Serum protein changes and their correlation with antibody production in rabbits following induced Setaria cervi infection.

The sera obtained from normal control and Setaria cervi infected rabbits were analysed weekly both qualitatively and quantitatively. Electrophoretic studies as well as biochemical estimation reveal an increase in total protein and globulin and a decrease in albumin level. The changes were more marked in the third week, followed by a declining phase reaching an almost normal level by the sixth week. The increased globulin level analysed electrophoretically also coincided with the appearance of 2 precipitin arcs in immunoelectrophoretic analysis indicating a correlation between antibody production and hyperglobulinaemia.

Protein kinases in different life stages of Brugia malayi and other filarial worms.

The investigation of protein kinase activity in different life stages of a few filarial worms indicates the presence of different protein kinases and of dissimilarities between the distribution pattern of these enzymes during the development. Phosvitin turned out to be the preferred exogenous acceptor protein, when homogenates of adults of Brugia malayi, Dipetalonema viteae, Setaria cervi and Litomosoides carinii were used as source of enzyme. In contrast to adults the L3-larval and the microfilarial stages demonstrated much lower phosvitin phosphorylating activity and the preference for acidic and basic proteins as phosphate acceptors was about equal. The occurrence of cyclic AMP-dependent protein kinase was established for adults of B. malayi.

Effect of anthelmintics on the enzyme activities of Setaria cervi (Nematoda: Filarioidea).

The present study deals with the comparative efficacy of Hetrazan, levamisole and tetramisole on the enzyme activities of adult Setaria cervi worms. The drugs were administered orally to white rats, intraperitoneally infected with the bovine filariid, Setaria cervi. Biochemical assays revealed the decreased activities of glucose-6-phosphatase, fructose-1,6-diphosphatase, lactate dehydrogenase and adenosine triphosphatase in the drug-treated worms. Hetrazan was relatively more effective than tetramisole and levamisole. A possible role of these enzymes in the energy supply and survival of the worm has been briefly discussed.

Studies on the parasites of zebras. III Nematodes of the mountain zebra from the farm "Kelpie" and the Namib-Naukluft Park, South West Africa/Namibia.

Twelve mountain zebra which were culled at monthly intervals on the farm " Kelpie " in South West Africa/Namibia were examined for helminths. The zebras varied in age from 2-15 years, the middle group of which, aged 4-7 years, had the highest worm burdens. Fourteen species of nematodes belonging to the families Atractidae , Strongylidae , Oxyuridae , Setariidae and Spiruridae were recovered. The highest worm burdens were those of Crossocephalus sp. with 692-61 066 680 and Probstmayria vivipara with 1 257 810-42 004 300. The predominance of the atractids is discussed. The nematodes consistently present were: Cylicodontophorus n. sp. (44-2 107), Triodontophorus spp. (2-934), Cylindropharynx spp. (20-2 332), Crossocephalus sp. and P. vivipara. Two new species, Cylicostephanus longiconus and Cylicodontophorus n. sp., were reported. An additional 3 mountain zebra, culled in the Namib - Naukluft Park, were also examined for helminths. Of 3 zebras ranging in age from 2-7 years, the 2 older animals had the highest helminth burden. Ten species of the nematodes belonging to the same families mentioned above were recovered. The only Spiruridae present were 3 Habronema majus in 1 zebra. The highest worm burdens were those of Crossocephalus sp. with 64 052-883 070 and P. vivipara with 50 720-220 200. The nematodes consistently present were the same as those in the " Kelpie " zebra. In addition, a 2nd, new species of Cylicodontophorus was reported.

Osmoregulation in a parasitic nematode, Setaria cervi.

Studies on osmotic and ionic regulation in Setaria cervi indicate that it can osmoregulate in hypertonic solutions but not in hypotonic solutions. The depression of the freezing point (delta degrees C) of host's peritoneal fluid was -0.71 degrees C.

Effect of diethylcarbamazine on Setaria cervi in vitro.

Diethylcarbamazine (DEC) produced an initial stimulation followed by depression of the movements of the intact worm and nerve-muscle preparation of Setaria cervi. The effective concentration of DEC was reduced to one hundredth in the nerve-muscle preparation as compared to the whole worm, suggesting that the cuticular barrier is highly effective in preventing the penetration of the drugs. The depressant effect of DEC was concentration dependent and was not reversed even after repeated changes of the bath fluid. The worms consumed 7.7 mg +/- 0.2 glucose/g wet weight/hr. The consumption of glucose was directly proportional to its motor activity; it increased during the stimulant phase with low doses of DEC and decreased during the depressant phase.

[Bancroft filariasis in Comores archipelago (immunologic study)].

An immunological study has complemented the epidemiological survey on Bancroft filariosis in the Mayotte island (Comores) carried out by BRUNHES et al., 1971. The search for antibodies to Dipatolonema viteae and Setaria labiotopapilosa was performed by means of electrophoresis and immunofluorescence in 87 individuals and 2 hydrocele fluids. Immunoelectrophoresis confirmed the high endemicity ratio: 57 o/o of the individuals presented 1 to 7 precipitating lines; with immunofluorescence 55 o/o of the subjects had a titer of 200 to 1.600. 46 to 50 o/o of the positivities were observed in people showing clinical symptoms without microfilaremia. On the other hand, 36 to 48 o/o of the proved filarian cases yielded negative results with immunofluorescence or electrophoresis. These figures emphasize the significance and insufficiencies of immunologic techniques for the diagnostic of bancroftoses. In spite of the frequency of certain well identified precipitating systems, an immunoelectrophoretic analysis did not allow to demonstrate a constant and characteristic scheme.

Development of Dirofilaria and Setaria nematodes in Aedes albopictus.

The development of Dirofilaria repens, D immitis and Setaria labiatopapillosa up to L3 stage was documented in a laboratory colony of Aedes albopictus originating from specimens collected in Civitavecchia (Central Italy). The susceptibility of Ae albopictus and its plastic trophic habits, strongly suggest that this mosquito may contribute to the spreading of these nematodes in the country. It is particularly emphasized the danger for human health of an increased probability of transmission of Dirofilaria in urban areas.

Prevalence and distribution of Setaria yehi in southeastern white-tailed deer.

Filarial worms, Setaria yehi, were found in 286 of 1,045 white-tailed deer (Odocoileus virginianus) from 13 of 16 southeastern states. Infections were widespread in the piedmont and coastal plain physiographic provinces, but rarely occurred in deer of the mountains or in southern Florida. Infections were significantly more prevalent among fawn and yearling deer. Infections with S yehi largely accounted for a mild fibrinous peritonitis commonly observed among southeastern deer.

Immune response of electroeluted detergent soluble 29 kDa antigen from Setaria digitata (Von Linstow).

Filariasis is one of the typical parasitic infections which cause immune suppression during the course of infection in both humans and experimental animals. A 29 kDa protein isolated from detergent soluble antigen of S. digitata showed maximum inhibition of cell mediated immune response. The heat inactivated 29 kDa protein was found to be devoid of property of suppression of immune response in the host. Histological study of spleen of BALB/C mice immunized with 29 kDa protein showed changes in regions of spleen such as follicle, trabeculae, capsule, reticuloendothelial cells and eosinophils. The 29 kDa protein, the most reactive of the detergent soluble proteins produced partial suppression of immune response, thereby contributing to the factors responsible for the survival of filarial parasites in hosts.
