Optimal modeling of corneal surfaces with Zernike polynomials.

Zernike polynomials are often used as an expansion of corneal height data and for analysis of optical wavefronts. Accurate modeling of corneal surfaces with Zernike polynomials involves selecting the order of the polynomial expansion based on the measured data. We have compared the efficacy of various classical model order selection techniques that can be utilized for this purpose, and propose an approach based on the bootstrap. First, it is shown in simulations that the bootstrap method outperforms the classical model order selection techniques. Then, it is proved that the bootstrap technique is the most appropriate method in the context of fitting Zernike polynomials to corneal elevation data, allowing objective selection of the optimal number of Zernike terms. The process of optimal fitting of Zernike polynomials to corneal elevation data is discussed and examples are given for normal corneas and for abnormal corneas with significant distortion. The optimal model order varies as a function of the diameter of the cornea.

Effect of individual amino acids and glucose on activation and germination of Rhizopus oligosporus sporangiospores in tempe starter.

AIM: To understand the conditions promoting activation and germination of spores, and to contribute to the control of tempe starters. METHODS AND RESULTS: Using microscopic counts of fluorescent labelled spores, the following results were obtained: (1) L-alanine plays an important role (of the same order as that of peptone) in stimulation of germination of dormant spores. Alanine can satisfy the requirements of carbon as well as nitrogen for spore germination; (2) L-proline, on the other hand, inhibits alanine uptake presumably by blocking/congesting transporters of spore cells, resulting in apparent low viability on agar media; (3) L-leucine and L-isoleucine slightly favour spore germination while L-arginine and L-lysine do not have any stimulating effect; (4) The stimulatory role of glucose was only evident in the presence of phosphate (in minimal medium); when glucose is used in the absence of phosphate, either alone or in combination with single amino acids its role is hardly distinguishable; (5) Phosphate plays a facilitating role in spore germination. CONCLUSIONS: Glucose and amino acids play important roles in activation and germination of sporangiospores of Rhizopus oligosporus in tempe starter (stored for 12 months). The ability and rate of germination of dormant/old sporangiospores of R. oligosporus, depend on their ability for uptake of individual amino acids and/or glucose. SIGNIFICANCE AND IMPACT OF STUDY: New light was shed on the counteractive role of proline and the stimulating effect of phosphate. Soybeans subjected to traditional preparation for tempe making are heavily leached; germination of starter spores on such beans is sub-optimal, and bean processing could be optimized.

Sodium hypochlorite: history, properties, electrochemical production.

This paper analyses the evolution of hypochlorite as strong disinfectant. The electrochemical production of hypochloric acid or sodium hypochlorite represents the best method to obtain a pure product. To have a good production (as quality and quantity), it is necessary to optimize the electrochemical process with the optimal of electrocatalytic electrodes (cathode and anode) the gap between electrodes, the temperature of electrochemical cell. It is very important for the product stability during a long period, avoid the presence of heavy metal ions and particulate as impurity-like carbon micro-powders in suspension. It is necessary a rigorous control of the pH of final product to have the optimal disinfection power of hypochlorite solution. The most stable sodium hypochlorite solutions are those that show the following characteristics: (1) low concentration of hypochlorite; (2) pH -> 11.5 and <13; (3) absence of graphite particulate and metallic ions; (4) storage at controlled temperature <30 degrees C. Enzymatic conversion of waste cooking oils into alternative fuel--biodiesel.

Production of biodiesel from pure oils through chemical conversion may not be applicable to waste oils/fats. Therefore, enzymatic conversion using immobilized lipase based on Rhizopus orzyae is considered in this article. This article studies this technological process, focusing on optimization of several process parameters, including the molar ratio of methanol to waste oils, biocatalyst load, and adding method, reaction temperature, and water content. The results indicate that methanol/oils ratio of 4, immobilized lipase/oils of 30 wt% and 40 degrees C are suitable for waste oils under 1 atm. The irreversible inactivation of the lipase is presumed and a stepwise addition of methanol to reduce inactivation of immobilized lipases is proposed. Under the optimum conditions the yield of methyl esters is around 88-90%.

Are communication activities shaped by environmental constraints in reverberating and absorbing forest habitats?

