Incorporation of tropical maize germplasm into inbred lines derived from temperate x temperate-adapted tropical line crosses: agronomic and molecular assessment.

Exotic maize ( Zea mays L.) germplasm may allow for increased flexibility and greater long-term progress from selection if it can be incorporated at high rates into U.S. breeding programs. Crosses were made between a temperate line, NC262A, and each of eight different lines consisting of 100% temperate-adapted tropical germplasm. Pedigree selection was used to generate a set of 148 F(5)S(2) lines that were evaluated in testcrosses with FR992/FR1064 in nine North Carolina environments. Several entries had grain yield, grain moisture content and standability that were comparable to three commercial checks. The best testcrosses outyielded the cross NC262A x FR992/FR1064 by 9.5 to 10.9%, suggesting that a significant amount of tropical germplasm was retained in these lines and that this germplasm combined well with the Stiff Stalk tester. Previous researchers had suggested that tropical alleles could be rapidly lost during inbreeding in populations derived from tropical x temperate bi-parental crosses, leading to the development of lines that possess significantly less than 50% tropical germplasm. F(5)S(5) sub-lines corresponding to the 14 best testcrosses were genotyped at 47 to 49 polymorphic simple sequence repeat (SSR) loci across all ten chromosomes to estimate the amount of tropical germplasm that was retained. The estimated genetic contribution from the tropical parent ranged from 32 to 70%, with the average being 49%. Only two of the 14 lines deviated significantly from a 50%-tropical/50%-temperate ratio, suggesting limited overall selection against germplasm from the tropical parents. These experiments collectively demonstrated that tropical maize germplasm can be incorporated at high rates into a temperate line via pedigree breeding methods in order to derive new inbred lines with acceptable agronomic performance.

Immunocytochemical characterization of cell lines from human malignant mesothelioma: characterization of human mesothelioma cell lines by immunocytochemistry with a panel of monoclonal antibodies.

A panel of nine monoclonal antibodies was used to characterize human mesothelioma cell lines that we established from human malignant mesothelioma. The antigens detected were cytokeratin, vimentin, epithelial membrane antigen, carcinoembryonic antigen, Leu-M1 (CD15), desmin, factor VIII-related antigen (von Willebrand factor antigen), OV632, and ME1, a specific monoclonal antibody directed against human malignant mesothelioma. The technique used was the alkaline phosphatase anti-alkaline phosphatase method. All 30 cell lines, either epithelial, sarcomatous, or mixed, showed strong reactivity with cytokeratin and vimentin antibodies. None of the cell lines demonstrated any reactivity with carcinoembryonic antigen, Leu-M1, or factor VIII antibodies; moreover, all of 22 cell lines studied were positive for ME1 antibody and 10 of 12 cell lines studied were positive for OV632. Some interesting features were noted: only two of the 30 cell lines presented a weak positive staining with epithelial membrane antigen, and nine of 19 cell lines tested demonstrated a cytoplasmic staining pattern with desmin antibody. These results show that established human mesothelioma cell lines still possess the immunocytochemical characteristics that are basically consistent with the immunohistochemical features described in tumor tissues of malignant mesothelioma. These characteristics can be used to identify the mesothelioma cells grown from human malignant mesothelioma. Hence, the mesothelioma cell lines will provide a useful tool for the investigation of the cell biology of the tumor and the mechanisms of mesothelial cell transformation, as well as the in vitro evaluation of the effects of some drugs in order to develop new therapies for malignant mesothelioma.

Newly established Askin tumor cell line and overexpression of focal adhesion kinase in Ewing sarcoma family of tumors cell lines.

Askin tumor is a malignant small round cell tumor that originates from the thoracopulmonary region and is a member of Ewing sarcoma family of tumors (ESFT). Only a few Askin tumor cell lines have been established. An Askin tumor cell line, designated MP-ASKIN-SA, was established from the left thoracic tumor of a 13-year-old Japanese boy. ESFT is known to have a high rate of distant metastases at diagnosis. The genes controlling the spread of ESFT cells, however, have not been elucidated. G-banding chromosome analysis revealed that the MP-ASKIN-SA cell line has complex chromosomal abnormalities including trisomy 8. The EWS/FLI1 chimeric transcript and c-myc overexpression were revealed by the reverse transcriptase-polymerase chain reaction and Northern blot analysis. Furthermore, we investigated the expression of the focal adhesion kinase (FAK) gene in the ESFT cell lines using Northern blot analysis. In addition to the MP-ASKIN-SA cell line, six Ewing sarcoma cell lines, one peripheral nerve sheath tumor cell line, and two Askin tumor cell lines were analyzed. All ESFT cell lines, including MP-ASKIN-SA, expressed five- to twenty-eight-fold-increased values of FAK, as compared with fibroblasts obtained from the bone marrow of a healthy volunteer. These results raise the possibility that the overexpression of c-myc and FAK are involved in the poor prognosis of ESFT.

