GABAergic excitatory synapses and electrical coupling sustain prolonged discharges in the prey capture neural network of Clione limacina.

Afterdischarges represent a prominent characteristic of the neural network that controls prey capture reactions in the carnivorous mollusc Clione limacina. Their main functional implication is transformation of a brief sensory input from a prey into a lasting prey capture response. The present study, which focuses on the neuronal mechanisms of afterdischarges, demonstrates that a single pair of interneurons [cerebral A interneuron (Cr-Aint)] is responsible for afterdischarge generation in the network. Cr-Aint neurons are electrically coupled to all other neurons in the network and produce slow excitatory synaptic inputs to them. This excitatory transmission is found to be GABAergic, which is demonstrated by the use of GABA antagonists, uptake inhibitors, and double-labeling experiments showing that Cr-Aint neurons are GABA-immunoreactive. The Cr-Aint neurons organize three different pathways in the prey capture network, which provide positive feedback necessary for sustaining prolonged spike activity. The first pathway includes electrical coupling and slow chemical transmission from the Cr-Aint neurons to all other neurons in the network. The second feedback is based on excitatory reciprocal connections between contralateral interneurons. Recurrent excitation via the contralateral cell can sustain prolonged interneuron firing, which then drives the activity of all other cells in the network. The third positive feedback is represented by prominent afterdepolarizing potentials after individual spikes in the Cr-Aint neurons. Afterdepolarizations apparently represent recurrent GABAergic excitatory inputs. It is suggested here that these afterdepolarizing potentials are produced by GABAergic excitatory autapses.

Improving charge capture to increase net revenue.

Although a decreasing percentage of hospital revenue continues to be based on total charges, providers can still significantly increase net revenue through a process of evaluating and improving current charge structures, charge capture procedures, and service delivery methods. After reviewing various reasons why hospitals fail to capture charges adequately, the authors present a systematic methodology for reviewing all revenue producing departments to identify opportunities for successfully addressing charge capture problems. This methodology, known as the clinical pricing approach, can improve the accuracy of internal cost accounting and have a positive net revenue impact of from one percent to three percent of gross revenue.

Peripheral blood lymphocyte immunocompetence in wild African green monkeys (Cercopithecus aethiops) and the effects of capture and confinement.

Capture and prolonged confinement of wild African green monkeys (AGM, C. aethiops) in single housing have been shown to result in high morbidity and mortality. The present study was designed to analyse immune modulation associated with these procedures in AGMs. Four wild resting AGMs were stunned with a rifle shot as controls (group I). Seven newly captured monkeys (group II) and seven laboratory conditioned monkeys (group III) housed singly were anaesthetised with ketamine hydrochloride on day 18 and 45, and 180 and 210 post-capture, respectively. Heparinized blood (10 ml) was drawn from each animal and lymphocytes were cultured in six wells per sample. A triplicate subset was stimulated with 10 microliters Concanavalin A for 48 hours. The cells were pulsed with tritiated thymidine and harvested on filter discs 18 hours later. Lymphocyte stimulation indices (stimulated cell count-unstimulated cell count) were 3598 in group I. 3843 and 4395 in group II and 2809 and 2196 in group III, respectively. This indicates that AGMs confined to single housing for a prolonged period exhibit immune suppression. The results confirm our previous findings that the AGM is more susceptible to stress associated with immune suppression and subsequent high frequency of infectious diseases than other East African nonhuman primates confined to laboratory housing.

Conditions under which immobilized platelets activate as well as capture flowing neutrophils.

Immobilized activated platelets present P-selectin and efficiently capture flowing neutrophils. We investigated how the treatment of the platelets affected whether adherent neutrophils rolled continuously or became immobilized. Washed platelets were maintained in a 'resting' state by Ca++ chelation, prostacyclin and theophylline. When these platelets were adhered to glass that had been coated with aminopropyltriethoxysilane (APES) they retained discoidal morphology. Compared to a confluent surface of spread platelets prepared by allowing heparinized platelet-rich plasma to settle on APES-glass, 'resting' platelets captured far fewer flowing neutrophils, which rolled rapidly. However, if neutrophils were perfused along with thrombin (>/= 0.2 U/ml), then the resting platelets rapidly changed shape, neutrophil binding increased markedly, rolling velocity decreased, and 40-70% of the neutrophils were immobilized via beta2-integrins. Similar effects could be induced using ADP perfused with the neutrophils. Thrombin did not itself activate neutrophils, and stationary adhesion could also be induced if platelets were treated with thrombin before addition of neutrophils. After thrombin treatment of platelets, rolling adhesion was only fully re-established after a prolonged period of washout. Thus, platelets presented a stable surface-bound agent able to activate neutrophils. Blockade of platelet-activating factor receptor, leukotriene B4 receptor, or CXC-chemokine receptor 1 (CXCR1) on neutrophils did not inhibit conversion from rolling to stationary adhesion, but blockade of CXCR2 maintained a higher proportion of rolling adhesion. Thus, platelets attached to damaged vessels may capture flowing neutrophils, but the stability of neutrophil deposition will depend on the scale of the local generation of platelet agonists such as thrombin and ADP.

