[Density-gradient centrifugation in lithium metatungstate and tris-(hydroxymethyl)aminomethane phosphotungstate].

New substances--lithium metatungstate (MTL) and tris-(hydroxymethyl)aminomethane phosphotungstate (PTT)--have been presented for density-gradient preparation. The buoyant densities of protein, RNA, DNA and some nucleoproteins were determined in solutions of these salts. Nucleic acids have been smaller buoyant density (1.1 g/cm3) than the proteins in contrast to CsCl-gradients. The protein in PTT solution have buoyant density 1.5 g/cm3 and in MTL solution 2.0-2,3 g/cm3. It was shown that MTL gradients allow to reach better resolution in nucleoprotein analysis than CsCl gradients.

Free energy changes in denaturation of ribonuclease A by mixed denaturants. Effects of combinations of guanidine hydrochloride and one of the denaturants lithium bromide, lithium chloride, and sodium bromide.

The denaturation of ribonuclease A by guanidine hydrochloride, lithium bromide, and lithium chloride and by mixed denaturants consisting of guanidine hydrochloride and one of the denaturants lithium chloride, lithium bromide, and sodium bromide was followed by difference spectral measurements at pH 4.8 and 25 degrees C. Both components of mixed denaturant systems enhance each other's effect in unfolding the protein. The effect of lithium bromide on the midpoint of guanidine hydrochloride denaturation transition is approximately the sum of the effects of the constituent ions. For all the mixed denaturants tested, the dependence of the free energy change on denaturation is linear. The conformational free energy associated with the guanidine hydrochloride denaturation transition in water is 7.5 +/- 0.1 kcal mol-1, and it is unchanged in the presence of low concentrations of lithium bromide, lithium chloride, and sodium bromide which by themselves are not concentrated enough to unfold the protein. The conformational free energy associated with the lithium bromide denaturation transition in water is 11.7 +/- 0.3 kcal mol-1, and it is not affected by the presence of low concentrations of guanidine hydrochloride which by themselves do not disrupt the structure of native ribonuclease A.

Microdosimetry and thermoluminescence.

The mechanism for production of light from a thermoluminescent phosphor requires the combination of a trapped electron and hole at a luminescent center. The kinetics is that of a second-order process. Accordingly one may expect a similarity in the formalism for describing thermoluminescent emission (TL) and the theory of dual radiation action in radiobiology (TDRA). The TDRA was developed to provide a framework for describing the effects of ionizing radiation on biological systems. It requires the interaction of two primary units of radiation damage called "sublesions" to form a "lesion" which may then be expressed as a biological effect. The physical parameters used in the TDRA come from microdosimetry, which deals with the deposition of energy by ionizing radiation in domains whose dimensions are of the order of micrometers. This paper explores the relationships between microdosimetric concepts and a description of TL properties, such as TL dose-response curves and TL LET dependence. This is then compared with similar quantities in biology to determine the possible relevance of TL as a biological model.

[Lithium ion modulation of the neuronal responses evoked by monoaminergic receptor agonists].

The authors studied the action of lithium ions on the responses of rat dorsal root ganglion neurons and frog spinal motoneurons evoked by monoamine agonists using the microelectrode technique. Lithium ions reversibly inhibit the depolarizing responses of spinal sensory neurons and motoneurons evoked by activation of muscarinic choline-, alpha 1-adreno-, and 5-hydroxytryptamine2 receptors, but enhance the hyperpolarizing neuronal responses evoked by activation of 5-hidroxytryptamine1A receptors.

Radiation dosage from use of the image intensifier in orthopaedic surgery.

The image intensifier is now commonly used in orthopaedic surgery for intraoperative assessment of fracture reduction and implant placement, especially with the increasing trend toward use of closed nailing devices. We conducted a study using lithium fluoride chips to measure the radiation dosage to the surgeon and the operating theatre personnel. Over a 6-month period we measured the cumulative radiation dosage over the eyes (0.83mSv), thyroid (with shield 0.51mSv, without 0.79mSv), waist (with apron 0.48mSv, without 0.86mSv), hands (right 0.7mSv, left 0.14mSv) and feet (0.62 mSv). These values were well within the ICRP safety guidelines. The use of protective lead shielding was effective in reducing radiation dosage to operators. A survey of the operating theatre area using a radiation counter showed that radiation scatter to OT personnel was low. This study hopes to allay the fears that use of the image intensifier is hazardous to OT personnel.

Synchrotron radiation in the study of the variation of dose response in thermoluminescence dosimeters with radiation energy.

