Synthesis of C-Glycosyl Lactones and Protected C-Glycosyl Amino Acids: Use of Radical Cyclization.

Protected carbohydrates 6a-13a, having one free hydroxyl and a phenylseleno group or halogen on the adjacent carbon, were condensed with (2,2-diphenylhydrazono)acetic acid (2); the resulting esters undergo radical cyclization to afford carbohydrates fused to a 2,2-diphenylhydrazino lactone unit (6c,d-13c,d). In suitable cases (9c,d) such carbohydrate lactones can be elaborated into C-glycosyl amino acids.

New Lewis-Acidic Molybdenum(II) and Tungsten(II) Catalysts for Intramolecular Carbonyl Ene and Prins Reactions. Reversal of the Stereoselectivity of Cyclization of Citronellal.

New Mo(II) complexes BnEt(3)N(+)[Mo(CO)(4)ClBr(2)](-) (A) and Mo(CO)(5)(OTf)(2) (B) and their W(II) congeners D and E have been developed as catalysts for the title reactions. Unlike other Lewis acids, the latter catalysts exhibit cis-stereoselectivity in the cyclization of citronellal (1 --> 3 with A and 1 --> 5 with B). Isotopic labeling allowed formulation of the reaction mechanism, according to which these complexes act as bulky Lewis acids, eta(1)-coordinated to the carbonyl oxygen. The stereochemistry appears to be controlled by the protruding ligand L(p), which dictates the boatlike transition state III. The kinetically formed cis-alkenol 3 can be equilibrated by [Mo(CO)(4)Br(2)](2) (C) or ZnCl(2) to its trans-epimer 2 via a retro-ene reaction.

Synthesis of Substituted 2-Pyridones via the Pummerer Cyclization-Deprotonation-Cycloaddition Cascade of Imidosulfoxides.

1-Ethanesulfinylacetylpiperidin-2-one was prepared from delta-valerolactam in two steps by heating with ethylsulfenylacetyl chloride followed by sodium periodate oxidation. Slow addition of the imidosulfoxide to a refluxing mixture of toluene, acetic anhydride (10 equiv), and p-toluenesulfonic acid (1 mol %) resulted in the formation of an isomünchnone dipole which underwent bimolecular trapping in good yield. The stereochemical assignment of the cycloadduct was made on the basis of X-ray crystallography and is the consequence of an endo cycloaddition with respect to the dipole. The regiochemistry of the cycloaddition is consistent with a HOMO-dipole controlled process. Several related imidosulfoxides containing a tethered alkenyl group were prepared and subjected to the Pummerer reaction conditions. The resulting mesoionic dipoles formed by the cyclization-deprotonation sequence undergo ready dipolar cycloaddition across the pendant olefin to afford intramolecular cycloadducts in high yield. Exposure of these cycloadducts to acetic anhydride in the presence of a trace of p-toluenesulfonic acid results in ring opening to give 5-acetoxy-substituted 2-pyridones. The lone pair of electrons on the amide nitrogen assists in opening the oxy bridge to generate a transient N-acyliminium ion, which subsequently loses a proton. In certain cases, the amide electron pair with the oxy bridge is partially twisted from an antiperiplanar arrangement and a competive ring cleavage also occurs to give 5-thioethyl-substituted 2-pyridones.

Asymmetric Synthesis of 1,2-Diamino-4,5-dimethylcyclohexanes by Zirconium-Catalyzed and -Promoted Reductive Cyclization Reactions.

The asymmetric synthesis of 1,2-diamino-4,5-dimethylcyclohexanes was achieved by the zirconium-catalyzed and -promoted reductive cyclization of N,N'-di[(S)-1-phenylethyl]-4(R),5(R)-diamino-1,7-octadiene. The reaction of the diene with 5 equiv of butylmagnesium chloride and 0.1 mol % of bis(cyclopentadienyl)zirconium dichloride in diethyl ether at 0-20 degrees C gave mainly the 1(R),2(R)-diamino-4(S),5(S)-dimethylcyclohexane derivative having C(2) symmetry, but the reaction with 4 equiv of dibutylzirconocene in tetrahydrofuran at -78 to -50 degrees C gave prevalently the diastereomer with the 4(R),5(S) configuration. By reductive cleavage of the auxiliary, followed by sulfonylation reaction, 1(R),2(R)-di(4-toluenesulfonyl)amino)-4(S),5(S)-dimethylcyclohexane was prepared.

