Protection from lethality and behavioral incapacitation resulting from intoxication by soman (pinacolyl methylphosphonofluoridate) and treatment with atropine sulfate and 2-PAM chloride in the guinea pig, cavia porcellus.

The lethal and incapacitating effects of the toxic organophosphorus (OP) agent, soman were evaluated in guinea pigs. The protective effects of the standard therapies atropine sulfate (ATR) and pralidoxime chloride (2-PAM) in minimizing or reducing soman-produced lethality and incapacitation (evaluated using a modification of the rat conditioned avoidance procedure) were also studied. At 0.75 and 1.5 LD50 soman was extremely toxic and fast-acting; its effects appeared within five minutes, and its lethal effects occurred within the first three hours. Therapeutic combinations of ATR (64 or 128 mg/kg) and 2-PAM (25 or 100 mg/kg) protected animals from the lethality of soman, but not from its incapacitating effects. However, therapeutic treatment with ATR and 2-PAM also produced a behavioral toxicity in its own right, an effect which lasted for at least three hours in the guinea pig. This behavioral toxicity was lessened by reducing ATR dosage from 128 to 64 mg/kg, but 2-PAM dosage did not influence the behavioral toxicity of the treatment combinations within the range of dosages studied.

[Rapid detection of toxic substances in water with ToxY-PAM].

A new type of dual-channel PAM fluorometer (ToxY-PAM) was developed for highly sensitive detection of toxic substances in water samples. In conjunction with suspension of unicellular algae as the biotest material, the ToxY-PAM provided a highly rapid and sensitive bioassay test system for the detection of toxic substances in water samples, which could directly or indirectly produce the inhibition of photosynthetic activity in algae. The development process of the detection of toxic substances in water with fluorescence technique, the basic principle and operation of the ToxY-PAM were reviewed in this paper.

Protective effects of oximes HI-6 and PAM-2 applied by osmotic minipumps in quinalphos-poisoned rats.

Protective and reactivating effects of oximes HI-6 and PAM-2, combined with atropine and diazepam, were investigated in quinalphos-poisoned rats. In protective experiments, atropine and diazepam decreased acute toxicity of the insecticide 3.3 times. Later administration of a single injection of oximes led to further improvement of protective indexes which were 1.45 (PAM-2) and 1.52 (HI-6) times larger. Plasma HI-6 concentrations below 1 microgram/ml, continuously maintained by osmotic minipumps and supported by a single administration of atropine and diazepam, protected animals from 18.6 LD50 of quinalphos, while its higher concentrations (ranging from 1 to 5.4 micrograms/ml) provided markedly better protection (up to 72 LD50). Corresponding plasma PAM-2 concentrations were even more effective in overcoming toxic effects of quinalphos. PAM-2 concentrations, continuously maintained in plasma, were distinctly better in protecting and reactivating peripheral cholinesterase activity than corresponding HI-6 concentrations in the case of quinalphos poisoning. On the basis of our findings we suggest that continuous maintenance of low oxime concentrations is preferred to single oxime administration in the therapy of organophosphate intoxications.

Evaluation of the blood level duration properties of procaine benzylpenicillin in oil with aluminium monostearate (PAM).

The study of procaine benzylpenicillin in oil with aluminium monostearate (PAM) described in this paper was undertaken to devise laboratory assay methods, based on animal tests, to evaluate those properties of PAM responsible for the persistence of penicillin in the circulation, as a preferable substitute for the existing test in man. Comparative studies of a number of batches of PAM in man proved, with statistical certainty, that the duration of the blood level of penicillin which they produced varied from batch to batch. The chemical and physical properties of the batches gave inadequate characterization and were of no value in assessing material for clinical use. A blood level duration test in rabbits was developed which reflected the blood level duration properties shown in man. The test was incorporated in the Requirements for Procaine Benzylpenicillin in Oil with Aluminium Monostearate that were set up by the WHO Expert Committee on Biological Standardization to ensure the suitability of batches for clinical use. An International Reference Preparation was necessary and was established. Batches of PAM in use in the field by WHO in anti-yaws campaigns were examined by the test described and found to be satisfactory.

Pregnancy-associated beta 1-macroglobulin (beta 1-PAM): a new serum protein associated with pregnancy serum-derived immune complexes and ovarian cancer.

A new pregnancy-associated serum protein, beta 1-PAM, was identified following low pH dissociation and separation, by gel chromatography, of pregnancy serum-derived immune complexes. A sandwich-type micro-enzyme immunoassay was developed and blood from various groups of subjects examined for the presence of beta 1-PAM. In addition to pregnant women, only subjects with ovarian cancer (70%) had consistently high levels of the antigen. beta 1-PAM isolated from both pregnancy and ovarian cancer serum was shown to be antigenically identical and to share the same isoelectric point (pH 5.25) and ammonium sulphate precipitation range (1.1--1.5 M). The serum protein had a molecular weight of 1.5 x 10(6), especially when obtained from ovarian cancer patients. However, it was apparent that other molecular weight forms were present, particularly in material isolated from pregnancy serum, which were probably related to the subunit nature of beta 1-PAM and its association with immune complexes.

