Results of IAEA intercomparisons in the frame of indirect methods for assessing intakes of radionuclides.

The IAEA safety activities to support the assessment of occupational exposure due to intake of radionuclides through a comprehensive set of Safety Guides, Safety Reports and other documents will soon be completed. The future IAEA activities in this field will focus more on training and international intercomparisons. In recent years extensive development in measurement techniques, phantoms and computational tools have occurred. Determination of the radionuclide activity in the body or excreta, the intake and the resulting internal dose can, therefore, be approached in many different ways, depending on the amount and quality of the data, the skill of the dosimetrist, computational tools available, the assumptions made and the methodology used. Thus, it is important for laboratories involved in internal dosimetry to undergo performance-testing procedures to demonstrate the correctness of the methods applied and also the consistency of their results with those obtained by other laboratories. Several intercomparison exercises were organized by the IAEA on the determination of radionuclides in human urine samples. These previous intercomparison exercises revealed significant differences in the approaches, methods and assumptions, and consequently in the results. This underlined the importance of this kind of intercomparison programmes as a key element of the harmonization process. The purpose of this paper is to present the current and future IAEA activities in support of assessment of occupational exposure due to intakes of radionuclides by organizing intercomparison runs focused on indirect methods for assessing intakes as well as the recommendations for setting up a Quality Management Systems for technical services in radiation safety.

Abnormal high natural radium concentration in surface waters.

Unexpected high 228Ra concentrations, up to 2 Bq 1(-1), were found in waters of a coastal lagoon close to a monazite sand separation plant. Due to their use as process waters in this plant, the initial supposition was a contamination related to its operation. However, it was concluded that these abnormal radium concentrations had a natural origin, springs at the lagoon head area with high 228Ra and 226Ra concentrations. The strong relationship among radium and light rare-earth elements (LREEs), the observed 228Ra/226Ra activity ratio and the rare-earth element pattern in the spring waters suggested that monazite is the main source of nuclides for water, indicating the disturbance of monazite chemical stability by the combined effects of low pH and high salinity. Both factors combined allow relatively low mobility of thorium, but, on the other hand, a relatively high mobility of radium and LREEs.

The suitability of transfer coefficients used for stochastic calculations in radioecology.

In radioecology, stochastic calculations are used to an increasing extent. It is shown that transfer coefficients which are defined as concentration ratios are not suited for stochastic calculations, at least for essential elements and their antagonists. The reason is that essential elements are regulated in plants and animals whereby their concentrations vary within narrow limits, rather independent of the concentrations in the source compartment. For a more accurate description of the transfer of radionuclides, it is proposed to include the concentrations of the essential elements in the source and target compartments into the transfer equation which leads to the specific activity model. This model has already been proposed by Comar et al. in 1956. It seems to work accurately for the transfer into animals and their products as well as into plants. Examples of its applicability are presented and discussed. Furthermore stochastic calculations seem to be more reasonable with this model.

2'-O-alkyloligoribonucleotides, synthesis and applications in molecular biology.

Oligo(2'-O-alkylribonucleotides) have been synthesized in which alkyl is methyl, allyl and butyl. The various phosphoramidite monomers of 2'-O-alkyl uridine, cytidine, adenosine, guanosine, inosine and 2,6-diaminopurine riboside have been synthesized from a minimum of key intermediates. Extra protection of the lactam function in uracil and hypoxanthine proves useful. The high stability of the oligomers combined with incorporation of non-radioactive reporter groups such as fluorophores, biotin and 2,4-dinitrophenylamino (DNP) moities renders them as excellent antisense tools for studying RNA processing, for locating and visualising RNA and RNP complexes in cells, for examining splicing complexes by electron microscopy and for the affinity chromatography of RNA or RNP complexes.

Expression of decorin mRNA in the skin of patients with ossification of the posterior longitudinal ligament.

