Cu- and Pd-catalyzed asymmetric one-pot tandem addition-cyclization reaction of 2-(2',3'-alkadienyl)-beta-keto esters, organic halides, and dibenzyl azodicarboxylate: an effective protocol for the enantioselective synthesis of pyrazolidine derivatives.

[reaction: see text] Optically active pyrazolidine derivatives have been constructed by the Cu- and Pd-catalyzed asymmetric one-pot tandem addition-cyclization reaction of 2-(2',3'-dienyl)-beta-ketoesters, organic halides, and dibenzyl azodicarboxylate. The absolute configurations of the final products depend on the structure of the ligand.

Intramolecular hydrogen abstraction promoted by amidyl radicals. Evidence for electronic factors in the nucleophilic cyclization of ambident amides to oxocarbenium ions.

A tandem 1,5-hydrogen atom transfer/radical oxidation/nucleophilic cyclization mechanism is proposed for the intramolecular hydrogen abstraction reaction promoted for primary carboxamidyl radicals. The electron-withdrawing capacity of the C-5 substituent can switch the reaction to give exclusively bicyclic spirolactams (6-oxa-1-azaspiro[4.5]decan-2-one) when R(1) = H or spirolactones (1,6-dioxaspiro[4.5]decan-2-one) when R(1) = OAc. With a substituent of medium polarity (R(1) = OMe), a mixture of lactones and lactams is formed.

Haloacetal radical cyclizations of alpha- and beta-hydroxyhydrazones.

[reaction: see text] Haloacetal radical cyclizations of alpha- and beta-hydroxyhydrazones provide a direct access to aminosugarlike compounds. Stereocontrol of this process is influenced by stereogenic centers of both the hydroxyhydrazone and the acetal. The outcomes are consistent with chair and twist transition states with the anomeric alkoxy group in pseudoaxial orientations.

Kinetic studies of the cyclization of singlet vinylchlorocarbenes.

[reaction: see text] Vinylchlorocarbenes (R = Me or Cl) cyclize to cyclopropenes with k = (2.4-5.1) x 10(7) s(-1) or (3.1-4.5) x 10(6) s(-1), respectively, in solution at room temperature. The corresponding E(a) values are 6.6 and 5.7 kcal/mol. Excited diazirines are also likely precursors of the cyclopropenes.

Gold-catalyzed cyclization of (Z)-2-en-4-yn-1-ols: highly efficient synthesis of fully substituted dihydrofurans and furans.

[reaction: see text] The gold-catalyzed cyclization of (Z)-enynols offers an efficient and straightforward route to stereodefined (Z)-5-ylidene-2,5-dihydrofurans and fully substituted furans under extremely mild reaction conditions. Importantly, the results indicated that both the oxyauration and the proto-demetalation steps are highly stereoselective.

Tropos or atropos nature of rhodium complexes bearing a tetrakis(phosphanyl)terphenyl ligand: highly enantioselective catalysis of ene-type cyclization.

[reaction and structure: see text] Not only axial but also helical chirality of tropos TETRAPHOS-Rh complexes can be controlled by chiral diamines. The flexibility of the TETRAPHOS-Rh complex is increased by association of DABN. In contrast, the diamine-free complex is chirally more stable than the BIPHEP counterpart. The higher levels of enantioselectivity in ene-type cyclization of 1,6-enynes can thus be achieved even at room temperature by the diamine-free TETRAPHOS-Rh complex.

Diastereoselective synthesis of 4-hydroxypiperidin-2-ones via Cu(I)-catalyzed reductive aldol cyclization.

[chemical reaction: see text]. 4-Hydroxypiperidin-2-ones may be prepared in highly diastereoselective fashion using a Cu(I)-catalyzed reductive aldol cyclization of alpha,beta-unsaturated amides with ketones. Used in combination with proline-catalyzed asymmetric Mannich reactions, this methodology enables the enantioselective synthesis of more highly functionalized piperidin-2-ones and hydroxylated piperidines.

The first examples of Nazarov cyclizations leading to annulated pyrroles.

[reaction: see text] Reactions between alpha-substituted unsaturated carboxylic acids 20 and N-tosylpyrroles [14, 23] in the presence of trifluoroacetic anhydride result in smooth alpha-acylation of the pyrrole, followed by Nazarov cyclization to give 50-80% yields of cyclopenta[b]pyrroles. The presence of an alpha-substituent in the unsaturated acid appears to be mandatory.

Mercuric chloride and iodide mediated cyclization of tethered alkynedithioacetals as a general route to five- and six-membered rings: tuning of regioselectivity by alkyne substitution.

[reaction: see text] Mercuric chloride mediated cyclization of tethered alkynedithioacetals has been established as a general route to five- and six-membered carbocycles and heterocycles. Substitution at the alkyne terminus leads to preferential formation of five-membered rings, whereas unsubstituted alkynedithioacetals give six-membered rings as the major products. Mercuric iodide interrupts the reaction at the intermediate dithioacetal stage.

Synthesis of 2-substituted 3-aroylindenes via palladium-catalyzed carbonylative cyclization of diethyl 2-(2-(1-alkynyl)phenyl)malonates with aryl halides.

[reaction: see text] The palladium-catalyzed reaction of readily accessible diethyl 2-(2-(1-alkynyl)phenyl)malonates with aryl halides under a balloon pressure of CO produced 2-substitued 3-aroylindenes in good yields. The reaction is believed to proceed via cyclization of the alkyne containing a proximate nucleophilic center promoted by an acylpalladium complex.

1,6-Stannatropic strategy: effective generation and cyclization of 1,5-dipoles from o-stannylmethylated thioanilides or phenyl isothiocyanates.

[reaction: see text] A new method for the generation of 1,5-dipoles from o-stannylmethylated thioanilides via 1,6-stannatropy under neutral conditions was developed. Cyclization of the 1,5-dipoles afforded indole derivatives effectively. The strategy has potential for application to the generation of alternative 1,5-dipoles from o-stannylmethylated aryl isothiocyanates leading to indole derivatives having a stannylthio group that was readily converted to other functional groups.