A comparison of the regulatory effects of human monocytes, pulmonary alveolar macrophages (PAMs) and spleen macrophages upon lymphocyte responses.

Human monocytes, pulmonary alveolar macrophages (PAMs) and spleen macrophages were concentrated by immobilization on cold insoluble globulins. These cell preparations were 90 +/- 3%, 95 +/- 1% and 83 +/- 3% esterase rich, respectively, 87 +/- 4%, 95 +/- 3% and 66 +/- 11% phagocytic and 78 +/- 3%, 79 +/- 9% and 68 +/- 5% reactive with OKM1 monoclonal antibody. Spleen macrophages differed from the other two cell preparations in that significantly fewer reacted with 61D3 or 63D2 monoclonal antibodies. Monocytes and PAMs promoted the mixed leucocyte response by autologous lymphocytes when added at low concentrations, but suppressed this response at high concentrations. Spleen macrophages only promoted the mixed leucocyte reaction but were required in much higher numbers than either monocytes or PAMs for optimal promotion. Likewise, the added presence of monocytes or PAMs in high numbers suppressed Ig synthesis stimulated with pokeweed mitogen, while spleen macrophages were not suppressive in this system. This study shows that the distribution of macrophages that differ in their regulatory effects upon lymphocyte responses varies in different tissues. The human spleen is deficient in macrophage related suppression.

HI-6 and 2-PAM in sheep: pharmacokinetics and effects on muscle tissue following intramuscular injection.

The pharmacokinetics of 2-PAM, a component of the current nerve agent antidote therapy for U.S. military forces was compared to the pharmacokinetics of another acetylcholinesterase reactivator HI-6. Additionally, the effects of these compounds on muscle tissue following intramuscular injection was examined. Plasma concentrations of the oximes were determined by HPLC. Plasma concentration-time profiles for both oximes fit a one-compartment open model with first-order absorption and elimination. The results demonstrate that the half-time of absorption of HI-6 was significantly higher than that for 2-PAM. Musculoirritancy was assessed on the basis of quantitative histological examinations of the injection sites and by the measurement of serum creatinine phosphokinase. Comparison of the scores from the histological sections demonstrate no difference between the two oximes. Serum creatinine phosphokinase values were elevated following injections of HI-6, but were not consistently elevated following the 2-PAM injections.

Fluorescence lifetime imaging in an optically sectioning programmable array microscope (PAM).

BACKGROUND: The programmable array microscopes (PAMs) are a family of instruments incorporating arbitrary control of the patterns of illumination and/or detection. The PAM can be used in sectioning and nonsectioning modes, thereby constituting a useful platform for fluorescence lifetime imaging. METHODS AND RESULTS: We used a PAM for acquisition of optically sectioned and widefield fluorescence lifetime images, in which contrast was increased predominantly by suppressing out-of-focus light contributions. We simulate, display, and discuss the effects of blurring and fluorophore heterogeneity on lifetime imaging in widefield and confocal configurations. CONCLUSION: Sectioning improves the quality of lifetime images of samples with multiple fluorophores or spatially varying Förster resonance energy transfer.

The origin of the genetic code cannot be studied using measurements based on the PAM matrix because this matrix reflects the code itself, making any such analyses tautologous.

Freeland et al. (Mol. Biol. Evol. 2000 a, 17, 511--518) have recently used a transformation of the PAM 74-100 matrix to study the level of optimization reached during genetic code origin. Since the PAM matrix counts the amino acid substitutions that occurred in families of homologous proteins during molecular evolution and as this process is mediated by the genetic code structure itself, it could be that the influence of the code on this matrix is such as to make any conclusion insignificant. As will be shown in the present paper, the transformation of the PAM matrix is affected in a non-marginal way by the organization of the genetic code and, thus, renders the analysis of Freeland et al. tautologous. Although, under the hypothesis of a highly optimized genetic code, some correlations may be expected between a measurement of similarity between amino acids and the genetic code structure, no certain conclusions can be drawn for the measurement used by Freeland et al.

Senile plaque-like structures: observation of a probably unknown type of senile plaque by periodic-acid methenamine silver (PAM) electron microscopy.

