Particle image correlation spectroscopy (PICS): retrieving nanometer-scale correlations from high-density single-molecule position data.

A new data analysis tool that resolves correlations on the nanometer length and millisecond timescale is derived. This tool, adapted from methods of spatiotemporal image correlation spectroscopy, exploits the high positional accuracy of single-particle tracking. While conventional tracking methods break down if multiple particle trajectories intersect, our method works in principle for arbitrarily large molecule densities and diffusion coefficients as long as individual molecules can be identified. The method is computationally cheap and robust and requires no a priori knowledge about the dynamical coefficients, as opposed to other methods. We demonstrate the validity of the method by Monte Carlo simulations and by application to single-molecule tracking data of membrane-anchored proteins in live cells. The results faithfully reproduce those obtained by conventional tracking. Upon activation, a fraction of the small GTPase H-Ras is confined to domains of <200 nm diameter, which further substantiates the prediction that membrane organization is a determinant in cellular signaling.

Intraclass correlation among measures related to alcohol use by school aged adolescents: estimates, correlates and applications in intervention studies.

School-based alcohol use prevention studies frequently employ designs in which schools are assigned to treatment conditions while observations are made on individuals. The nesting of schools within treatment conditions requires that the treatment effect be assessed against the between-school variance; unfortunately, that variance is usually larger than for randomly constituted groups and its precision is usually less than that for the within-school variance. These factors often combine to reduce power substantially. To address these problems, investigators need good estimates for the intraclass correlation which together with the number of observations per school determines the magnitude of the extra variation in the nested design. This article presents estimates of school-level intraclass correlation for measures related to alcohol use among ninth and twelfth grade students and discusses their use in planning new studies and analyzing previous or current studies.

Positive illusions: positively correlated with subjective well-being, negatively correlated with a measure of personal growth.

Psychologists have long debated the benefits and costs of self-deceptive enhancement or positive illusions. Accurate perception of reality is central to the definitions of mental health proposed by many personality and clinical psychologists, but Taylor and Brown have suggested that having positive illusions is associated with increased happiness and satisfaction with life. One explanation for the conflicting assertions is that mental health, broadly defined, includes both subjective well-being and personal growth, distinguishable factors which are differentially related to positive illusions. For this study, 81 college students completed measures of positive illusions (Balanced Inventory of Desirable Responding Self-deceptive Positivity and How I See Myself Questionnaire), subjective well-being (Satisfaction With Life Scale and Existential Anxiety Scale), and moral reasoning (Defining Issues Test) as an index of personal growth. As predicted, positive illusion composite scores were positively correlated with scores on the subjective well-being composite (r=.40) but negatively correlated with Defining Issues Test scores (r=-.25). The quadratic relationship between these measures of positive illusion and subjective well-being composites was not significant, indicating no support for an "optimal margin of illusion."

Sensitization to hyperthermia by 3,3'-dipentyloxacarbocyanine iodide: a positive correlation with DNA damage and negative correlations with altered cell morphology, oxygen consumption inhibition, and reduced ATP levels.

The cyanine dye 3,3'-dipentyloxacarbocyanine iodide (DiOC5(3)) (concentrations of 0.5 microgram/ml to 5.0 micrograms/ml) was shown to be a potent sensitizer of Chinese hamster ovary (CHO) cells to hyperthermic cell killing at 43.0 degrees C or 45.5 degrees C, while exhibiting no cytotoxicity at 37.0 degrees C. Sensitization to hyperthermic cell killing was accompanied by an increase in damage to the DNA, as measured by DNA unwinding. The increased DNA damage correlated qualitatively with the enhanced heat killing induced by DiOC5(3). This correlation was better in cells heated at 43.0 degrees C than in those heated at 45.5 degrees C. DiOC5(3) is known to affect other cellular functions. It inhibits electron transport, uncouples oxidative phosphorylation, and inhibits calcium ATPases. The effects of DiOC5(3) on oxygen consumption and ATP content were therefore measured at 37.0 degrees C and at hyperthermic temperatures. The results demonstrated that inhibition of oxygen consumption and reduction of cellular ATP levels played no role in inducing heat sensitization in DiOC5(3)-treated cells, or in causing cell death in cells treated with heat alone.