Evasion mechanisms to tumor necrosis factor alpha (TNF-alpha) of small cell lung carcinoma and non-small cell lung carcinoma cell lines: comparison with the erythroleukaemia K-562 cell line.

The tumour necrosis factor alpha (TNF-alpha) is produced by mononuclear phagocytes as a defence mechanism against malignant cells. However, these cells can evade destruction by TNF-alpha. The present study evaluates in three lung cancer cell lines (small cell carcinoma NCI-H69, adenocarcinoma A-427, squamous carcinoma SK-MES-1) and one erythroleukaemia (K-562) cell line the following evasion mechanisms: (1) inhibition of TNF-alpha production, in indirect and direct co-cultures with monocytes; (2) the expression of type I and type II receptors for TNF-alpha (TNFRI and TNFRII) by tumour cell lines, using indirect immunofluorescence and flow cytometry; (3) the sensitivity of tumour cell lines to the toxic action of recombinant human TNF-alpha (rhTNF-alpha). With the exception of cell line NCI-H69, the other tumour cell lines liberated soluble factors that inhibited TNF-alpha production in monocytes. This effect occurred even after membrane contact with the A-427 and SK-MES-1 cell lines. Erythroleukaemia K-562 cells expressed both types of receptors for TNF-alpha, whereas the NCI-H69 cells expressed only TNFRI, and the A-427 and SK-MES-1 cells expressed no receptors. Lines NCI-H69, A-427 and K-562 were insensitive to the cytotoxic action of rhTNF-alpha. In conclusion, different lung cancer cell lines may evade destruction by TNF-alpha by various mechanisms that range from blocking TNF-alpha production by monocytes to blocking the cytotoxic action of this molecule. For selecting the most effective immunotherapy, knowledge of the evasion mechanisms would be useful.

Expression of cytokeratins (CKs) 8, 13 and 18 and their mRNA in epithelial linings of radicular cysts: implication for the same CK profiles as nasal columnar epithelium in squamous epithelial lining.

OBJECTIVE: Our objective was to specify cytokeratin (CK) patterns in lining epithelia of radicular cysts which are sometime lined with ciliated columnar epithelia as seen in the nasal epithelia. MATERIALS AND METHODS: We examined the CK expression in 52 radicular cysts obtained from 32 maxillary and 20 mandibular lesions and investigated CK-mRNA expression using in situ hybridization in 24 maxillary and 13 mandibular cysts and reverse transcription-polymerase chain reaction (RT-PCR) in 24 maxillary cysts. RESULTS: Of the maxillary cysts, 20, 29 and 19 squamous epithelial linings were positive for CK8, CK13 and CK18, respectively; of the mandibular cysts, 10, 20 and 11 linings were positive for these CKs, respectively. The expression patterns of CK18(+)-CK13(-), CK18(+)-CK13(+) and CK18(-)-CK13(+) were observed in 3, 16 and 13 linings of the maxillary cysts and 0, 11 and 9 linings of the mandibular cysts, respectively. In situ hybridization revealed the expression of CK18-mRNA in 9 and 4 linings of 24 maxillary and 13 mandibular cysts examined, respectively. With RT-PCR, we explored that both CK18- and CK13-mRNA were expressed not only in the normal nasal and gingival epithelia but also in the examined maxillary cyst linings although their expression levels differed correlating with the difference in CK staining. CONCLUSION: It is concluded that CK13- and CK18-mRNA are constitutively expressed in columnar and squamous epithelial cells, respectively, and that the variant CK expression patterns with CK18-mRNA expression in maxillary radicular cysts are indicative of the possibility of phenotypic transformation in the cyst linings.