In the dense vegetation of temperate or tropical forests, communication processes are constrained by propagation-induced modifications of the transmitted sounds. The presence of leaves, trunks and branches induces important sound reverberation and absorption leading to diminution of the signal energy as well as qualitative modifications. The aim of this paper is to briefly review the different strategies used by birds to manage with these constraints. At the emitter's level, an adapted emission behavior which takes into account both the physical heterogeneities of the forest environment and the temporal variations of the acoustic constraints, is especially useful to control the active space of signaling. The coding of information into acoustic parameters that have different susceptibility to propagation constraints is also of great interest. At the receiver's level, an adaptive reception behavior (listening post) and a great tolerance to sound degradation during the decoding process are the keys to an optimal communication process.

[Optimization of technical parameters for processing radix Aconiti coreani].

Based on the determination of guanfu A, hypaconitine, and total alkaloids, along with the experiment of acute toxicity of sliced Radix Aconiti Coreani and in compliance with the quality standard stipulated in pharmacopeia-surface features cross section colour and odor of sliced Radix Aconiti Co-reani the technology of processing Radix Aconiti Coreani has been optimized to be steaming the drug for four hours.

Bone remodeling and structural optimization.

Bone remodeling has been viewed both as a process which adapts bone tissue to the mechanical environment at each point in the structure, and as a process which optimally adjusts the tissue distribution within bones to bear the loads placed on them. We have developed a connection between these two views of bone remodeling, in a restricted sense. We start with a remodeling rate equation based on strain energy density. We then define an indicator function which is a weighted sum of total strain energy and a measure of bone mass, and we show that finding bone density distributions in which the remodeling rate equation predicts no changes with time is the same as finding density distributions in which the indicator function is insensitive to small changes in density. The set point in the remodeling rate equation corresponds to a parameter in the indicator function which determines the relative importance of bone mass and strain energy in the optimization indicator function. We have not assessed whether the density distributions which make the density rate of change zero are actually local or global minima for the indicator function in this study, but a related study shows that there is a single unique minimum for the indicator function developed here, implying that a unique solution exists for the bone remodeling rate equations considered in this study.

Application of model-predictive control based on artificial neural networks to optimize the fed-batch process for riboflavin production.

The fed-batch process for commercial production of riboflavin (vitamin B2) was optimized on-line using model-predictive control based on artificial neural networks (ANNs). The information required for process models was extracted from both historical data and heuristic rules. After each cultivation the process model was readapted off-line to include the most recent process data. The control signal (feed rate), however, was optimized on-line at each sampling interval. An optimizer simulated variations in the control signal and assessed the forecasted model outputs according to an objective function. The optimum feed profile for increasing the product yield (YB2/S) and the amount of riboflavin at the time of harvesting was adjusted continuously and applied to the process. In contrast to the control by set-point profiles, the novel ANN-control is able to react on-line to variations in the process and also to incorporate the new process information continuously. As a result, both the total amount of riboflavin produced and the product yield increased systematically by more than 10% and the reproducibility of seven subsequently optimized batches was enhanced.

Optimizing the APC residue washing process to minimize the release of chloride and heavy metals.

An optimization of the air pollution control (APC) residue washing process was carried out to minimize the release of chloride and heavy metals. Taking into account economic parameters such as the consumption of water and reaction time, the best relation found was a S/L ratio of 1/3 during 1 h. At a laboratory scale and according to the values obtained for chloride and heavy metals, the APC residue is classified as non-special according to Catalonian Regulations (Spain). Moreover, the pH of the solution, when MgSO(4) is added during the washing process, may be controlled by the formation of gypsum. In these conditions, the concentration of heavy metals will decrease as a consequence of the formation of their respective insoluble hydroxides. Therefore, the counter-current batch washing process with the addition of small amounts of MgSO(4) is revealed as an economically feasible treatment of the APC residue. This washed residue is ready to be used as secondary material or to be landfilled safely.

Optimization of medium and process parameters for the production of inulinase from a newly isolated Kluyveromyces marxianus YS-1.

A newly isolated strain of Kluyveromyces marxianus YS-1 was used for the production of extra cellular inulinase in a medium containing inulin, meat extract, CaCl2 and sodium dodecyl sulphate (SDS). Fermentation medium pH 6.5, cultivation temperature 30 degrees C and 5% (v/v) inoculum of 12 h-old culture were optimal for enzyme production (30.8 IU/ml) with a fermentation time of 72 h at shake flask level. Raw inulin (2%, w/v) extracted from dahlia tubers by processing at 15 kg/cm2 for 10 min was optimum for bioreactor studies. Maximum enzyme production (55.4 IU/ml) was obtained at an agitation rate of 200 rpm and aeration of 0.75 vvm in a stirred tank reactor with a fermentation time of 60 h.

