Eccrine sweat gland carcinoma of the eyelid with orbital involvement.

A 70-year-old man had a sweat gland carcinoma in his right upper eyelid that was slowly growing, but it clinically remained confined to the lid tissues for a period of 25 years. The tumor ultimately extended deeper into the orbit and invaded the sclera, the optic nerve sheath, and the ethmoidal sinuses. Histopathologic examination disclosed malignant features with a variety of cellular patterns. The results of electron microscopic studies confirmed the origin of the tumor from the eccrine sweat glands.

Sweat gland tumors of the head and neck.

Surgeons performing soft-tissue surgery in the head and neck area occasionally will encounter other varieties of neoplasms, such as skin-appendage tumors, in addition to basal cell carcinoma and squamous cell carcinoma. Sweat gland tumors are the most common variety of the skin-appendage tumors and the most easily confused with basal cell carcinoma and squamous cell carcinoma. We report this case to illustrate this fact. The classification of sweat gland tumors and their suggested management are reviewed.

Sweat gland carcinoma. Current concepts of surgical management.

In three new cases of sweat gland carcinoma that we observed within recent years, the sites were the axilla, back, and arm. Axillary lymph node dissections were performed in two of the patients and the nodes were normal. Preoperative diagnoses were hydroadenitis, squamous cell carcinoma of the skin, and pyogenic granuloma. In one patient who was followed up for four years, there has been no recurrence; the follow-up period for the other two has been short. Sweat gland carcinoma is an uncommon neoplasm that occurs mostly in the older age groups. It may be very slow growing and is extremely difficult to diagnose preoperatively. Lymph node metastases are frequent and overall survival is poor. Prognosis is related to histologic cell type and presence or absence of lymph node metastases. Treatment by wide local excision of the lesion and primary regional node dissection is recommended.

Sweat gland carcinoma with syringomatous features: a light microscopic and ultrastructural study.

A 76-year-old man presented with a 10-year history of a slowly enlarging lesion of his upper lip. An excisional biopsy revealed a diffusely infiltrating, well-differentiated, sweat gland carcinoma with syringomatous features. The ultrastructural characteristics corresponded to those described for benign syringoma and normal eccrine ducts. A review of the literature emphasized the extreme rarity of this lesion. Sweat gland carcinomas tend to have an indolent history but often manifest aggressive behavior at the time of clinical presentation. The preponderance of anaplastic lesions accounts for a generally unfavorable prognosis.

Adenocarcinoma of the sweat gland of the vulva: light and electron microscopic study.

A sweat gland carcinoma, a rare tumor of the skin which arose in the posterior fourchette in a 53-year-old white woman, was studied by light and electron microscopy. The tumor formed cords or small acini infiltrating into the stroma with foci of Pagetoid appearance. The cells showed poor intercellular cohesions. PAS and mucin strains were negative. Ultrastructurally the tumor cells in the periphery of the acini were firmly attached to the collagenous matrix with a few pseudopodia. A basal lamina was seen in some areas. The cytoplasm was filled with many ribosomes and moderately developed mitochondria. The bundles of microfilaments in the perinuclear zone were characteristic findings in this tumor. The ultrastructure of this tumor confirmed that this was carcinoma of the sweat gland, which closely resembled intraductal carcinoma of the breast.

Direct electrochemical determination of pyruvate in human sweat by capillary zone electrophoresis.

Capillary zone electrophoresis was employed for the determination of pyruvate in human sweat using electrochemical detection with a carbon fiber microdisk bundle electrode at a constant potential of 1.60 V vs. saturated calomel electrode. The optimum separation conditions are 3.6 x 10(-3) mol/L Na2HPO4-1.4 x 10(-3) mol/L NaH2PO4 (pH 7.2) for the buffer solution, and 18 kV for the separation voltage. The limits of detection of pyruvate are 8.0 x 10(-6) mol/L or 24 fmol (S/N = 3) for the injection voltage of 6 kV and the injection time of 10 s. The relative standard deviation is 2.0% for the migration time and 5.7% for the electrophoretic peak current. The method was applied to determining pyruvate in human sweat.

Identification of new odoriferous compounds in human axillary sweat.

3-Hydroxy-3-methylhexanoic acid (1) and the 3-sulfanylalkan-1-ols 2-5 were identified to contribute to the odor of human axillary sweat. Quantitative analyses of axillary sweat extracts from 50 healthy men showed an unambiguous correlation between the detected levels of 1 and the intensity of the axillary odor. Chiral-GC analyses revealed 1 to be a 72:28 mixture of the (S)/(R)-isomers. Optically pure (S)-1 (>97% ee) emanated a strong spicy note, which recalled typical axillary odors. 3-Methyl-3-sulfanylhexan-1-ol (2), the enantiomeric ratio of which equaled that of 1, was present in greater quantity than any of the other 3-sulfanylalkanols. Optically pure (S)-2 (>97% ee) had a strong meaty, fruity note, also reminiscent of axillary odor. The compounds identified, in particular (S)-1 and (S)-2, contribute significantly to the olfactory impression of human axillary odor.