Fat storage capacity in growth-selected and control mouse lines is associated with line-specific gene expression and plasma hormone levels.

OBJECTIVE: For a detailed understanding of the complex traits growth and fat storage, a dissection into single genetic entities is mandatory. Therefore, blood plasma concentrations of hormones and the expression of selected genes were measured in extremely differentiated mouse lines. Genes were selected as candidates which might influence the complex traits body weight and fat accumulation, and which are located in chromosomal regions recently identified to affect trait differences between the lines. SUBJECTS AND MEASUREMENTS: The mouse lines were selected for high body weight (DU6), high carcass protein content (DU6P) and unselected controls (DUKs). In the selected lines DU6 and DU6P, mean body weights at the age of six weeks were about twice as high as the DUKs, whereas total fat weight was increased 2.2-fold in DU6 mice compared to DU6P and 3.2-fold in comparison to DUKs. Blood plasma concentrations of insulin-like growth factor 1 (IGF-1), growth hormone (GH), insulin and leptin, were measured in all lines at three weeks and at six weeks of age. Expression patterns of the genes encoding growth hormone (Gh), insulin-like growth factor 1 (Igf1), lipoprotein lipase (Lpl), glycerolphosphate dehydrogenase 1 (GDC-1), and adipocyte protein 2 (Ap2) were analyzed by Northern blot hybridization. RESULTS: In line DU6, highly significant increased concentrations of insulin and leptin were observed at six weeks of age; at this stage, IGF-1 concentrations were elevated in the two selected lines compared to controls with maximal concentrations of IGF-1 and GH in DU6P. The amount of mRNA for GH in the pituitary gland, for Igf1 in the liver and for LPL in epididymal fat tissue was significantly elevated in the two selected lines compared to controls at the age of three weeks, but not at six weeks. IGF-1 and GDC-1 mRNA concentrations were significantly higher in the DU6 mice than in the DU6P (P < 0.01) and the DUKs (P < 0.001) mice examined at both ages. CONCLUSIONS: The results prove line-specific concentrations of the analyzed hormones and the transcription amounts of Gh, Igf1, GDC-1 and Lpl. The measured differences are either direct genetic effects or secondary changes, resulting from different food consumption.

The organization of titin filaments in the half-sarcomere revealed by monoclonal antibodies in immunoelectron microscopy: a map of ten nonrepetitive epitopes starting at the Z line extends close to the M line.

mAbs specific for titin or nebulin were characterized by immunoblotting and fluorescence microscopy. Immunoelectron microscopy on relaxed chicken breast muscle revealed unique transverse striping patterns. Each of the 10 distinct titin antibodies provided a pair of delicate decoration lines per sarcomere. The position of these pairs was centrally symmetric to the M line and was antibody dependent. The results provided a linear epitope map, which starts at the Z line (antibody T20), covers five distinct positions along the I band (T21, T12, T4, T1, T11), the A-I junction (T3), and three distinct positions within the A band (T10, T22, T23). The epitope of T23 locates 0.2 micron before the M line. In immunoblots, the two antibodies decorating at or just before the Z line (T20, T21) specifically recognized the insoluble titin TI component but did not recognize TII, a proteolytic derivative. All other titin antibodies recognized TI and TII. Thus titin molecules appear as polar structures lacking over large regions repetitive epitopes. One physical end seems related to Z line anchorage, while the other may bind close to the M line. Titin epitopes influenced by the contractional state of the sarcomere locate between the N1 line and the A-I junction (T4, T1, T11). We discuss the results in relation to titin molecules having half-sarcomere lengths. The three nebulin antibodies so far characterized again give rise to distinct pairs of stripes. These locate close to the N2 line.

M-line protein preparations from frog skeletal muscle: isolation and localization of an M-line protein and a 105 000 dalton polypeptide contaminant.