Colorimetric capture assay for human-immunodeficiency-virus-I reverse transcriptase activity.

The development of a colorimetric capture assay for HIV-1 reverse transcriptase (RT) activity is described. This assay consisted of three basic steps: enzyme purification, RT reaction and product detection, which were all performed in the same microtitre plate. Mouse monoclonal anti-RT antibodies of subclass G2a were bound by polyclonal goat anti-(mouse IgG2a) immobilized in the wells of a microtitre plate. The monoclonal antibodies (mAbs) were selected for their ability to bind HIV-1 RT without hampering the polymerase activity. The assay system first involved the RT's adherence to the immobilized mAbs. Non-specific enzymes and other impurities were removed by a simple wash, after which an RT reaction mixture containing BrdUTP as nucleotide substrate was added. After the RT reaction substrate and product had been separated by washing of the plate, the amount of BrdUMP-DNA in the wells was finally detected with alkaline-phosphatase-conjugated mouse anti-BrdU antibodies of subclass IgG1. The background signal in this system was similar to the signals obtained with control wells coated with BSA only. A detection limit of 1.2 micro-units of RT activity, corresponding to 0.3 pg of RT protein, was obtained for the capture assay when applying colorimetric product detection. The assay detected RTs from HIV-1 subtypes A and B and one of the two D type isolates tested. None of the five non-HIV-1 RTs tested was found positive. At least 50 microl of human serum or plasma per sample could be included in the capture assay without adverse effects on the recovery of the RT activity.

Formaldehyde generators and capturers as influencing factors of mitotic and apoptotic processes.

There is a growing amount of evidence pointing to the fact that several endogenous and exogenous methylated compounds are potential formaldehyde generators in their biological reactions. N(G)-methylated lysines, N(G)-methylated as well as hydroxymethylated arginines, and 1'-methyl-ascorbigen have been examined in this respect. The apoptosis-inducing effect of formaldehyde molecules formed from methyl groups was earlier first published by our group. Dimedone, an artificial capturer molecule for formaldehyde, has been found to prevent the apoptosis-inducing effect of 1'-methyl-ascorbigen as well as N(G)-hydroxymethylated arginines. More recently resveratrol, present in grapes and wines, has been shown to have cardioprotective and cancer chemopreventive effect. Our group has been successful in demonstrating that this natural formaldehyde capturer molecule can also influence cell proliferation and apoptosis. The apoptosis-inducing or -preventing effect of formaldehyde generators and capturers seems to be dose-dependent and may be utilized in various disturbances of cell proliferation and active cell death.

Detection of antibody to turkey coronavirus by antibody-capture enzyme-linked immunosorbent assay utilizing infectious bronchitis virus antigen.

An antibody-capture enzyme-linked immunosorbent assay (ELISA) for detection of antibody to turkey coronavirus (TCV) utilizing infectious bronchitis virus (IBV) antigen was developed. Anti-TCV hyperimmune turkey serum and normal turkey serum were used as positive or negative control serum for optimization of the ELISA system. Goat anti-turkey immunoglobulin G (light plus heavy chains) conjugated with horseradish peroxidase was used as detector antibody. The performance of the ELISA system was evaluated with 45 normal turkey sera and 325 turkey sera from the field and the cutoff point was determined. Serum samples of turkeys experimentally infected with TCV collected sequentially from 1 to 63 days postinfection were applied to the established antibody-capture ELISA using IBV antigens. The optimum conditions for differentiation between anti-TCV hyperimmune serum and normal turkey serum were serum dilution at 1:40 and conjugate dilution at 1:1600. Of the 325 sera from the field, 175 were positive for TCV by immunofluorescent antibody (IFA) assay. The sensitivity and specificity of the ELISA relative to IFA test were 93.1% and 96.7%, respectively, based on the results of serum samples from the field turkey flocks using the optimum cutoff point of 0.18 as determined by the logistic regression method. The ELISA values of all 45 normal turkey sera were completely separated from that of IFA-positive sera. The ELISA results of serum samples collected from turkeys experimentally infected with TCV were comparable to that of the IFA assay. Reactivity of anti-rotavirus, anti-reovirus, anti-adenovirus, or anti-enterovirus antibodies with the IBV antigens coated in the commercially available ELISA plates coated with IBV antigens could be utilized for detection of antibodies to TCV in antibody-capture ELISA.

