The total eosinophil count and adrenal function in asthmatic children.

In 36 asthmatic children 8:00 a.m. plasma cortisol levels were compared with changes in their total eosinophil count (TEC) from 8:00 a.m. to 9:30 a.m. All children with low cortisol levels had decreases in TEC of less than 2%, whereas 78% of children with normal cortisol levels had decreases greater than 15%. All children with decreases greater than 15% had normal cortisol levels.

Circadian variations of the absolute eosinophil count and serum histaminase activity in tropical pulmonary eosinophilia.

Nine healthy volunteers and 25 tropical pulmonary eosinophilia (TPE) patients were used to study circadian variations of absolute eosinophil count (AEC) and serum histaminase activity (SHA). A marked circadian variation was found in AEC for healthy volunteers and TPE patients with the worst symptoms in the late evening and morning hours only; no rhythm could be detected in SHA for healthy subjects. However, TPE patients with worst symptoms in the late evening hours did exhibit a significant rhythm in SHA. Increased SHA in all TPE patients at all time-points of the 24 hour day-night cycle, irrespective of the worsening hours of symptoms in comparison to healthy controls, could be due to increased histamine production in such situations.

The use of methacholine inhalation, methacholine skin testing, distilled water inhalation challenge and eosinophil counts in the evaluation of patients presenting with cough and/or nonwheezing dyspnea.

Twenty-four patients presenting with cough and/or nonwheezing dyspnea were evaluated with methacholine inhalation challenge (MC), distilled water inhalation challenge (DC), intracutaneous tests to varying concentrations of methacholine, total eosinophil counts (TEC), sinus and chest x-rays. We found a statistically significant difference (P less than .005) in the mean TEC in those patients with a positive MC test and those with a negative test. Hyperreactivity of the airways to methacholine in asthmatics is not found in the skin. Distilled water inhalation did not serve to substitute for MC as a test of hyperreactive airways. The TEC is an excellent screening test as a predictor of patients with cough or dyspnea who have hyperreactive airways.

Peripheral lymphocyte and eosinophil counts as indicators of prognosis in primary breast cancer.

As a part of a major study on the pathophysiologic indices for recurrence of human breast cancer, preoperative eosinophil and lymphocyte counts were determined on 419 and 581 primary breast cancer patients, respectively. Patients with lymphocyte counts less than or equal to 1500/mm3 and/or eosinophil counts of less than 55/mm3 had significantly higher risk of recurrent disease than those patients who had normal or high levels of eosinophils and/or lymphocytes. These findings may indicate that the immunologic activities of eosinophils and lymphocytes enhance the patients' ability to respond against disease.

Elevation of serum IgE level and peripheral eosinophil count during T lymphocyte-directed gene therapy for ADA deficiency: implication of Tc2-like cells after gene transduction procedure.

We have successfully carried out T-cell-directed gene therapy for a boy with severe combined immunodeficiency due to adenosine deaminase deficiency (ADA SCID) and unexpectedly found an elevation of serum IgE level and peripheral eosinophil count during the course. More than 90% of transduced cells cultured for 7-11 days before infusion into the patient were positive for CD8 and expressed Th2-type cytokine genes such as IL-4, IL-5 and IL-13. Furthermore, CD4(+) T-depleted PBMC (peripheral blood mononuclear cells) from the patient synthesized IgE in vitro by stimulation with IL-4. Collectively, these results suggested that Tc2-like cells in the transduced cells have distinct immunological functions to help IgE synthesis and activate eosinophils.
