A new quality-of-life measure for men experiencing involuntary childlessness.

BACKGROUND: Infertility may considerably reduce quality-of-life. Many of the existing generic quality-of-life measures, which often focus on physical impairments, do not represent the specific complaints of infertile patients. In this article, we report on the development and validation of the TLMK (Tübinger Lebensqualitätsfragebogen für Männer mit Kinderwunsch), an instrument for measuring quality-of-life in male patients with involuntary childlessness. METHODS: The first version of the questionnaire, which consisted of 91 items, was administered to 275 men who attended andrology and gynaecology clinics for fertility evaluations. After the questionnaires were scored, item analysis and reduction, principal component analysis and internal consistency analyses were conducted. RESULTS: The final version of the TLMK consists of 35 items in four scales and provides an internally consistent quality-of-life profile for men experiencing involuntary childlessness. Convergent and discriminant validity was supported through the correlation of the TLMK scales with established questionnaires on life satisfaction (FLZ) and partnership (PFB). CONCLUSION: The TLMK provides information about the quality-of-life in men experiencing involuntary childlessness and was found to be easy to administer and acceptable to patients. It may be used to assess patients' baseline and ongoing quality-of-life during fertility treatment and as an outcome variable in the evaluation of integrated psychological counselling.

Structure of the rat behaviour in the forced swimming test.

Forced swimming test (FST) or 'behavioural despair' test is a useful screening for antidepressant drugs. The FST predictability has been improved by a number of procedural modifications. Description of the behavioural microstructure in FST may help to delineate innovative protocols. Thus, counts of all behaviours emitted during FST in rats (four-month-old Wistar male, n = 63) were recorded and examined by Markovian sequential analysis (MSA) and principal components analysis (PCA). In a second experiment, rats (n = 28) were tested in an open field test (OFT) followed a week later by FST; behaviours in both tests were recorded and analysed by two correlation methods (Pearson's test and sliding window correlation). The descriptive ethological analysis displayed counts of swimming and immobility increased over the course of the test, whereas climbing behaviour decreased. The MSA revealed the occurrence of immobility was predicted by swimming, climbing, and diving behaviours whereas the immobility predicted the occurrence of swimming behaviour and headshakes. The PCA showed duration of immobility and climbing loaded into one component and duration of immobility and swimming loaded into another one. Low as well high levels of climbing behaviour were positively correlated with motor activity in the OFT. In brief, the present data suggest there are at least two different factors that grouped variables related to the behavioural despair in the FST. In addition, altered motor activity could be predicted by the frequency of climbing behaviour recorded in the FST.

A nonparametric quantification of neural response field structures.

The response fields of higher cortical neurons are usually approximated with smooth mathematical functions for the purpose of population parameterization or theoretical modeling. We used instead two nonparametric methods (principal component analysis and independent component analysis), which provided a basis for the response field clustering. Although both methods performed satisfactorily, the principal component analysis space is more straightforward to calculate. It also gave a clear preference toward the smallest number of functional response field classes. Clustering was performed with both K-means and superparamagnetic clustering algorithms with similar results. We also show that the shapes of the eigenvectors remain consistent regardless of the response field data sets size. This finding reflects the fact that the response fields were generated by the same neural network and encode the same underlying process.

Time-oriented hierarchical method for computation of principal components using subspace learning algorithm.