[Spatial distribution of sharks captures in the Pacific of Nicaragua and its relationship with several oceanographic variables].

Between August 1995 and August 1997 long line fishing techniques and a bathythermograph were used to correlate some physical variables with the spatial distribution of four shark species in 26 fishing cruises off the Nicaraguan Pacific Coast. They were the thresher (Alopias vulpinus), blue (Prionace glaucea), gray (Carcharhinus falciformis) and hammer (Sphyra lewini). All species concentrated in the southeastern Nicaraguan Pacific, at the seasonal upwelling area of Papagayo Gulf. The range of sea surface temperatures in which the sharks were captured was 25-28 degrees C. We could clearly associate this physical parameter with shark availability. The vertical distribution of the captured sharks suggests that they occupy termocline levels above the 15 degrees C isotherm. Although these species are oceanic, the blue shark was captured in ocean waters over the 1800 m isobar, while the grey and thresher sharks where close to the continental shelf. Body length in decreasing order are: thresher (210-290 cm, SN = 21), blue (60-240 cm, SN = 13) and gray (80-200 cm, SN = 17).

Duplex DNA capture.

This article describes the sequence-specific isolation and purification of intact double-stranded DNA (dsDNA) by oligonucleotide/PNA-assisted affinity capture (OPAC). The OPAC assay is based on selective tagging of a DNA duplex by biotinylated oligodeoxyribonucleotide (ODN) through formation of a so-called PD-loop. The PD-loop is assembled with the aid of a pair of PNA "openers", which allow sequence-specific targeting with a Watson-Crick complementary ODN probe in the exposed region of the dsDNA. The protocol involves three steps. First, two cationic bis-PNAs locally pry the DNA duplex apart at a predetermined site. Then, the exposed DNA single strand is targeted by a complementary biotinylated ODN to selectively form a stable PD-loop complex. Finally, the capture of dsDNA is performed using streptavidin covered magnetic beads. The OPAC procedure has many advantages in the isolation of highly purified native DNA over other affinity capture and amplification techniques.

A cardiac evoked response algorithm providing automatic threshold tracking for continuous capture verification: a single-center prospective study.

BACKGROUND: The AutoCapture algorithm as implemented in Regency and Microny pacemakers (Pacesetter Inc., Sylmar, CA, USA) provides beat-by-beat monitoring of capture based on proper detection of the evoked response, provides high output back-up pulse when loss of capture occurs, performs periodic threshold evaluations and acquires the capture threshold data in a time-based event counter for later retrieval. The safety and efficacy of this algorithm was prospectively evaluated at a tertiary care hospital of north India. METHODS AND RESULTS: Fifty-four patients (38 males, mean age 66+/-13 years) received a ventricular pacemaker model Regency SC+ with low polarization bipolar lead for high-grade atrioventricular block (n=42) and sick sinus syndrome (n=12). Evoked response and polarization signal were assessed initially at 24 hours postimplant, and follow-up measurements were systematically conducted at week 1 and months 1, 3 and 6. Further evaluation of eligible patients was performed at 6-monthly intervals. Lead implantation parameters were optimum in all patients. At 6 months, the algorithm was functional in 51 patients. The pacing threshold increased to 0.89+/-0.36 V (p<0.001) in the first month and stabilized thereafter. Significant saving of energy was accomplished by a constant output safety margin of 0.3 V instead of the traditional 100%. While the evoked response signal remained stable throughout the study period, the potential signal increased significantly from 0.6+/-0.7 mV to 1.0+/-0.6 mV (p<0.001) in the first month and remained steady subsequently. Back-up pacing in the event of exit block was confirmed in all 25 patients who underwent a 24-hour Holter test. Based on the suggested sense margins, ventricular undersensing was observed in 7 (28%) patients, the majority of whom had competitive cardiac rhythms. An elderly patient with pneumonic illness succumbed to pulmonary embolism at 6 months. CONCLUSIONS: This large single-center experience on AutoCapture demonstrates the success of this algorithm in low-energy ventricular pacing without compromising the patient's safety.