Optimal structure for automatic processing of DNA sequences.

The faithful recovery of the base sequence in automatic DeoxyriboNucleic Acid (DNA) sequencing fundamentally depends on the underlying statistics of the DNA electrophoresis time series. Current DNA sequencing algorithms are heuristic in nature and modest in their use of statistical information. In this paper, a formal statistical model of the DNA time series is presented and then used to construct the optimal maximum-likelihood (ML) processor. The DNA-ML algorithm that is derived in this paper features Kalman prediction of peak locations, peak parameter estimation, whitened waveform comparison and multiple hypothesis processing using the M-algorithm. Properties of the algorithm are examined using both simulated and real data. Model parameters of critical importance and their impact on different types of error mechanisms, such as insertions and deletions, are pointed out. The statistical model of the DNA time-series and the structure of the DNA-ML algorithm provides a basis for future investigation and refinement of DNA sequencing techniques.

What process is glycolytic stoichiometry optimal for?

It has been proposed that the glycolytic stoichiometry of 2 ATP per glucose is the result of an optimization that maximizes the rate of ATP production. However, using a nonequilibrium thermodynamic approach, we show here that glycolysis operates under optimal output power and not at optimal flow of ATP production. Furthermore, it can be proved that the same maximal output power can be achieved with different stoichiometries. However, changes in the glycolytic stoichiometry would dramatically affect the efficiency of all those cellular processes powered by ATP. Our results suggest that the stoichiometric coefficient, as found in most contemporary cells, may be the outcome of an evolutionary process leading to yield an operative quantum energy for the hydrolysis of ATP.

Multivariable optimization of a multiproduct continuous chemicals facility.

Multivariable optimization (MVO) is a powerful nonlinear steady-state flowsheet simulation technique used widely in the chemical process industry to optimize plant performance by increasing plant capacity and/or reducing energy usage. The user supplies the objective function(s), constraints, and variable limits based on operating heuristics, prior experience, and observed process behavior. In this paper we describe how we used MVO in conjunction with improved automation and statistical process monitoring to increase capacity and reduce energy consumption. This project was conducted over a two-year period in a methylamines facility that produces three products: monomethylamine (MMA), dimethylamine (DMA), and trimethylamine (TMA). It led to a 14% improvement in MMA capacity and a 7% improvement in TMA capacity. Energy consumption per pound of main product decreased by approximately 20% for the six-month period (April-September, 2000) when compared to a three-year average (1997-2000). It was accompanied by a 60% improvement in process capability and 31% improvement in product quality. We attribute this improvement to several factors: smoother transition between optimum set points determined by the process optimizer, improved disturbance rejection due to controller installation and tuning, and reduced operator intervention because of controlled operation at or near the optimum set points.

Validation of an integrated metagenomic pipeline combining optimized wet-lab processing and tiered reporting for CSF pathogen detection.

UNLABELLED: Metagenomic next-generation sequencing (mNGS) in the infectious disease diagnostic space has been gaining traction and is popular for aiding in the diagnosis of central nervous system infections. However, many challenges and obstacles remain in making this technology a gold standard for infectious disease diagnostic testing. One major challenge is being able to distinguish between the clinically relevant organisms from background contamination. We performed a validation study for mNGS on cerebrospinal fluid (CSF) that utilized positive clinical samples and contrived samples that incorporated a bioinformatics pipeline that can better distinguish between background contamination and clinically relevant organisms and used a three-tiered reporting algorithm meant to decrease the inherent subjectivity that comes with interpreting and reporting data from clinical metagenomic sequencing. The validation of this assay and category-based reporting pipeline revealed an overall concordance of 91.8%, with a sensitivity of 100% and a specificity of 72.4%. In addition, we improved the detection of clinically relevant RNA viruses to almost 100% in the CSF by modifying the wet lab processing of the sample. This bioinformatics pipeline with a category-based reporting algorithm will provide more confidence in reporting microorganisms detected with this technology, mNGS, and improving patient care. IMPORTANCE: Metagenomic next-generation sequencing (mNGS) can offer a broad, unbiased approach for the detection of infectious pathogens and has shown promise in diagnosing central nervous system infections. Despite its potential, clinical implementation remains limited by challenges in distinguishing clinically relevant organisms from background contamination. This study validated an mNGS assay for cerebrospinal fluid that incorporates an optimized bioinformatics pipeline with a three-tiered reporting algorithm designed to reduce subjectivity and enhance diagnostic confidence. The assay also has improved detection of clinically relevant RNA viruses through modified wet-lab processing. These findings support the clinical utility of a structured, category-based reporting approach for mNGS, advancing its reliability as a diagnostic tool in infectious disease testing.