A 165 000 dalton polypeptide was purified and characterized from high salt extracted crude M-line protein preparations of frog skeletal muscle. It has an isoelectric point of 5.8-6.2 and an amino acid composition similar to that reported for the putative M-line protein in chicken muscle (termed component I) and rabbit muscle. Antibodies prepared against the 165 000 dalton polypeptide not not react with purified frog debranching enzyme, and indirect immunofluorescent studies localize this polypeptide wholly in the middle of the A band (presumably the M line) of frog skeletal muscle. We conclude that this 165 000 dalton polypeptide is a basic constituent of the M line in frog skeletal muscle. Crude M-line protein preparations from frog muscle invariably contained a 105 000 dalton polypeptide contaminant. In crude M-line protein preparation from chicken muscle, a polypeptide with a similar molecular mass was identified as phosphorylase b. Characterization of the 105 000 dalton polypeptide isolated from crude M-line protein preparations establishes that in frog muscle extracts this component has an amino acid composition which is different from phosphorylase b, but similar to frog alpha-actinin. Since it also exhibits immunochemical cross-reactivity with alpha-actinin and not with purified phosphorylase b, and is localized wholly at the Z line, we conclude that the 105 000 dalton polypeptide contaminant in crude M-line protein preparations from frog muscle is alpha-actinin.

Second-line chemotherapy of advanced disseminated gastric cancer after cisplatin, infusional 5-fluorouracil, folinic acid (PLF): benefit dependent on progression-free interval after first-line therapy.

BACKGROUND: The role of second-line chemotherapy in gastric cancer is not yet established. We analyzed patients receiving second-line treatment after chemotherapy with cisplatin, infusional 5-fluorouracil, folinic acid (PLF). PATIENTS AND METHODS: 27 patients took part in this retrospective analysis. The second-line chemotherapy consisted of different regimens depending on the recommendations of the attending physician based on response to and toxicity of first-line treatment. RESULTS: In 3 of 27 patients, partial response was achieved (11%, 95%-confidence interval 0-23%). The progression-free survival and overall survival was 3.1 (2.1-4.3) and 5.1 (3.8-6.6) months, respectively. Progression-free intervals (PFI) of more than 7 months after first-line therapy were a predictor for patients who may benefit from the treatment (median survival from the start of second-line therapy: 10.6 (6.6-13.1) months). CONCLUSION: Second-line chemotherapy after cisplatin / 5-FU rarely induced tumor response, and progression-free survival as well as overall survival remained short. Based on our data, we hypothesize that the PFI after first-line treatment may be a selection criterion for patients suitable for second-line chemotherapy.

The development of lateral-line receptors in Eigenmannia (Teleostei, Gymnotiformes). I. The mechanoreceptive lateral-line system.

The South American weakly electric fish of the genus Eigenmannia were induced to spawn by simulating the conditions of the rainy season. Whole animals were viewed using scanning electron microscopy, and skin from embryos and larvae of different ages was prepared for histological examination. Additional live fish were stained with vital dyes. Neuromasts develop within the epidermis and then rise to the surface, at which time a cupula is forming. The first neuromasts appear on the head, forming the temporal, mechanoreceptive lateral line, at 3.5 days after spawning, and 1 day later neuromasts appear on the trunk as a ventral trunk line. On day 8 all the cephalic neuromasts have appeared and a secondary, medial trunk line begins to form. A dorsal trunk line forms when the fish are juvenile. Eight neuromasts of the cephalic lines, 7 neuromasts of the medial trunk line and all neuromasts of the ventral and dorsal trunk lines remain at the surface and do not become enclosed in canals. The opercular neuromasts and 7 neuromasts of the ventral trunk line degenerate later. The formation of the head canals begins on day 17, whereas the canal of the medial trunk line starts to develop on day 25, and both head and trunk canal systems are completed by day 33. The mechanosensory system develops before the electrosensory system. Behavioral observations also indicate that the mechanoreceptive system is functional as early as day 5.

Responses to 19 generations of litter size selection in the Nebraska Index line. I. Reproductive responses estimated in pure line and crossbred litters.

