Cascade radical cyclizations of benzannulated enyne-allenes. Unusual cleavage of a benzene ring leading to twisted 1,1'-dialkyl-9,9'-bifluorenylidenes and spiro[1H-cyclobut[a]indene-1,9'-[9H]fluorenes].

Treatment of the benzannulated enediynyl propargylic alcohol 16 (isomer ratio = 2:1) with thionyl chloride induced a sequence of reactions leading to the twisted 1,1'-dipropyl-9,9'-bifluorenylidene 17, the polycyclic compounds 18 and 19, and the spiro[1H-cyclobut[a]indene-1,9'-[9H]fluorene] 20 (trans/cis = 5:1). The transformation from 16 to the unexpected 17 presumably involved an initial formation of the benzannulated enyne-allene 21 followed by a C(2)-C(6) cyclization reaction and an intramolecular radical-radical coupling reaction, giving rise to the formal Diels-Alder adduct 23. Repeat of this sequence then furnished 24. Cleavage of the bond connecting the two carbons having the propyl substituent afforded 25. A subsequent rotation of the carbon-carbon bond joining the two central five-membered rings then gave the trans isomer 26. Oxidation of 26, presumably by oxygen, followed by hydrolysis then produced 17. Interestingly, the pathway leading to 17 involved an unusual cleavage of a benzene ring. The X-ray crystal structure of 17 reveals that it has a twist angle of 45.2 degrees for the carbon-carbon double bond connecting the two bifluorenylidene fragments. The spiro[1H-cyclobut[a]indene-1,9'-[9H]fluorene] 20 apparently was produced via two intramolecular [2 + 2] cycloaddition reactions of the benzannulated enyne-allene moieties, generated in situ from the benzannulated enediynyl propargylic alcohols. The twisted 1,1'-dimethyl-9,9'-bifluorenylidene 33 and the spiro[1H-cyclobut[a]indene-1,9'-[9H]fluorene] 39 (trans/cis = 3:1) were likewise produced from 32 and 38, respectively.

Intramolecular cyclization with nitrenium ions generated by treatment of N-acylaminophthalimides with hypervalent iodine compounds: formation of lactams and spiro-fused lactams.

N-Phthalimido-N-acylnitrenium ions are generated from N-acylaminophthalimides, a new class of precursors, by treatment with hypervalent iodine compounds (PIFA and HTIB). In HFIP, the nitrenium ions undergo intramolecular electrophilic substitution reactions to afford N-aminonitrogen heterocycles in high yields. In TFEA, spirodienones bearing the 1-azaspiro[4.5]decane skeleton are obtained by treatment of N-phthalimido-3-(4-halogenophenyl)propanamides with HTIB as a result of ipso attack of the intermediate nitrenium ion. Similarly, using PIFA in TFEA, ipso cyclization of unactivated benzenoid compounds occurs to afford spirodiene derivatives. This involves loss of aromaticity despite the absence of other activating substituents on the phenyl group.

Reductive cyclizations of hydroxysulfinyl ketones: enantioselective access to tetrahydropyran and tetrahydrofuran derivatives.

The stereocontrolled formation of cis-2,5-disubstituted tetrahydrofurans and cis-2,6-disubstituted tetrahydropyrans is achieved from enantiopure ketosulfinyl esters by reduction, Weinreb's amide, and ketone formation, followed by the reductive cyclization (Et3SiH/TMSOTf) of the resulting hydroxysulfinyl ketones. The sulfoxide-bearing heterocycles were transformed into two natural products, (-)-centrolobine (1) and both enantiomers of cis-(6-methyltetrahydropyran-2-yl)acetic acid (2).

An approach toward azacycles using photochemical and radical cyclizations of N-alkenyl substituted 5-thioxopyrrolidin-2-ones.