Ossification of the posterior longitudinal ligament (OPLL) is an intractable disease developing into severe myelopathy and often accompanied by ossification of several other spinal ligaments and articular ligaments. Therefore, it is highly probable that predisposition to systemic ossification underlies the onset of this disease. Terayama et al(1981) reported that the nuchal skin of OPLL patients tended to be tougher than that of healthy individuals. Decorin is a component of the extracellular matrix which antagonistically regulates the action of Transforming growth factor-beta (TGF-beta). Imamura et al(1995) demonstrated immunohistochemically that decorin increased in the epidermis of the nuchal skin of OPLL patients. This suggests an abnormal expression of the extracellular matrix in the skin of OPLL patients. In the present study, in situ hybridization with non-radioactive synthetic oligodeoxynucleotide probes revealed an enhanced expression of decorin mRNA in the epidermal keratinocytes of OPLL patients. The increased expression of decorin mRNA in the epidermis of OPLL patients may be interpreted as reflecting abnormalities of the matrix associated with ossification of the ligaments. Studies on the role of decorin in ossification will contribute to clarification of the pathophysiology and pathogenesis of OPLL.

[Kinetics of pulmonary defense in deep lung (author's transl)].

The association of different experimental methods using radioactive particulate aerosols with bronchial washing and morphometric techniques reveals the importance of alveolar macrophages in antibacterial defense and the interest of their quantitative evaluation. The study of the kinetics of pulmonary phagocytic systems has shown a "capillary compartment", which consists of a reserve of monocytes which are temporarily present in the lung and in transit through it. This reserve represents 4.6% (sigma=1.4) of the cells from deep in the lung and is much larger than the systemic monocytic pool. 0.5% of these monocytes are in a phase of DNA synthesis. This capillary compartment must be clearly separated from interstitial phagocytes, the number of which is insignificant under physiological conditions. The phagocytic relay by polynuclear elements appears during bacterial aggression and is interdependent with macrophagic population. The particularities of the immune defenses of the distal parts of the lung are stressed especially in comparison with systemic defenses.

Light and electron microscopic characterization of the proliferative response induced by tobramycin in rat kidney cortex.

Administration of aminoglycoside antibiotics is frequently associated with tubular necrosis which can eventually lead to renal dysfunction. Previously, we have shown that renal tissue injury due to aminoglycoside nephrotoxicity elicits a process of tissue repair characterized by stimulation of cell proliferation. The present study was undertaken to examine both quantitatively and qualitatively the cell proliferation associated with renal tissue repair. Female Sprague-Dawley rats (180-200 g body weight) were treated ip for 10 days with various doses of tobramycin (10, 20, or 50 mg/kg twice daily). Each animal received 200 microCi [3H]thymidine 1 hr before sacrifice to evaluate the extent of cell proliferation in renal cortex. The rate of DNA synthesis in renal cortex was estimated by measuring the specific radioactivity of the nucleic acid. The frequency and localization of S-phase cells in cortex tissue were determined on paraffin and plastic tissue sections processed for histoautoradiography. In addition, the ultrastructure of proliferating cells was characterized by electron microscopic examination of consecutive ultrathin sections. An excellent correlation (r = 0.993) was found between the rate of DNA synthesis and the frequency of S-phase cells evaluated in rats receiving various doses of tobramycin. The stimulation of cell proliferation involved mostly proximal tubular cells and interstitial cells. The latter cells had the ultrastructural appearance of fibroblasts at various stages of differentiation. Similarly, S-phase cells in proximal tubules were either fully differentiated epithelial cells or immature elements. Taken together, the present experimental data illustrate the capacity of the kidney to trigger complex tissue reactions in response to nephrotoxic injury.

Determination of detection efficiency curves of HPGe detectors on radioactivity measurement of volume samples.

An efficiency calibration method has been developed to measure radioactivity of volume samples with HPGe detectors. The method has two main procedures. One is a point-source calibration at a representative point near the detector. The position of the representative point is determined by Monte Carlo simulations. The other is correction for absorption by a sample matrix and a sample container. This paper presents application of the method for obtaining efficiency curves for volume samples in the energy range from 22 to 1836 keV. In addition to the standard volume sources of cement and water matrices, measurement was made with some kinds of environmental samples that are often used in environmental monitoring for radiation control works. The composition of the environmental sample was determined by elemental analysis. For these samples, measured absorption correction factors were compared with those by the Monte Carlo simulations. As a result, detection efficiency curves were obtained with good accuracy for variety types of volume samples.

Molecular epidemiology of Shigella flexneri in a diarrhoea-endemic area of Lima, Peru.

