We directly compared risk factors between 214 histologically confirmed melanomas (CMM), 215 basal-cell carcinomas (BCC) and 139 squamous-cell carcinomas (SCC) in a multiple case-case-control study with 349 controls from patients without dermatological disease admitted to the same hospitals. Subjects with fair hair had a significant risk increase for all types of tumours at a comparable level (OR(adj) for blonde hair: CMM 2.3; SCC 2.4; BCC 2.3). The effect of pale eyes was significant and similar for CMM and BCC (OR(adj) 2.6). Intermittent sun exposure measured in hours spent at beach during holidays was significant for both CMM (OR(adj) 2.6 for more than 7000 lifelong hours) and BCC (OR(adj) 2.1 for more than 7000 lifelong hours), while SCC exhibited a significant risk increase for chronic exposure to sunlight measured in hours of outdoor work (OR(adj) 2.2 for more than 6000 lifelong hours). In the case-case comparison using a multinomial logistic regression model, we found a statistically significant risk difference for pale eyes, and number of naevi in the CMM group, compared to other skin cancers. For intermittent sun exposure, there was a significant risk difference of BCC when compared to the risk of SCC. Factors influencing risk of SCC are different, with chronic exposure to sun playing a major role in causing this type of carcinoma.
The occuloalbinism 2 (OCA2) gene, localized at 15q11, encodes a melanosomal transmembrane protein that is involved in the most common form of human occulo-cutaneous albinism, a human genetic disorder characterized by fair pigmentation and susceptibility to skin cancer. We wondered whether allele variations at this locus could influence susceptibility to malignant melanoma (MM). In all, 10 intragenic single-nucleotide polymorphisms (SNPs) were genotyped in 113 patients with melanomas and in 105 Caucasian control subjects with no personal or family history of skin cancer. By comparing allelic distribution between cases and controls, we show that MM and OCA2 are associated (p value=0.030 after correction for multiple testing). Then, a recently developed strategy, the 'combination test' enabled us to show that a combination formed by two SNPs was most strongly associated to MM, suggesting a possible interaction between intragenic SNPs. In addition, the role of OCA2 on MM risk was also detected using a logistic model taking into account the presence of variants of the melanocortin 1 receptor gene (MC1R, a key pigmentation gene) and all pigmentation characteristics as melanoma risk factors. Our data demonstrate that a second pigmentation gene, in addition to MC1R, is involved in genetic susceptibility to melanoma.
PURPOSE: To report the therapeutic outcome of transpupillary thermotherapy (TTT) for subfoveal choroidal neovascularizarion (CNV) in brown retina using a diode-laser with the setting of lower energy level compared to the previous studies on light-pigmented Caucasian patients. METHODS: A total of 19 subfoveal CNVs in 18 patients were treated with TTT. The power of diode-laser was set 160 mW for 1.2 mm beam, 270 mW for 2.0 mm beam, and 400 mW for 3.0 mm beam, and the laser was delivered for 1 min through a slit-lamp mounted-delivery system. Patients were followed up for a mean of 8.8 months (4-12 months). Visual acuity and the fundus change as judged by funduscopic examination and simultaneous fluorescein and indocyanine green angiography were evaluated. Visual acuity was measured by a Japanese standard Landolt visual acuity chart and converted to logarithm of the minimal angle resolution (log MAR) visual acuity for statistical analysis. Improvement or decline in vision was defined as change of more than 0.2 in log MAR visual acuity. RESULTS: In eyes with minimally classic or occult only CNV, visual acuity improved in two eyes (18%) stabilized in seven eyes (64%) and worsened in two eyes (18%). In eyes with predominantly classic CNV, visual acuity improved in two eyes (25%), stabilized in four eyes (50%) and worsened in two eyes (25%). In all, 15 (84%) eyes of all studied subjects had improvement in exudation. Two (11%) and one (5%) eye(s) were noted to have a significant post-treatment haemorrhage and retinal pigment epithelial tear, respectively. CONCLUSION: In patients with brown retinal colour, the treatment outcome of TTT was comparable to that of light-pigmented Caucasian patients with approximately half the laser power energy. Further randomized control studies are warranted.
