Human alpha-fetoprotein and its purification by chromatography on immobilized estrogens.

Human alpha-fetoprotein (AFP) was isolated from human abortive tissue by biospecific chromatography on immobilized estrogens. The most effective sorbents were: estrone-0-3-hemisuccinyl-hexamethylenediamine-Sepharose CL 4B and diethylstilbestrol-diasoanisole-sulfonyl-oxyethyl-Sepharose CL 4B. As elution solution the most optimum was 10% buffered aqueous butanol. Taking into consideration the data obtained, one can conclude that AFP in human biological fluids is bound to immobilized estrogens. Butanol extraction deestrogenizes AFP, and as a result human AFP acquires affinity to immobilized estrogens. During rechromatography on immobilized diethylstilbestrol, it was possible to obtain AFP preparations of about 95% purity. The present results provide the opportunity to work out new methodological approaches to human AFP isolation using biospecific chromatography on immobilized estrogens.

Simultaneous comparison of relative damage to chromatic pathways in ocular hypertension and glaucoma: correlation with clinical measures.

PURPOSE: To use a new methodological approach, based on luminance noise, to assess without bias the relative damage of blue-yellow and red-green pathways in ocular hypertension and glaucoma and to correlate obtained measurements with clinical markers of disease progression. METHODS: A psychophysical procedure modified from Cambridge color test was used to assess color discrimination in three different groups: patients with primary open-angle glaucoma (n = 51 eyes), patients with ocular hypertension (n = 95 eyes), and control subjects (n = 46 eyes). Viewing conditions were such that the function of the macula was being tested, using a discrimination task under noisy conditions. Confusion vectors, and parameters obtained from discrimination ellipses were correlated with perimetric and clinical data taken from the same groups. RESULTS: The lengths of the major axis of chromatic discrimination ellipses and all confusion vectors were significantly different between the groups. These measures followed a significant gradient of worsening performance from the control to hypertensive and glaucoma groups, which was steeper for the tritan axis. There was a significant global positive correlation between test parameters and cup-to-disc ratio and a significant negative correlation with the perimetric mean deviation index. Ellipse length in patients with ocular hypertension correlated significantly with the duration of their hypertensive state. CONCLUSIONS: This psychophysical test can detect visual dysfunction in a significant subset of patients with long-term hypertension with preserved visual fields. Macular function is damaged earlier than previously believed, in both the blue-yellow and red-green pathways.

Antinuclear antibody profile in Italian patients with connective tissue diseases.

In the present work we report data on the specificity of antinuclear antibodies (ANA) in a large series of Italian patients suffering from a broad spectrum of connective tissue diseases (CTD), by using a series of homogeneous and validated techniques. The present study confirms, on the one hand, generally accepted concepts, i.e. that certain autoantibodies are strictly associated to certain disease states (such as anti-PCNA and anti-Sm in systemic lupus erythematosus, Jo 1 in polymyositis, and ACA and Scl-70 in scleroderma); the presence of 'marker' antibodies is, however, restricted to a relative minority of CTD patients. The application of a new methodological approach that considers the entire profile of ANA can greatly augment their diagnostic relevance and may provide useful indications for their interpretation, allowing us to establish for the first time the diagnostic usefulness not only of marker autoantibodies but also of certain associations between non-marker autoantibodies. Finally, the application of a more appropriate and powerful statistical tool (multiple correspondence analysis) has further emphasized the clear relationship existing between antibody specificities and certain disease states.

Relation between heart rate and problem behaviors.

A new methodological approach for understanding self-injury, aggression, and property destruction exhibited by individuals with severe developmental disabilities was evaluated in this descriptive study. Measures of heart-rate changes before, during, and after episodes of problem behavior were obtained in real time. Unique patterns of heart rate and problem behavior were documented for each participant. Heart rate, identified as an indicator of negative arousal, was reliably associated with higher scores of perceived distress. These results indicate that further research is needed to examine the link between problem behavior and physiology. The study also suggests that physiological measurement can be included in the functional assessment process to further our understanding of the mechanisms underlying problem behavior.

Fluorescence studies on association of human translation initiation factor eIF4E with mRNA cap-analogues.

