[Development of the mitochondrial apparatus and blood supply of skeletal muscle fibers during ontogenesis of domestic fowl].

The diameter, length, and numerical density of capillaries, diameter of muscle fibers, size and numerical density of their profiles, and relative volume of mitochondria in them were determined in the chicken red oxidative gastrocnemius and white glycolytic pectoral muscle during development from day 10 of embryogenesis to six month of postnatal life. The bulk blood flow was measured in these muscles by hydrogen clearance during postembryonic development. During embryogenesis, the fibers of gastrocnemius muscle develop and grow at a higher rate, while during postembryonic development, those of the pectoral muscle develop faster. The density of mitochondrial profiles increases during embryogenesis and decreases after hatching, while their mean size increases, especially in the oxidative fibers, but it somewhat decreases in 6-month old chicks. Redistribution of mitochondria by the fiber section during development takes place in both muscles: they are localized predominantly in the center in 18-day embryos and in the periphery, especially in the gastrocnemius fibers, in 6-month old fowl. At hatching, the lengths of capillaries are similar in both muscles, but as chicks grow, the proportion of longer (more than 600 microm) capillaries in the pectoral muscle sharply increases, while their density and bulk blood flow decrease. Ratios were determined between structural parameters of the capillary bed and mitochondria, on the one hand, and oxygen consumption (ml/min per 1 mm fiber and 100 g muscle mass), on the other.

Embryonic expression of beta-actin-lacZ hybrid gene injected into the fertilized ovum of the domestic fowl.

An experiment was carried out to investigate the expression of cloned DNA injected into the germinal disc of the chick fertilized ovum. The beta-actin-lacZ hybrid gene, MiwZ, was injected, in the closed circular form, into the cytoplasm of the germinal disc at the single-cell stage. The embryos were cultured in vitro, then in recipient eggshells up to day 4 of incubation. The survival rate of the embryos at day 4 was 42% (55/130), and the rate of embryos expressing MiwZ was 64% (35/55). Twenty-two embryos expressed the MiwZ in both embryonic and extraembryonic tissues, while the remainder expressed the MiwZ in only extraembryonic tissues. Mosaic expression was observed in most of the embryos expressing MiwZ in embryonic tissues. Expression throughout all tissues of the embryo including blood cells occurred in one case. In this case, the injected DNA was assumed to have integrated at an earlier stage. The results indicate that it is now possible to investigate the promoter activities of introduced exogenous genes as well as the effect of introduced genes on embryogenesis in early chick embryos. This technique may also facilitate the production of transgenic chicks.

Serologic examination of birds from the area of Southern Moravia for the presence of antibodies against arboviruses of the groups alfa, flavo, Bunyamwera supergroup, and the virus Yaba 1-Lednice 110. I. Domestic fowls.

196 geese, 141 ducks, and 100 hens from 9 localities of the South Moravian region (District of Breclav) were examined by means of the haemagglutination-inhibition test (HIT) for the presence of antibodies against 12 arbovirus antigens of the groups Alfavirus (Western Equine Encephalitis (WEE), Eastern Equine Encephalitis (EEE), Semliki, Sindbis, Chikungunya, O'nyong-nyong), Flavovirus (Tick-borne Encephalitis (TBE), Dengue and West Nile Virus (WN), Bunyamwera Supergroup (Tahna and Beta Calovo), and the Yaba 1-Lednice 110 Virus. In tested fowls antibodies were mostly found against Yaba 1-Lednice 110 Virus, namely 3.6% in geese and 17.7% in ducks. Antibodies against Calovo Virus were found in only 1 of the tested ducks. All sera of hens were negative. Antibodies against arboviruses of the Alfa-and Flavovirus groups were not detected.

Development of the cloacal bursa in the domestic fowl. II. A quantitative and fine structural analysis of the follicular cortex and medulla.

