Immunomodulatory and antitumoral effects of triterpenoid saponins.

Genuine saponins of Solidago virgaurea L., Heteropappus altaicus (Willd.) Novopokr., Heteropappus biennis (Ldb.) Tamamsch. and Helianthus annuus L. (Asteraceae) as well as related carbohydrate modified glycosides of polygalacic acid and echinocystic acid and some commercial available triterpenoid saponins were investigated in view of their immunomodulating and antitumoral effects. Mitogeneic effects on murine spleen and thymus cells, as well on human mononuclear cells in vitro could be demonstrated. The activity of murine bone marrow macrophages was stimulated in a chemoluminescence assay, and further an induction of cytotoxic macrophages and a TNF alpha release from murine macrophages was observed. The mitogeneic and TNF alpha releasing virgaureasaponin E (1) showed in vivo antitumoral effects in the allogeneic sarcoma 180 tumor model and in the syngeneic DBA/2-MC.SC-1 fibrosarcoma tumor model. In mice, treated with 1, phagocytosis of bone marrow cells and proliferation of spleen and bone marrow cells were stimulated in an ex vivo assay whereby the TNF alpha concentration in blood considerably increased compared to the control group.

A primary pulmonary sarcoma in a rhesus monkey after inhalation of plutonium dioxide.

A pulmonary fibrosarcoma of bronchial origin was discovered in a Rhesus monkey that died of pulmonary fibrosis 9 years after inhalation of plutonium-239 dioxide and with a radiation dose to lung of 1400 rad (14 Gy). It grew around the major bronchus of the right cardiac lung lobe and extended into the bronchial lumen and into surrounding pulmonary parenchyma. It also readily invaded muscular pulmonary arteries, resulting in infarction and scarring in the right cardiac lobe. Despite this aggressive growth, the tumor did not metastasize. The primary cause of death was severe pulmonary fibrosis involving the alveolar septa and and perivascular and peribronchial interstitium. Bullous or pericitrical emphysema was prominent. The initial lung burden of plutonium in this monkey was 270 nCi (10 kBq) which is equivalent to approximately 500 times the maximum permissible lung burden for man on a radioactivity per unit body weight basis. The time-dose relationship for survival is consistent with that of dogs and baboons that inhaled plutonium dioxide and died with lung tumors.

Surface glycoproteins of human sarcoma- and fibroblastic cells.

A comparison was made of the cell surface glycoproteins of four human cell lines, namely a giant tumor of bone cell line, an osteosarcoma line, a fibrosarcoma line and a human fibroblast line. The cells were labeled by lactoperoxidase catalyzed iodination and the glycoproteins extracted by 0.5% Triton X-100 were bound to lentil-lectin and subsequently analyzed by SDS gel electrophoresis. While the cell lines examined shared a series of common glycoproteins, it was found that the giant cell tumor line and the fibrosarcoma lines exhibited a higher degree of homology than the other cell lines.

Current soft-tissue sarcoma classifications.

In order to analyse the impact of modern sarcoma classification criteria, pathological material from 281 extremity soft-tissue sarcomas (STS) was reviewed. The cases were originally diagnosed between 1972 and 1994, and the most frequent diagnoses then were malignant fibrous histiocytoma (MFH) (26%), liposarcoma (21%), fibrosarcoma (11%), and leiomyosarcoma (10%). After reclassification, the proportions had changed significantly, with the largest group now being leiomyosarcomas (20%), liposarcomas (17%), synovial sarcomas (14%), and sarcomas "not otherwise specified" (NOS) (11%). The original diagnosis was changed in 57% of the cases; in particular, the number of fibrosarcomas was reduced from 32 to 6, and MFHs from 72 to 2, with 22 renamed as myxofibrosarcomas; 20 (7%) were found not to be sarcomas. The main reasons for these results are the recent advances in immunohistochemistry (IHC) together with changes in nomenclature. The findings have obvious implications, in particular for retrospective research.

Histologic variation among experimental tumors of common origin: relationships to viral p21 expression and to tumor-derived extracellular matrix.

Transformed lines, obtained by infection of adult rat adrenocortical fibroblast-like cells with Kirsten murine sarcoma virus, produce s.c. tumors which range histologically from fibrosarcomas to carcinomas to undifferentiated neoplasms. This study was undertaken to determine whether the histology was a stable characteristic of individual lines and whether it was related to oncogene expression and to differentiation in culture. Three lines were infected at passage 1 and tumor histology, differentiation markers and expression of the transforming protein v-p21 were evaluated periodically over a 1-year period. The original stromal characteristics of the target cells (metachromatic extracellular matrix (ECM), a high rate of collagen production and collagen incorporation into ECM, and a fibroblastic morphology) diminished with transformation. In contrast, parenchymal differentiation markers (lipid, steroidogenic enzymes) were enhanced and constitutively expressed in all lines. The two lines in which ECM production was most reduced formed carcinomas in vivo, while the third line showed a smaller shift in these characteristics and formed sarcomas. Thus, histopathology seemed to be influenced by tumor-derived ECM but unrelated to the expression of steroidogenic properties. The tumors became less differentiated with time, but retained distinct line-specific phenotypes. Throughout the study, v-p21 remained overexpressed in all lines to a similar degree, but the subcellular localization of v-p21 differed between fibroblastic and epithelial cells, suggesting a possible correlation with differentiation.

Amplification of the c-myc proto-oncogene in soft tissue sarcomas.

Soft tissue sarcoma fresh specimens from 23 patients were screened for genetic abnormalities of 3 cellular oncogenes, i.e., c-myc and c-myb and c-Ha-ras. Southern blot hybridization analysis demonstrated amplification of the c-myc gene (4- to 8-fold) in 7 cases, i.e., 3 malignant fibrous histiocytomas, 1 liposarcoma, 1 fibrosarcoma, 1 lymphoma of the soft tissues and 1 synovial sarcoma. Amplification of the c-myc gene was not associated to other genetic alterations such as gene rearrangement. Hybridizations with c-myb and c-Ha-ras probes showed no gene abnormalities in this series. Our results indicate that c-myc oncogene amplification may play an important role in the development of certain soft tissue sarcomas as it has been found for other human malignancies. The clinical significance of these features remains to be established.