A year-long community-based study of diarrhoeal diseases was conducted in Canto Grande, a periurban community in Lima, Peru. In 109 (34%) houses out of 323 that were visited, at least one individual was detected with shigellosis. The frequency of the 161 shigella isolates obtained was as follows: 117 S. flexneri (73%), 21 S. boydii (13%), 15 S. dysenteriae (9%), and 8 S. sonnei (5%). Using a non-radioactive ipaH gene probe as a molecular epidemiological tool, a total of 41 S. flexneri strains were shown to be distributed in 25 intra-family comparisons by pairs (icp). Further subdivision, based on a comparison of the serotype, plasmid profile, antibiotic resistances and ipaH hybridization patterns indicated that Group I, with 11 icp (44%), had strains that were identical. Group II with 8 icp (32%), had strains that were different and Group III with 6 icp (24%), had strains with the same serotype and identical ipaH profiles but with differences in other markers. This data indicates that a diversity of shigella clones circulated in this community resulting from both clonal spread and horizontal transfer of genetic elements. Furthermore, ipaH profiling of isolates can be used not only to differentiate between closely related shigella strains but also with other parameters, help to understand the dynamics of the generation of new clones of pathogenic bacteria.

Environmental isotopes as a useful tool for studies at mixed uranium mill tailings sites.

Groundwaters in the area of a mixed landfill (domestic waste above uranium mill tailings) in Dresden (Saxony, Germany) were investigated for their isotope signatures to distinguish between different groundwater types. To determine between the two contamination sources (waste and uranium mill tailings) a multi parameter interpretation was done using both, the main hydrochemical parameters the radionuclides 234U, 238U, 226Ra and 222Rn as well as the environmental isotopes of the elements hydrogen, oxygen, sulphur and carbon. The seepage water from the landfill shows higher delta34S, delta18O and tritium values as the inflow. The tritium values give an idea about water movement in the dump and mean residence time of the groundwater. The water in the dump shows varying delta13C values which indicate different processes occurring in the dump.

Methanococcus vannielii selenium metabolism: purification and N-terminal amino acid sequences of a novel selenium-binding protein and selenocysteine lyase.

Selenium is an essential component of several enzymes and proteins in a number of methane-producing archae. Information concerning accessory proteins that function in selenium transport processes, however, is limited. A novel selenium-binding protein with a potential transport role and a selenocysteine lyase that serves as a selenium delivery protein are present in Methanococcus vannielii. The selenium-binding protein was purified from extracts of 75Se-labeled cells. Although there was gradual loss of 75Se during purification, the isolated protein still could be detected as a radioactive 42 kDa species on native PAGE gels and as a 33 kDa species on SDS PAGE gels. The N-terminal amino acid sequence of residues 1 - 63 of the protein was determined by automated Edman degradative analysis. The only homologous sequence detected in the recorded data base was that of a gene encoding an unknown protein located in the genomic sequence of Methanococcus maripaludis. Cloning and expression of the corresponding gene from M. vannielii are described in a manuscript in press (Self et al.). A 47 kDa selenocysteine lyase isolated from M. vannielii extracts exhibited sequence homology to the NIFS family of proteins that transport sulfur. The purified selenocysteine lyase catalyzed the elimination of an elemental form of selenium from free selenocysteine and delivered this selenium directly to selenophosphate synthetase. The synthetase converted the selenium to selenophosphate in an ATP-dependent reaction.

Sodium iron EDTA [NaFe(III)EDTA] as a food fortificant does not influence absorption and urinary excretion of manganese in healthy adults.

NaFe(III)EDTA is a promising iron (Fe) compound for food fortification programs because of its high Fe bioavailability from meals containing dietary inhibitors of Fe absorption such as phytic acid. However, this Fe compound is not currently used in any large-scale fortification program because of concern over its possible negative influence on the metabolism of other essential minerals or its possible influence on the absorption of potentially toxic elements, such as manganese (Mn). In this study, Mn absorption and urinary excretion were studied in adults after intake of an Fe-fortified weaning cereal labeled with 54Mn. In a crossover design, the fortification of the weaning cereal with Fe as NaFeEDTA was compared with ferrous sulfate. Manganese absorption was measured by extrapolation from whole-body retention data 10-30 d after intake, and urinary excretion of 54Mn was measured over 7 d. No significant differences in 54Mn absorption or urinary excretion were found; 1.1 +/- 0.15 and 0.91 +/- 0.35% of the ingested dose was absorbed from the cereal fortified with NaFe(III)EDTA and FeSO4, respectively. Urinary excretion of 54Mn was very low; the total radioactivity in urine represented 1.1 +/- 0.55% of the absorbed dose with NaFe(III)EDTA and 0.72 +/- 0.53% of the absorbed dose with FeSO4. Until now, Fe-fortification programs have met with only limited success. The introduction of NaFeEDTA as a food fortificant could be a useful tool to provide bioavailable Fe to vulnerable groups in the population and thus aid in combating Fe deficiency.