The photochemical reactions of a series of cyclic N-alkenyl-substituted thioimides have been examined. Irradiation of N-3-methylbut-3-enyl-5-thioxo-pyrrolidin-2-one (16) results in intramolecular [2 + 2] cycloaddition to give the highly strained thietane 17, whose structure was confirmed on the basis of its X-ray analysis. Treatment of cycloadduct 17 with dimethyl(methylthio)sulfonium tetrafluoroborate gave 2,5,6,7-tetrahydropyrrolizin-3-one (20) in good yield. Further reduction of 20 with Raney-Ni afforded 5,5-dimethylhexahydro-pyrrolizin-1-one (21). This sequence of reactions demonstrates the facility with which the 2 + 2 photoadduct can be converted into the pyrrolizidine alkaloid core skeleton. The photochemistry of the closely related N-butenyl thioxopyrrolidin-one (22) proceeded in a slightly different fashion and produced 7-mercaptomethyl tetrahydropyrrolizin-3-one (24) in 68% yield. In contrast to the above results, irradiation of the thioxaphthalimido system containing an N-cycloalkenyl group in the side chain gave rise to products derived by gamma-hydrogen abstraction from the n-pi triplet excited state. The photobehavior of the related N-3-alkenyl pyrrolidine-2,5-dithione system (62) was also studied and found to give products derived from both a 2 + 2 cycloaddition (63) and hydrogen atom transfer (64). Finally, the reaction of several N-alkenyl substituted thioimides (71-73) with tributylstannane in the presence of AIBN gave cyclized products derived from transient radical intermediates.

Unexpected migration and oxidative cyclization of substituted 2-acetophenone triflates under basic conditions: synthetic and mechanistic insights.

Oxidative ring closure of alkyl-substituted 2-hydroxyacetophenone trifluoromethanesulfonate esters (triflates) occurs upon exposure to base in anaerobic DMF at 20-90 degrees C. Alkyl substitution is required for ring closure. A migrated enol triflate product forms at lower temperature in high yield via migration of the trifluoromethanesulfonate in the unsubstituted and monoalkyl-substituted cases. The alkyl-substituted enol triflates also enter into the benzofuran-3-one ring-forming process under thermal cyclization conditions. Potential mechanistic pathways are evaluated.

Stereoselection at the steady state in radical cyclizations of acyclic systems containing one radical acceptor and two precursors in a 1,5-relationship under pseudo-first-order conditions.

The first example of a successive kinetic resolution of acyclic diastereomeric radical intermediates in a 1,5-relationship under pseudo-first-order conditions is reported. A mechanistic model involves nonselective generation of the radical intermediates followed by different partitioning of these between two different chemical pathways. The "2,5-cis" selectivity in the radical cyclization step arises from transition geometries with the substituents aligned in pseudoequatorial positions.

Alkanethioimidoyl radicals: evaluation of beta-scission rates and of cyclization onto S-alkenyl substituents.

Thioimidoyl radicals were generated by addition of alkylsulfanyl radicals to alkyl isonitriles and were characterized by electron paramagnetic resonance (EPR) spectroscopy. The beta-scissions of their C.S-C bonds were studied by variable-temperature EPR spectroscopy and the fragmentation rate constants and activation energies were calculated. The scission rates depend on the stability of the released alkyl radicals but in any case, at room temperature, the processes were fast. Data collected on similar oxyimidoyls showed that their fragmentations are slightly slower compared to those of analogous thioimidoyls. The scission rates of selenoimidoyls could not be studied by EPR and were evaluated by theoretical calculations. EPR experiments also enabled both beta-scission and 5-exo ring closure rate constants of two S-but-3-enyl-substituted imidoyl radicals to be determined, showing that cyclization prevails only at low temperatures. Density functional theory (DFT) theoretical calculations predicted that the fragmentation process preferentially occurs from the s-cis rotamers (X-C bond) of the imidoyl radicals. Thio- and seleno-imidoyls (but not oxyimidoyls) prefer s-trans conformations so that their fragmentations involve prior rotation about the X-C bond.

Synthesis of 3-substituted 4-piperidinones via a one-pot tandem oxidation-cyclization-oxidation process: stereodivergent reduction to 3,4-disubstituted piperidines.