Correlation with Cerec 3 for end teeth: single-side correlation.

Correlation is not necessarily a problem for end teeth any longer. All the advantages of Correlation, anatomical occlusal surfaces, and very slight vertical occlusion adjustment are also available with the described method for end teeth and gaps.

[Correlation between quantitative indicators of the bioelectrical activity of the heart atria and central hemodynamics in patients with ischemic heart disease based on correlation analysis data].

Atrial bioelectrical activity (according to large-scale ECG findings) and central hemodynamics were studied in 43 coronary patients. A correlation analysis showed an increased correlation between the studied values in patients with diffuse and postinfarctional cardiosclerosis and those in healthy subjects. The authors stressed the most significant large-scale ECG values. A conclusion was made of the appropriateness of a study of atrial bioelectrical activity by the method of large-scale electrocardiography and hemodynamics for diagnostics in coronary patients with the initial stage of cardiac insufficiency.

[Correlation of the quantitative ECG and echocardiogram indices in left ventricular hypertrophy according to correlational analysis data].

In 47 patients with arterial hypertension and clinicoroentgenological signs of hypertrophy of the left ventricle the echo- and electrocardiographic study (the McFee-Parungao system) was carried out. Highly significant (p less than 0.001) direct correlation was revealed between the index of the corrected ECG, such as sigma R x,y,z + + sigma S x,y,z and Rx + Sz with echocardiographic indices of mass r = +0.851 and +0.848, respectively, and the thickness of the left ventricle of the myocardium (r = +0.806 and +0.794, respectively). The above electrocardiographic indices do not correlate to the main echocardiographic indices of the contractile function of the left ventricle.

Ex vivo two-dimensional proton nuclear magnetic resonance spectroscopy of smooth muscle tumors: advantages of total correlated spectroscopy over homonuclear J-correlated spectroscopy.

Two-dimensional total correlated proton nuclear magnetic resonance spectroscopy (TOCSY) at 600 MHz was used for an ex vivo biochemical analysis of a benign human smooth muscle tumor (leiomyoma) and high grade malignant smooth muscle tumor (leiomyosarcoma). While there are a number of applications of proton nuclear magnetic resonance magnitude-mode two-dimensional correlated spectroscopy (COSY) to the ex vivo study of tissues and cells in the literature, to our knowledge this is the first application of TOCSY for the ex vivo study of biological tissue. Comparison of TOCSY and purged COSY data demonstrate the potential sensitivity advantages of the TOCSY method for the study of heterogeneous biological tissues. These TOCSY spectra were used to identify and quantitate a wide range of metabolites such as amino acids, peptides, triglycerides, phospholipid precursors and degradation products, bound fucose, and other saccharides. The leiomyosarcoma was found to have 5-fold higher levels of triglycerides and a 7-fold increase in the glycerophosphocholine:choline ratio compared to the leiomyoma. These metabolite changes may enhance membrane fluidity in the leiomyosarcoma compared to leiomyoma and thus may be of fundamental importance to cell motility, recognition, sarcoma tumorigenesis, and metastatic potential.

[Correlation between DNA content and amplification of oncogenes (c-myc, L-myc, c-erbB-2) and correlation with prognosis in 143 cases of resected lung cancer].