Automated charge capture at the point of care increases revenue.

Group practices can reduce lost charges, denied claims, and time-to-billing rates by using automated charge capture at the point of service. One orthopedic department had been experiencing many process-related problems associated with physician charge capture. To improve revenues, the physicians began using hand-held devices to input data during patients' office visits. A 12-month analysis of charge data from the orthopedic physicians' outpatient visits using a paper-based charge process showed that 824 outpatient charges had been missed or lost, representing 6 percent of the department's annual outpatient activity and more than $54,000 in projected additional revenue. Another analysis conducted after the automated charge-capture process at the point of care was implemented showed that the orthopedic department had not missed any charges.

Bedside vital signs capture for the non-ICU setting--an open source, PC-based solution.

The efficient and reliable capture of vital signs and other bedside data in the non-ICU setting has been a challenging problem for the medical informatics community. The problem is compounded by the complexities associated with storage of this data into an electronic medical record system (EMRS). There are a lack of off-the-shelf solutions that satisfy the basic system requirements of bedside data capture, user authentication, data validation prior to storage, error handling, and convenience. With the current state of technology available, we feel the solution to this problem requires the presence of a PC with custom interface software at the bedside. This allows for the successful interface between available vital signs capture devices, existing EMRS s, and the user. This report summarizes the alternatives we found and our proposed solution to this important problem.

Loss of heterozygosity in primary lung cancer using laser capture microdissection and WAVE DNA fragment analysis techniques.

BACKGROUND: A number of molecular changes observed by varied conventional methods, including loss of heterozygosity (LOH) on chromosome 3, have been associated with primary lung cancer. To further define the locus of chromosome 3p allele loss in lung cancer, we performed LOH study by using innovative laser capture microdissection and WAVE DNA Fragment Analysis. MATERIAL/METHODS: Thirty-eight paired specimens from patients with adenocarcinoma of the lung were used for this study. Formalin-fixed, paraffin-embedded tissue from normal stromal cells or lymphocytes and adenocarcinoma were collected using laser capture microdissection. DNA was extracted and amplified by PCR using six polymorphic DNA markers for chromosome 3. PCR products were analyzed by both gel electrophoresis and WAVE DNA Fragment Analysis. RESULTS: LOH at 3p22-24 was found in tumor cells from twelve out of thirty-eight patients (32%) when analyzed by WAVE DNA Fragment Analysis and LOH was found in tumor cells from nine out of thirty-eight patients (23%) when analyzed by gel electrophoresis. LOH was found in normal control from one out of thirty-eight patients. CONCLUSIONS: 1. Our results suggest putative tumor suppressor gene(s) is present in a region at 3p22-24, which may play a role in carcinogenesis of lung cancer. 2. Laser capture microdissection is essential tool for defined LOH studies. 3. WAVE DNA Fragment Analysis is an accurate, sensitive and automated tool for analysis of DNA fragments.

Electron microscopic study on the response of the normal canine brain to boron-neutron capture therapy.

The response of normal cerebral tissue of dogs to boron-neutron capture therapy by the currently available improved method was studied by electron microscopy. Peripheral blood capillaries of the neutron-irradiated area in the left cerebral hemisphere were compared with their counterparts in the shielded and non-irradiated right hemisphere. No ultrastructural changes, as those noted in classical method of boron-neutron irradiation by the improved technique of boron-neutron capture therapy. There was no swelling of endothelial cells, disappearance of cristae of mitochondria, increased pinocytosis, disappearance of ribosomes, enlargement of Golgi apparatus, or increased appearance of endoplasmic reticulum. Basement membrane was not disrupted and was uniform. Pericytes, synaptosomes, and other glial elements remained intact. In contrast to the old clinical trials up to 1961, the renewed boron-neutron capture therapy is regarded not to cause serious damage to the central nervous system.

Using the two-source capture-recapture method to estimate the incidence of acute flaccid paralysis in Victoria, Australia.