5-Endo-Trig Radical Cyclizations of Bromomethyldimethylsilyl Diisopropylpropargylic Ethers. A Highly Diastereoselective Access to Functionalized Cyclopentanes.

An efficient radical sequence involving a 5-exo-dig, a diastereoselective 1,5-H transfer, and a rarely observed in an all-carbon system 5-endo-trig cyclization allows the construction of cyclopentyl derivatives 2 bearing four controlled stereogenic centers from diisopropyl precursors 1. Olefins 3 were also isolated as minor side products. The effect of the acetylenic substituent Y has been investigated, and the scope and the limitations of the cascade have been delineated.

Titanium-Mediated Cyclization of omega-Vinyl Imides in Alkaloid Synthesis: Isoretronecanol, Trachelanthamidine, 5-Epitashiromine, and Tashiromine.

A new method for the stereocontrolled synthesis of pyrrolizidine and indolizidine alkaloids by means of titanium-mediated cyclization of omega-vinyl imides is described. The general procedure involves treatment of readily available omega-vinyl imides 9 and 10 with 2.5 equiv of cyclopentylmagnesium chloride in the presence of ClTi(O-i-Pr)(3) (1.1 equiv) and subsequent stereoselective reduction of the N-acylaminal group. The cis and trans ring junction stereoisomers can be stereoselectively prepared by catalytic hydrogenation (H(2), PtO(2), EtOAc) and NaCNBH(3) reduction (TFA, MeOH), respectively. Finally, treatment of the resulting lactams with LAH or diborane afforded the target alkaloids 1-8 in good yields.

Cyclizations of substituted benzylidene-3-alkenylamines: synthesis of the tricyclic core of the martinellines.

The martinellines (1 and 2) are natural products that possess both interesting biological activity and chemical structure. During the investigation of a hetero Diels-Alder route to these molecules, alternate Lewis acid-dependent cyclizations of (2'-amino-N'-tert-butoxycarbonyl-5'-chlorobenzylidene)-3-butenylamine (10) were observed. The reaction of a variety of imines with TMSOTf or TiCl(4) led to the formation of different heterocycles including iminodibenzo[b,f][1,5]diazocines, hexahydropyrido[1, 2-c]quinazolin-6-ones, tetrahydropyrrolo[1,2-c]quinazolin-5-ones, 2-arylpiperidines, and 2-arylpyrrolidines. Tetrahydropyrrolo[1, 2-c]quinazolin-5-one 54, obtained via this new methodology, was used as an intermediate in the synthesis of the tricyclic ring system (65) of the martinellines.

Synthesis of 3-trifluoroethylfurans by palladium-catalyzed cyclization-isomerization of (Z)-2-alkynyl-3-trifluoromethyl allylic alcohols

Hydroiodonation of trifluoromethyl propargylic alcohols 1 regio- and stereoselectively produce (Z)-2-iodo-3-trifluoromethyl allylic alcohols 2. (Z)-2-Alkynyl-3-trifluoromethyl allylic alcohols 5, available through Pd(PPh3)4-mediated coupling of 2 and terminal alkynes 4, cyclize and subsequently isomerize to 3-trifluoroethylfurans 6 upon catalysis under PdCl2(CH3CN)2 in THF at 5-10 degrees C.

Dearomatization of naphthalene: novel stereoselective cyclization reactions promoted by Osmium(II)

A series of Michael acceptors has been combined with the Os(II) eta(2)-naphthalene complex (1) to form stable 1H-naphthalenium species. Under acidic conditions, these complexes undergo ring closure at C2 to form the phenanthrenone core. In contrast, the corresponding 1-methylnaphthalene complex (15) upon addition of MVK at C8 undergoes ring closure at C5 to form a bridged tricyclic complex (18). Michael addition of MVK to the naphthalene complex (1) followed by deprotonation, an inter-ring linkage isomerization, and ring closure forms a 9-methylphenalene complex (21). In all cases, the organic cyclization products may be decomplexed by heating with silver triflate and isolated in moderate yield.

Cyclization into hydrindanones using samarium diiodide

Samarium(II) iodide has been employed to promote vinylogous pinacol coupling reaction of aldehyde onto alpha,beta-unsaturated ketones. The diastereoselectivity of 6-endo products was changed by addition of a proton source and/or HMPA and by the reaction temperature. The cyclization reactions described herein provide a general approach to the syntheses of 3,3-dimethylhydrindanes with a cis-relationship between the OH at C-4 and the proton at C-3a with good diastereoselectivity and under mild reaction conditions.