Binding of a long series of mono- and dinucleotide analogues of the 7-methylguanosine containing 5'-mRNA-cap to human protein translation initiation factor eIF4E has been investigated by means of fluorescence. A new methodological approach in gathering and analysis of the fluorescence data provided us with very accurate values of the association equilibrium constant K and normalized, maximal quenching of the protein fluorescence delta Fmax, during titration of eIF4E by various cap-analogues. The results confirm participation of at least two conserved tryptophan residues of eIF4E in interaction with 7-methylguanine, as has been described recently for murine eIF4E, complexed with 7-methyl-GDP in crystal (Marcotrigiano et al., 1997, Cell 89, 951), and for yeast eIF4E, complexed with the same ligand in solution (Matsuo et al., 1997, Nature Struct. Biol. 4, 717). On the other hand binding by eIF4E of unmethylated guanine nucleotides and N2,N2,7-trimethylguanine containing nucleotides differ substantially from the way of binding of the regular mRNA-cap. Influence of the structural features of the cap-analogues, especially the type of the second nucleoside in the dinucleotide caps, on their association with eIF4E and biological activities in in vitro protein translation systems has been discussed in light of the known structures of the eIF4E-7-methyl-GDP complexes in crystal and solution.

Speed, sex, gay men, and HIV: ecological and community perspectives.

Fifteen years into the HIV/AIDS epidemic, a great deal is now known about the different populations impacted by the disease, including those affected directly or indirectly by drug use. Anthropology has played a critical role in assisting with this task by identifying hidden populations, developing new methodological approaches, and targeting outreach efforts. In spite of this considerable body of ethnographic knowledge, men who have sex with other men (i.e., MSM, or gay and bisexual men) who use drugs have not received the same research attention as other drug users, despite the fact that they represent nearly one-fifth of AIDS cases in the U.S. with injection drug histories. In response to the alarming increase in HIV seroprevalence among this population, this ethnographic project provides preliminary data about those who are at dual risk for HIV through both homosexual behavior and injection drug use.

A second opinion: rethinking the public-private dichotomy for health insurance.

Does the public-private dichotomy effectively describe health insurance systems in the advanced industrialized democracies? Is the boundary separating the public and private sectors accurate for studies o f social policy formation and cutback? This article has three goals. The first is to discuss reasons for reconsidering the public-private dichotomy, as it applies to health insurance systems. The second is to offer a reconceptualization of the public-private demarcation useful for analyses of health insurance systems; the author presents four sectors that may illuminate patterns of health insurance for different OECD countries: the social, individual, public, and market sectors. The third goal is to present results using a new methodological approach useful for studying complex social phenomena: the fuzzy-set approach, which allows researchers to treat social phenomena as partially belonging to more than one category. This approach is employed to demonstrate that health insurance provision rarely is solely public or private, but is formed by a combination of sectors. Underlying these three goals is the contention that comparative and historical sociological researchers can offer innovative approaches to the study of health insurance and the interests served by public and nonpublic health insurance programs through reconceiving the public-private dichotomy.

Application of mass spectrometry to structural identification of flavonoid monoglycosides isolated from shoot of lupin (Lupinus luteus L.).

Flavonoid glycosides constitute important group of plant secondary metabolites. This class of natural products play significant role in different physiological processes. A new methodological approach where mass spectrometric techniques are applied to structural studies of this class of compounds is presented. Four flavonoid O-monoglycosides and one C-monoglycoside were isolated from green parts of lupin (Lupinus luteus L.). Several different mass spectrometric techniques were applied to structural elucidation of isolated compounds. Desorption ionization mass spectrometry was used for registration of mass spectra of intact and derivatized (permethylated) flavonoid glycosides. In some cases electron impact mass spectra of permethylated compounds were also recorded. Methylated samples after methanolysis and further derivatization of free hydroxyl groups (methylation or acetylation) were analyzed with gas chromatography-mass spectrometry. Combined information drawn from the registered mass spectra enabled us to define molecular mass, structure of aglycones and sugars, and positions of glycosidic bonds on the aglycon. Structures of four flavonoid monoglycosides were elucidated as follows: genistein 7-O-glucoside (1), genistein 4'-O-glucoside (2), 2'-hydroxygenistein 7-O-glucoside (3), and apigenin or genistein 8-C-glycoside (5). For the fourth O-glycoside (4) only molecular mass and masses of the aglycone and sugar were estimated.

[Isolation of a panel of HIV-positive serum by blocking them with various HIV antigens].

A new methodological approach to preparation of a panel of positive sera containing antibodies to individual viral proteins has been developed. The method consists in exhaustion of initial sera from HIV-infected patients with the known titers of antibodies by HIV antigens differing by the spectrum of virus proteins. Antigen preparations containing the least amounts of envelope proteins were selected for exhaustion. A set of HIV-positive sera containing antibodies mainly to envelope proteins and differing in ELISA was obtained.

[The rehabilitation of middle-aged and elderly patients in a pulmonary tuberculosis clinic].