DNA content analysis using flow cytometry and amplification of c-myc, L-myc, and c-erbB-2 oncogenes in 143 cases of resected lung cancer were analyzed using the same specimen, and we examined the correlation with prognosis of DNA content and amplification of oncogenes. There were 54 DNA diploid cases (38%), 81 DNA aneuploid cases (57%) and 8 DNA multiploid cases. Analysis of oncogene amplification revealed 22 cases of c-myc, 4 cases of L-myc, and 22 cases of c-erbB-2. In curatively resected cases, the 5-year survival rate was 65% in 31 DNA diploid cases, and 36% in 40 DNA aneuploid cases. There was a statistically significant difference between the two groups (p < 0.02). However, in non-curatively resected cases, the 5-year survival rate was 11% in 23 DNA diploid cases, and 33% in 49 DNA aneuploid cases. There were no statistically significant differences among these groups. The correlation between DNA content and amplification of oncogenes was as follows. In DNA diploid cases, there were 4 cases of c-myc, and 6 cases of c-erbB-2. In DNA aneuploid cases, there were 15 cases of c-myc, 4 cases of L-myc, and 15 cases of c-erbB-2. In DNA multiploid cases, there were 3 cases of c-myc, and 1 cases of c-erbB-2. Amplification of oncogenes was seen more frequently in DNA aneuploid and multiploid cases than in DNA diploid cases. In 71 curative resected cases, the 5-year survival rate for amplified cases of c-myc (10 cases) was 0%, and that of cases with no amplification was 61% (no statistically significant difference). The 5-year survival rate for amplified cases of c-erbB-2 (10 cases) was 40%, against 52% for cases with no amplification. DNA content analysis using flow cytometry was more convenient than analysis of amplification of oncogenes, and reflects the prognosis of resected lung cancer better than oncogenes. There was no relation between DNA content and gene amplification.

Correlation of selected serum constitutents: 2. Consistency of intra-individual correlation values, means, and variances during four months.

We examined whether inter-individual differences in correlation coefficients previously found among subjects truly reflect consistent inter-individual differences or are time-related within an individual. The consitutents studied in this investigation were (a) the enzmes aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase; and (b) the non=protein nitrogen-containing constituents urea, uric acid, and creatinine. Ten healthy women were each subjected to 15 venipunctures over a five-week period (Series I), and, after a two-month interval, were again subjected to 15 venipunctures over a second five-week period (Series II). Before statistical analysis, the data were corrected for the batch-to-batch (day-to-day) arnalytical variation. There was a signiificant (P less than .05) change in the covariance structure (variances or correlation coefficients, or both) between the two series in four of the 10 subjects for the combination of enzymes, and in three other subjects for the combination of nonprotein nitrogen constitutents. Although we found a significant (P lees than .05) average intra-individual variation in the mean values from series to series in the cases of the three enzymes and urea, the magnitude of the inter-series variation in means was relatively small. CV's were: alkaline phosphatase, 3.4%; lactate dehydrogenase, 2.3+; aspartate aminotransferase, 3.3%; urea, 5.0%; uric acid, 1.0%; and creatinine, 1.2%.

Fluorescence correlation microscopy (FCM)-fluorescence correlation spectroscopy (FCS) taken into the cell.

Confocal fluorescence correlation spectroscopy (FCS) and other confocal spectroscopic techniques are ideally suited for the analysis of molecular interactions at the subcellular level. However, one requires exact positioning in three dimensions within the cell. Our instrument integrates FCS with high sensitivity digital imaging microscopy and high precision positioning. We present first measurements of intracellular FCS, with specification of the instrumental requirements and methods of data analysis. We propose the term fluorescence correlation microscopy (FCM) for this extended modality of FCS.

Comparison of correlated correlations.

We consider a problem where kappa highly correlated variables are available, each being a candidate for predicting a dependent variable. Only one of the kappa variables can be chosen as a predictor and the question is whether there are significant differences in the quality of the predictors. We review several tests derived previously and propose a method based on the bootstrap. The motivating medical problem was to predict 24 hour proteinuria by protein-creatinine ratio measured at either 08:00, 12:00 or 16:00. The tests which we discuss are illustrated by this example and compared using a small Monte Carlo study.

Cytotoxicity of 3'-azido-3'-deoxythymidine correlates with 3'-azidothymidine-5'-monophosphate (AZTMP) levels, whereas anti-human immunodeficiency virus (HIV) activity correlates with 3'-azidothymidine-5'-triphosphate (AZTTP) levels in cultured CEM T-lymphoblastoid cells.