Principal Component Analysis (PCA) and Principal Subspace Analysis (PSA) are classic techniques in statistical data analysis, feature extraction and data compression. Given a set of multivariate measurements, PCA and PSA provide a smaller set of "basis vectors" with less redundancy, and a subspace spanned by them, respectively. Artificial neurons and neural networks have been shown to perform PSA and PCA when gradient ascent (descent) learning rules are used, which is related to the constrained maximization (minimization) of statistical objective functions. Due to their low complexity, such algorithms and their implementation in neural networks are potentially useful in cases of tracking slow changes of correlations in the input data or in updating eigenvectors with new samples. In this paper we propose PCA learning algorithm that is fully homogeneous with respect to neurons. The algorithm is obtained by modification of one of the most famous PSA learning algorithms--Subspace Learning Algorithm (SLA). Modification of the algorithm is based on Time-Oriented Hierarchical Method (TOHM). The method uses two distinct time scales. On a faster time scale PSA algorithm is responsible for the "behavior" of all output neurons. On a slower scale, output neurons will compete for fulfillment of their "own interests". On this scale, basis vectors in the principal subspace are rotated toward the principal eigenvectors. At the end of the paper it will be briefly analyzed how (or why) time-oriented hierarchical method can be used for transformation of any of the existing neural network PSA method, into PCA method.

Translation invariance in the responses to faces of single neurons in the temporal visual cortical areas of the alert macaque.

1. The responses of single neurons in the inferior temporal cortex and the cortex in the banks of the anterior part of the superior temporal sulcus of three awake, behaving macaques were recorded during a visual fixation task. Stimulus images subtending 17 or 8.5 degrees were presented in the center of the display area, and fixation was either at the center of the display area, or at one of four positions that were on the stimulus, or several degrees off the edge of the test stimulus. The experiments were performed with face-selective cells, and the responses were compared for fixation at each position for both effective and noneffective face stimuli for each cell. 2. The firing rates of most neurons to an effective image did not significantly alter when visual fixation was as far eccentric as the edge of the face, and they showed only a small reduction when the fixation point was up to 4 degrees from the edge of the face. Moreover, stimulus selectivity across faces was maintained throughout this region of the visual field. 3. The centers of the receptive fields of the cells, as shown by the calculated "centers of gravity," were close to the fovea, with almost all being within 3 degrees of the fovea. 4. The receptive fields of the cells typically crossed the vertical midline for at least 5 degrees. 5. Information theory procedures were used to analyze the spike trains of the visual neurons. Nearly six times more information was carried by these neurons' firing rate about the identity of an image than about its position in the visual field. Thus the information theory analysis showed that the responses of these neurons reflected information about which stimulus had been seen in a relatively translation invariant way. 6. Principal component analysis showed that principal component 1 (PC1) is related primarily to firing rate and reflected information primarily about stimulus identity. (For identity PC2 added only 14% more information to that contained in PC1.) Principal component 2 (PC2) was more closely related to neuronal response latencies, which increased with increasing eccentricity of the image in the visual field. PC2 reflected information about the position of the stimulus in the visual field, in that PC2 added 109% more information to that contained in PC1 about the position of the stimulus in the visual field.(ABSTRACT TRUNCATED AT 400 WORDS)

Analysis of mandible of newly developing strain of Japanese white rabbit (Nib:JWNS).

In the present paper, we applied analysis of the mandible to a new strain of Japanese White rabbit being developed (Nib: JWNS), at 5th and 8th generations (F5 and F8), and evaluated the process of establishment, making comparison with the parent colonies (I, K and L), and other established JW colonies (A, E, O and R). The mandibles were measured at 12 sites and the data obtained were calculated by principal component analysis and discriminant analysis. As a result of principal component analysis, the parent colonies were found to be close to one another with intermediate size of the mandible and height of condyloid and angular processes, but in F5 and F8 the mandibles were slightly shorter in height and different from those in the parent colonies. On the other hand, results of discriminant analysis revealed that mandibles were discriminated correctly 100% in colonies A, O and R, and 90% in E and I. Colonies A, O, R, E and I were therefore regarded as established strains. In the developing colonies, the discriminant rate was 70% in F5 and 80% in F8. One mandible from F5 and two from F8 were wrongly classified to the parent colony L, which was the lowest discriminant rate among the colonies examined (61.5%). The results of both statistical analyses suggest that the JWNS are almost established at 8th generation as a new strain.

Correlation and prediction of trunk fat mass with four anthropometric indices in Chinese males.