This study examined the association between numbers of benign melanocytic naevi in 7-year-old children in Oxfordshire born in 1988-9 with their mother's arm naevus count, and maternal and child pigmentation factors. We believe this is the first time that the relationship between child and maternal naevus counts has been reported. A high naevus count in the child was associated with male sex (P = 0.009), freckling (P = 0.001) and propensity of the child to burn in the sun (P = 0.05). A low naevus count was observed in red-haired children (P = 0.02). The strongest association of child's naevus count was with a high maternal arm naevus count, independent of the child's pigmentation factors (trend P < 0.0001). Maternal pigmentation factors were not associated with child's naevus count independent of the child's own pigmentation factors. Maternal arm naevus counts may be a better predictor of child naevus count than the child's own pigmentation factors and children. There has not been examination, however, of the relationship between naevus counts in children and those in their parents. We therefore conducted a study of the occurrence of naevi in children aged 7-8 years in Oxfordshire, examining, in addition to sex and pigmentation factors in the child, the relationship of maternal pigmentation factors and maternal naevus counts with naevi in their offspring.
This study aimed to assess longitudinal associations between iris, hair and skin colour, plus skin sensitivity to sun and the 5-year incidence of age-related maculopathy (ARM). Of 3654 baseline Blue Mountains Eye Study participants (aged 49+ years), 2335 survivors (75.1%) were re-examined after 5 years. Retinal photographs were graded using the Wisconsin ARM Grading System and incident ARM lesions confirmed using side-by-side grading.Iris/skin/hair colour was assessed and skin sensitivity questions were asked at baseline. After adjusting for age, sex and smoking, no significant associations were found between iris or hair colour and incident late or early ARM. Compared to persons with fair skin, those with very fair skin had an increased risk of developing geographical atrophy (odds ratio [OR] 3.5,95% confidence interval [CI] 1.2-10.4).However, persons with sun-related skin damage were less likely than those without to develop indistinct soft drusen (OR 0.6, 95% CI 0.4-0.9). Longitudinal data provide no support for the previously reported cross-sectional association between iris colour and ARM.
AIM: To investigate the relationship between iris colour, ethnic origin and the progression of age-related macular degeneration (AMD). METHODS: Participants were recruited from the population-based Melbourne Visual Impairment Project or the prospective, randomized, double-masked Vitamin E, Cataract and Age-Related Macular Degeneration study. From these two cohorts, 171 participants aged between 52 and 93 years who were identified as having early AMD features at their baseline examination (1992-1995) were followed for an average of 6.8 years (until 2001) to determine the progression rate of early AMD. The participants' iris colour was categorized as light, intermediate or dark. Ethnic origin was categorized as Anglo-Saxon or non-Anglo-Saxon, according to the participants' grandparents' country of birth. RESULTS: In total, 53 (31%) of the 171 participants showed signs of AMD progression. Participants with light iris colour had twofold the risk of AMD progression of those with dark or intermediate iris colours, although the age-adjusted and multivariate-adjusted associations were not significant (both P = 0.13). Age-adjusted and multivariate comparisons of Anglo-Saxon ethnic origin to non-Anglo-Saxon ethnic origin showed a noticeable but non-significant association with progression of AMD (P= 0.22 and P= 0.14, respectively). CONCLUSION: Individuals with light iris colour or of Anglo-Saxon ethnic origin had a strong tendency to greater progression of AMD. A larger sample is required to confirm these clinically important, but statistically non-significant, associations.
Fluorescein angiography of the iris vascular system is more difficult in heavily pigmented irides of the coloured races; in light skinned patients it is easy to perform and even more details are demonstrated in patients with atrophies of the mesodermal iris structure. In congenital albinism very fine vascular structures are demonstrated in younger patients; a similar picture is seen in pupillotonia if there is atrophy of the iris stroma. In senile iris atrophies, Adie's syndrome and congenital albinism the iris vascular structure is basically normal, but atrophy of the iris stroma may lead to increased fluorescein permeability in the elderly.
