Microbioassay system for an anti-cancer agent test using animal cells on a microfluidic gradient mixer.

We developed a novel microbioassay system equipped with a gradient mixer of two solutions, and we applied the microfluidic system to an anti-cancer agent test using living animal cells on a microchip. A microchannel for the gradient mixing of two solutions and eight other microchannels for cell assay were fabricated on a poly(dimethylsiloxane) substrate using a soft-lithography method. The functions necessary for this bioassay, i.e., cell culturing, chemical stimulation, cell staining, and fluorescence determination, were integrated into the microfluidic chip. Eight gradient concentrations of the fluorescein solution, ranging from 1 to 98 microg/ml, were archived at 0.1 microl/min on a microchip. A stomach cancer cell line was cultured, and a cell viability assay was conducted using 5-Fluorouracil as an anti-cancer agent on the microchip. Cell viability changed according to the estimated concentration of the agent solution. With the microbioassay system, an anti-cancer agent test was conducted using living cells simultaneously in eight individual channels with the gradient concentration of the agent on a microchip.

Studies of wet granulation in small scale high shear mixer of 20 mg tablets of policosanol.

It was studied the influence of three factors in the manufacturing process of tablets containing 20 mg of policosanol in a small scale when was used a high shear mixer. A factorial design without repetitions was used, considering as independent variables: volume of binder solution, kneading time and the use or not of chopper. Data analysis was carried out using the Yates algorithm, considering the effect of these three factors over the diameter of the granules, the percent of fine powders in granulate and the compression forces. The diameter of the granules was increased with an increase in the volume of the binder solution added and with an increase in the kneading time and was decreased when the chopper was used. The percent of fine powders, present in granulate, was significantly diminished with an increase of the volume of binder solution and diminished with an increase in the kneading time, the influence of the use of chopper was insignificant. The compression forces diminished with the use of chopper, with the increase of the volume of binder solution used and are not affected with the changes in the kneading time.

Preparation of sustained release hydrophilic matrices by melt granulation in a high-shear mixer.

PURPOSE: The objective of this work was to prepare theophylline sustained release matrix tablets based on the combination of hydroxypropyl methylcellulose (HPMC K4M and K100M) and different meltable binders by melt granulation in a high-shear mixer. METHODS: Dissolution profiles of each formulation were compared to those of TheoDur 200 mg tablets and the mean dissolution time (MDT) and similarity factor (f2 factor) were calculated. The matrices swelling behavior was investigated by texture analysis. RESULTS: The results obtained show that the type of excipient influenced the drug release rate. In particular, the dissolution rate was delayed when lipophilic binders were used and only formulations containing Gelucire 50/13 or PEG 6000 with HPMC K4M had a profile similar to the commercial formulation. The release mechanism of theophylline from the formulations was described by Peppas's equation showing a non-Fickian release mechanism. The investigation of matrices swelling behavior showed that the gel layer thickness increased continuously over the time period studied. Moreover, a correlation between gel layer thickness and strength with the percentage released was found. CONCLUSIONS: These results suggest that melt granulation could be an easy and fast method to formulate sustained release tablets.

[An amalgam mixer for dental work].

A stomatologic amalgam mixer with an automated dosage device ACD-02 is destined for measuring the powdered alloy of silver and mercury out, preparation of amalgams from the mentioned components or filling material "Gallodent-M" in conditions of stomatological polyclinics. Constant mixture of powder layers in the scale body reservoir and immediate measuring out the powdered alloy on switching on the electric drive are provided. The apparatus is completed by a cup with a forked holder and a special reusable capsule for manufacture of filling material "Gallodent-M".

Sample preparation of some shelled treenuts and peanuts in a vertical cutter-mixer for mycotoxin analysis.

A procedure has been devised for more rapid preparation of large samples of nutmeats (10-80 lb) for mycotoxin analysis without the use of grinding aids, by combining the grinding and mixing steps into one operation. Ease of cleaning equipment facilitates the preparation of several samples in a short time. The procedure was tested using shelled raw and roasted peanuts, and raw almonds, walnuts, and pecans. The reduction of particle size to pass a 20 mesh screen was attained using a 25 qt Hobart vertical cutter-mixer (VCM) equipped with standard serrated blades and a 40 qt VCM using either a standard serrated blade or a smooth-edge blade modified with sharp-edge notches to increase its cutting ability. Portions of ground composite were removed at various time intervals and particle sizes were measured. Original and check analyses of 12 naturally contaminated samples over a 3 year period indicate that this procedure is practical and reproducible.

Reduction of pesticide exposure with protective clothing for applicators and mixers.

Systemic pesticide illness in agricultural workers may result from excessive dermal exposure to pesticides. Workers who apply and mix pesticides (applicators and mixers) are not at special risk. Both acute and chronic exposures can occur from spillage or by environmental contamination of clothing. Two exposure assessment studies were conducted of Central Florida citrus grove workers who applied ethion daily. Measuring the percentage of penetration of ethion through clothing and measuring the daily urinary excretion of diethyl phosphate (DEP) were the techniques used to assess the protection afforded by changing daily to freshly laundered 100% cotton coveralls. Coveralls provided significantly greater protection than did regular clothing and the use of respirators.

[Stomatological amalgam mixer].

For preparing silver amalgam an amalgam mixer AC-01 has been constructed with a time relay. This is a table apparatus in whose body and oscillating movement mechanism with an electric drive and a time relay are arranged. Mixing the amalgam is done in capsules. The set is made up together with proportioners for mercury, powder and capsules for the preparation of the amalgam.

Coaxial flow mixer for real-time monitoring of cellular responses in flow injection cytometry.

Improved time resolution of kinetic cellular events in flow cytometry is demonstrated by using a coaxial flow-mixing device integrated within a flow-injection (FI) system. The instrument is used in combination with a Becton Dickinson FACS Analyzer for on-line reagent addition, rapid sample mixing, and temperature control of cell suspensions. The coaxial flow device can instantaneously (< 60 ms) mix reagent and sample streams, allowing cytometric analysis of subsecond events to be performed. Kinetic measurements can be performed on the FACS analyzer in a variable time range of from 100 ms to 3 min. The system also allows the collection of unlimited cellular events at a specific incubation time point. Because the system operates continuously and no boost in core flow is required, disturbances of flow conditions are avoided. The capabilities of the flow injection cytometer have been demonstrated by the determination of internal [Ca2+]i mobilization in Jurkat T lymphocytes perfused internally with INDO-1 and stimulated by ionomycin.