To increase our understanding of the relationships of trunk fat mass (FMtrunk) and four anthropometric indices in Chinese males, 1090 males aged 20-40 years were randomly recruited from the city of Changsha, China. Waist circumference (WC) and hip circumference (HC) were measured using standardized equipment, and three other anthropometric indices of BMI, waist:hip ratio (WHR) and conicity index (CoI) were calculated using weight, height, HC and WC. FMtrunk (in kg) was measured using a Hologic QDR 4500 W dual-energy X-ray absorptiometry scanner. There was an increasing trend of FMtrunk, %FMtrunk (percentage of FMtrunk) and BMI, WC, WHR, CoI in successively older age groups (e.g. the mean FMtrunk values were 4.63 (SD 2.58), 5.39 (SD 2.74), 5.93 (SD 2.82), 6.57 (SD 2.94) in four 5-year age groups, respectively). FMtrunk and %FMtrunk were significantly correlated with four anthropometric indices with the Pearson's correlation coefficients ranging from 0.25 to 0.86. Principal component analysis was performed to form three principal components that interpreted over 99.5% of the total variation of four related anthropometric indices in all age groups, with over 65% of the total variation accounted by principal component 1. Multiple regression analyses showed that three principal components explained a greater variance (R(2) 70.0-80.1%) in FMtrunk than did BMI or WC alone (R(2) 57.8-74.1%). The present results suggest that there is an increasing trend of FMtrunk and four anthropometric indices in successively older age groups; that age has important effects on the relationships of FMtrunk and studied anthropometric indices; and that the accuracy of predicting FMtrunk using four anthropometric indices is higher than using BMI or WC alone.

[Structural analysis for psychosocial factors including health locus of control (HLC) and sense of coherence (SOC) associated with lifestyle-related diseases].

The purpose of this study was to clarify psychosocial characteristics associated with preventive health behavior for lifestyle-related diseases. The author performed objective health examinations and gave questionnaires to 289 men (39.7 +/- 11.8 years, mean +/- SD) and 80 women (32.8 +/- 10.4 years) engaged in office work. Psychosocial factors included lifestyle and perceived stress, as well as the health locus of control (HLC) and sense of coherence (SOC) as newly developed indicators for health behavior. The principal component analysis for men did not extract lifestyle from the psychosocial structures. Multiple regression analysis showed that internal HLC (IHLC), chance HLC (CHLC), professional HLC (PHLC) and stress significantly contributed to SOC. Principal component analysis using psychosocial factors in women showed two psychosocial structures, i.e. the second principal (high SOC, high lifestyle, and low stress) and the 4th principal components (high supernatural HLC, and high PHLC). Both components were negatively correlated with systolic blood pressure. SOC was recognized to be negatively associated with age, stress, and total cholesterol, and positively with IHLC, FHLC, lifestyle, and gamma-GTP using multiple regression analysis for women. These results indicated a distinguishable sex difference regarding the involvement of psychosocial factors including HLC and SOC in objective health. SOC seems likely to be involved not in objective health, but closely with stress, suggesting a direct influence on mental health. Lifestyle should be divided into more detailed categories such as smoking and salt intake. Structural analysis of women suggests that SOC is involved directly or indirectly through lifestyle in objective health, different from men. To further clarify causal relationships between psychosocial factors and risk factors for lifestyle-related diseases, a longitudinal study is necessary based on these results.

Chemical rank estimation by noise perturbation in functional principal component analysist.

Some kinds of chemical data are not only univariate or multivariate observations of classical statistics, but also functions observed continuously. Such special characters of the data, if being handled efficiently, will certainly improve the predictive accuracy. In this paper, a novel method, named noise perturbation in functional principal component analysis (NPFPCA), was proposed to determine the chemical rank of two-way data. In NPFPCA, after noise addition to the measured data, the smooth eigenvectors can be obtained by functional principal component analysis (FPCA). The eigenvectors representing noise are sensitive to the perturbation, on the other hand, those representing chemical components are not. Therefore, by comparing the difference of eigenvectors obtained by FPCA with noise perturbation and by traditional principal component analysis (PCA), the chemical rank of the system can be achieved accurately. Several simulated and real chemical data sets were analyzed to demonstrate the efficiency of the proposed method.