In certain blue-eyed patients with shallow anterior chambers an irregular whitish saw-tooth pattern is visible at the root of the iris, especially on the temporal side. This finding does not appear to have been described previously. The anatomical reasons for this iris root pattern are discussed.

Patients with oculocutaneous or ocular albinism have misrouting of optic fibers, with fibers from 20 degrees or more of the temporal retina crossing at the chiasm instead of projecting to the ipsilateral hemisphere. Misrouting can result in strabismus and nystagmus. Because patients with the Prader-Willi syndrome may also have hypopigmentation and strabismus, we wondered whether they too might have misrouting of optic fibers. We therefore studied six patients with Prader-Willi syndrome selected for a history of strabismus, using pattern-onset visually evoked potentials with binocular and monocular stimulation to look for evidence of misrouted retinal-ganglion fibers. Four had hypopigmentation, and three of these four had abnormal evoked potentials indistinguishable from those recorded in human albinos. The two with normal pigmentation had normal responses. These findings indicate that patients with Prader-Willi syndrome who have hypopigmentation have a brain abnormality characterized by misrouting of retinal-ganglion fibers at the optic chiasm--a finding previously reported only in forms of albinism.

This article describes a predictable method for indexing the ocular portion of an orbital prosthesis. Because indexing is performed from the anterior aspect of the cast, damage is minimized, fewer modifications are required, and the mold can be preserved for repeated use.
Blur or degrading of the retinal image has been shown to be a factor in the formation of visual-geometric illusions where intersecting line elements are present. Light irises allow more scattered light within the eye, which results in more image degradation than found in dark-eyed subjects. Measurements on 755 observers show that illusion magnitude varies as a function of iris pigmentation for a configuration with intersecting line elements (Müller-Lyer illusion), but not for a configuration devoid of such features (Ebbinghaus illusion).
The results of progeny tests of males and females captured during two field trials of sex-linked translocation strains for genetic control of L. cuprina are presented. Males released as mature larvae survived to adulthood and mated with field females. However, the levels of genetic death introduced into the population were insufficient to suppress the native population. This was due partly to seasonal ineffectiveness of the release method, and partly to poor performance of the released males. On average, the mating competitiveness of the released males was only one-third that of field males, whereas their field-reared, translocation-bearing sons were fully competitive with native males.
Sex allocation theory offers excellent opportunities for studying the precision of adaptation. One of the best-supported areas in the field of sex allocation is Hamilton's theory of local mate competition, which predicts female-biased offspring sex ratios when populations are structured such that mating takes place locally before females disperse. As predicted by local mate competition theory, females of numerous species, especially parasitoid wasps, have been shown to lay a less female-biased sex ratio as the number of females simultaneously laying eggs on a patch increases. It has usually been assumed that this sex ratio adjustment comes through individuals adjusting their behavior directly in response to the presence of other females. Here we show that in the parasitoid wasp Nasonia vitripennis, this shift in offspring sex ratios is primarily caused by the presence of eggs laid by other females and to a lesser extent by the presence of other females. We confirm that females are behaving as predicted by theory, but the way in which they do so is not as straightforward as is often assumed. Instead, even when there are multiple females on a patch, individuals still use the cues that are more commonly associated with sex ratio adjustment in response to sequential visits to a patch by females. This result provides a possible explanation for the observed variation in N. vitripennis sex ratios. More generally, it confirms the need to consider the mechanistic basis of a behavior to understand fully its adaptive value.
Retroviruses and related genetic elements generate terminally redundant RNA products by differential polyadenylylation within a long terminal repeat. Expression of the white-apricot (wa) allele of Drosophila melanogaster, which carries an insertion of the 5.1-kilobase retrovirus-like transposable element copia in a small intron, is influenced by signals within copia. By using this indicator, we have isolated a 518-base-pair deletion, 312 base pairs upstream of the copia polyadenylylation site, that is phenotypically like much larger deletions and eliminates RNA species polyadenylylated in copia. This requirement of distant upstream sequences for copia polyadenylylation has implications for the expression of many genetic elements bearing long terminal repeats.
