Identification of steviol glucuronide in human urine.

Stevioside (250 mg capsules) was given three times daily to 10 healthy subjects. Steviol glucuronide (steviol 19-O-beta-D-glucopyranosiduronic acid; MM, 494.58; melting point, 198-199 degrees C) was characterized in the 24 h urine as the only excretion product of oral stevioside by MS, NMR, IR, and UV spectroscopy. This is the first report on the unambiguous identification of steviol glucuronide in human urine.

Hydroquinone antioxidants from the Indian ocean tunicate Aplidium savignyi.

Three hydroquinone compounds have been isolated from the Indian Ocean tunicate Aplidium savignyi. Two are already known: geranylhydroquinone and the 2-(3-hydroxy-3,7-dimethyloct-6-enyl)-1,4-benzenediol as a new stereoisomer 2-(2Z)-(3-hydroxy-3, 7-dimethyloct-2,6-dienyl)-1,4-benzenediol. The structures were determined using spectroscopic data ((1)H NMR, (13)C NMR, MS, UV, and IR). These compounds are potential natural antioxidants.

Antioxidative phenolic compounds from Japanese barnyard millet (Echinochloa utilis) grains.

Three antioxidative phenolic compounds, one serotonin derivative and two flavonoids, were isolated from an ethanol extract of Japanese barnyard millet (cv. Kurohie) grains by Sephadex LH-20 chromatography and preparative high-performance liquid chromatography. Their structures were established to be N-(p-coumaroyl)serotonin, luteolin, and tricin on the basis of spectrometric data from (1)H and (13)C and two-dimensional nuclear magnetic resonance techniques, fast atom bombardment mass spectrometry, and Fourier transform infrared spectrophotometry. N-(p-Coumaroyl)serotonin exhibited a strong antioxidant activity almost equivalent to that of butylated hydroxyanisole at the same concentration (w/v). Although the antioxidant activity of luteolin was lower than that of N-(p-coumaroyl)serotonin, it was nearly equal to that of quercetin, whereas the activity of tricin was lower than that of luteolin. All of them were newly isolated from Japanese barnyard millet grains.

Novel synthetic oleanane and ursane triterpenoids with various enone functionalities in ring A as inhibitors of nitric oxide production in mouse macrophages.

We initially randomly synthesized about 60 oleanane and ursane triterpenoids as potential anti-inflammatory and cancer chemopreventive agents. Preliminary screening of these derivatives for inhibition of production of nitric oxide induced by interferon-gamma in mouse macrophages revealed that 3-oxooleana-1, 12-dien-28-oic acid (B-15) showed significant activity (IC(50) = 5.6 microM). On the basis of the structure of B-15, 19 novel olean- and urs-12-ene triterpenoids with a 1-en-3-one functionality having a substituent at C-2 in ring A have been designed and synthesized. Among them, 3-oxooleana-1,12-diene derivatives with carboxyl, methoxycarbonyl, and nitrile groups at C-2 showed higher activity than the lead compound B-15. In particular, 2-carboxy-3-oxooleana-1, 12-dien-28-oic acid (3) had the highest activity (IC(50) = 0.07 microM) in this group of triterpenoids. The potency of 3 was similar to that of hydrocortisone (IC(50) = 0.01 microM), although 3 does not act through the glucocorticoid receptor. Interesting structure-activity relationships of these novel synthetic triterpenoids are also discussed.

Contraceptive agents from cycloaddition reactions of diarylcyclopropenones and diarylthiirene 1, 1-dioxides.

The potential for compounds with antifertility activity from the reactions of diphenylcyclopropernone (1) and 2, 3-diphenylthiirene 1, 1-dioxide (2) with enamines is described. In certain instances, a marked dissociation of antifertility from estrogenic activity was possible. Two series were studied extensively, one was stilbene amides (7) and the other stilbene amino ketones (8). The latter series (8) afforded several materials from which, on further biological work-up, was singled out compound 21 as a potent antifertility agent in rats and hamsters.

Purinylhydantoins. Facile conversion of the naturally occurring N-(purin-6-ylcarbamoyl)-L-amino acids into 3-pruin-6-ylhydantoins and 3-cyclohexyl-1-(purin-6-ylcarbamoyl)hydantoins.

The naturally occurring N-(purin-6-ylcarbamoyl)-L-threonine (PCT, 1b), N-(purin-6-ylcarbamoyl)glycine (PCG, 1a), and some of their analogs were converted into novel purine derivatives, the purinylhydantoins. The PCT and PCG underwent intramolecular cyclization in the presence of N,N-dicyclohexylcarbodiimide (CDD) to give the 3-purin-6-ylhydantoins (2a-c). The same hydantoins were also obtained when the PCT and PCG were allowed to react through the mixed anhydride formed from cyclohexyl isocyanate or ethyl chloroformate. 1,3-Dicyclohexyl-1-(N-(purin-6-ylcarbamoyl)aminoacyl)ureas 3a and 3c, by-products obtained from the DCC reaction, were rapidly converted in aqueous NaOH to another type of purinylhydantoins, the 3-cyclohexyl-1-(purin-6-ylcarbamoyl)hydantoins 4a and 4b. Compound 4a when heated in base underwent hydrolysis of the hydantoin ring giving biuret N-(cyclohexylcarbamoyl)-N-(purin-6-ylcarbamoyl)glycine (5a) and N-(purin-6-ylcarbamoyl)glycine cyclohexylamide (6a). The characterization of these hydantoins was carried out by uv, nmr, and mass spectrometry. The 3-purin-6-ylhydantoins and 3-cyclohexyl-1-(purin-6-ylcarbamoyl)hydantoins showed growth inhibitory activity in the cultured leukemic cells, while the parent amino acid compounds were inactive.

Narcotic antagonists. 4. Carbon-6 derivatives of N-substituted noroxymorphones as narcotic antagonists.

A series of new narcotic antagonists has been synthesized by modifying the C-6 carbonyl group in naloxone (Ia) and naltrexone (Ib). New functional units were introduced by reaction with various phosphorus and sulfur ylides and alkyllithium reagents. The activity of the new compounds were measured by the hot-plate and tail-clip tests after oral administration to mice. The majority of the new narcotic antagonists exhibited oral potencies considerably superior to the parent compounds, with 6-methylene derivatives IIa and IIb showing the most impressive increases.

Synthesis of 15-keto-6beta, 7beta-methyleneprogesterone. Effect of the 6beta, 7beta-methylene group on mineralocorticoid activity.

15-Ketoprogesterone is as active as spironolactone in blocking the mineralocorticoid effect of deoxycorticosterone acetate. This activity is reduced when a methylene group is attached to the 6beta, 7beta position. The title compound was prepared from 15alpha-acetoxy-6-dehydroprogesterone. Methylenation of the delta6 double bond with dimethyloxosulfonium methylide proceeds steroselectively from the beta side of the molecule.

7-N-amidinocephalosporins.

7-Aminocephalosporanic acid tert-butyl ester reacts quantitatively at--20 degrees with iminium chlorides to give amidino derivatives. Removal of the tert-butyl protecting group with trifluoroacetic acid and treatment with 1 equiv of triethylamine yield the corresponding zwitterions. These compounds were less active than their penicillin analogs.