A novel approach to 3-substituted 4-piperidinones is described. The one-pot tandem oxidation-cyclization-oxidation of unsaturated alcohols 1a-e by PCC or PCC and trifluoromethanesulfonic acid affords piperidinones 2a-e in good yield. Reduction of 2a-e by L-Selectride gives the corresponding cis 3,4-disubstituted piperidines with diastereomeric ratios of >99:1. By contrast, reduction of 2a-e by Al-isopropoxydiisobutylalane gives the trans products with diastereomeric ratios of up to 99:1.

Synthesis of diversely functionalized hexahydropyrrolo[2,3-b]indoles using domino reactions, olefination, isomerization and Claisen rearrangement followed by reductive cyclization.

Hexahydropyrrolo[2,3-b]indoles 6 were synthesized in five steps from indolin-3-one 8 by a general and efficient method, in which elements of molecular diversity were readily added onto the 3a-position of the pyrrolo[2,3-b]indole ring system. Horner-Wadsworth-Emmons reaction of 2-allyloxyindolin-3-ones 7, derived from indolin-3-one 8 and a variety of allylic alcohols, smoothly proceeded with successive Claisen rearrangement to give the corresponding 3-allyl-3-cyanomethylindolin-2-ones 15. Indolin-2-ones 15 were converted into pyrrolo[2,3-b]indoles 6 using partial hydrolysis followed by reductive cyclization with LiAlH(4). Synthesis of N-methylated pyrrolo[2,3-b]indole derivatives 23 and 26 is also described.

Synthesis of 2-alkylidenepyrrolidines, pyrroles, and indoles by condensation of silyl enol ethers and 1,3-bis-silyl enol ethers with 1-azido-2,2-dimethoxyethane and subsequent reductive cyclization.

The condensation of 1,3-bis-silyl enol ethers with 1-azido-2,2-dimethoxyethane and subsequent reductive cyclization allowed an efficient regio- and diastereoselective synthesis of a variety of 2-alkylidene-4-methoxypyrrolidines. The thermal elimination of methanol resulted in the formation of functionalized pyrroles. Similarly, 2,3,3a,4,5,6-hexahydro-2,3-benzopyrroles were prepared and transformed into 4,5,6,7-tetrahydro-2,3-benzopyrroles. In contrast, treatment of 2-alkylidenepyrrolidines with trifluoroacetic acid resulted in formation of indoles by [4 + 2] cycloaddition and subsequent extrusion of a nitrogen atom.

Ketyl-allene cyclizations promoted by samarium(II) iodide.

[reaction: see text] Samarium(II) iodide has proven to be an effective reagent for intramolecular reductive coupling reactions. Previous investigations of intramolecular ketyl-olefin coupling reactions provided carbocycles in excellent yield and good diastereoselectivity. This method has been extended to ketyl cyclizations with allenes. Substrates leading to both carbocycles and heterocycles in a selective manner are explored.

Domino "[3+3]-cyclization-homo-Michael" reactions of 1,3-bissilyl enol ethers with 1,1-diacylcyclopropanes.

The Lewis acid mediated domino "[3+3]-cyclization-homo-Michael" reaction of 1,3-bissilyl enol ethers with 1,1-diacylcyclopropanes allows an efficient one-pot synthesis of functionalized salicylates containing a halogenated side chain. A great variety of substitution patterns could be realized by variation of the starting materials and of the Lewis acid. The mechanism of the domino process was studied.

Diastereoselectivities realized in the amino acid catalyzed aldol cyclizations of triketo acetonides of differing ring size.

A study designed to assess the diastereoselectivity of the intramolecular aldol reaction of two differently sized monocyclic 1,3-diketones bearing a chiral, oxygenated side chain has been undertaken. The cyclizations were brought about under catalysis by pyrrolidine, a series of D- and L-amino acids including proline, and several proline derivatives. The levels of selectivity were found to be consistently higher with the six-membered ring system than its cycloheptane counterpart.
