Inactivation of the opiate receptor in bovine caudate nucleus by azide enkephalin analogs.

Photoaffinity labels have been prepared to react with the opiate receptor. The compounds are based on analogs of the enkephalins but involve significant changes in structure which allow introduction of azido substituents while still retaining binding affinity. The compounds designed have been shown to have biological activity in guinea pig ileum tests. A good correlation is obtained between the Kd for stereospecific binding and the Kd for activity in the guinea pig test. Affinity to and inactivation of the opiate receptor is obtained with two of the compounds which have affinities and activities comparable to morphine.

Photocatalyzed labeling of adipocyte plasma membranes with an aryl azide derivative of glucose.

The photoreactive glucose derivative, N-(4-azido-2-nitrophenyl)-2-amino-2-deoxy-D-glucose, has been synthesized and used as a site-directed probe to label adipocyte plasma membranes. In the absence of photoactivation, the glucose derivative was shown to inhibit D-glucose and 3-O-methyl-glucose uptake in the isolated adipocyte system. Glucose was also shown to partially inhibit the uptake of the photoreactive probe. Photolysis of this reagent with light of wavelength greater than 300 nm in the presence of intact adipose cells resulted in the covalent incorporation of radioactivity into the plasma membrane fraction. Analysis of this material by sodium dodecyl sulfate-polyacrylamide gel electrophoresis indicated that four membrane components were significantly labeled and that two of these components corresponded in electrophoretic mobility to the major glycoproteins with apparent molecular weights of 100,000 and 81,000. Photolysis of the glucose analog in the presence of the particulate pellet derived from homogenized adipocytes resulted in the incorporation of the radioactive label into most of the membrane proteins. Although the cells appear to be permeable to the photoreactive probe, only a few membrane components are labeled when the cells are intact, suggesting that the probe has a high affinity for these membrane components which may be functionally involved in the glucose transport system.

A Metamagnetic Two-Dimensional Molecular Material with Nickel(II) and Azide.

Bridging azido ligands transmit both ferro- and antiferromagnetic interactions. The sheetlike structured complex [Ni(µ-N(3))(2)(N,N-Et(2)-N'-Me-en)](n) (see picture) exhibits a range of magnetic behaviors at low temperature. Short-range ferromagnetic and three-dimensional antiferromagnetic ordering compete, giving metamagnetic behaviors. N,N-Et(2)-N'-Me-en=N,N-diethyl-N'-methylethylenediamine.

Controlled synthesis and magnetic properties of 2D and 3D iron azide networks infinity2[Fe(N3)2(4,4'-bpy)] and infinity3[Fe(N3)2(4,4'-bpy)].

Controlled synthesis of transition metal complexes with mixed ligands has led to two new compounds with the same empirical formula [Fe(N3)2(4,4'-bpy)] (4,4'-bpy=4,4'- bipyridine). The compound 2D-[Fe(N3)2(4,4'-bpy)] (I) contains end-on (EO) bridging azido ligands. It crystallizes in the orthorhombic crystal system, space group Cmmm (No. 65): a=11.444(2) A, b=15.181(3) A, c=3.458(1) A, V=600.8(2) A(3), and Z=2. The compound 3D-[Fe(N3)2(4,4'-bpy)] (II) contains end-to-end (EE) azido bridges. It belongs to the tetragonal crystal system, space group P4(1)2(1)2 (No. 92): a=8.132(1) A, b=8.132(1) A, c=16.708(3) A, V=1104.9(5) A(3), and Z=4. Crystals of I and II have been grown by the diffusion method. Phase-pure samples of both compounds have been obtained by means of an optimal solution synthesis. Spontaneous long-range magnetic ordering was found in both I and II, with I being a metamagnet, and II being a ferromagnet. For I, in the low-field region, multiple transitions at TN1=20 K and TN2=5 K were observed, and these indicated the existence of Fe moment reorientation. Heat capacity measurements on II confirmed ferromagnetic transition at TC=20 K.