The estimated volumes of the follicular medulla (x) and cortex (y) from 14-day-old embryos till 28-day-old White Leghorn chicks were associated through the allometric formula y = bxk or log y = log b + k log x. Two successive allometric growth stages (I and II) are discernable, being hatching the transition region between them. The volumetric growth of the cortex is 2.49 times greater than that of the medulla in stage I, whereas cortex and medulla grow isometrically in stage II. The curve fitting procedure analysis of the absolute cortical and medullary growth confirmed these results. The fine structure of the cell types in the follicular medulla revealed that: a) in the allometric stage I pre-existing, bud precursor (Pr) cells appear to give rise to basal (Ba) and medullary epithelial (ME) cells, in both cases showing lucent and dark varieties. A medullary cytoreticulum is established at the onset of this stage. b) The marked lymphocyte proliferation during stage II occurs among the thin and short cytoplasmic processes of BA cells. These processes extend towards the centre of the medulla and also show many lateral interdigitating processes. During this same stage, the cytoplasmic processes of ME cells elongate and become thinner promoting a widening of the cytoreticulum interstices. The fine structural analysis of the cortical cytoarchitectural arrangement showed that: a) before the onset of stage I (14-day-old embryos) the cortex consists mainly of typical fibroblasts (F) and a few blastic (Pr?) cells. Later in this stage I, a poorly defined cortical framework is made up of typical fibroblasts, few cortical branching (CB) cells of the epithelial variety (which seem to be derived from Pr cells) and CB cells of the fibroblast-like variety. These cells are interspersed with mature and immature lymphocytes. b) Allometric stage II of the cortex is characterized by the presence of very thin and long cytoplasmic processes from CB cells of both epithelial and fibroblast-like varieties. The arrangement of CB cell profiles, visualized in electron micrographic montages, is remarkably similar to that of the ME cells profiles which are known to form a cytoreticulum. We thus propose that the mature follicular cortex is endowed with a cellular framework forming wide interstices in which packed mature lymphoid cells are lodged.

Subclinical pneumonia associated with an experimental adenovirus infection in the domestic fowl and the effect of concurrent infectious laryngotracheitis virus.

A CELO-type adenovirus (AV) isolated from fowls with respiratory disease was inoculated experimentally into the tracheas of young birds. No symptoms referable to respiratory infection were evident. Post mortem examination between days 2 and 5 after inoculation revealed pneumonia involving up to 30 per cent of the surface of the lungs. Histologically, a focal to diffuse interstitial lymphocytic infiltration and bronchiolar degeneration were present. Concurrent infections with a mild strain of infectious laryngotracheitis virus (ILT) failed to enhance the pathogenicity of either the AV or ILT infections.

An avian model for the study of acute hemolytic anemia in the domestic fowl (Gallus domesticus).

Acute hemolytic anemia was experimentally induced in chickens with the oxidant chemical phenylhydrazine. From results of the graded doses trial, subcutaneously injected 6 mg/100 g body weight single dose was designated as standardised. The anemic response, characterized by marked reductions in hemoglobin, packed cell volume and total erythrocyte count, was perceptible on day 1 post-injection. Peak anemia was observed on day 3. Compensatory erythropoiesis was discernible on day 5. The hematological profile indicated near complete recovery on day 11 post-injection. This avian model is proposed for study of the basic mechanisms of hemolytic anemia in chickens.

Use of cyproterone acetate to lower circulating corticosterone and its effects on lymphocytes in domestic fowl.

Chickens were given daily injections of cyproterone acetate (CA) and the effects on plasma corticosterone, bodyweight, weights of the adrenals and lymphoid organs, numbers of circulating peripheral blood lymphocytes and their proliferation in the presence of lectins, concanavalin A (Con A) and phytohaemagglutinin (PHA), were investigated. Five daily doses of 10 or 30 mg CA kg-1 bodyweight each week over a three-week period caused a decrease in weight gain and a reduction in the relative weights of the bursa and thymus but not the spleen. There was a small decrease in the adrenals after treatment with 10 mg CA kg-1. When daily injections of CA were given over a seven-day period doses of 6 and 10 mg CA kg-1 bodyweight caused a significant (P less than 0.05) decrease in plasma corticosterone concentration after four days. However, after eight daily injections of CA a single injection of corticotrophin (10 iu ACTH kg-1) increased circulating corticosterone indicating CA had not completely blocked adrenal synthesis. CA had no effect on numbers of circulating peripheral blood lymphocytes or their ability to proliferate in the presence of Con A or PHA. The results indicate that CA is effective in lowering circulating corticosterone in the fowl but this did not affect the numbers or responsiveness of peripheral blood lymphocytes.

Responsiveness to dietary cholesterol in the domestic fowl is not related to the degree of body fatness.

The hypothesis that chickens of a lean rather than a fat line are hyperresponsive to dietary cholesterol was tested. Birds of the fat line showed higher initial plasma total cholesterol levels. The addition of 1% (w/w) of cholesterol to the diet significantly increased plasma cholesterol concentrations but there was no difference in response between the two lines. Dietary cholesterol increased liver cholesterol in both lines, the effect being more pronounced in birds of the lean line. It is concluded that the responsiveness of plasma cholesterol to dietary cholesterol in chickens is not related to the degree of body fatness.

Effects of repeated eastward and westward synchronizer phase-shifts on growth, blood morphology and biochemical variables in male domestic fowls.