In respect of other age groups the number of elderly and old patients constantly increases and today it constitutes the 5th part of population in many countries. That's why the "ageing" of tuberculosis is a natural process. The urgency of the problem of pulmonary tuberculosis in the elderly and senile age is determined by the high epidemiologic indices of disease incidence, by the difficulties of diagnosis and clinical course and by the peculiarities of rehabilitative measures in the patients of these age groups. The article deals with some medical aspects of the hospital period of rehabilitation in the patients with pulmonary tuberculosis. These aspects concern the new methodological approaches to the evaluation of therapeutic efficiency and to the formation of patient populations of the elderly and senile age, who need the restorative treatment under conditions of the specialized rehabilitative department.

[Comparative evaluation of neurovirulence of domestic and foreign live mumps vaccine].

Morphological and immunofluorescent study of changes in the central nervous system of monkeys with mumps was carried out in order to determine the criteria of neurovirulence of different mumps virus strains. Quantitative evaluation showed a lower residual neurovirulence of L-3 strain vs. Jeryl Lynn and Urabe Am9 strains. Use of new methodological approaches to evaluation of mumps vaccine strain neurovirulence will improve the safety control of live mumps vaccines.

[Ultrasound transabdominal study of the gastrointestinal tract].

It has been conventionally believed for a long time that echography of the intestines and other hollow organs containing gas could give very little information and would be even impossible at times due to the acoustic noise created by the gas medium in their lumen. Nevertheless, it is high time to elaborate new methodological approaches to diagnostics of these diseases and assessment of efficiency of the conducted therapy due to the wide spread of gastrointestinal tract diseases, complexity of their pathogenesis, insufficient effectiveness of the current methods of treatment, and the inclination to the prolonged recurrent course.

[Methodological aspects of gas-mercury monitoring of non-industrial areas].

The paper presents new methodological approaches to gaseous mercury monitoring whose key link is to record the territorial background level of air pollution, which is calculated by using the frequency distribution diagrams of single measurements for uniform objects within an area by comparing them with logarithmically normal random distribution. The methodological approach has been used during surveys of schools and preschool children establishments in Moscow, Kaluga, and the Kaluga Region.

[The role of liver antioxidant system in predisposition of rats to hepatotoxic effect of ethanol].

A new methodological approach capable of revealing factors responsible for the susceptibility of rat liver to ethanol hepatotoxicity has been developed. Using the correlation, dispersion, iteration, multifactor regression, and canonical analyses, a relation was established between the initial state of the liver antioxidant system and the character and degree of the subsequent ethanol-induced damage. In particular, it was found that intact animals with initially low level of reduced glutathione and retinols in the liver, as well as those with enzymopathy of cytosol HDNB-glutathione-8-transferase, are more susceptible to the ethanol liver damage.

[Capillary electrophoresis for monitoring stability of pharmaceutical proteins].

Like chemical drugs, pharmaceutical proteins have to be of strictly controlled purity to become biopharmaceuticals. Detection, identification and characterization of impurities and compounds present in purified biopharmaceuticals are of central interest. Simultaneous development of structural biology, biotechnologies and of highly resolutive analytical tools, allow significant progress in this field to be achieved. Thus, capillary electrophoresis can be advantageously used for monitoring protein stability. As illustrated by the examples of potential enzymes for detoxification of organophosphates, prophylaxis and treatment of poisoning by pesticides and nerve agents or for skin decontamination, the present feature article shows that this new methodological approach allows: to determine purity and homogeneity of proteins;to analyze their resistance to denaturing conditions such as heat or high electric fields;to detect the presence of unwanted hidden protein-bound ligands capable of altering functional conformation and stability of enzymes, and moreover susceptible to be released and to induce side effects, such as immunologic response; and to establish the role of parameters controlling the "good compromise" between conformational stability and plasticity for allowing optimal functional efficiency of enzymes. Finally, capillary electrophoresis has proved to be a pertinent tool to validate the conformity of purified enzymes to a status of biopharmaceutical.

Histochemical localization of capillary enzyme activities in brain smears.

Since the regulation of the vascular permeability in the CNS is dependent partly on the enzymes associated with the wall of brain capillaries, the histochemical demonstration of these enzymes may furnish further data on the function of the blood brain barrier (BBB), a new methodological approach, using brain smears was developed for the histochemical demonstration of several enzymes participating in the function of the BBB. The method presented renders possible also the subsequent demonstration of monoamines and the activity of different enzymes in the same tissue preparation. The usefulness of this simple technique in the study of brain capillary functions is discussed.