Corrosion behavior of nickel-containing alloys in artificial sweat.

The corrosion resistance of various nickel-containing alloys was measured in artificial sweat (perspiration) using the Tafel extrapolation method. It was found that Ni, CuNi 25 (coin alloy), NiAl (colored intermetallic compounds), WC + Ni (hard metal), white gold (jewelry alloy), FN42 and Nilo Alby K (controlled expansion alloys), and NiP (electroless nickel coating) are in an active state and dissolve readily in oxygenated artificial sweat. By contrast, austenitic stainless steels, TiC + Mo2C + Ni (hard metal), NiTi (shape-memory alloy), Hastelloy X (superalloy), Phydur (precipitation hardening alloy), PdNi and SnNi (nickel-containing coatings) are in a passive state but may pit under certain conditions. Cobalt, Cr, Ti, and some of their alloys were also investigated for the purpose of comparison. Cobalt and its alloys have poor corrosion resistance except for Stellite 20. Chromium and high-chromium ferritic stainless steels have a high pitting potential but the latter are susceptible to crevice corrosion. Ti has a pitting potential greater than 3 V. Comparison between the in vitro measurements of the corrosion rate of nickel-based alloys and the clinical observation of the occurrence of contact dermatitis is discussed.

Metastatic sweat gland carcinoma: response to 5-fluorouracil infusion.

A 78-year-old white male presented with metastatic sweat gland carcinoma involving regional lymph nodes, pleura, pericardium, and chest wall. A complete response of three months duration was achieved by using a 96-hour infusion of 5-fluorouracil. This is the first report of a complete response to systemic chemotherapy in metastatic sweat gland carcinoma.

Measurement of chloride concentration in microvolume samples of sweat.

Measuring sweat Cl concentration requires multiple steps. Following pilocarpine iontophoresis we compared Cl concentrations of sweat obtained on gauze pads to microvolumes (5 microL) collected into capillary tubes (N = 111). Chloride concentrations obtained by these two methods correlated significantly (r = 0.95; P < 0.001), indicating that Cl concentration can be measured accurately and easily on small volumes.

Electrophysiological responses to oxytocin and ATP in monolayers of a human sweat gland cell line.

It was shown that oxytocin (OT) elicits electrophysiological responses in cultured monolayers of NCL-SG3, a human immortalized sweat gland cell line. The response to OT was greater for basal applications. It was also found that monolayers respond to ATP with a transient transepithelial-potential change, with a more pronounced response to apical than to basal applications. The IC50 for the response to OT was 180 nM at room temperature. The response to OT was not due to effects of OT on vasopressin (AVP) receptors as evidenced by three tests: (a) The response was completely blocked by the selective OT-receptor antagonist [Mpa1,D-Tyr(Et)2,Thr4,Orn8]-OT (CAP) applied at equal concentrations (100-1000 nM) to that of OT. (b) The response to OT was similar to that of ionomycin (2 microM) or ATP (150 microM). In contrast, the response to AVP (500 nM) or cAMP (2 mM) were smaller and of a different time course. (c) OT increased but AVP had no effect on the intracellular free calcium. It is suggested that OT may have a role in the regulation of salt balance in sweating.

The role of acetylcholine in regulating secretory responsiveness in rat sweat glands.

While retrograde regulation of neuronal development by target-derived factors in the autonomic nervous system is well established, the importance of anterograde influences on target development is unclear. Previous studies suggest that sympathetic innervation of sweat glands plays a critical role in the acquisition and maintenance of their secretory function. To define the signal(s) responsible, we disrupted muscarinic cholinergic transmission in developing and adult rats. Treatment of young rats with the nonselective antagonist, atropine, or an antagonist selective for the glandular muscarinic subtype, 4-DAMP, delayed the development of secretory responsiveness. Treatment of adult animals with atropine caused its loss. Further, following denervation, treatment with the muscarinic agonist, pilocarpine, largely preserved responsiveness while untreated animals lost function. Thus, acetylcholine, whose presence in sweat gland innervation is retrogradely specified by developmental interactions with the target tissue, in turn plays an important role in inducing and maintaining target tissue responsiveness through muscarinic receptor activation.

Morphological evidence for the innervation of apocrine sweat glands in the general hairy skin of the goat.