The development of a standardized neighborhood deprivation index.

Census data are widely used for assessing neighborhood socioeconomic context. Research using census data has been inconsistent in variable choice and usually limited to single geographic areas. This paper seeks to a) outline a process for developing a neighborhood deprivation index using principal components analysis and b) demonstrate an example of its utility for identifying contextual variables that are associated with perinatal health outcomes across diverse geographic areas. Year 2000 U.S. Census and vital records birth data (1998-2001) were merged at the census tract level for 19 cities (located in three states) and five suburban counties (located in three states), which were used to create eight study areas within four states. Census variables representing five socio-demographic domains previously associated with health outcomes, including income/poverty, education, employment, housing, and occupation, were empirically summarized using principal components analysis. The resulting first principal component, hereafter referred to as neighborhood deprivation, accounted for 51 to 73% of the total variability across eight study areas. Component loadings were consistent both within and across study areas (0.2-0.4), suggesting that each variable contributes approximately equally to "deprivation" across diverse geographies. The deprivation index was associated with the unadjusted prevalence of preterm birth and low birth weight for white non-Hispanic and to a lesser extent for black non-Hispanic women across the eight sites. The high correlations between census variables, the inherent multidimensionality of constructs like neighborhood deprivation, and the observed associations with birth outcomes suggest the utility of using a deprivation, index for research into neighborhood effects on adverse birth outcomes.

Tissue inhibitors of metalloproteinases level and collagenase activity in gingival crevicular fluid: the relevance to periodontal diseases.

OBJECTIVES: To provide an overall assessment of levels of tissue inhibitors of metalloproteinases (TIMPs), collagenase activities, and of immuno-reactivities for matrix metalloproteinases (MMP)-1 and -8 in gingival crevicular fluid (GCF) obtained from healthy subjects, and gingivitis and periodontitis patients, and to analyse the relationships between periodontal tissue destruction and the GCF components in periodontal diseases by principal component analysis. MATERIALS AND METHODS: GCF was sampled with sterile paper strips from 10 gingivitis and 11 periodontitis patients. Ten volunteers served as clinically healthy controls. TIMP-1 and -2 protein amounts in GCF were measured by ELISA, and active and APMA-activatable collagenase activities were determined by functional assays using image-analysis after SDS-PAGE. RESULTS: GCF TIMP-1 level and both active and latent collagenase activities were significantly higher in the diseased groups than in the healthy group. TIMP-2 was detectable in only 29% of all subjects (mean: 2.06 ng). Western blot analysis showed that MMP-8 was the major interstitial collagenase in the GCF of the diseased groups. Principal component analysis using clinical parameters and the GCF components has indicated components one to three account for 87% of total variation when evaluating the relevance of their measurements to periodontal diseases. CONCLUSIONS: We conducted the functional and immunological characterization of MMPs and TIMPs in the GCF of periodontally diseased patients. Principal component analysis indicated components one to three explaining 87% of total variation, and further suggested that higher collagenase activity (especially in active collagenase) would be an important marker in evaluating the pathogenesis of periodontitis. Consequently, these observations may have significant therapeutic and diagnostic implications.

Construction and validation of a questionnaire to measure the health beliefs of general practice patients.

The health belief model has been widely used as a conceptual framework for understanding and explaining compliance behaviour. A weakness characterizing work in the area has been lack of standardization of measurement of the components of the health belief model. This paper describes the development and validation of a questionnaire to measure these components. The questionnaire was designed for use with general practice patients who have a wide range of different illnesses, therefore the nature of the patients' illness is not mentioned in the content of the items. Principal components analysis was used to determine the dimensions underlying patients' beliefs. Principal components analysis and application of Cronbach's alpha statistic identified four reliable sub-scales of the questionnaire. The sub-scales measured patient's beliefs about: the threat caused by illness, the efficacy of traditional medical care, the way illness is dealt with and the barriers to taking medications.