Activation of the anti-human immunodeficiency virus (HIV) compound 3'-azido-3'-deoxythymidine (AZT) is dependent on its 5'-phosphorylation by cellular nucleoside and nucleotide kinases. Azidothymidine 5'-triphosphate (AZTTP) is considered to be the metabolite responsible for both the anti-HIV effect of AZT, via inhibition of reverse transcriptase, and cytoxicity by interference with cellular DNA polymerases. During the characterization of AZT metabolism in cultured human T-lymphoblastoid CEM cells, a spontaneously occurring variant cell line, CEM/Ag-1, was found that showed approximately 10-fold resistance to AZT growth inhibition as compared to wild type (wt) cells (EC50 = 2 mM as compared to 350 microM for wt cells). CEM/Ag-1 cells had a 3-fold reduced capacity to accumulate azidothymidine monophosphate (AZTMP) compared to wt cells whereas similar levels of AZTTP were found in both cell lines. The intracellular half-life of AZTMP was approximately 70 min in both wt and CEM/Ag-1 cells. A 3-fold lower specific activity of cytoplasmic thymidine kinase was observed in CEM/Ag-1 extracts as compared to wt. The reduced thymidine kinase activity was not correlated to a decreased level of thymidine kinase mRNA. Syncytium formation of CEM/Ag-1 cells infected with HIV-2 as well as HIV-1 antigen production was inhibited at the same concentrations of AZT (approx. 0.01 microM) as were HIV-1 and HIV-2 infected wt cells. Thus, minor decreases in cellular thymidine kinase levels may markedly affect the cytoxicity of AZT but have no major effect on the antiviral activity of AZT. Our results strongly suggest that AZTMP is responsible for a major part of the growth inhibitor effects, while AZTTP mainly mediates the antiviral activity of AZT.

Adolescents with learning disorders have atypical EEG correlation indices. I. Correlation indices during visual discrimination.

OBJECTIVE: To find out whether 14-16-year-old pupils with visual perception disabilities have atypical EEG activation patterns during visual discrimination. METHODS: EEG correlation indices (EEGCIs), based on the waveform characteristics of two EEG signals, were used as measures of slow joint activation of cortical regions during visual discrimination in pupils with visual perception disabilities. RESULTS: During visual discrimination low EEGCIs were seen between the left temporal and both parieto-occipital EEG channels in pupils with visual perception disabilities and in pupils with a poor occupational outcome. The pupils with low performance intelligence and those with difficulties in the visual discrimination task had low EEGCIs within the left hemisphere. The left hemispheric dominance of the findings is suggested to reflect the psychophysiology of the task since visual discrimination demands attention to details (local processing) and is thus supposed to be more strongly represented in the left hemisphere. CONCLUSIONS: During visual discrimination, low EEGCIs were seen in the posterior and left hemispheric regions of pupils with disabilities in visual perception and visual discrimination and with a poor occupational outcome. SIGNIFICANCE: Low EEGCIs in posterior and left hemispheric regions during visual discrimination can to some extent be seen as neurophysiological markers for visual perception disabilities and a poor occupational outcome and imply an increased need for adjustment of the educational curriculum and a need for occupational guidance.

Adolescents with learning disorders have atypical EEG correlation indices. II. Correlation indices during reading.

OBJECTIVE: To find out whether 14-16 year old reading and writing impaired pupils have atypical EEG activation patterns during reading. METHODS: EEG correlation indices (EEGCIs), based on the waveform characteristics of two EEG signals, were used as measurers of slow joint activation of cortical regions during reading in pupils with reading and writing impairment. RESULTS: Reading was associated with high EEGCIs within the right hemisphere in reading and writing impaired pupils. The finding is analogous to the results of an earlier study [Byring, Electroencephalogr. Clin. Neurophysiol. 63 (1986) 1] in boys with spelling disabilities. The activation in the right hemisphere might represent a compensation for a left hemisphere dysfunction in pupils with reading and writing impairment during reading, as suggested by a number of functional neuroimaging studies. This interpretation was corroborated by high EEGCIs especially in those impaired pupils who had a good occupational outcome. CONCLUSIONS: EEGCIs during reading are high within the right hemisphere in pupils with reading and writing impairment. SIGNIFICANCE: High EEGCIs within the right hemisphere during reading might be considered neurophysiological markers for reading and writing impairment.