Thermoluminescence dosimetry (TLD) is a versatile technique with many applications for dosimetry of ionising radiation. However, in the range of kilovoltage x-rays which is widely used for diagnostic and therapeutic medical applications, problems arise from the differing dose response of most TL dosimeters with the radiation energy. The dose response of various TL detector types was investigated in mono-energetic x-ray beams of 26.8, 33.2, 40, 80.4 and 99.6keV from a synchrotron radiation source at the National Laboratory for High Energy Physics in Japan. This response was studied as a function of TL material (LiF:Mg,Ti, LiF:Mg,Cu,P and Al2O3), the detector geometry and size, and their thermal history. Due to the asymmetric diffraction from a Si crystal employed to produce monoenergetic photons there was more than 50% dose inhomogeneity in some of radiation fields used. Therefore, the different TL dosimeter types were rotated around and the results related to the reading of a set of "standard" LiF:Mg,Ti ribbons which were included in all experiments as reference detectors. No significant influence of the detector shape (physical size, thickness) on the dose response with energy could be found. However, the pre-irradiation thermal history influences the dose response with radiation energy: a fast cool down of LiF:Mg,Ti after a high temperature anneal will increase the sensitivity by more than a factor of two. The relatively new TLD material LiF:Mg,Cu,P (GR-200, obtained from Solid Dosimeter & Detector Laboratories, Beijing) was found to be approximately 100 times more sensitive than the standard LiF:Mg,Ti. In addition it proved to be more tissue equivalent for photon radiation between 27keV and 40keV. The performance of LiF:Mg,Cu,P makes it a very interesting TL material deserving further evaluation for applications in diagnostic and therapeutic x-rays.

[Serum digitoxin in concomitant use of antiepileptics in routine therapy].

Concomitant use of digitoxin and enzyme-inducing antiepileptics may lower serum levels, and accordingly the effect of digitoxin, unless the higher metabolic clearance is compensated for by higher dosage. Use of digitoxin is almost always guided by serum concentration measurements. Information on a possible enzyme-inducing effect of phenobarbital, phenytoin and carbamazepine is easily accessible. Compilation of serum level measurements for digitoxin showed that serum levels shifted towards lower values during concomitant use of phenytoin or carbamazepine than when digitoxin was used alone. As a consequence, the fraction of patients with serum levels below the therapeutic range was doubled. Concomitant use of phenobarbital did not cause a shift in the levels of digitoxin. In fact, in this group, a larger fraction of the serum level measurements were within the therapeutic range. Thus, the dosage of digitoxin appears to be fully compensated during concomitant use of phenobarbital, but obviously deserves attention during concomitant use of phenytoin or carbamazepine.

Changes in the redox potential and catalase activity of Mn2+ ions during formation of Mn-bicarbonate complexes.

Changes in the redox potentials of Mn2+ ions and Mn-bicarbonate complexes were studied due to their possible participation in the photosynthetic oxidation of water in plant photosystem 2 and in H2O2 decomposition. Electrochemical oxidation of Mn2+ ions was demonstrated by voltammetry on a platinum electrode in 0.1 M LiClO4 solution at a potential of 1.19 V (vs NHE). When NaHCO3 was added, the oxidation peak of Mn2+ ions disappeared. New oxidation peaks appeared at 0.92 V and 0.63 V which corresponded to the oxidation of Mn(HCO3)+ and Mn(HCO3)2, respectively, generated by complex formation between Mn2+ and HCO3- ions. The effect of the Mn(2+)-bicarbonate complex on H2O2 redox decomposition was studied. It was shown that the addition of MnSO4 to the H2O2 solution did not affect the H2O2 oxidation peak height thereby indicating the absence of H2O2 decomposition by Mn2+ ions. At the same time, subsequent addition of NaHCO3 resulted in the disappearance of the oxidation peaks of both H2O2 and Mn2+. As at pH 7 the thermodynamic potential of H2O2 reduction is 1.1 V and the Mn2+ oxidation potential is 1.19 V, the redox reaction between them is hampered. Formation of the Mn(2+)-bicarbonate complex shifted the Mn2+ oxidation potential to 0.63 V, thereby inducing the decomposition of H2O2. It is suggested that the decrease in the Mn2+ oxidation potential resulting from the formation of the bicarbonate complex determines the bicarbonate capability to enhance the Mn2+ ability to donate electrons for PS 2 reaction centres.

[Synthesis of triterpene 2,6-dideoxy-alpha-L-arabino-hexopyranosides from L-rhamnal. Their pharmacological properties].

Triterpene 2,6-dideoxy-alpha-L-arabino-hexopyranosides were synthesized by the glycosylation of oleanane triterpene alcohols with L-rhamnal acetate in the presence of cationite KU-2-8 and lithium bromide. 2,6-Dideoxy-alpha-L-arabino-hexopyranosides of allobetulin and methyl glycyrrhetinate showed pronounced antiulcerous activity; the latter also stimulated reparative skin regeneration in rats and was effective as a hepatoprotectant.

Nonerasing dose readout in lithium fluoride crystals by positron lifetime spectroscopy.

The measurement of the mean lifetime of the positron (tau) in a lithium fluoride (LiF) chip which had been irradiated with up to 40 Gy from a 60Co gamma-ray source demonstrates a correlation between the positron mean lifetime and the dose which has been delivered to the crystal. This result points out the possibility of performing the readout of gamma-irradiated LiF thermoluminescent dosimeters by positron lifetime spectroscopy without changing the information stored in the chip significantly.