Numerous diffuse senile plaque-like structures (SPLSs) were found in the cerebral cortex from cases with dementia of the Alzheimer type by means of the methenamine-Bodian method. SPLSs varied in shape and size. They were never recognized in the original Bodian, PAS and Congo red preparations, but were positive with anti-beta-protein immunostaining and periodic-acid methenamine silver (PAM) methods, which are thought to specifically stain amyloid substance. With PAM electron microscopy, we found sparse aggregations of amorphous, often ramified, structures with fine granular silver deposits in SPLS. Routine electron microscopic examination on the same portion where SPLS were confirmed by PAM electron microscopy revealed amorphous, partially fibrous structures. These structures might be amyloid or amyloid-precursor substance. In SPLSs only a few degenerated neurites and astrocytic processes with glycogen granules were seen. We consider SPLSs to be a kind of senile plaque.

A comparative study between the PAM and the laser interferometer in cataracts.

We conducted a prospective study on 35 consecutive eyes undergoing cataract extraction to compare the Guyton-Minkowski Potential Acuity Meter (PAM) and the Laser Interferometer (LI) in determining potential visual acuity. The eyes were divided into: group A, which had good visualization of optic disc details, and group B, whose media allowed only disc outline or less to be seen. We found that in group A eyes, both instruments were reliable (with accuracies of 94% for the PAM and 88% for the LI). However, in group B eyes (advanced cataracts), both instruments were unreliable (accuracies of 33% for the PAM and 53% for the LI). Interestingly, in a subgroup of high myopes (axial length greater than 29 mm) with moderate cataracts and poor visual acuties, both instruments were very reliable and helpful in determining how much of the vision loss was due to the cataracts. We therefore strongly recommend potential acuity testing in high myopes with moderate cataracts and poor visual acuity.

Synthesis and biological activity of conformationally controlled 2-PAM derivatives.

A series of conformationally controlled 2-PAM derivatives were prepared from 2-acetylpyridine and 2,3-pyrido[b]cycloalkenones in two steps and their reactivities towards parathion poisoned AChE were evaluated. The most planar 2,3-pyrido[b]cyclohexanone oxime methiodide showed an activity comparable to 2-PAM implying E-syn is that the most active conformation of 2-PAM in the biological system.

PAM fluorometry in the determination of the sensitivity of Chlorella vulgaris, Selenastrum capricornutum, and Chlamydomonas reinhardtii to copper.

The pulse-amplitude-modulation (PAM) fluorometric method was used in the past as a sensitive and rapid method for assessing toxic effect of pollutants in plants. Here, we used the advantages of this method to evaluate the difference in sensitivity of Chlorella vulgaris, Selenastrum capricornutum, and Chlamydomonas reinhardtii to copper toxicity. We found by using the PAM-fluorescence parameters (phiM, phi'M, QP, and QN), that C. reinhardtii is the most sensitive species, followed by S. capricornutum and C. vulgaris. Indeed, the copper 5-, 48-, and 96-h EC50 for phiM, phi'M, and QP vary between 11 and 34 microg/L for C. reinhardtii, while for S. capricornutum this value is between 25 and 50 microg/L. However, the same parameters obtained from C. vulgaris did not show susceptibility to copper, even at very high concentrations (100 microg/L). In this study, the estimated 5-h EC50 for C. reinhardtii and S. capricornutum are lower than the 24-h EC50 reported earlier for these species with other bioassays, indicating that the use of PAM fluorometry in copper bioassay provides a more sensitive method than the other biotests in aquatic toxicology.

[Management of conjunctival malignant melanoma associated with primary acquired melanosis (PAM) using 0.02% mitomycin C eyedrops].

BACKGROUND: The therapy of malignant diseases of the conjunctiva with local chemotherapy is an extension of the therapeutic options in this field. We report on our experience in the therapy of malignant melanomas of the conjunctiva associated with primary acquired melanosis (PAM). METHODS AND PATIENTS: Between March 1998 and April 2001, 13 patients with malignant melanoma of the conjunctiva associated with PAM (6 female; 7 male; mean age 57+/-13 years) were treated with local chemotherapy. The tumor was classified as stage pT2 (pN0, pM0) in seven patients, stage pT3 (pN0, pM0) in three patients and in the remaining three patients the lid was involved in the malignant process (pT4, pN0, pM0). Local chemotherapy (mitomycin C 0.02% eyedrops 5 times a day) was applied after incisional biopsies in 2 cycles for 14 days with a 14-day break. In 4 patients a third cycle was included. RESULTS: Regression of the tumor was observed after completion of the therapy in all cases. Severe ocular or systemic secondary effects were not seen in our patients. Nine patients were without recurrence within the follow-up time. In three patients, a recurrence of the disease was observed. In these cases, the eyelid was involved in the process. CONCLUSION: Local chemotherapy with mitomycin C is a useful option in the treatment of malignant melanomas of the conjunctiva associated with PAM if the tumor stage is pT3 or less. From our point of view the combination with incisional biopsy is of great benefit. Prognosis of conjunctival malignant melanomas involving the lid margin (pT4) is poor.

