Microelectrode studies of facilitated O2 transport across hemoglobin and myoglobin layers.

Experimentally measured PO2 profiles across layers of hemoglobin and myoglobin solutions were compared with profiles predicted from facilitated transport theory assuming chemical equilibrium. Measurements across myoglobin layers were in excellent agreement with theory, but measurements across hemoglobin layers departed from theory at low PO2. This departure was greatest for salt-free hemoglobin solution, which may be caused by an electrical potential formed by a pH gradient in the layer as oxyhemoglobin is deoxygenated.

Microelectrode depth study of the electroretinographic oscillatory potentials in the frog retina.

The depth profile of the electroretinographic oscillatory potentials was studied in the isolated frog retina. The intraretinal electrode was introduced from the receptor side, and the reference electrode was placed on the vitreal side. The electroretinogram, recorded either transretinally or with the electrode tip in the receptor layer, showed 4 to 10 oscillatory potential wavelets. As the electrode was advanced proximally, the wavelets disappeared as a function of retinal depth. The wavelets with longer peak latencies disappeared earlier, and only the first one or two wavelets could be identified when the electrode was in the inner plexiform layer. These findings indicate that the oscillatory potentials are generated between the inner and outer plexiform layers and that the earlier wavelets originate in the more proximal retina. The results are consistent with the notion that the oscillatory potentials are generated by synaptic feedback circuits.

Simultaneous recording with 30 microelectrodes in monkey visual cortex.

A 30-fold multielectrode for extracellular recording of neuronal spikes is described. Single neuronal spikes were isolated simultaneously by about half of the electrodes. The technique has been applied to demonstrate the spatial distribution of ocular dominance and orientation preference in striate cortex.

Evidence for an acid pH in rat renal inner medulla: paired measurements with liquid ion-exchange microelectrodes on collecting ducts and vasa recta.

We reevaluated the pH in the renal medulla in rats. pH of the vasa recta blood was about 1 pH unit acidic in comparison to the pH of renal artery blood. During furosemide-induced diuresis pH of vasa recta blood increased whereas pH of collecting duct urine further decreased. The acidic pH in the rat renal inner medulla during antidiuresis raises important questions about the source of H+ in inner medulla.