A theoretical study on the reaction mechanism for the bergman cyclization from the perspective of the electron localization function and catastrophe theory.

The reaction mechanism associated with the Bergman cyclization of the (Z)-hexa-1,5-diyne-3-ene to render p-benzyne has been analyzed by means of a combined use of the electron localization function (ELF) and the catastrophe theory on the basis of density functional theory (DFT) calculations (B3LYP/6-31G(d)). The complex electronic rearrangements of this reaction can be highlighted using this novel quantum mechanical perspective. Five domains of structural stability of the ELF occurring along the intrinsic reaction path as well as four catastrophes (fold-cusp-fold-cusp) responsible for the changes in the topology of the system have been identified. The multiple factors that occur along the intrinsic reaction coordinate path are presented and discussed in a consistent way. The topological analysis of ELF and catastrophe theory reveals that mechanical deformation of the C1-C2-C3 unit and closed-shell repulsion between terminal acetylene groups lead to an early formation of diradicaloid character at C2 and C5 atoms. Immediately after the transition structure (TS) is reached, the open-shell-singlet biradical becomes stable. Meanwhile, C1 and C6 atoms are preparing to be covalently bonded; that will finally occur at a distance of 1.791 A. In addition, a separation of the ELF into in-plane (sigma) and out-of-plane (pi) contributions allows us to discuss the aromaticity profiles; sigma-aromaticity appears in the vicinities of the TS, while pi-aromaticity takes place in the final stage of the reaction path, once the ring has been formed.

The p53 inhibitor pifithrin-alpha forms a sparingly soluble derivative via intramolecular cyclization under physiological conditions.

The transcription factor p53 coordinates cell cycle arrest and apoptosis in response to DNA damage. Pifithrin-alpha (PFT-alpha) is a small molecule inhibitor of p53 activity that is frequently used in cell culture studies of p53 function. Here we report an investigation of the stability of this compound. PFT-alpha rapidly converts to a planar tricyclic derivative, with a half-life of 4.2 h under physiological conditions. This spontaneous conversion greatly alters the structural and physicochemical properties of the drug. PFT-alpha has a pKa of 9.11 and is an ionic species in physiological medium, whereas the tricyclic derivative has a pKa of 4.36 and exists as the neutral free base at pH 7. The tricyclic derivative is very hydrophobic, with a log P of 4.26. Although PFT-alpha is generally used at 10-30 microM concentration, the aqueous solubility of its derivative is only 0.2 microM, and it can form a visible precipitate under conditions of typical use. The conversion of PFT-alpha proceeds via an intramolecular cyclization reaction involving the imine and carbonyl groups. Modification of the carbonyl function creates a stable analogue of PFT-alpha that remains soluble indefinitely. These results provide a strategy for the rational design of PFT-alpha analogues that exhibit predictable stability, hydrophobicity, and aqueous solubility.

A new route to cyclopentenones via ruthenium-catalyzed carbonylative cyclization of allylic carbonates with alkenes

[RuCl2(CO)3]2/Et3N and (eta 3-C3H5)RuBr(CO)3/Et3N are highly effective catalyst systems for carbonylative cyclization of allylic carbonates with alkenes to give the corresponding cyclopentenones in high yields. For example, treatment of allyl methyl carbonate (1a) with 2-norbornene (2a) in the presence of a catalytic amount of [RuCl2(CO)3]2 (2.5 mol %) and Et3N (10 mol %) at 120 degrees C for 5 h under 3 atm of carbon monoxide gave the corresponding cyclopentenone, exo-4-methyltricyclo[5.2.1.0(2,6)]dec-4-en-3-one (3a), in 80% yield with high stereoselectivity (exo 100%).

Suppressed beta-hydride elimination in palladium-catalyzed cascade cyclization-coupling reactions: an efficient synthesis of 3-arylmethylpyrrolidines

[formula: see text] A novel type of palladium-catalyzed cascade cyclization-coupling reaction that proceeds with suppressed beta-hydride elimination has been found. One of the N-sulfonyl oxygens is suggested to coordinatively stabilize an alkylpalladium intermediate, thus preventing the intermediate from the usual beta-elimination. This is the first sequential palladium-catalyzed coupling reaction where the Suzuki and Heck reactions can compete. The reaction provides an efficient synthetic route to 4-methylene-3-arylmethylpyrrolidines, which are not readily available by other routes.
