[Assay of micronuclei in binucleated T-lymphocytes. Analysis of the distribution and variation factors in a population of 100 subjects].

The authors analysed micronuclei levels distribution in lymphocytes of 100 non occupationally exposed subjects and studied the effect of age, sex and smoking of donors on the distribution. Results showed that micronucleated cells were distributed according to a normal distribution (average = 9.5 +/- 4 micronucleated cells in 1,000 binucleated lymphocytes). Age and sex of donors had no effect on the distribution but, concerning smoking, the results showed that micronuclei levels were correlated to the number of cigarettes daily smoked.

PREFABRICATION DESIGN CONSIDERATIONS FOR A LONG-TERM ELECTRICALLY-ACTUATED ABDOMINAL LEFT VENTRICULAR ASSIST DEVICE (E-TYPE ALVAD).

The conceptual design and development of a long-term, low-profile intracorporeal left ventricular assist device is a multifaceted project involving a series of technical, anatomic and physiologic considerations. Patients with severe left ventricular failure refractory to all other forms of therapy could benefit from such a device. Prior to fabrication of such a blood pump, consideration must be given to physiologic parameters of the projected patient population. The pump must be designed to meet physiologic demands and yet conform to the anatomic constraints posed by the patient population. We measured the body surface area (BSA) of a group of patients (n=50) and found the mean BSA for this group to be 1.804 +/- 0.161 m(2). Using 25 ml/m(2) as a stroke volume index indicative of left ventricular failure and a stroke volume index of 45 ml/m(2) as normal, distributions of stroke volumes (normal and in left ventricular failure) were plotted for a potential population and demonstrated that 63% of the projected population can be returned to normal by a pump with a stroke volume >/= 83 ml. Cadaver fitting studies established that 73% of the potential population can accommodate an ALVAD 10.8 cm in diameter. In-vitro tests demonstrated that a pump stroke volume >/= 83 ml could be achieved by the proposed pump with a 15 mmHg filling pressure at rates up to 125 B/min. A pusher-plate stroke of 0.56 inches would be necessary to provide a stroke volume >/= 83 ml. The percent of the patient population that could be served was determined by excluding those in whom the pump would not fit or in whom it would provide less than a normal resting stroke volume. Approximately 73% of the projected patient population would accommodate this pump and be returned to normal circulatory dynamics.

The contribution of collective attack tactics in differentiating handball score efficiency.

The prevalence of 19 elements of collective tactics in score efficient and score inefficient teams was analyzed in 90 First Croatian Handball League--Men games during the 1998-1999 season. Prediction variables were used to describe duration, continuity, system, organization and spatial direction of attacks. Analysis of the basic descriptive and distribution statistical parameters revealed normal distribution of all variables and possibility to use multivariate methods. Canonic discrimination analysis and analysis of variance showed the use of collective tactics elements on attacks to differ statistically significantly between the winning and losing teams. Counter-attacks and uninterrupted attacks predominate in winning teams. Other types of attacks such as long position attack, multiply interrupted attack, attack with one circle runner attack player/pivot, attack based on basic principles, attack based on group cooperation, attack based on independent action, attack based on group maneuvering, rightward directed attack and leftward directed attack predominate in losing teams. Winning teams were found to be clearly characterized by quick attacks against unorganized defense, whereas prolonged, interrupted position attacks against organized defense along with frequent and diverse tactical actions were characteristic of losing teams. The choice and frequency of using a particular tactical activity in position attack do not warrant score efficiency but usually are consequential to the limited anthropologic potential and low level of individual technical-tactical skills of the players in low-quality teams.

AgNOR distribution in normal and dysplastic laryngeal mucosa and in laryngeal epidermoid carcinomas.

