The respiratory pigments of rabbit polymorphonuclear leukocytes (PMN) and alveolar macrophages (A-MO) were compared in a spectrophotometric study conducted at liquid nitrogen temperature. In the difference spectra of the KCN-treated minus the untreated states and of the reduced minus the oxidized states, the cellular and granular fractions of PMN showed sharp absorption maxima identical with the absorption bands of myeloperoxidase (MPO). Even though A-MO revealed some guaiacol-peroxidation activity, the difference spectra of cellular and subcellular A-MO fractions did not show an MPO spectrum. The enzyme catalyzing the guaiacol peroxidation by A-MO appears to be an enzyme other than the MPO found in PMN. Despite the absence of the MPO spectrum, the difference spectra of A-MO granules when measured with KCN, dithionite, and succinate showed characteristic absorption bands of mitochondrial cytochromes, in contrast to the difference spectra of PMN, which showed far smaller cytochrome absorption bands. The results obtained suggest that the phagocytic metabolism of rabbit A-MO depends on mitochondrial respiration for its energy supply.
Vitamin E concentrations were determined by high-performance liquid chromatography in different anatomical regions of the brain from 3-month-old Fischer 344 rats. Gray matter from cerebellum and cervical spinal cord contained the lowest concentrations, while gray matter from the frontal cortex and thalamus had the highest concentrations of vitamin E. Radioactive alpha-tocopherol injected intravenously into the rat was readily taken up by brain although the level of uptake was very low compared with the liver. The ratios of brain-to-serum radioactivities ranged from 0.011 to 0.016 depending upon the brain region. Cerebellar gray matter is characterized by a low concentration of unlabeled alpha-tocopherol and a high level of uptake of radioactive alpha-tocopherol and thus is particularly active in the metabolism of vitamin E. Concentrations of unlabeled alpha-tocopherol were highest in microsomal and mitochondrial fractions and were the lowest in cytosol and nuclear fractions.
Myelin and a heavy membrane fraction (1.0/1.2 fraction) were isolated from rabbit white matter by a slight modification of the procedure for bovine CNS. The specific activities of acetylcholinesterase and Na+, K+-ATPase were higher in the 1.0/1.2 fraction than in myelin. In contrast, the cerebroside content and 2'3'-cyclic nucleotide 3'-phosphohydrolase activity in the 1.0/1.2 fraction were 4.5 and 3.4 times lower than in myelin. Total lipids accounted for only 30% of the 1.0/1.2 fracton's dry weight; for myelin, they represented 70%. Polacrylamide gel electrophoresis showed the presence of many high molecular weight proteins and glycoproteins in the 1.0/1.2 fraction but myelin components were practically missing. Cytochrome c oxidase and NADPH-cytochrome c reductase activities suggested about 15% contamination in the 1.0/1.2 fraction but less than 5% for myelin. In electron micrographs of the 1.0/1.2 fraction, there were many membraneous profiles that varied in size, some mitochondrial fragments, and only a few lamellar whorls of compact myelin. The results suggest that the 1.0/1.2 fraction is different from other myelin-related fractions and is probably enriched in axolemma.
