The effect of long-term penfluridol treatment on the sensitivity of the dopamine receptors in the nucleus accumbens and in the corpus striatum.

The effect of local application of dopamine to the nucleus accumbens or corpus striatum on locomotor activity was studied in rats 4 days after withdrawal from a 6 weeks term of penfluridol medication. The bilateral application of dopamine into the nucleus accumbens of penfluridol-treated rats produced a very marked increase in coordinated locomotor activity which was 3-5 times higher than that of rats not treated with penfluridol. This effect of dopamine in both penfluridol-treated and control rats was antagonized by intraperitoneally administered haloperidol. The bilateral application of dopamine into the corpus striatum of penfluridol-treated animals produced a marked stereotyped behavioural syndrome in all rats studied, whereas no signs of stereotyped behaviour were observed in any of the rats not treated with penfluridol. The results indicate that long-term treatment of rats with the dopamine receptor blocking agent penfluridol produces an increase in the sensitivity of the dopamine receptors in the nucleus accumbens and corpus striatum and that the nucleus accumbens may play a role in locomotor activity.

Histochemical characterization of the monoamine-containing cells of the adenohypophysis in the Chinese quail.

The demonstration of biogenic amine-containing cell is classically performed by means of the formaldehyde-induced fluorescence technique after administration of the monoamine precursors, L-dopa or 5-hydroxytryptophan (5-HTP. The injection of these two two compounds rapidly leads to the formation of large amounts. The biochemical pathway evoked above is studied in the avian adenohypophysis by two different methods involving enzyme...

Histofluorescence study on monoamine entry into the brain before and after opening of the blood-brain barrier by various mechanisms.

The relationship between exogenous, circulating monoamines to the wall of cerebral microvessels, and the entrance of these amines into the cerebral parenchyma was studied by the formaldehyde histofluorescence technique in rats. No monoamine fluorescence could be detected in the wall tissue of the microvessels (pericytes and andothelial cells) unless either MAO or COMT were inhibited; these are integral to the blood-brain barrier mechanisms to monoamines. After transient opening of the morphologic blood-brain barrier by either a hypertonic of hypertensive insult, the amine fluorescence in the walls of the microvessels was intensified compared to that which was noted after monoamine oxidase inhibition by itself. Following opening of the structural blood-brain barrier, the circulating amines also passed through into the neuropil where they were concentrated within neurons, as demonstrated by prior depletion of endogenous monoamine transmitters by reserpine. Thus, both enzymatic and morphologic mechanisms in the blood-brain barrier ar involved in impeding the passage of monoamines into the cerebral parenchyma.

Some cytochemical and cytological features of the so-called SIF cells of the superior cervical ganglion of the rat.

The properties of uptake and storage of dopamine and noradrenaline of the SIF cells of the superior cervical ganglion of the rat were tested, using the radioautographic method. Their ability to store exogenous catecholamines appears very poor under the present experimental conditions, in spite of the great number of storage vesicles they usually contain. This situation may be related either to the absence of a high affinity uptake mechanism in the SIF cell membrane or to a normal saturation of SIF cells in catecholamine. Cytological peculiarities of nerve terminals on SIF cells were pointed out. Dense patches attached to the inner face of the SIF cell membrane suggest a local release of vesicular contents, with further functional implications.
