppressive agent, cyclosporin-A, on natural killer cell and antibody-dependent cell-mediated cytotoxic activity.

Athymic nude mouse homozygotes (nu/nu) and the heterozygote nu/nu+ were injected intraperitoneally with 2 mg of cyclosporin-A (CS-A) twice, 72 and 1 h prior to analysis. We have found that in vivo administration of CS-A results in a significant suppression of natural killer (NK) cell and antibody-dependent cell-mediated cytotoxic (ADCC) activity which probably play an important role in tumor and graft rejection. T cells were also suppressed, in contrast, B cells were found to be immunocompetent as determined by cell proliferation and antibody-forming capacity in response to mitogens and antigens. The identity of cells mediating NK cell and ADCC activity and its relationship to T lymphocytes are also discussed.

Flow cytometric analysis of lymphocyte populations in guinea pig blood and spleen. Ia antigen expression and the effects of immunosuppressive agents.

A panel of novel mouse monoclonal antibodies was used, in an indirect immunofluorescence procedure, to identify subsets of guinea pig lymphoid cells which were quantified by flow cytometric analysis. The staining characteristics of the antibodies were also examined by alkaline phosphatase-anti-alkaline phosphatase staining of cryostat sections of spleen and cytocentrifuge preparations of blood and spleen mononuclear cells. The values obtained for T and B cell populations were similar to those previously described using rosetting procedures, and the study also confirmed the high incidence of constitutive Ia antigen expression on guinea pig lymphocytes. Compared with animals receiving immunization (ovalbumin) alone, guinea pigs treated with ciclosporin A showed only minor changes in lymphocyte subsets, whereas those pretreated with cyclophosphamide revealed striking reductions in circulating T and B lymphocytes and in splenic B cells. Both ciclosporin A and cyclophosphamide markedly reduced Ia antigen expression on lymphocytes in blood, whilst cyclophosphamide also inhibited Ia antigen expression in the spleen.

Cardiovascular effects of an immunosuppressive agent cyclosporin A.

Cyclosporin A (CsA) is now routinely used for transplantation of all solid organs, bone marrow transplantation, and for an increasing number of immunological diseases. However, treatment with CsA is an important iatrogenic cause of post-transplant hypertension, hyperlipidemia, and diabetes, which may contribute to the high cardiovascular morbidity in transplant recipients. Recently, the calcineurin inhibitor CsA has been employed in vivo and in vitro to examine the role of calcineurin in the signal transduction leading to cardiac hypertrophy. A cell culture study demonstrated the inhibitory effect of CsA on cytokine production by cardiac myxoma cells, the most common primary tumor of the heart. This review discusses recent data on the cardiovascular effects of CsA.

A sensitive and reproducible assay for the quantitation of erythrophagocytosis and its correlation with reduction in tritiated thymidine uptake in a tumor target cell system modified by immunoenhancing or immunosuppressive agents.

A new and sensitive assay procedure for studying erythrophagocytosis is described. The assay technique permits quantitation of the in vivo and in vitro effects of chemicals, hormones, and cell, or microbrial products, on the level of phagocytic activation of glass-adherent cells. The effect of intraperitoneal injection of BCG, Zymosan, Vitamin A, B. pertussis, cortisone, estrone, and thioglycollate on phagocytic activation of peritoneal exudate cells harvested from two days up to 28 days following drug injection was examined by this assay. Erythrophagocytosis was compared to the effect of "activated" spleen cells on tritiated thymidine uptake of a tumor target cell suspension.

[Production of anti-Aeromonas salmonicida agglutinins by the rainbow trout. Influence of temperature, an adjuvant and an immunosuppressive agent].

Adult rainbow trout were immunized intraperitoneally with a chloroform killed Aeromonas salmonicida suspension. Agglutinating antibodies were titrated using a microtiter system. Antibody activity was associated with IgM fraction obtained from Sepharose 4 B gel filtration. The kinetics of antibody formation were established at three temperatures. At 50C detectable antibodies were found at day 50 post immunization. At 10degrees C and 16 degrees C the agglutinin titer rose before day 20. The antibody titer was significantly raised by Freund's complete adjuvant, but cyclophosphamide at a total dose of 500 mg/kg of body weight administered in 10 injections before and after immunization had no effect.