The effects of repeated synchronizer phase-shifts on growth (increment in body weight), blood morphology and growth related biochemical variables in 3-day-old juvenile male chicks were evaluated till they become 20 weeks old. The control birds were kept under a fixed LD 12:12 light schedule throughout, while the LD 12:12 regimen of experimental birds was shifted weekly either by delaying (westward shift) or by advancing (eastward shift) the time of light onset by 8 hr. The rate of growth was more in fowls which were exposed to repeated advancement of synchronizer schedule than the controls and those exposed to westward shifts of the synchronizer schedule. Hematological indices, viz., hemoglobin concentration, number of circulating erythrocytes, hematocrit and biochemical variables, such as concentration of glucose and protein in plasma; glycogen, protein and lipid in liver; and muscle tissues were significantly more in chicks exposed to repeated eastward shifts, when compared with the control birds (LD 12:12). It is suggested that the eastward shift of the synchronizer may be beneficial for the poultry birds.

Relationship between the saccus endolymphaticus and the fourth ventricle of the brain of the domestic fowl (Gallus gallus f. domestica).

The fine structure of the wall of the SE was determined exactly and its relationship to the cisternae (the evaginations of the roof of the fourth ventricle extending to the SE) was defined. The way in which the cisterna is formed was defined and the development of its fine structure was described by comparing serial sections from 19-day embryos and adult fowls. Like the SE, the cisternae are lodged in the angle between the cerebellum and the medulla oblongata, in the subarachnoid space. The terminal segment of the cisterna lies in the immediate vicinity of the mesenchymal epithelium bordering the basal labyrinth of the SE cells. Collagen trabeculae keep the SE and the cisternae suspended in the subarachnoid space. The cisternae and trabeculae are wrapped in mesenchymal epithelium. The cisterna is avascular and does not communicate with the SE. The cisterna is lined internally with simple squamous epithelium (modified neural epithelium of the roof of the fourth ventricle). The bodies of the cells bulge into the lumen of the cisterna in the region of localization of their nucleus. The epithelium is seated on a pronounced basal lamina. The surface turned towards the subarachnoid space is lined continuously with mesenchymal epithelium without a basal lamina. The cells of the cisternal epithelium are connected by tight junctions of the type of zonulae occludentes and desmosomes. The basal lamina is continuous and distinct. The mesenchymal epithelium of the subarachnoid space has no basal lamina, as on the subarachnoid surface of the SE, the cisternae, the trabeculae, the pia mater and the arachnoidea.(ABSTRACT TRUNCATED AT 250 WORDS)

An allometric study of the development of the cloacal bursa in the domestic fowl.

The allometric relation between the increment of the body weight and the relative growth of the cloacal bursa shows that the bursal growth is greater than the body growth, during the period analysed (from the 16th d of embryonic development through the 28th post-hatched day). No statistical difference in the mean relative bursal weight between male and female birds was achieved in the above mentioned period analysed. The advantages of using an allometric study for the evaluation of an organ's growth during development are discussed.

Development of the cloacal bursa in the domestic fowl. I. An allometric and morphological study of the interfollicular surface epithelium (ISE).

The allometric relation applied between the body weight and the volume increment of the interfollicular surface epithelium (ISE) showed just one developmental stage during the period analysed (from 16d old embryos through 28 old d chicks). In this single allometric developmental stage the ISE growth is relatively more pronounced than that of the bursa as a whole due to profound cellular modifications which that epithelium undergoes from the primitive undifferentiated epithelial cells through goblet and prismatic cells. The latter could have an endocytic capability due to their primary and secondary lysosomes content. A possible cellular differentiation of the ISE during the histogenesis of the cloacal bursa is also discussed.

Male sterility in the domestic fowl (Gallus domesticus) after 1-(2-chloroethyl)-3-(4-methylcyclohexyl)-1-nitroso-trans-urea (NSC-95441) treatment of chicks.

Of 11 male baby chicks treated twice with 3 mg of 1-(2-chloroethyl)-3-(4-methylcyclohexyl)-1-nitroso-trans-urea (once on the day of hatching and once on the day after), 9 showed absolute lack of spermatogenesis and 2 showed severe inhibition when examined at the age of 6 months. Plasma testosterone level was not altered, however, and histologic examination revealed well-organized testicular interstitial tissue with an abundance of Leydig's cells. In the lining of the seminiferous tubules only Sertoli's cells were present. The weight of the testes from treated animals was only about 10% of control weight. These findings emphasized the possibility that the nitrosoureas cause severe damage to the reproductive system in man.