Our objective was to estimate responses in reproductive traits in the Nebraska Index line (I) after 19 generations of selection for increased litter size. Responses were estimated in dams producing pure line, F1, and three-way cross litters. A total of 850 litters were produced over six year-seasons, including 224 pure line litters, 393 F1 litters produced from I and C females mated with Danbred NA Landrace (L) or Duroc-Hampshire (T) boars, and 233 litters by F1 L x I and L x C females mated with T boars. Contrasts of means were used to estimate the genetic difference between I and C and interactions of line differences with mating type. Farrowing rates of lines I (u = 91.0%) and C (u = 92.8%) did not differ. Averaged across all genetic groups, mean number born alive per litter was 10.1 pigs, and number and weight of pigs weaned per litter, both adjusted for number nursed and weaning age of 12 d, were 9.7 pigs and 34.4 kg, respectively. Averaged across mating types, direct genetic effects of I were greater than C (P < 0.05) for total born (3.53 pigs), number born alive (2.53 pigs), number of mummified pigs (0.22 pig), and litter birth weight (2.14 kg). The direct genetic effect of line I was less than C (P < 0.05) for litter weaning weight (-1.88 kg). Interactions of line effects with crossing system were significant (P < 0.05) for total number born, number of stillborn pigs, number weaned, and litter weaning weight. In pure line litters, I exceeded C by 4.18 total pigs and 1.76 stillborn pigs per litter, whereas the estimate of I-C in F1 litters was 2.74 total pigs and 0.78 stillborn pig per litter. The contrast between I and C for number weaned and litter weaning weight in pure litters was 0.32 pig and -0.28 kg, respectively, compared with 0.25 pig and -2.14 kg in F1 litters. Crossbreeding is an effective way to use the enhanced reproductive efficiency of the Index line.

Does intensive treatment with high dose chlorambucil and prednisone as first line and cladribine as second line influence the survival of the patients with chronic lymphocytic leukemia?

Cladribine (2-CdA) and fludarabine are the new purine analogs introduced in the treatment of chronic lymphocytic leukemia (CLL). Despite the high response rate, their influence on survival is still uncertain. The aim of this study was a retrospective analysis and comparison of the response rate and survival of CLL patients treated with high dose chlorambucil (HDChl) as first and 2-CdA as second line, with an historical group of patients never treated with purine analogs who received standard doses of chlorambucil (SDChl). We analyzed 347 patients with CLL treated between January 1985 and January 2000. Group A (190 patients) received HDChl (12 mg/m(2) ) with prednisone (P) 30 mg/m(2) daily for 7 days monthly as first line and in refractory or early relapsed patients 2-CdA (0.12 mg/kg/day) for 5 days with or without P (30mg/m(2) ) as second line. Group B (157 patients) received continuous SDChl (4-8 mg/m(2) /day) and P as first line and COP or CHOP as second line. The overall response rate (OR) for the first line was 48,4% in group A and 38,9% in group B (p=0.09). 148 patients in group A and 52 in group B received the second line treatment and the second OR was 19.6% and 13.5%, respectively (p=0.4). At the time of analysis, 124 patients died in group A and 139 in group B. Median survival was 65 months and 50 months, respectively. In group A, survival was longer in advanced Rai stage patients (p=0.001) but in early Rai stage was similar for both groups (p=0.4). We suggest that intensive treatment with HDChl as first line and 2-CdA as second line should be applied in more advanced rather than in less advanced stages of CLL until the final results of randomized clinical trials are available.

Deaths and disposals in lines selected for milk yield and control lines of Jersey cattle.

Deaths and disposals of Jerseys from birth to 120 d of first lactation during a selection experiment were summarized. Cows were randomly assigned to the high milk yield and young sire lines. Service sires were bulls with the highest estimated breeding value for milk for the high milk line and a collection of all young unproven bulls available in AI in 1967. Cattle that were unsuitable for conditions of normal herd management were candidates for culling. Culling of uncalved heifers and of lactating cows was also permitted to maintain group sizes. Culling for herd reduction was random. Culling for low milk yield was permitted after cows reached 120 d of first lactation. Death and disposal data were analyzed through 120 d of first lactation. By 120 d of first lactation, 29.6% of females born to the young sire line had died or been involuntarily removed compared with 29.8% born to the high milk yield line. Deaths were 54.8% of total losses for the high milk yield line compared with 58.4% for the young sire line. Stillbirths were a larger proportion of total losses for animals in the high milk yield line (21.4%) than for those in the young sire line (15.3%). The most frequent cause of death was scours and pneumonia during 1st mo of life for animals in both lines. Selection of sires for high milk yield does not decrease total survival rates through the early first lactation.

Endogenous viral genes in thirteen highly inbred chicken lines and in lines selected for immune response traits.