Development and validation of questionnaire measuring attitudes towards sexual health among university students.

AIM: To develop a questionnaire on attitudes towards sexual health and validate it on a sample of medical and non-medical students and adult women. METHODS: For the purpose os constructing a Likert-type scale, four medical studentsgenerated 130 statements reflecting clearly positive or negative attitudes towards sexual health. The scale had five scoring points (1 - strongly disagree, 2 - disagree, 3 - undecided, 4 - agree, and 5 - strongly agree). We revised the items to ensure that all were clearly positive or clearly negative in their form, as well as straightforward in their meaning, leaving 100 items in the raw questionnaire, which was then applied to the convenience sample (total n=336) of female medical students (n=93), male medical students (n=75), female non-medical students (n=93) and adult working women with at least high-school education (n=75). After the administration of the questionnaire, we performed content examination, discriminant validation of the statements, and principal-components analysis. RESULTS: After discriminant validation of the statements and principal-components analysis limited to three factors, the final scale contained 50 items (20 positive and 30 negative), distributed in three subscales: "Personal sexual health care," "Principles of sexual behavior," and "Safe sex behavior." The Cronbach alpha of the whole scale was 0.910, indicating good internal consistency. When the scores of the final scale were analyzed for the study sample, all participants had positive attitude towards sexual health, with a mean score (+/-standard deviation) of 4.1+/-0.4 out of maximum 5. The same was true for all threee subscales. Female groups, regardless of their student status or age, did not differ in their attitude scores on the whole scale, as well as on any of the subscales. Male medical students had lower scores on the whole scale and all subscales than the female groups. CONCLUSIONS: It is posible to construct a reliable instrument to assess attitudes towards three important aspects of sexual health: personal sexual health care, sexual behavior, and safe sex behavior. Future studies with this instrument, on different populations and in different cultural and socio-economic settings, are needed to make generalized conclusions on sexual health attitudes and behavior.

Three-way partial least-squares regression for the simultaneous kinetic-enzymatic determination of xanthine and hypoxanthine in human urine.

The performance of three-way principal component analysis and three-way partial least-squares regression when applied to a complex kinetic-enzymatic system is studied, in order to investigate the analytical potential of the combined use of these chemometric technologies for non-selective enzymatic systems. A enzymatic-kinetic procedure for the simultaneous determination of hypoxanthine and xanthine in spiked samples of human urine is proposed. The chemical system involves two consecutive reactions catalyzed by xanthine oxidase (EC 1.17.3.2). This enzyme catalyzes the oxidation of hypoxanthine, first to xanthine and then to uric acid, a competitive inhibitor of the reactions. The influence of uric acid during quantitative determination was considered in the design of the calibration set. The sample and enzyme solution were mixed in a stopped-flow module and the reaction was monitored using a diode array spectrophotometer. The recorded data have an intrinsical three-component structure (samples, time and wavelength). This data array was studied via three-way principal component analysis and was modeled for quantitative purposes using a three-way partial least-squares calibration procedure. Results are compared with those obtained by applying classical bilinear PLS to the previously unfolded data matrix.

Evaluation of the spatial aspects of T-wave complexity in the long-QT syndrome.