Simultaneous collection of rapid chlorophyll fluorescence induction kinetics, fluorescence quenching parameters, and environmental data using an automated PAM-2000/CR10X data logging system.

The PAM-2000 portable chlorophyll fluorometer represents one of the first commercially available instruments utilizing the Pulse Amplitude Modulation (PAM) measurement principle, and has become a widely used platform for measuring chlorophyll fluorescence in a wide range of study systems. In this paper, we describe a new method for externally driving and gathering data from the PAM-2000, a method that allows the user to execute a pre-defined user run (or runs) and capture (1) rapid induction kinetics (at 2 ms frequency) during all saturating pulses, (2) measures of F, F(o), F(o)', F(m), and F(m)' associated with those same pulses, and (3) changes in fluorescence F at user-defined intervals between pulses, for the entire user run, with all data compressed into a single, manageable data logger file. Practically, the method makes possible, for example, a post-hoc evaluation of the appropriateness of saturation pulse lengths and intensities during a user run. More importantly it captures, during entire user runs, the varied information contained in slow changes in fluorescence between saturating pulses, as well as rapid induction kinetics, quenching coefficients and quantum yields all gathered simultaneously from all saturating flashes.

Applying Pulse Amplitude Modulation (PAM) fluorometry to microalgae suspensions: stirring potentially impacts fluorescence.

The use of microalgae suspensions in PAM-fluorometers such as the Water-PAM (Walz GmbH, Germany) presents the problem of maintaining a homogeneous sample. The Water-PAM is marketed with an optional accessory for stirring the sample within the cuvette while in the emitter-detector (ED) unit. This stirring device can help to prevent cells from settling out of suspension over the time-course of chlorophyll-a fluorescence measurements. The ED unit was found to provide a vertically heterogeneous light environment and, therefore, cells within a single sample can exist in different quenched states. Enhancing cell movement by stirring was found to substantially influence measured fluorescence yield while performing induction curve and rapid light curve analyses. This is likely to result from relatively unquenched cells outside the main light-path moving into a higher light region and thus emitting disproportionately more fluorescence than quenched cells. Samples containing cells with high sinking rates or motile species may encounter similar (but reduced) problems. This effect can be mitigated by: (a) reducing analysis time to minimise the distance cells can sink/swim during the measurement procedure and avoiding the necessity of stirring; (b) limiting the proportion of sample outside the light path by minimising sample volume or; (c) by activating the stirrer only for short periods between saturation pulses and allowing enough time after stirring for quenching to stabilise before activation of the saturation pulse. Alternatively, modifications to the instrument providing a vertical dimension to the LED-array could resolve the issue by providing a more homogeneous light environment for the sample.

Amidating processing enzyme complex for bioactive peptides (PAM) shows differences in specific activity and form in secretory granules isolated from the proximal and distal parts of the hypothalamo-neurohypophyseal tract in rats.

In rats the PAM specific activity in hypothalamic and neurohypophyseal extracts was 0.58 +/- 0.8, respectively 1.78 +/- 0.6 nmol.mg prot.-1 x h-1 (n = 5). PHM specific activity in the soluble part of the granules was higher in the neurohypophyseal than in the hypothalamic granules, and the fraction of total PHM and PAL present in the soluble part increased with the distance from the hypothalamus from some 45% to approx. 85%. Western blots of membrane and soluble granule fractions showed prevalence of higher mol. wt. forms in hypothalamic granules. It would appear that higher mol. wt. forms of PAM are processed by proteolytic enzymes during transport in the neuron and that non-neural cells in the neurohypophysis have a considerable PAM activity.

Lethal photosensitization by endogenous porphyrins of PAM cells--modification by desferrioxamine.

The effects of exogenous delta-aminolevulinic acid (ALA) on cultured PAM 212 cells were investigated. PAM cells exposed to ALA produce an excess of products of the heme biosynthesis pathway within hours. The endogenous porphyrins render the cells photosensitive in a time-, ALA dose- and irradiation-dependent manner. Independently of the ALA-induced photosensitized processes, ALA itself reduces the proliferation rate of PAM cells during the exponential growth phase. Iron deprivation by addition of the chelating agent desferrioxamine (df) accelerates the photosensitizing process, and thus makes it more efficient at lower ALA concentrations. The effects of df were compared with the effects of manganese-df and iron-df. The results indicate that iron trapping is the most important factor for the potentiation of ALA-stimulated photodynamic sensitization as well as for the dark toxicity. Iron complexing agents may thus be used to optimize ALA effects in the photodynamic treatment of cutaneous neoplasms with endogenous porphyrins.