Thirteen highly inbred lines of chickens of Leghorn, Spanish, and Egyptian Fayoumi origin, four partly inbred Leghorn lines selected for MHC alleles and immune response to GAT (Ir-GAT), and two replicated, noninbred Leghorn lines divergently selected for multiple immune response traits were subjected to molecular genotyping for endogenous viral (ev) gene sequences. In all highly inbred lines of Leghorn origin, ev1 alone or both ev1 and ev2 were observed. The Spanish and Fayoumi lines had three and five ev genes, respectively, most of which were not readily identifiable with standard Leghorn ev gene loci. The Leghorn lines selected for MHC and Ir-GAT had ev1 fixed in the population. Differences in ev3 and ev5 gene frequency were associated with Ir-GAT in the B1 haplotype, but not in the B19 haplotype. In the noninbred lines, which were divergently selected for multiple traits of immune responsiveness, ev6 and ev9 differed in frequency between lines, and both were in lower frequency in the lines selected for high immunoresponsiveness. These two ev genes are the only ones known in White Leghorns that have the gs-chf+ phenotype [expressing chicken helper factor (chf) but not expressing group-specific antigen (gs)].

Establishment and characterization of immortalized non-transplantable mouse mammary cell lines cloned from a MMTV-induced tumor cell line cultured for a long duration.

During the culturing of a mouse mammary tumor cell line, MuMT73, maintained in vitro for more than a decade, we found morphological heterogeneity in its cells; some showed contact inhibition in their growth, some formed domes and some grew criss cross and piled up. In trying to clone the cell line to isolate cells showing contact inhibition or dome formation, we were able to establish six clonal cell lines. These six cell lines were categorically divided into three groups according to their phenotypical behavior, Groups A, B and C. Group A (clones 1, 5 and 7) cells had a property of contact inhibition. They induced no tumor when injected into the subcutaneous tissue of the back, nor even when injected into the mammary fat pads or under the kidney capsule of syngenic or nude mice, and therefore were thought to be non-malignant in nature. They were positively stained by anti-keratin antiserum and had mouse mammary tumor viruses (MMTVs). Group B (clone 6) cells grew in a crisscross pattern and piled up, and they induced tumors when injected into the subcutaneous tissue of the back of mice. Group C (clones 3 and 4) cells formed domes in their growth and induced some tumors in the mammary fat pads and under the kidney capsule of KSN nude mice. In Southern blots with MMTV-env probe, numerous exogenous MMTV proviruses were detected in these cell lines. The insertion patterns of these proviruses in cells of non-malignant clonal lines (Group A) resembled those of malignant lines (Group B), except one band (about 26 Kb), but were considerably different from those of intermediate lines (Group C). On the other hand, no difference was detected in Southern blots with int-1 or int-2 probes among the non-malignant, intermediate and malignant clonal cell lines.

Induction of metastatic ability in a stably diploid benign rat mammary epithelial cell line by transfection with DNA from human malignant breast carcinoma cell lines.

Transfection of the stably diploid rat mammary epithelial cell line, Rama 37, which yields nonmetastasizing, adenomatous tumors in syngeneic rats with HindIII-fragmented DNA from malignant or nonmalignant human breast epithelial cell lines and the drug-resistance plasmid pSV2neo, yields transformants with a frequency of 10(-4) to 10(-5). The resultant cell lines form tumors with varying frequencies when injected s.c. into the mammary fat pads of syngeneic rats. Cells transfected with DNA from the malignant human breast carcinoma cell line, Ca2-83, or DNA from the human pleural effusion-derived cell lines, MCF-7 or ZR-75-1, yield transformants which metastasize to lungs and/or lymph nodes at high frequency, whereas transfection of HindIII-fragmented DNA from nonmetastatic human mammary epithelial cell lines, transfection of the drug-resistance plasmid pSV2neo alone, or nonspecific DNA such as salmon sperm DNA fails to yield transformants expressing the metastatic phenotype. Transfectants which metastasized were reestablished in culture and reinjected into syngeneic rats to confirm their metastatic properties. These transfectants yield rapidly growing tumors with reduced latent periods, which give rise to significant numbers of metastases. The karyotype of selected transfectants after passage in vivo remains stably diploid. Hybridization of a 32P-labeled oligonucleotide probe specific for the human Alu family of sequences to DNA from these transfectants reveals the presence of human-specific DNA sequences integrated into the genome. It is suggested that transfection of specific genomic DNA sequences from the malignant human cell lines can induce the metastatic phenotype in the nonmetastatic Rama 37 cell line in a genetically dominant manner, whereas genomic DNA from the nonmetastatic cells cannot confer metastatic properties to the Rama 37 cell line.