Samples of normal and dysplastic laryngeal mucosa and of laryngeal epidermoid carcinomas were submitted to the AgNOR silver staining technique. AgNORs were subdivided in large (LN) and small (SN) and counted separately along their sum (TN). Overlap of values is great between normal and mildly dysplastic mucosa and between moderate and severe dysplasia. Grouping together normal with mildly dysplastic mucosa, and moderate with severe dysplasia, their mean values show minimal overlap and the differences between them are highly significant. Plotting the mean values of these two groups and of carcinomas, the linear interpolations show a clearcut increase of values.

Progesterone-binding cyst protein (PBCP = GCDFP-24) and steroid hormone receptors as markers of differentiation in breast cancer. Inverse relation of distribution in normal and malignant tissue of the same breast.

Progesterone-binding cyst protein (PBCP, identical to Gross Cystic Disease Fluid Protein; GCDFP-24) has been evaluated as a possible marker of differentiation in breast tissues. In 17 women with verified operable breast cancer, we have quantitated the cytosol content of PBCP, steroid hormone receptors (estrogen receptor[ER], progesterone receptor[PR] and androgen receptor[AR] as well as albumin in specimens from the primary tumor and from the adjacent non-malignant tissue of the same breast. A significantly higher amount of PBCP (p less than 0.001) and albumin (p less than 0.003) was found in the non-malignant tissue. Conversely, the content of steroid receptors was significantly higher (p less than 0.001) in the malignant tumor, compared to the non-malignant breast tissue. A significant correlation (p = 0.005) between PBCP content in the malignant tumor and in the non-malignant tissue was found. In malignant tissue, ER was significantly correlated with AR (p = 0.007) and to age at operation (p = 0.006). Our results are in agreement with recent reports on other tissue parameters, which indicate qualitative and quantitative differences between the malignant and the non-malignant counterpart with regard to regulation of cell growth and the expression of differentiation markers. This study provides evidence for PBCP as a marker of differentiation to be implemented in further clinical and basic research on breast cancer.

Characterization and distribution in normal and tumoral human tissues of breast cancer-associated antigen defined by monoclonal antibody 7B10.

Monoclonal antibody 7B10, raised against the human breast cancer cell line T47D, identifies an antigen found in human breast carcinomas and in normal breast. Western blot and immunoprecipitation studies detected a Mr 76,000 antigen in cytosol, cell membrane, and cell culture supernatants of T47D cells. 7B10 binding to T47D cell extracts was affected by proteolytic digestion with protease type VI, trypsin, and subtilisin while it was not altered by neuraminidase digestion. Adsorption of breast cancer cell line extracts with concanavalin A reduced 7B10 immunoreactivity more than 70%. These results suggest that the antigen is a glycoprotein and that the epitope does not contain sialic acid. 7B10 was reactive with neither human milk fat globule membrane, nor skimmed milk, nor the milk-derived HBL 100 cell line. Conversely binding was detected in more than 50% of normal breast epithelial cells in culture. 7B10 immunostaining was positive on frozen sections of normal breast and nonmalignant mastopathies in 30 to 90% cells. In frozen sections of other normal tissues, 7B10 immunoreactivity was detected only in colon, apocrine glands of skin, parotid ducts, and luteal phase endometrium, confirming previous data on paraffin sections. Strong, homogeneous immunostaining was observed on frozen sections of intraductal and invasive lobular breast carcinomas (100% of cases), while more heterogeneous staining was found on invasive ductal carcinomas. Colon and rectal carcinomas, one carcinoma of the esophagus, and some cells in serous ovarian carcinomas also showed 7B10 reactivity. Immunoblotting of the 7B10-immunoreactive fraction isolated by Sepharose CL-6B chromatography of a breast carcinoma tissue sample extract identified the Mr 76,000 antigen, which was also detected in several breast cancer specimens, in colon adenocarcinomas, and in serous ovarian carcinoma fresh tumor extracts. The Mr 76,000 glycoprotein described here represents a breast cancer-associated antigen previously undescribed, mainly expressed in normal breast and breast tumors.

Discriminant analysis of trace element distribution in normal and malignant human tissues.

