Antiplatelet aggregation principles of Dendrobium loddigesii.

The MeOH extract of the stem of Dendrobium loddigesii was found to inhibit the aggregation of rabbit platelets induced by arachidonic acid and collagen. Two active compounds, moscatilin [1] and moscatin [2], were isolated. Moscatilin diacetate [3] also exhibited antiplatelet aggregation activity.

In vitro and in vivo antitumoral phenanthrenes from the aerial parts of Dendrobium nobile.

Two phenanthrenes were isolated from the aerial part of Dendrobium nobile Lindl. and their structures were identified to be 4,7-dihydroxy-2-methoxy-9,10-dihydrophenanthrene (1) and denbinobin (2), among which the former has been first isolated from this plant. These two compounds were found to be cytotoxic against A549 (human lung carcinoma), SK-OV-3 (human ovary adenocarcinoma), and HL-60 (human promyelocytic leukemia) cell lines. Compound 1 also showed antitumor activity on the life span of ICR mice intraperitoneally implanted with 1 x 10(6) cells of sarcoma 180.

Dihydroayapin, a new coumarin compound from Dendrobium densiflorum.

A new coumarin named dihydroayapin (1) together with seven known compounds were isolated from the stems of Dendrobium densiflorum. On the basis of physicochemical and spectral evidences, the structure of 1 was established as 6,7-methylenedioxy-3,4-dihydrobenzopyran-2-one.

[On the best time for harvesting Dendrobium candidum Wall. ex Lindl].

OBJECTIVE: To explore the theory on the best time for harvesting Dendrobium candidum so that the production of the herb can be directed more effectively. METHOD: Based on a component analysis in combination with mathematical calculation, a synthetical appraisal on the yield, contents of effective components and efficacy of D. candidum in different growing periods has been made, and different appraisal indexes and synthetical harvesting indexes have been worked out. RESULT: The harvesting indexes from the first year to the fourth year are as follows: 5.69, 9.54, 17.9, 22.2 (curing throat and eye diseases); 11.2, 14.6, 21.6, 17.6(strengthening immunity); 8.44, 12.1, 19.7, 19.9(the above-mentioned indexes and yield factor considered together). CONCLUSION: The best time for harvesting D. candidum is in the fourth year for curing throat and eye diseases; in the third or first year for strengthening immunity; and in autumn of the third year if the above-mentioned indexes and yield factor are to be considered together.

In vitro propagation of Dendrobium macrostachyum Lindl.--a threatened orchid.

In vitro propagation of Dendrobium macrostachyum, a threatened and endemic species was achieved through nodal explants. The nodal explants were cultured on Murashige and Skoog (MS) basal medium and MS medium supplemented with N6-benzyladenine (BA-2.22, 4.44 and 8.88 microM), Kinetin (KN-2.32, 4.65, and 9.29 microM) and Coconut water (CW, 5, 10 and 15%) individually or in combination with 2.69 microM alpha-naphthalene acetic acid (NAA). Axillary shoots were induced directly from nodal explants in medium containing BA, KN or CW. Optimal shoot induction (6 shoots/explant) was attained from nodal explants cultured on medium supplemented with 15% CW. Well developed shoots rooted at an average 5 roots per shoot in half strength MS medium devoid of any growth regulators.

[Studies on the chemical constituents of Dendrobium loddigesii rolfe].

According to IR, UV, MS and NMR spectra of the compounds isolated from the fresh stem of Dendrobium loddigesii Rolfe (a Chinese medicine), 3 constituents were identified. They are shihunidine, shihunine and dendrophenol (4,4'- dihydroxy-3,3',5-trimethoxybibenzyl). Chemical reactions showed that shihunidine was derived from shihunine during isolation. Shihunidine and shihunine were shown to be inhibitors of Na+, K+-ATPase of the rat kidney.

[Medicinal plant resources of Dendrobium in Sichuan Province].

Eleven species of Dendrobium grown in Sichuan Province were studied including their classifications and distributions. All of them are used as the merchandise Shihu. Among them, there are 3 species which are found in Sichuan for the first time and 2 new species whose medical effects are described here in. Problems about the exploitation, utilization, quality standards, etc. of these resources are preliminarily discussed. A key for the identification of all these 11 species has been compiled and presented.

[Effect of Dendrobium nobile Lindl. on gastric acid secretion, serum gastrin and plasma somatostatin concentration].

The patients with chronic superficial gastritis were perfused in the stomach with 20 g of Dendrobium nobile to observe the variations in gastric acidity output, serum gastrin and plasma somatostatin concentration. The result showed a significant increase in both acidity output and serum gastrin concentration (P < 0.01). No significant change occurred in plasma somatostatin concentration (P > 0.05).

[Studies on population difference of Dendrobium officinale II establishment and optimization of the method of ISSR fingerprinting marker].

OBJECTIVE: To establish and optimize ISSR-PCR system of Dendrobiwn officinale according to the ISSR-PCR characters of D. officinale. METHOD: The effects of ISSR-PCRs was examined by selecting primers and designing different concentrations of the factors in the ISSR-PCRs, the reliable ISSR-PCR systems for D. officinale populations researching was established by analyzing the reasons for occurrence of differential bands and optimizing reaction conditions. RESULT AND CONCLUSION: The optimal ISSR-PCR system in D. officinale was reported for the first time, and 25 15327012 microL ISSR-PCR system contained 10 x Taq buffer, 1.5 U Taq DNA polymerase, 1.2-1.8 mmol x L(-1) MgCl2, 80 micromol x L(-1) dNTP, 0.2 micro mol x L(-1) primer and 20 ng template DNA. The appropriate annealing temperature was among 52-60 degrees C. ISSR PCRs were significantly influenced by Taq DNA polymerase, template DNA quantity and annealing temperature, etc. The ISSR-PCR systems, which were established in this paper for studying D. officinale, could provide clear reliable abundant polymorphisms molecular markers and were proved suitable for studying population authentication and population molecular ecology of D. officinale.

[The influence of abscisic acid treatment on desiccation tolerance in Dendrobium candidum protocorm-like bodies].

Effect of ABA on desiccation tolerance in the protocorm-like bodies (PLBs) of D. candidum was studied. The best result was obtained while ABA precultured at 5 micromol/L for 24 h (Fig.2). PLBs vitality rate was 10 times of the control after 2 h desiccation (Table 1). Detection of cytoplasmic osmoticums showed that ABA preculture could not induce the accumulations of soluble sugar, proline, etc (Tables 3, 4 and 7). But the content of soluble polysaccharide rose conspicuously (Table 5). So we think that the accumulation of soluble polysaccharide may play an important role in desiccation tolerance induced by ABA preculture.