[Effect of Hypericum perforatum extract on in vitro labelling of blood elements with technetium-99m and on biodisponibility of sodium pertechnetate in Wistar rats].

PURPOSE: To evaluate the effect of a hiperico extract (Hypericum perforatum) on the labeling of blood elements with technetium-99m (99mTc) and in the bioavailability of the radiopharmaceutical sodium pertechnetate in Wistar rats. METHODS: Blood (heparinized) withdrawn from Wistar rats is incubated with a hiperico extract, with a stannous cloride and with 99mTc, as sodium pertechnetate (99mTcONa). Plasma (P) and cells (C) are isolated by centrifugation. Samples of P and C are also precipitated with trichloroacetic acid (TCA 5%) and soluble (FS-P; FS-C) and isoluble (FI-P; FI-C) fractions are separated. In the bioavailability analysis, the extract or NaCl 0.9% solution is administrated into Wistar rats (gavage) during 15 days. Sodium pertechnetate was administered and after 10 min, the animals are sacrificed, the organs were isolated, the radioactivity determined in a well counter, and the percentages of radioactivity per gram (%ATI/g) in the organs are calculated. RESULTS: The hiperico extract decreasedsignificantly (P < 0.05) the %ATI in the cells, cellular insoluble fraction and plasma insoluble fraction. The biodistribution was significantly (P < 0.01) decreased in bone, muscle and thyroid and significantly (P < 0.05) increased in pancreas. CONCLUSION: The analysis of the results indicates that in studied extract should have substances that should oxidize the stannous ion, reducing the fixation of the 99mTc on the erythrocytes and plasma and cellular proteins. Moreover, it could produce metabolic alterations with influence in the uptake of the radiopharmaceutical 99mTcO4Na in bone, muscle, pancreas and thyroid.

Performance of an array of large-volume germanium detectors for whole-body counting.

An array of four large-volume coaxial germanium detectors was installed in a shielded room for the in vivo measurement of gamma emitters in the body. All four detectors are in a single 50-L dewar of liquid nitrogen and are positioned underneath the supine subject with the axes of the detector elements parallel with the length of the body. This orientation gives approximately 15% more counting efficiency than having the axes of the detectors perpendicular to the body length. The detectors have resolutions that are less than 2 keV with an average of 1.86 keV for the 60Co 1.33-MeV gamma-ray peak. The efficiencies of the four detectors range from 59% to 77% of a 7.62- x 7.62-cm (3- x 3-in.) NaI (Tl) detector. The detectors are interfaced to a multichannel analyzer such that spectra from the detectors are stored both as a sum of all counts from the four detectors and as a spectrum from each detector. The sensitivity of the germanium array for measuring radioactivity in the body exceeds that of multiple large NaI (Tl) detectors for measuring single radionuclides in the body and is much superior for measuring internal depositions of radionuclide mixtures. Another major advantage of this system compared to NaI(Tl) systems is that the results can be calculated by computer analysis with minimal human interpretation. The sensitivity of the array configuration is described and applications of the array in other configurations, such as that used for measuring the very small quantities of 24Na induced in the body during air travel, are also discussed.

Lipid synthesis in chick epidermis.

Lipid synthesis in newborn chick epidermis was studied by electron microscopic autoradiography after injection of tritiated palmitate. The labeled lipid product in the tissue was identified as mostly triglyceride. At the earliest time after injection (6 hr), the radioactive precursor was taken up by all viable cells of the epidermis. Grain density was heaviest over basal cells, moderate over spinous cells, and slight over granular cells; thus lipid incorporation is highest in the basal and spinous regions of the chick epidermis. As time after injection progressed, the increasing amounts of grains over the granular and horny cells and decreasing amounts over the basal and spinous cells reflected the continuous upward displacement of cells from one layer into the next. From the distribution of silver grains within the epidermal cells, it has been concluded that, with the passage of time, triglycerides synthesized by the epidermal cells were mainly located in lipid droplets. The numerous grains associated with the elements of the endoplasmic reticulum indicated that this organelle is involved in aggregating triglyceride molecules into lipid droplets. The fact that grains were seen within the horny cells indicated that part of the horny cell consists of lipid probably derived from the lipid droplets retained by the cells during keratinization.