OBJECTIVE: To estimate the incidence and the completeness of ascertainment of acute flaccid paralysis (AFP) in Victoria, Australia, in 1998-2000 and to determine its common causes among children aged under 15 years. METHODS: : The two-source capture-recapture method was used to estimate the incidence of cases of AFP and to evaluate case ascertainment in the routine surveillance system. The primary and secondary data sources were notifications from this system and inpatient hospital records, respectively. FINDINGS: The routine surveillance system indicated that there were 14 cases and the hospital record review identified 19 additional cases. According to the two-source capture-recapture method, there would have been 40 cases during this period (95% confidence interval (CI) = 29-51), representing an average annual incidence of 1.4 per 100000 children aged under 15 years (95% CI = 1.1- 1.7). Thus case ascertainment based on routine surveillance was estimated to be 35% complete. Guillain-Barré syndrome was the commonest single cause of AFP. CONCLUSIONS: Routine surveillance for AFP in Victoria was insensitive. A literature review indicated that the capture-recapture estimates obtained in this study were plausible. The present results help to define a target notification rate for surveillance in settings where poliomyelitis is not endemic.

[Detection of TTV DNA from high background nuclear acid samples using specific nuclear acid captured-polymerase chain reaction].

OBJECTIVE: To improve the amplification of non-specificity of TTV DNA detection using PCR from high background nuclear acids because of its low level in samples. METHODS: Specific nuclear acid captured PCR(SNAC-PCR) was established through capturing TTV DNA onto microplate with specific nuclear acid by means of avidin-biotin system, then amplifying captured TTV DNA as usual. Its specificity was evaluated by comparing with the detection of routine extraction samples using nested-PCR with or without subsequent DNA sequencing in 10 samples of sera and liver tissues in pairs. RESULTS: Positive results were obtained using routine protocol in 4 and 9 out of 10 samples of the sera and the liver tissues respectively. The PCR products were proved to be specific using RFLP analysis with Kpn I. However, positive results were found in only 2/10 and 3/10 of samples of sera and liver tissue using DNA sequencing. The results of SNAC-PCR were the same as those of routine protocol with subsequent DNA sequencing. Furthermore, sharper electrophoresis bands were obtained. CONCLUSION: SNAC-PCR could dramatically increase the specificity of the detection of TTV DNA, especially in high background nuclear acid samples. It can be widely used in the detection of other pathogens with low seral level or in high background nuclear acid samples. The level of TTV infection might be over-evaluated by currently used TTV DNA detection when the PCR products were not confirmed by DNA sequencing.

Focal distraction: spatial shifts of attentional focus are not required for contingent capture.

Contingent capture occurs when distractors that share the target's defining attribute capture attention and slow down target identification. This slowdown has been attributed to an involuntary attentional shift to the location of a pertinent distractor. The present study examined an additional source of delay: the time spent in processing pertinent distractors. In 7 experiments, distractors were presented at fixation, and targets were presented either at fixation or peripherally. Contingent capture invariably occurred when a salient distractor was presented within about 600 ms before the target, even when spatial shifts in attentional focus were ruled out. A 2-stage model is proposed in which stimuli must pass an input filter tuned to the target's defining attribute before gaining access to a high-level stage that is unavailable while a distractor is being processed.

Evaluation of an IgG cystatin capture enzyme-linked immunosorbent assay for the detection of anti-cysteine proteinase antibodies in asymptomatic trichomoniasis patients.

All isolates of T. vaginalis release cysteine proteinases proteolytic enzymes that are shed into the vagina or culture medium. Cystatin has been used successfully as a capture agent in ELISA to detect cysteine proteinase antibodies without the need for purified proteinases. The ELISA was evaluated in comparison to wet mount microscopy and culture techniques. IgG cystatin capture ELISA proved to be a sensitive and highly specific (100%) assay that could rapidly detect anti-cysteine proteinase antibodies in both vaginal washouts and sera of asymptomatic patients with a sensitivity of 100% and 86.7%, respectively. A defined discrimination between sero-positive and sero-negative individuals was markedly observed for ELISA-vaginal washouts providing a more conclusive diagnosis by this technique. The results demonstrated that in trichomoniasis patients (52 cases) whether symptomatic or asymptomatic. T. vaginalis infection was detected in 19 (36.5%), 49 (94.2%), 50 (96.1%) and 48 (92.3%) by wet mount, culture, cystatin capture ELISA-vaginal washouts and ELISA-sera, respectively. The assay was reliable also as a test of cure with a specificity of 94.4% in the vaginal washouts and 83.3% in sera.