BACKGROUND: The duration of the QT interval is only a gross estimate of repolarization. Besides its limited accuracy and reproducibility, it does not provide information on the morphology of the T wave; thus, morphologic alterations such as notches can be only qualitatively described but not objectively quantified. METHODS AND RESULTS: To measure the complexity of repolarization in the long-QT syndrome (LQTS) patients, we previously applied principal component analysis to body surface mapping and found it useful in distinguishing normal from abnormal repolarization patterns (sensitivity, 87%). In the present study, we applied principal component analysis to 12-lead Holter recordings. The index of complexity of repolarization that we have developed (CR24h) reflects the average 24-hour complexity of repolarization and is mathematically defined as the average ratio between the second and the first eigenvalue. We studied 36 LQTS patients and 40 control subjects. A mean of 22+/-1.3 ECG recordings at 1-hour intervals was used in each patient, and a total of 1655 recordings were analyzed. CR24h was significantly higher in LQTS than in control subjects (34+/-12% versus 13+/-3%; P<.0001). A CR24h exceeding 2 SD above the mean of the control group (>20%) was present in 32 of 36 patients (88%). The negative predictive value of CR24h in LQTS was 88%, and the combination of prolonged QT and abnormal CR24h identified all LQTS patients from normal subjects, including 4 affected symptomatic individuals with a normal QT interval duration, suggesting that CR24h provides information independent of QT duration. CONCLUSIONS: Our data suggest that principal component analysis applied to 24-hour, 12-lead Holter recording adequately quantifies the complexity of ventricular repolarization and may become a useful noninvasive diagnostic tool in LQTS.

The use of multivariate methods in the identification of subtypes of Alzheimer's disease: a comparison of principal components and cluster analysis.

Two contrasting multivariate statistical methods, viz., principal components analysis (PCA) and cluster analysis were applied to the study of neuropathological variations between cases of Alzheimer's disease (AD). To compare the two methods, 78 cases of AD were analyzed, each characterised by measurements of 47 neuropathological variables. Both methods of analysis revealed significant variations between AD cases. These variations were related primarily to differences in the distribution and abundance of senile plaques (SP) and neurofibrillary tangles (NFT) in the brain. Cluster analysis classified the majority of AD cases into five groups which could represent subtypes of AD. However, PCA suggested that variation between cases was more continuous with no distinct subtypes. Hence, PCA may be a more appropriate method than cluster analysis in the study of neuropathological variations between AD cases.

Casein haplotype structure in five Italian goat breeds.

The aim of this work was to investigate the genetic structure of the casein gene cluster in 5 Italian goat breeds and to evaluate the haplotype variability within and among populations. A total of 430 goats from Vallesana, Roccaverano, Jonica, Garganica, and Maltese breeds were genotyped at alphas1-casein (CSN1S1), alphas2-casein, (CSN1S2), beta-casein (CSN2), and kappa-casein (CSN3) loci using several genomic techniques and milk protein analysis. Casein haplotype frequencies were estimated for each breed. Principal component analysis was carried out to highlight the relationship among breeds. Allele and haplotype distributions indicated considerable differences among breeds. The haplotype CSN1S1*F- CSN1S2*F-CSN3*D occurred in all breeds with frequencies >0.100 and was the most common haplotype in the Southern breeds. A high frequency of CSN1S1*0-CSN1S2*C-CSN3*A haplotype was found in Vallesana population (0.162). Principal component analysis clearly separated the Northern and Southern breeds by the first component. The variability of the caprine casein loci and variety of resulting haplotypes should be exploited in the future using specific breeding programs aiming to preserve biodiversity and to select goat genetic lines for specific protein production.

Subjective experiences and the psychopathological dimensions in schizophrenia.

The aim of this article was to explore the relationships between subjective experience (SE) and objective symptoms in schizophrenia. Exploratory principal component analysis was used to determine the latent organization of the variables. A group of 310 chronic patients with schizophrenia who met ICD-10 criteria for F20 schizophrenic disorder were included in the study. SE and objective symptoms were rated using, respectively, the 24-item version of the Frankfurt Complaint Questionnaire (FCQ-24) and the Positive and Negative Syndrome Scale (PANSS). An exploratory principal component analysis was performed on the correlation matrix comprising items from both the FCQ-24 and the PANSS. Using several guidelines to select the number of factors, the exploratory principal component analysis yielded a six-factor solution with no overlap of the significant factor loadings for the items from each scale. These six factors represent, respectively, the subjective, negative, positive, disorganization, hostility and anxious-depressive dimensions of the schizophrenic phenomenology. The findings support the view that SE is a construct that is separate and distinct from the objective symptomatology in schizophrenia.
