Cloning and characterization of a ribosomal P protein from Taenia solium, the aetiological agent of human cysticercosis.

A cDNA encoding the complete open reading frame of the Taenia solium acidic ribosomal phosphoprotein P2 has been cloned. The 417 bp cDNA (arpP2) was isolated from a T. solium cDNA expression library by PCR using P protein specific pimers. The ORF encodes a protein of 121 amino acids exhibiting 40-55% identity to mammalian, fungal, insect, and parasite P2 proteins. The inferred molecular mass of the protein is 12,592 Daltons, similar to that reported for other ribosomal P2 proteins. After subcloning and expression, the recombinant protein was purified by affinity chromatography under non-denaturing conditions and was shown to have a molecular mass of 15 kDa which is equivalent to the expected size of the full length recombinant fusion protein, comprising the 12.5 kDa arpP2 plus an additional 2 kDa for the N-terminal fusion peptide incorporating the six histidine residues required for purification.

[The efficacy of praziquantel against experimental cysticercosis and hydatidosis (author's transl)].

Praziquantel is active against cysticerci in experimentally infected mice, rabbits, sheep and cattle. After single or repeated application the infectious stages of Hymenolepis nana, Cysticercus fasciolaris, C. pisiformis, C. tennuicollis and C. bovis were killed. The compound is more active against older than against younger stages as demonstrated with H. nana-cystecercoids and C. fasciolaris in mice. Against the tetrahyridia of Mesocestoides corti in mice praziquantel is only effective after an intraperitoneal application of an oily suspension. Praziquantel shows also activity in rodents against the infectious protoscolices of Echinococcus multilocularis but failed to inhibit the growth of the hydatid cysts.

Praziquantel treatment of muscle Taenia solium cysticercosis. 4. Reversible in vitro effect.

The in vitro effect of Praziquantel (PZQ) on Taenia solium cysticerci was analyzed. The oxygen consumption rate of the parasites was inhibited and the release of proteins was enhanced, but no statistically significant differences were found between the control group and the experimental groups. The drug had a significant, dose-dependent negative effect on the evagination ability of the larvae; 50% effect was seen at concentrations of between 10(-9) and 10(-8) M PZQ. The drug also induced morphological disturbances in the tegument of the worm and of the bladder wall. Finally, a very drastic effect was the induction of spastic paralysis in evaginated cysticerci at high drug doses and of flaccid paralysis at lower PZQ concentrations. The dose inducing these effects was various orders of magnitude lower than that inhibiting the evagination ability of intact cysts. Moreover, the latter effect was reversible after had been washed out the drug and the parasites had been cultured. We suggest that PZQ alters the Ca2+ flux in T. solium as it does in other helminths. Furthermore, we corroborated the protective role of the bladder for the invaginated worm. Finally, we think that in vivo the drug must act synergistically with the immune system components so as to eliminate the parasite.

Cerebral cysticercosis in a European patient: problems of disease activity and therapeutic implications.

The case of a German patient who was treated with two courses of praziquantel for parenchymal cerebral cysticerosis (CC) is reported. Efficacy of the treatment could not reliably be distinguished from the natural course of the disease. After a brief review of the literature concerning problems with assessment of the activity of CC, the criteria recommending praziquantel therapy are discussed.