Perception of facial emotions in chronic schizophrenia does not correlate with negative symptoms but correlates with cognitive and motor dysfunction.

BACKGROUND: Appropriate expression of emotions and correct perception of emotional expression in others are important social skills which may be impaired in schizophrenia and contribute to poor social adjustment. We examined the relationship between expression of emotions as measured by affective flattening and other negative symptoms and their perception. We compared performance on tests of perception of facial emotions with that in other cognitive areas. METHODS: 36 chronic schizophrenic patients on stable doses of atypical antipsychotics were assessed using tests of identification (FID) and discrimination (FDIS) of facial emotional expressions, visual retention (BVRT) and general cognitive function (Mini Mental State Examination, MMSE). Clinical symptoms were assessed with scales for the assessment of negative symptoms (SANS) and positive symptoms (SAPS). Motor symptoms were assessed with side effects (SA) and AIMS scales and Finger Tapping Test. RESULTS: Negative symptoms showed no relation to FID or FDIS. FID showed significant correlation with Visual Retention and Finger Tapping but not MMSE. CONCLUSION: The ability to identify facial emotional expressions is not related to negative symptoms in chronic schizophrenia and shares common mechanisms with visual reproduction and ability to make rapid motor movements. This suggests common defects in perceptual, timed processes consistent with postulated dysfunction of cortico-subcortical circuits.

Extraction of correlated gene clusters from multiple genomic data by generalized kernel canonical correlation analysis.

MOTIVATION: A major issue in computational biology is the reconstruction of pathways from several genomic datasets, such as expression data, protein interaction data and phylogenetic profiles. As a first step toward this goal, it is important to investigate the amount of correlation which exists between these data. RESULTS: These methods are successfully tested on their ability to recognize operons in the Escherichia coli genome, from the comparison of three datasets corresponding to functional relationships between genes in metabolic pathways, geometrical relationships along the chromosome, and co-expression relationships as observed by gene expression data.

Multiple patterns of MHC class II antigen expression on cellular constituents of rat heart grafts. Lack of correlation with graft survival, but strong correlation with vasculitis.

The patterns of induced major histocompatibility antigen expression on indigenous cellular elements of heterotopic rat cardiac grafts were determined by immunohistologic methods in a variety of donor-recipient combinations. Heart grafts were studied in combined full-MHC- and non-MHC-disparate combinations, isolated intra-MHC-disparate combinations, and non-MHC-disparate combinations. The pattern of class II expression on cellular constituents of the grafts was highly variable and critically dependent upon the nature of the specific unidirectional donor-recipient combination. No uniform pattern of class II expression emerged that was clearly predictive of rapidity of rejection or of protracted survival. However, vasculitis was confined to grafts in combinations in which induced class II expression on graft large vessel endothelium was present. Sites of vasculitis were never encountered in the absence of induced class II expression on overlying endothelium. Vasculitis and associated induced class II expression on large vessel endothelium were present in rapidly rejecting grafts and in grafts with indefinite survival. In the latter, vasculitis was shown to progress to a late phase of occlusive intimal thickening. Induced class I expression on graft cardiac myofibers was present in all the genetically disparate donor-recipient combinations examined in this study, irrespective of the length of graft survival. This investigation has shown that no uniform stereotyped pattern of MHC antigen expression on cellular constituents correlates with the length of graft survival. However, induced class II expression on graft large vessel endothelium is closely associated with vasculitis, which can directly progress to occlusive intimal thickening in grafts with prolonged survival.
