Metabolism and excretion of the quaternary ammonium compound thiazinamium methylsulfate (Multergan) in man. I. Parenteral administration.

The quaternary ammonium compound thiazinamium was found to be metabolized by sulfoxidation solely. No ring hydroxylation products, nor demethylation products (e.g. promethazine) could be found. Thiazinamium sulfoxide was found both in urine and bile, but thiazinamium is-after parenteral administration - mainly excreted in the unchanged form. After intravenous injection about 40% of the dose was excreted, unchanged, in the urine. The excretion was very rapid and almost complete within eight hours. About 9% of the dose was excreted in the urine in the form of thiazinamium sulfoxide cations. After intramuscular injection virtually the same figures for urinary excretion were found. No correlation could be observed between urine production or pH and the amount of drug excreted in urine. In a study involving bile-fistula patients it was found that both thiazinamium cations and thiazinamium sulfoxide cations are excreted to a considerable extent in bile. The amount of unchanged drug in bile was almost equal to that in urine, but the amount of sulfoxide was slightly higher.

Biological and physicochemical properties of gemini quaternary ammonium compounds in which the positions of a cross-linking sulfur in the spacer differ.

We synthesized two novel gemini quaternary ammonium compounds (gemini QACs), 4,4'-[1,6-(2,5-dithiahexane)]bis(1-alkylpyridinium bromide) and 4,4'-[1,6-(3,4-dithiahexane)]bis(1-alkylpyridinium bromide), which are essentially two dimerized pyridinium salts. Three gemini QACs in which the positions of a cross-linking sulfur in the spacer differ, in addition to the previously described 4,4'-[1,6-(1,6-dithiahexane)]bis(1-alkylpyridinium bromide) to both gemini compounds, were determined for their antimicrobial, hemolytic and surface activities and molecular hydrophobicity. Comparative biological and physicochemical studies concluded that the position of sulfur in the spacer chain for three gemini QAC series influences the surface activity, the hydrophobicity and the electron density of the ammonium nitrogen, and that their biological properties are ascribable to the variation of these parameters caused by the position of the sulfur.

Identification of unknown quaternary ammonium compounds in corneal epithelium and aqueous humor.

Bovine corneal epithelium and bovine aqueous humor are investigated for their content of quaternary ammonium compounds. In total, four compounds are found. Three of these are identified. For the fourth compound, a proposal for its structure is made on the basis of tandem mass spectrometry fragmentation spectra. The compounds investigated have m/z values of 146, 160, and 174. The compounds with m/z 146 are confirmed as acetylcholine (in corneal epithelium) and (3-carboxypropyl)-trimethylammonium (in both corneal epithelium and aqueous humor). The compound with m/z 174 is identified as butyrylcholine (in corneal epithelium). The compound with m/z 160 is probably acetyl-g-homocholine (in both corneal epithelium and aqueous humor). For both butyrylcholine and acetyl-g-homocholine, it is the first time the presence of these compounds in corneal epithelium or aqueous humor (or both) is described. Both acetylcholine and butyrylcholine are unstable compounds, which are probably susceptible to enzymatic degradation by acetylcholine-esterase and butytrylcholine- esterase, respectively.

Changes in fatty acid composition of Pseudomonas aeruginosa ATCC 15442 induced by growth conditions: consequences of resistance to quaternary ammonium compounds.

The effects of changes in the fatty acid composition of Pseudomonas aeruginosa induced by growth conditions on its resistance to two quaternary ammonium compounds (QAC) were investigated. The temperature and growth phase were the most influential parameters affecting the fatty acid composition of this bacterium. Furthermore, the formation of saturated fatty acids and cyclopropane fatty acids was stimulated by increasing the temperature, whereas the proportion of unsaturated fatty acids fell. The degree of saturation and the proportion of cyclopropane fatty acids increased in the course of the exponential and stationary phases. These modifications mostly concerned the inner membrane of the bacterium. Resistance of P. aeruginosa to both QAC tested was not significantly influenced by temperature and growth phase variations. Thus, resistance to the two QAC did not seem to be dependent on modifications of the fatty acid composition of the inner membrane.

[Comparative study of fungicidal activity in vitro of various quaternary ammonium compounds used in ophthalmology].

A study was conducted to evaluate the antifungal activity in vitro of 5 quaternary ammonium compounds. N cetylpyridinium was found to be particularly active against Candida albicans and Aspergillus fumigatus. The association of N cetylpyridinium and of neosynephrine, found in various eye drop compositions, resulted in potentation of the antifungal activity of this quaternary ammonium.

Clinical reduction of gingivitis through the use of a mouthwash containing two quaternary ammonium compounds.

A commercially available antimicrobial mouthrinse was evaluated among students in Mexican boarding schools. The application was made daily for 3 months under the direct supervision of the project personnel. The observations indicated that the use of this mouthrinse decreased gingivitis substantially in comparison to the use of a control rinse which consisted of flavored water. It is concluded that this mouthwash, containing the known antimicrobial agents cetylpyridinium chloride and domiphen bromide, can be of clinical benefit if conscientiously applied as an adjunct in an oral hygiene regimen of regular toothbrushing with a dentifrice.