Effect of curing regime on the cytotoxicity of resin-modified glass-ionomer lining cements applied to an odontoblast-cell line.

OBJECTIVE: The aim of this in vitro study was to evaluate the cytotoxicity of resin-modified glass-ionomer lining cements submitted to different curing regimes and applied to an immortalized odontoblast-cell line (MDPC-23). METHODS: Forty round-shaped specimens of each experimental material (Fuji Lining LC and Vitrebond) were prepared. They were light-cured for the manufacturers' recommended time (MRT = 30 s), under-cured (0.5 MRT = 15 s), over-cured (1.5 MRT = 45 s) or allowed to dark cure (0 MRT). Sterilized filter papers soaked with either 5 microL of PBS or HEMA were used as negative and positive control, respectively. After placing the specimens individually in wells of 24-well dishes, odontoblast-like cells MDPC-23 (30,000 cells/cm2) were plated in each well and incubated for 72 h in a humidified incubator at 37 degrees C with 5% CO2 and 95% air. The cytotoxicity was evaluated by the cell metabolism (MTT assay) and cell morphology (SEM). RESULTS: Fuji Lining LC was less cytotoxic than Vitrebond (p < 0.05) in all the experimental conditions. However, the cytotoxicity of Fuji Lining LC was noticeably increased in the absence of light-curing while the same was not observed for Vitrebond. The length of light-curing (15, 30 or 45 s) did not influence the toxicity of both lining materials when they were applied on the odontoblast-cell line MDPC-23. SIGNIFICANCE: The light-activation plays an important role in reducing the cytotoxicity of Fuji Lining LC. Following the manufacturer' recommendation regarding the light-curing regime may prevent toxic effect to the pulp cells.

Glial cell line-derived neurotrophic factor receptor alpha1 availability regulates glial cell line-derived neurotrophic factor signaling: evidence from mice carrying one or two mutated alleles.

Glial cell line-derived neurotrophic factor receptor alpha1 (GFRalpha1, also known as GDNFR-alpha) is a glycolipid-anchored membrane protein of the GFRalpha family, which binds glial cell line-derived neurotrophic factor [Jing S. et al. (1996) Cell 85, 1113-1124; Treanor J. J. et al. (1996) Nature 382, 80-83], a survival factor for several populations of central and peripheral neurons, including midbrain dopamine neurons [Lin L. F. et al. (1993) Science 260, 1130-1132], and mediates its ligand-induced cell response via a tyrosine kinase receptor called Ret [Takahashi M. et al. (1988) Oncogene 3, 571-578; Takahashi M. and Cooper G. M. (1987) Molec. Cell Biol. 7, 1378-1385]. In this paper, we show that mice with a null mutation of the GFRalpha1 gene manifest epithelial-mesenchymal interaction deficits in kidney and severe disturbances of intestinal tract development similar to those seen with glial cell line-derived neurotrophic factor or Ret null mutations. There is a marked renal dysgenesis or agenesis and the intrinsic enteric nervous system fails completely to develop. We also show that newborn GFRalpha1-deficient mice display no or minimal changes in dorsal root and sympathetic ganglia. This is in contrast to the deficits reported in these neuronal populations in glial cell line-derived neurotrophic factor and Ret null mutations. Mesencephalic dopaminergic neurons in the substantia nigra and ventral tegmental area appear intact at the time of birth of the mutated mice. Mice homozygous for the GFRalpha1 null mutation die within 24 h of birth because of uremia. Heterozygous animals, however, live to adulthood. There is a significantly reduced neuroprotective effect of glial cell line-derived neurotrophic factor in such heterozygous animals, compared with wild-type littermates, after cerebral ischemia. Taken together with previous data on glial cell line-derived neurotrophic factor and Ret, our results strongly suggest that GFRalpha1 is the essential GFRalpha receptor for signaling in the glial cell line-derived neurotrophic factor-Ret pathway in the kidney and enteric nervous system development, and that GFRalpha2 or GFRalpha3 cannot substitute for the absence of GFRalpha1. Moreover, neuroprotective actions of exogenous glial cell line-derived neurotrophic factor also require full GFRalpha1 receptor expression.