The role of PopCel1 and PopCel2 in poplar leaf growth and cellulose biosynthesis.

Poplar calli transcribed two cellulase (endo-1,4-beta-glucanase) genes, PopCel1 and PopCel2, whose mRNAs were differentially located in the growing leaves of poplar during cell wall synthesis. Histochemical and RT-PCR analyses of promoter-GUS fusion gene activities in transgenic poplar demonstrated that PopCel1 promoter-derived GUS activity was localized in the petiole and leaf veins, whereas PopCel2 was confined to mesophyll cells and disappeared from the tip during the development of leaves. Autoradiography of the leaf showed that the radioactivity of [14C]sucrose incorporated into cellulose corresponded to the combination of the sucrose-induced tissue-specific patterns of PopCel1 and PopCel2. Interestingly, 2,6-dichlorobenzonitrile (DCB) not only inhibited the incorporation of the radioactivity into cellulose, but also repressed the induction of both cellulase genes. Suppression of cellulases by expression of PopCel1 antisense cDNA or co-suppression of PopCel1 mRNA by overexpression of PopCel1 sense cDNA reduced leaf growth. Therefore, we came to the conclusion that PopCel1 and PopCel2 probably function to promote leaf growth in poplar by the endohydrolysis of 1,4-beta-glucan.

Comparative analysis of heavy metal and radionuclide contaminants in Black Sea green and red macroalgae.

A comparative analysis about the accumulation of heavy metal, natural and technogenic radionuclides from the Black Sea environment in different Bulgarian coastline regions is made. The possibilities to use Chlorophyta and Rhodophyta algae phylums as bioindicators in marine environment are investigated. Environmental contamination in the Black Sea alga species (green and red) was studied from 1992 to 2003. Sampling sites were selected to cover the whole coastal region. Low level gamma spectrometry was used to determine the natural and technogenic nuclide concentrations. The heavy metal concentrations were measured by Atomic Absorption Spectrometry (AAS). The obtained data show that radionuclide and metal concentrations depend on the macrophyte type. Tendencies in the concentration of pollutant variations during the studied period are examined and all data give information about different macrophytic species' ability to accumulate certain elements from one and the same sampling location. All obtained results show that use of macroalgae in marine environmental monitoring reduces the need for complex studies on chemical speciation of aquatic contaminants and makes algae valuable indicators for seawater quality assessment. All data show the lack of serious pollution along the Bulgarian Black Sea coast.

Analysis of the effects of CRABP I expression on the RA-induced transcription mediated by retinoid receptors.

The involvement of cellular retinoic acid-binding protein I (CRABP I) in the RA signaling was investigated by examining its effects on the interaction of retinoid receptors (RARs and RXRs) with RA-response elements (RAREs) as well as on the RA-induced transcription mediated by retinoid receptors. Analysis of the expression of mouse CRABP I from a cDNA expression plasmid in COS-1 cells revealed that this protein was about 5-fold more abundant in cytosol than in nuclei. The identity and the localization of CRABP I in the cytoplasm as well as the nuclei were also confirmed by the immunoperoxidase staining of the transfected COS-1 cells with CRABP I-specific antibody. When the nuclear extract containing a 10-fold molar excess of CRABP I was incubated with RAR alpha extract in the presence of [3H]RA and resolved on an FPLC size-exclusion column, a 20% decrease in the bound radioactivity in the RAR alpha fraction was accompanied by a proportional increase in the CRABP I fraction. In contrast, the addition of CRABP I did not significantly affect the interaction of RAR alpha or RAR alpha-RXR alpha heterodimers with RAREs. Moreover, the coexpression of CRABP I in CV-1 cells did not markedly inhibit or enhance the transcription activated by RARs and RAR alpha-RXR alpha heterodimers under RA concentrations ranging from 10(-10) to 10(-6) M. These results demonstrate that CRABP I, while it might be important for RA homeostasis, is not directly involved in the retinoid receptor-mediated RA-signaling pathway.