A histomorphological and histochemical study was made on the nerve supply to the apocrine sweat glands in the general hairy skin of the goat. In harmony with the previous report that the sweat glands in the goat are functionally under the control of sympathetic nervous system, the present study clearly demonstrates cholinesterase-reactive nerve fibers that closely surround the secretory portion of these glands in most of the hairy skin area, though the nerve network is fairly coarse. Analysis with cholinesterase inhibitors indicated that the sudomotor nerves in the goat contain both specific and non-specific cholinesterase.

Calcitonin gene-related peptide-like immunoreactive cells in the secretory portions of rat sweat glands.

We found cells with calcitonin gene-related peptide-like immunoreactivity and with many cored vesicles in the secretory portions of sweat glands of rat foot pads. About 10% of sweat glands contained single immunoreactive cells. The immunoreactive cells were flask-shaped, with a narrow apex facing the glandular lumen and the bulk of the cell body in the basal half of the glandular wall. In the cytoplasm, there were many vesicles, 100-250 nm in diameter, with cores of various electron densities. These cytochemical and cytological characteristics suggest that the immunoreactive cells are homologous to gastrointestinal basal granulated cells.

Chloride and potassium conductances of cultured human sweat ducts.

The purpose of this study was to characterize the ion conductances, in particular those for Cl- and K+, of human sweat duct cells grown in primary culture. Sweat duct cells from healthy individuals were grown to confluence on a dialysis membrane, which was then mounted in a mini-Ussing chamber and transepithelial and intracellular potentials were measured under open-circuit conditions. Under control conditions the epithelia developed mucosa-negative transepithelial potentials, Vte, of about -10 mV. The apical membrane potential, Va, was -25 mV to -30 mV (n = 97) in most cells, but several cells had a higher potential of about -55 mV (n = 29). Mucosal amiloride (10 mumol/l) hyperpolarized Va from -31 +/- 1 mV to a new sustained level of -46 +/- 2 mV (n = 36). These changes were accompanied by increase in the fractional resistance of the apical membrane, fRa, and decreases of Vte and the equivalent short-circuit current, Isc. In amiloride-treated tissues an increase in mucosal K+ concentration (5 mmol/l to 25 mmol/l) depolarized Va by 5 +/- 1 mV (n = 8), while the same step on the serosal side depolarized Va by 20 +/- 2 mV (n = 8). A Cl- channel blocker 3',5-dichlorodiphenylamine-2-carboxylate DCl-DPC; 10 mumol/l) depolarized Va by 5 +/- 1 mV (n = 6), an effect that was lost after amiloride application. The blocker had no effect from the serosal side. Reduction of mucosal Cl- (from 120 to 30 or 10 mmol/l) depolarized Va by 9-11 mV (n = 35), an effect that was often followed by a secondary hyperpolarization of 10-30 mV (n = 27).(ABSTRACT TRUNCATED AT 250 WORDS)

Pathologic S-phase in the cells of eccrine sweat glands and of hair follicles in psoriasis.

The prolongation of the S-phase, characteristic of psoriatic keratinocytes, can be observed in cells of the eccrine sweat glands as well as in those of the infundibular part of hair follicles. In healthy individuals the average duration of DNA-synthesis time (ts) in the cells of the hair follicle is 6.8 h, in unaffected psoriatic skin 11.6 h, and in affected psoriatic skin 8.4 h. Ts in the proliferating cells of the eccrine sweat glands reveals no difference between normal skin and unaffected psoriatic skin, but is markedly prolonged in affected psoriatic skin.

Sweat gland function in dermatophytosis.

In 54 cases of dermatophytosis, sweat gland disturbances and their possible mechanisms were studied. The basic perspiratory malfunction was intense hypohidrosis, with some hyperhidrotic phenomena being observed at particular points of the exanthem (margin and disc) in a few cases. It is worth noting that in 51.85% of the cases, hypohidrotic phenomena were also observed in areas of normal skin adjacent to the lesions. One of the most important forms of perspiratory disturbances was the closure of sweat ducts at the keratin level. The inflammatory process in the dermis may also affect gland function. Finally, there is a discussion of the significance of perspiratory disturbances in relation to special aspects of the disease.

[Immunological demonstration of beta-glucuronidase in eccrine sweat (author's transl)].

Electrophoretic and immunological investigations on highly concentrated sweat have shown that human eccrine sweat contains 6 different esterases. Two of them were found to hydrolyze naphthol-AS-Bi-beta-D-glucuronide. One of the two glucuronic acid ester splitting enzymes is likely to be a substrate-specific beta-glucuronidase. The other one is a non-specific carboxyl-esterase occurring in numerous organs and glandular secretions. This non-specific enzyme of widespread distribution can be mistaken for beta-glucuronidase because it hydrolyzes glucuronic acid esters as well as other synthetic substrates.