Discriminant analysis of 16 trace element levels measured by ultramicro energy dispersive X-ray fluorescence in malignant and histologically normal human breast, colon, and lung tissues is shown to be a potentially valuable methodology for making malignant-normal and tissue-type classifications. Linear composites of trace elements producing optimal malignant-normal discriminations are found to differ with respect to the number and identity of elements included in the composite for breast, colon, and lung tissues. Nine-, 10-, and 11-element discriminant functions produced overall classification accuracies of 98% for breast, 100% for colon, and 100% for lung tissues, respectively. Elements found to be most important in distinguishing between malignant and normal tissues are Ca, Rb, and Zn in breast, Ca, Zn, and Fe in colon, and Fe, Mn, and Cu in lung samples. Three-group discriminations between breast, colon, and lung tissues were 85% accurate using trace element levels in paired malignant-normal tissues and 91% accurate using trace element levels in tumor tissues only.

Class I and II HLA antigen distribution in normal mucosa, adenoma and colon carcinoma: relation with malignancy and invasiveness.

HLA class I and II antigen expression was studied in normal mucosa, adenoma and colon carcinoma. Alkaline phosphatase anti-alkaline phosphatase (APAAP) staining techniques were used in cryostatic sections with anti-HLA-ABC and DR,DP,DQ monoclonal antibodies. All normal mucosa were intensely positive for HLA class I antigen expression, while failing to express class II molecules, except in mucosa adjacent to tumors (15/19 cases). All adenomatous polyps expressed HLA class I antigen, while the intensity of class II expression (DR greater than DQ greater than DP) was paralleled by the degree of dysplasia. In colon carcinoma, the loss of class I expression was seen in 4 out of 32 cases, and class II expression was found to be heterogeneous in 16 of these 32 cases (DR greater than DP greater than DQ). No relationship was noted between class II expression and degree of differentiation. However a correlation was seen between HLA-DR antigen expression and degree of invasiveness, mononuclear infiltrate and prognosis, according to Jass's criteria.

[Temperature distribution in normal and tumor tissues of animals subjected to local SHF-hyperthermia].

Temperature gradients are studied in normal and tumour tissues of animals under different duration of treatment (6-30 min) with microwave field which has a frequency of 460 MHz and 40 W power range. It is shown that in animal muscles and tumours an increase in the temperature is directly dependent on the time of microwave heating, the maximum temperatures being obtained in the depth of tissues, while there is no overheating in subcutaneous fatty tissues.

Rifampin and trimethoprim distribution in normal and hydronephrotic kidney and in prostate: an experimental study in dogs.

Concentrations of rifampin and trimethoprim, administered in the ratio 3.75:1 were measured in renal hilar lymph, renal interstitial fluid, prostatic interstitial fluid and prostatic secretion in dogs during constant infusion of the antibiotic combination. The lymph was obtained by direct cannulation of the lymphatics and the interstitial fluids from small plastic tissue chambers implanted 4 weeks before the experiments. Unilateral hydronephrosis was produced by ureteral ligation in dogs with tissue chambers in both kidneys to compare concentrations of the drug combination in renal interstitial fluid and urine from normal and hydronephrotic kidneys. Rifampin produced lower concentrations in both prostatic interstitial fluid and prostatic secretion than in plasma, while trimethoprim concentrations were much higher in prostatic secretion and prostatic interstitial fluid than in plasma. Renal lymph/plasma ratios and renal interstitial fluid/plasma ratios were significantly higher for trimethoprim than for rifampin, but only the lymph concentrations of trimethoprim exceeded the corresponding plasma concentrations. High urine concentrations were achieved for both drugs except in pelvises of hydronephrotic kidneys, where virtually no drug activity could be detected. Renal interstitial fluid concentrations of both drugs were similar in hydronephrotic and normal kidneys. The ratios between rifampin and trimethoprim concentrations varied considerably among plasma, various tissues and tissue fluids.
