Separation and analysis of 4'-epimeric UDP-sugars by borate high-performance liquid chromatography.

Separation of UDP-glucose from UDP-galactose, of UDP-N-acetylglucosamine from UDP-N-acetylgalactosamine, of UDP-glucuronate from UDP-galacturonate, or of UDP-glucosamine from UDP-galactosamine was achieved within 10-45 min by isocratic anion-exchange high-performance liquid chromatography (HPLC) using a flow rate of 2 ml/min. The eluants were composed of borate as complex-forming and eluting agent and of glycerol for protection of the alkali-labile silica packing of the column. This borate HPLC was suitable for the analysis of 4'-epimeric UDP-sugars in the range of 2 to 100 nmol. The applicability of this technique was demonstrated by determination of the relative amounts of 4'-epimeric UDP-amino sugars formed in rat liver after administration of D-galactosamine. Since a high salt content of UDP-sugar samples can interfere with borate HPLC, desalting was performed on a 1-ml C18 cartridge using triethylammonium hydrogen carbonate buffer. This procedure enabled the complete separation of various nucleotides from salts within 10 min prior to HPLC.

Tris-tricine and Tris-borate buffer systems provide better estimates of human mesothelial cell intermediate filament protein molecular weights than the standard Tris-glycine system.

Human mesothelial cells contain a number of well defined intermediate filament proteins (IFs) that have been completely sequenced including vimentin and the cytokeratins (K7, K8, K18, and K19). The electrophoretic migration of these IFs was monitored as a function of second dimension gel buffer composition using various systems including Tris-glycine (pH 8.3 or 9.2), Tris-glycine with 20% methanol, Tris-borate, Tris-tricine, and sodium phosphate. All of the second dimension buffer chemistries yielded patterns of sufficient resolution to identify the major cytoskeletal proteins but differed in the relative mobilities of the IFs. Using gene sequence calculated molecular weight data, the major cytoskeletal polypeptides of human mesothelial cells were ranked from highest molecular weight to lowest molecular weight. This rank order of sequence calculated molecular weights was then compared to the rank order determined form the actual migration of the polypeptides in the different gel systems. With the Tris-tricine and the Tris-borate gel systems as well as gene sequence data, KS = vimentin greater than beta-tubulin = K7 greater than K18 greater than K19 greater than actin. With the pH 8.3 and 9.2 Tris-glycine systems, as well as the sodium phosphate gel system, the rank order of the polypeptides did not correspond to gene sequence data. Adding 20% methanol to the Tris-glycine system resulted in IF migration that more closely corresponded to the gene sequence derived data. Migration position of the IFs depended upon the temperature of the second dimension separation as well. In mesothelial cells, the migration of a total of 15-25% of the polypeptides was influenced by differing buffer systems.(ABSTRACT TRUNCATED AT 250 WORDS)

Primary amines and chloroquine inhibit cytotoxic responses to Shigella toxin and permit late antibody rescue of toxin treated cells.

The effect of lysosomotrophic agents and primary amine inhibitors of transglutaminase on Shigella toxin activity in HeLa cells was determined by measuring cytotoxicity and late antibody rescue. All agents tested resulted in significant antibody rescue, but only ammonium chloride, dansylcadaverine, putrescine, bacitracin, serine/borate buffer, and chloroquine were inhibitory in the absence of added antibody. These compounds appear to be acting within the endocytic vacuole and/or inhibiting translocation of toxin from the cell surface to the cytosol. These data are consistent with a mechanism of translocation of ST from the cell surface to the cytosol by the process of receptor mediated endocytosis.

Capillary electrophoresis of heparin and dermatan sulfate unsaturated disaccharides with triethylamine and acetonitrile as electrolyte additives.

Capillary electrophoresis at constant voltage with the addition of triethylamine as electrolyte to a running buffer containing borate using fused-silica capillaries permits the complete resolution in less than 30 min of 11 standard heparin and 8 standard dermatan sulfate disaccharides, which represent degradation products of heparin and dermatan sulfate by specific lyases. Triethylamine influences the migration time of disaccharides by reducing both their electrophoretic mobility towards the anode and the electroosmotic flow towards the cathode. A modulated combination of these effects together with borate-disaccharide complex formation is responsible for separation, especially in the case of isomers which differ in the position of the sulfate groups. The addition of acetonitrile did not introduce any favourable effect in the separation of disaccharide mixtures. Under these conditions different dermatan sulfates were analysed to assess the source of the preparations.

Exploring borate-activated electron-rich glyoxals as the arginine-reactivity probes. The reactivities of functionally critical arginines in some representative enzymes.

4-Substituted styrylglyoxals have been synthesized and found to benefit from borate catalysis to serve as highly reactive arginine modifiers. A study of the mechanism in arginine modification has affirmed: (i) the role of electronic effects in modulating the arginine reactivity in alpha-dicarbonyls, (ii) the intermediacy in the reaction of a reversible arginine-alpha-dicarbonyl complex and (iii) the identity of the nucleophilic attack at ketone as the rate determining step in the arginine reaction with arylglyoxals. 4-Methoxystyrylglyoxal and 4-(N,N-dimethylamino)phenylglyoxal were used as the probes in analyzing some representative enzymes for the reactivities of their susceptible arginyls. The susceptible arginyls in lactate and malate dehydrogenases were thus found to be appreciably reactive while those in carboxypeptidase-A and pyruvate kinase were only moderately reactive.