Experimental implementation of identification-based optimizing control of a simulated moving bed process.

Experimental implementation of an optimizing controller based on identified model for the separation of nucleosides in a laboratory scale simulated moving bed (SMB) unit is reported in this study. The manipulative variables are the three external and one internal flow rates while the outputs are productivity, solvent consumption, and purities of extract and raffinate streams averaged over a switching period. The feedback information is the concentration profile of extract and raffinate measured online using two ultraviolet (UV) detectors. Experimental results show that the designed controller is able to operate the SMB units under optimal condition fulfilling the purity requirements. Besides, the controller demonstrated excellent performance in terms of rejecting disturbances that may occur during SMB operations.

Integrated widely targeted metabolomics and GC-IMS reveal dynamic flavor, nutritional, functional, and metabolic profiles in macadamia kernels during processing.

Different processing stages influence the color, flavor, and antioxidant activities of macadamia kernels. However, the biochemical mechanisms that occur during processing are not well known. This study integrated widely targeted metabolomics (UPLC-MS/MS) with GC-IMS to systematically characterize non-volatile and volatile compounds in macadamia kernels across key three sample groups: fresh kernels (FMN), low-temperature-dried kernels (DMN), and roasted kernels (BMN). A total of 622 non-volatile metabolites and 52 volatile compounds were identified. Low-temperature drying promoted the accumulation of phenolic acids and flavonoids, enhancing antioxidant capacity. Roasting degraded heat-sensitive nutrients but generated flavor compounds via Maillard reaction and lipid oxidation, shifting aroma from green to nutty notes. Nutritional assessment confirmed that roasting significantly reduced antioxidant activities and bile acid binding capacity. Pearson correlation analysis verified the key metabolite-antioxidant relationships. These findings provide critical insights into metabolic dynamics during nut processing and establish a scientific basis for optimizing thermal processing strategies.

Reaction pathways and kinetics of butylated hydroxyanisole with UV, ozonation, and UV/O(3) processes.

The chemical degradation of the endocrine disrupting chemical, butylated hydroxyanisole (BHA), was investigated by different treatment processes including ultraviolet (UV) irradiation, ozonation (O(3)), and UV/O(3). O demethylation, dimerization, and oxidation have been found to be the main degradation mechanisms. A systematic decay pathway was proposed based on ten identified intermediates in the studied processes, including a unique pathway leading to the formation of precipitates in the ozonation process. An unconventional minimum-type variation of BHA decay rate constants from acidic to caustic range has been found for both ozonation and UV/O(3) processes. The precipitates formed during ozonation can be removed during the process to optimize the treatment, while the UV/O(3) process can offer a relatively fast and clean process to degrade BHA and its associated intermediates.

A life span model of successful aging.

To lay the foundation for our model, we first describe existing conceptions of successful aging, underlying assumptions of development, and criteria for success. The model presented extends the discourse on this topic in three directions: (a) It frames the discussion of successful aging in the broader context of life course development; (b) it accounts for both normative and nonnormative (i.e., exceptional) success; and (c) it integrates motivational processes into a theory of successful aging. Successful aging is equated with the development and maintenance of primary control throughout the life course, which is achieved through control-related processes that optimize selection and failure compensation functions. Selection processes regulate the choice of action goals so that diversity is maintained and positive and negative trade-offs between performance domains and life stages are taken into account. Compensation mechanisms serve to maintain, enhance, and remediate competencies and motivational resources after failure experiences. Both compensation and selection processes are motivated by desires for primary control and can be characterized in terms of primary and secondary control processes.
